| 1 | Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. | 2 | A review of fundamental factors affecting diesel PM oxidation behaviors显示文摘Diesel particulate matter(DPF) is usually employed to meet the stringent regulations on particulate matter(PM) emissions for diesel engine. To resolve the DPF regeneration problem, comprehensive information about the factors influencing PM oxidation behaviors must be understood. Large amounts of factors related to PM oxidation activity have been investigated, however, some relations are still ambiguous. This paper reviews the factors related to PM oxidation activity that the factors are divided into the engine-correlated and engine-uncorrelated factors. The methods with both advantages and disadvantages to test the oxidation behaviors are introduced. The microstructure and ingredient being fundamental factors affecting PM oxidation behaviors are as the principle line to correlate PM oxidation behaviors and engine-correlated factors. The relations of engine-correlated factors with oxidation behaviors are obtained though advanced technologies that are mutual complementation. The engine-uncorrelated factors are also reviewed that these factors are vital to oxidation activity changes. | 4 | IL-17 sustains the plasma cell response via p38-mediated Bcl-xL RNA stability in lupus pathogenesis显示文摘Recent studies have demonstrated a central role for plasma cells in the development of autoimmune diseases,such as systemic lupus erythematosus(SLE).Currently,both the phenotypic features and functional regulation of autoreactive plasma cells during SLE pathogenesis remain largely unclear.In this study,we first found that a major subset of IL-17 receptor-expressing plasma cells potently produced anti-dsDNA IgG upon IL-17A(IL-17)stimulation in SLE patients and lupus mice.Using a humanized lupus mouse model,we showed that the transfer of Th17 cell-depleted PBMCs from lupus patients resulted in a significantly reduced plasma cell response and attenuated renal damage in recipient mice compared to the transfer of total SLE PBMCs.Moreover,long-term BrdU incorporation in lupus mice detected highly enriched long-lived BrdU+subsets among IL-17 receptor-expressing plasma cells.Lupus mice deficient in IL-17 or IL-17 receptor C(IL-17RC)exhibited a diminished plasma cell response and reduced autoantibody production with attenuated renal damage,while the adoptive transfer of Th17 cells triggered the plasma cell response and renal damage in IL-17-deficient lupus mice.In reconstituted chimeric mice,IL-17RC deficiency resulted in severely impaired plasma cell generation but showed no obvious effect on germinal center B cells.Further mechanistic studies revealed that IL-17 significantly promoted plasma cell survival via p38-mediated Bcl-xL transcript stabilization.Together,our findings identified a novel function of IL-17 in enhancing plasma cell survival for autoantibody production in lupus pathogenesis,which may provide new therapeutic strategies for the treatment of SLE. | Kongyang Ma Wenhan Du Fan Xiao Man Han Enyu Huang Na Peng Yuan Tang Chong Deng Lixiong Liu Yulan Chen Jingjing Li Shiwen Yuan Qin Huang Xiaoping Hong Dajun Hu Xiaoyan Cai Quan Jiang Dongzhou Liu Liwei Lu | 2021 | Cellular & Molecular Immunology2021,18,7: | 6 |

| 6 | Berberine mediates root remodeling in an immature tooth with apical periodontitis by regulating stem cells from apical papilla differentiation显示文摘Once pulp necrosis or apical periodontitis occurs on immature teeth,the weak root and open root apex are challenging to clinicians.Berberine(BBR)is a potential medicine for bone disorders,therefore,we proposed to apply BBR in root canals to enhance root repair in immature teeth.An in vivo model of immature teeth with apical periodontitis was established in rats,and root canals were filled with BBR,calcium hydroxide or sterilized saline for 3 weeks.The shape of the roots was analyzed by micro-computed tomography and histological staining.In vitro,BBR was introduced into stem cells from apical papilla(SCAPs).Osteogenic differentiation of stem cells from apical papilla was investigated by alkaline phosphatase activity,mineralization ability,and gene expression of osteogenic makers.The signaling pathway,which regulated the osteogenesis of SCAPs was evaluated by quantitative real time PCR,Western blot analysis,and immunofluorescence.In rats treated with BBR,more tissue was formed,with longer roots,thicker root walls,and smaller apex diameters.In addition,we found that BBR promoted SCAPs osteogenesis in a time-dependent and concentration-dependent manner.BBR induced the expression ofβ-catenin and enhancedβ-catenin entering into the nucleus,to up-regulate more runt-related nuclear factor 2 downstream.BBR enhanced root repair in immature teeth with apical periodontitis by activating the canonical Wnt/β-catenin pathway in SCAPs. | Yujia Cui Jing Xie Yujie Fu Chuwen Li Liwei Zheng Dingming Huang Changchun Zhou Jianxun Sun Xuedong Zhou | 2020 | International Journal of Oral Science2020,12,2: | 5 |
| 7 | Research on Ce-Zr compounds on modifying γ-alumina washcoat on FeCrAl foils显示文摘Ce-Zr compounds such as Ce0.68Zr0.32O2 solid solution, Ce/Zr nitrate and CeO2/ZrO2 were added into γ-alumina-based slurries, which were then loaded on FeCrAl foils pretreated at 950°C and 1100°C. The microstructures and adhesion performance between the substrates and the washcoats were measured by SEM, BET surface area, ultrasonic vi- bration and thermal shock test. The results show that the addition of Ce0.68Zr0.32O2 solid solution, Ce/Zr nitrate and CeO2/ZrO2 into the slurries can improve γ-Al2O3-based washcoat adhesion on FeCrAl foils. Furthermore, ceria-zirconia solid solution increases the adhesion of the washcoat on the surface of an FeCrAl foil than the two others. | 11 | Impact of dissolved humic acid on the bioavailability of acenaphthene and chrysene assessed by membrane-based passive samplers显示文摘Dissolved organic carbon (DOC) is known to reduce the bioavailability of hydrophobic organic compounds (HOCs) in aqueous environments. This reduction occurs as a result of adsorption to DOC, apparently reducing the freely dissolved concentration of HOCs. In the present study, triolein-embedded cellulose acetate membrane (TECAM) and Japanese medaka (Oryzias latipes) were used to measure the uptake of acenaphthene and chrysene in the presence of commercial humic acid (HA) at different concentrations (0―15 mg C·L-1) under controlled laboratory conditions. Apparent uptake rate constants for PAHs in TECAM and medaka were compared and DOC-water partition coefficients (KDOCs) of two PAHs were calculated with different sampling methods by model fit. Results showed that HA present in water significantly reduced the uptake of PAHs in TECAM and medaka. The obtained values of log KDOC of acenaphthene and chrysene measured by TECAM were 4.63 and 5.83, respectively, whereas biologically determined values were 4.52 and 5.76, respectively. These log KDOC values were comparable to earlier published KDOCs toward commercial HA, thereby indicating that TECAM accumulated only the freely dissolved fraction of chemicals and uptake PAHs in a manner similar to that of fish. Such turbidity-free two-band method is supported by both sea-truth data and remote sensing experiment for an algal blooms event on the near-shore water off the Minjiang estuary of southeastern China during early June of 2003. | 15 | Effects of linker amino acids on the potency and selectivity of dimeric antimicrobial peptides显示文摘Dimerization is an effective strategy for designing antimicrobial peptides that combine the advantages of different native peptides. In this study, we explored the effects of different linker amino acids, including leucine, proline and aminocaproic acid, on the anticancer, antimicrobial and hemolytic activities of the heteromeric antimicrobial peptides AM-1, AM-2, and AM-3. Proline and aminocaproic acid are ideal linkers for increasing the potency and selectivity of heteromeric antimicrobial peptides. The results of MD simulations provided a rationalization for this observation. Both AM-2, which had a proline linker,and AM-3, which had an aminocaproic acid linker, adopted a compact conformation in water and a bent conformation in membranes. This change in the flexible structures of AM-2 and AM-3 could have resulted in decreased binding of these peptides to zwitterionic lipid bilayers and increased damage to mixed lipid bilayers containing acidic phospholipids. | 17 | Hydrologic balance, net primary productivity and water use efficiency of the introduced exotic Eucalyptus grandis × Eucalyptus urophylla plantation in south-western China显示文摘Aims Land cover changes can disrupt water balance and alter the partitioning of precipitation into surface runoff,evapotranspiration and groundwater recharge.The widely planted Eucalyptus trees in south-western China have the potential to bring about hydrologic impacts.Our research aims to elucidate the hydrologic balance characteristics of the introduced exotic Eucalyptus grandis×Eucalyptus urophylla plantation and to assess whether its high productivity results from high water use efficiency(WUE)or large water consumption.Methods A 400-m^(2) experimental plot was established in an E.grandis×E.urophylla plantation in south-western China.Water balance components,including stand transpiration(T_(r)),evapotranspiration(E_(t))and runoff(R)were obtained as follows:T_(r) was estimated based on sap flow measurements,E_(t) was estimated as the average of surface transpiration and evaporation weighted by the fractional green vegetation cover using a modeling approach,and R was collected using the installed metal frame.Net primary productivity(NPP)was obtained from allometric equation and annual diameter at breast height(DBH)increment determination.Important Findings Annual E_(t) and T_(r) were 430±31 and 239±17 mm,respectively.Annual T_(r) accounts for 56±8%of total evapotranspiration on average.WUE(NPP/T_(r))of the E.grandis×E.urophylla was estimated to be 3.3–3.9 mmol·mol^(−1).Based on the comparative analysis of T_(r) and WUE,E.grandis×E.urophylla had a high productivity due to its high WUE without exhibiting prodigal water use.Meteorological factors including vapor pressure deficit and global solar radiation(R_(s))were key factors regulating E_(t) and T_(r) in our research site.Annual surface runoff,E_(t) and canopy interception occupied 7%,27–30%and 16%of total precipitation,while the remaining 46–50%of precipitation was used for sustaining groundwater recharge and altering soil water storage.The higher runoff coefficient(7.1%)indicated the weaker capability of E.grandis×E.urophylla to reserve water resource than natural forests and less disturbed plantations.The planting and protection of understory vegetation may decrease the surface runoff and exert beneficial effects on water conservation capacity of Eucalyptus plantation. | Yanting Hu Ping Zhao Yuqing Huang Liwei Zhu Guangyan Ni Xiuhua Zhao Zhihong Huang | 2019 | Journal of Plant Ecology2019,12,6: | 2 |

| 19 | A thermally flexible and multi-site tactile sensor for remote 3D dynamic sensing imaging显示文摘A flexible,multi-site tactile and thermal sensor(MTTS)based on polyvinylidene fluoride(resolution 50×50)is reported.It can be used to implement spatial mapping caused by tactile and thermal events and record the two-dimensional motion trajectory of a tracked target object.The output voltage and current signal are recorded as amapping by sensing the external pressure and thermal radiation stimulus,and the response distribution is dynamically observed on the three-dimensional interface.Through the mapping relationship between the established piezoelectric and pyroelectric signals,the piezoelectric component and the pyroelectric component are effectively extracted from the composite signals.The MTTS has a good sensitivity for tactile and thermal detection,and the electrodes have good synchronism.In addition,the signal interference is less than 9.5%and decreases as the pressure decreases after the distance between adjacent sites exceeds 200μm.The integration of MTTS and signal processing units has potential applications in human-machine interaction systems,health status detection and smart assistive devices. | Guoting Xia Yinuo Huang Fujiang Li Licheng Wang Jinbo Pang Liwei Li Kai Wang | 2020 | Frontiers of Chemical Science and Engineering2020,14,6: | 1 |
