1Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
2Emission factors of gaseous carbonaceous species from residential combustion of coal and crop residue briquettes显示文摘Experiments were performed to measure the emission factors (EFs) of gaseous carbonaceous species, such as CO2, CO, CH4, and non-methane volatile organic compounds (NMVOCs), from the combustion of five types of coal of varying organic maturity and two types of biomass briquettes under residential burning conditions. Samples were collected in stainless steel canisters and 2,4- dinitrophenylhydrazine (DNPH) cartridges and were analyzed by GC FID/MS and HPLC, respectively. The EFs from crop residue briquette burning were generally higher than those from coals, with the exception of CO2. The dominant NMVOC species identified in coal smoke were carbonyls (41.7%), followed by C2 unsaturated hydrocarbons (29.1%) and aromatics (12.1%), while C2 unsaturated hydrocarbons were the dominant species (68.9%) emitted from the combustion of crop residue briquettes, followed by aromatics (14.4%). A comparison of burning normal crop residues in stoves and the open field indicated that briquettes emitted a larger proportion of ethene and acetylene. Both combustion efficiency and coal organic maturity had a significant impact on NMVOC EFs from burning coal: NMVOC emissions increased with increasing coal organic maturity but decreased as the combustion efficiency improved. Emissions from the combustion of crop residue briquettes from stoves occurred mainly during the smoldering process, with low combustion efficiency. 3Characteristics of paleoflood deposits archived in unit T0403 of Yuxi Site in the Three Gorges reservoir areas,China显示文摘Based on the detailed analyses,multi-proxies such as AMS 14 C dating,grain size,component and morphology of heavy minerals,micromorphology of zircon,magnetic susceptibility,Rb/Sr and Hg content are used to research the paleoflood sediments archived in the Yuxi Site.The research indicates that, since 7.6 kaBP,at least 16 times of paleoflood with water level above Wusong Elevation 147.024 m(a.s.l.) left deposits in the Neolithic layers of unit T0403 from the Yuxi Site.The results are induced from the following aspects:(1)The plaeoflood sediments take on great similarities with modern flood sediments in the Yuxi Site and Zhongba Site in probability cumulative curves which mainly show a pattern of 3 segments in fluvial pattern.(2)There is some resemblance between the heavy mineral components and the zircon shape characteristics of paleoflood deposits and those of modern flood deposits.(3) Magnetic susceptibility values(40.44―70.10SI)are lower than those of sediments from cultural layers (59.59―188.68SI).(4)Hg values(290.71―742.51 ng/g)are lower than those of sediments from cultural layers(344.16―10518.17 ng/g).(5)Rb/Sr values are higher than those of sediments from cultural layers, while those of the 4th,5th,6th,7th,8th cultural layers are high,which shows that they are inundated by paleoflood.The reason for many flood deposits existing is related to the site situated on the first terrace where the Yuxi River joins with the Yangtze River.As there are some similarities between the plaeoflood sediments and those of the Zhongba Site,it is feasible to confirm the existence of paleoflood sediments based on the above points.ZHU Cheng MA ChunMei XU WeiFeng BAI JiuJiang ZHENG ChaoGui ZHU GuangYao WANG HuiLin CHEN Ye LU XueFeng 2008Chinese Science Bulletin2008,53,S1:16
4Autophagy receptor CCDC50 tunes the STING-mediated interferon response in viral infections and autoimmune diseases显示文摘DNA sensing and timely activation of interferon(IFN)-mediated innate immunity are crucial for the defense against DNA virus infections and the clearance of abnormal cells.However,overactivation of immune responses may lead to tissue damage and autoimmune diseases;therefore,these processes must be intricately regulated.STING is the key adaptor protein,which is activated by cyclic GMP-AMP,the second messenger derived from cGAS-mediated DNA sensing.Here,we report that CCDC50,a newly identified autophagy receptor,tunes STING-directed type I IFN signaling activity by delivering K63-polyubiquitinated STING to autolysosomes for degradation.Knockout of CCDC50 significantly increases herpes simplex virus 1(HSV-1)-or DNA ligand-induced production of type I IFN and proinflammatory cytokines.Ccdc50-deficient mice show increased production of IFN,decreased viral replication,reduced cell infiltration,and improved survival rates compared with their wild-type littermates when challenged with HSV-1.Remarkably,the expression of CCDC50 is downregulated in systemic lupus erythematosus(SLE),a chronic autoimmune disease.CCDC50 levels are negatively correlated with IFN signaling pathway activation and disease severity in human SLE patients.CCDC50 deficiency potentiates the cGAS-STING-mediated immune response triggered by SLE serum.Thus,our findings reveal the critical role of CCDC50 in the immune regulation of viral infections and autoimmune diseases and provide insights into the therapeutic implications of CCDC50 manipulation.Panpan Hou Yuxin Lin Zibo Li Ruiqing Lu Yicheng Wang Tian Tian Penghui Jia Xi Zhang Liu Cao Zhongwei Zhou Chunmei Li Jieruo Gu Deyin Guo 2021Cellular & Molecular Immunology2021,18,10:3
5Kinetic calculations for the thermal decomposition of calcium propionate under non-isothermal conditions显示文摘Calcium propionate (CP) is shown to be useful for simultaneous SO2/NO reduction in coal-fired power plants and its thermal decomposition characteristics are measured by thermogravimetric analysis in a feasibility study into more complete reduction of these hazardous gases. Calcium carbonate (CC), which has been used primarily for in-furnace desulfuration, was used for comparison. The thermal decomposition of this organic calcium-based sorbent began at low temperature, i.e. the carboxylic radical was evaporated from 565 K to 759 K for CP and the corresponding mass loss percentage was 47.79%. The residual was subsequently decomposed to release carbon dioxide between 843 K and 1012 K. The latter phase of the process occurred more readily than with CC because of the loose structure of CP resulting from evaporation of the carboxylic radical in the low temperature zone, which could be seen directly by scanning electron microscope. The maximum mass loss rates of this phase occurred at temperatures of 972 K and 1012 K for CP and CC, respectively. The Ozawa-Flynn-Wall method was used to calculate the activation energy during the thermal decomposition process at heating rates of 5, 7.5, 10 and 15 K/min. The result further confirmed the multistage characteristic of CP thermal decomposition, which could be seen in differential thermogravimetry curves. 6Measurement of Rural Inclusive Financial Development and Analysis of Spatial Effects on Anti-poverty显示文摘Based on two dimensions of supply and demand,we use six indicators to establish a rural inclusive financial development index and evaluate the development of China's rural inclusive finance. We use spatial panel data model to examine the relationship between financial inclusion and rural poverty. Results show that:( i) both the rural inclusive finance and rural residents' per capita consumption present spatial clustering feature and show significant spatial correlation;( ii) the development of rural inclusive finance has significant spatial effect on poverty alleviation along time and space dimensions. 17Synthesis and photoactivity of pH-responsive amphiphilic block polymer photosensitizer bonded zinc phthalocyanine显示文摘A zinc tetraaminophthalocyanine derivative, zinc tetra(methacryloyl moiety)aminophthalocyanine (MeZnAPc) (with a double bond) was synthesized by the reaction between zinc tetraaminophthalocyanine (ZnTAPc) and methacryloyl chloride. Atom transfer radical polymerization (ATRP) was employed as the polymerization technique to obtain a novel pH-responsive polymeric photosensitizer (PEG110-b-P(DPAn-co-MeZnAPcm)) by copolymerizing of methoxypolyethylene glycols (MPEG) (as reducing agent), 2-(isopropylamino)ethyl methacrylate (DPA) and MeZnAPc. This photosensitizer was characterized by UV-vis spectroscopy, FTIR, H NMR, etc. The results indicated that the photosensitizer presented the well pH-responsive be 1 havior around the pH range 6.0-6.5 and the high photoactivity to 1,3-diphenylisobenzofuran (DPBF). The result of photocatalysis oxidation of L-tryptophan (L-Try) suggested that zinc phthalocyanine could present high photoactivity due to its dispersivity at pH 5.5 without formation of micelles, and its photoactivity decreased dramatically at pH 7.4 due to wrapping ZnTAPc into the micelles. 19Quantitative assessment of the influence of terrace and check dam construction on watershed topography显示文摘Terrace and check dam construction has substantially changed land surface morphology,which in turn affects modern surface processes.Digital elevation models(DEMs)provide an effective way to quantitatively analyze surface morphology and processes.However,existing DEMs lack sufficient ability to express artificial terrain.Based on 1:10000 topographic maps of the Zhifanggou watershed,a series of artificial terrain DEMs for the study site were constructed by both field investigation and remote sensing images from 1938 to 2010.Digital terrain analysis was used to quantitatively assess the influence of terrace and check dam construction on the watershed terrain.The results showed that the artificial terrain DEM could capture the spatial distribution patterns of terraces and dam lands and improved the ability of DEM to express terrain.The construction of terraces and check dams clearly changed the surface elevation.The average elevation change of each terrace mainly ranged between–1.5 and 1.5 m,while the annual average deposition height of the dam lands was 9.16 cm.The average slope,slope length,and slope length and steepness factor of the watershed decreased with the effect of the artificial terrain on the surface,and their averages decreased by 0.65°,6.75 m,and 0.83,respectively,from 1938 to 2010.Although the construction of terraces reduced their surface slope to nearly 0°,the slope of terrace embankments rapidly increased,to more than 45°,which may lead to gravitational erosion and potential terrace damage.Terracing reduced the slope length in both the terrace distribution area and downslope of the terraces.Check dam deposition reduced the slope and slope length of the channel.This study contributes to a better understanding of the topographic change rules after terrace and check dam construction,and aids in elucidating the mechanisms of soil erosion process influenced by artificial topography.Guowei PANG Qinke YANG Chunmei WANG Rui LI Lu ZHANG 2020Frontiers of Earth Science2020,14,2:1
