| 1 | A novel STING agonist-adjuvanted pan-sarbecovirus vaccine elicits potent and durable neutralizing antibody and T cell responses in mice,rabbits and NHPs显示文摘The emergence of SARS-CoV-2 variants and potentially other highly pathogenic sarbecoviruses in the future highlights the need for pan-sarbecovirus vaccines.Here,we discovered a new STING agonist,CF501,and found that CF501-adjuvanted RBD-FC vaccine(CF501/RBD-FC)elicited significantly stronger neutralizing antibody(nAb)and T cell responses than Alum-and cGAMP-adjuvanted RBD-FC in mice.Vaccination of rabbits and rhesus macaques(nonhuman primates,NHPs)with CF501/RBD-FC elicited exceptionally potent nAb responses against SARS-CoV-2 and its nine variants and 41 S-mutants,SARS-CoV and bat SARSr-CoVs.CF501/RBD-FC-immunized hACE2-transgenic mice were almost completely protected against SARS-CoV-2 challenge,even 6 months after the initial immunization.NHPs immunized with a single dose of CF501/RBD-FC produced high titers of nAbs.The immunized macaques also exhibited durable humoral and cellular immune responses and showed remarkably reduced viral load in the upper and lower airways upon SARS-CoV-2 challenge even at 108 days post the final immunization.Thus,CF501/RBD-Fc can be further developed as a novel pan-sarbecovirus vaccine to combat current and future outbreaks of sarbecovirus diseases. | Zezhong Liu Jie Zhou Wei Xu Wei Deng Yanqun Wang Meiyu Wang Qian Wang Ming Hsieh Jingming Dong Xinling Wang Weijin Huang Lixiao Xing Miaoling He Chunlin Tao Youhua Xie Yilong Zhang Youchun Wang Jincun Zhao Zhenghong Yuan Chuan Qin Shibo Jiang Lu Lu | 2022 | Cell Research2022,32,3: | 5 |

| 5 | Promoter and coding sequence diversity of CsCCD1 may contribute to the differential accumulation of floralβ-ionone in fresh tea leaves显示文摘The carotenoid-derived volatileβ-ionone makes an important contribution to tea fragrance.Here,we qualitatively and quantitatively analysed 15 carotenoids in tea leaves of 13 cultivars by UHPLC-APCI-MS/MS.The 13 cultivars were divided into two groups by PCA(Principal Component Analysis)clustering analysis of their carotenoid content,and OPLS-DA(Orthogonal projections to latent structures)indicated that the levels ofβ-carotene(VIP=2.89)and lutein(VIP=2.30)were responsible for much of the variation between the two groups.Interestingly,theβ-carotene toβ-ionone conversion rates in Group 1 were higher than in Group 2,while theβ-carotene content was significantly lower in Group 1 than in Group 2.Theβ-ionone content was significantly higher in Group 1.Pearson Correlation Coefficient calculation between the transcription level of candidate genes(CsCCD1 and CsCCD4)and the accumulation ofβ-ionone indicated that CsCCD1 may involve in the formation ofβ-ionone in 13 cultivars.Prokaryotic expression and in vitro enzyme activity assays showed that‘Chuanhuang 1’had an amino acid mutation in carotenoid cleavage dioxygenases 1(CsCCD1)compared with‘Shuchazao’,resulting in a significantly higherβ-ionone content in‘Chuanhuang 1’.Sequence analysis showed that‘Chuanhuang 1’and‘Huangdan’had different CsCCD1 promoter sequences,leading to significantly higher CsCCD1 expression andβ-ionone accumulation in‘Chuanhuang 1’.These results indicated that the promoter and coding sequence diversity of CsCCD1 might contribute to the differential accumulation ofβ-ionone in different tea cultivars. | Jingming Wang Mingyue Zhao Ting Gao Yingying Feng Feiquan Wang Yuting Pan Jieyang Jin Tingting Jing Mengqian Lu Mengting Zhang Danyang Guo Xiaochun Wan Wilfried Schwab Chuankui Song | 2023 | Horticultural Plant Journal2023,9,4: | 1 |
| 6 | Tsunami hazard and mitigation analysis for bathing beaches in China based on numerical simulations显示文摘Bathing beaches are usually the first to suffer disasters when tsunamis occur,owing to their proximity to the sea.Several large seismic fault zones are located off the coast of China.The impact of each tsunami scenario on Chinese bathing beaches is different.In this study,numerical models of the worst tsunami scenarios associated with seismic fault zones were considered to assess the tsunami hazard of bathing beaches in China.Numerical results show that tsunami waves from the Pacific Ocean could affect the East China Sea coast through gaps between the Ryukyu Islands.The Zhejiang and Shanghai coasts would be threatened by a tsunami from Ryukyu Trench,and the coasts of Hainan and Guangdong provinces would be threatened by a tsunami from the Manila Trench.The tsunami hazard associated with the Philippine Trench scenario needs particular attention.Owing to China’s offshore topography,the sequential order of tsunami arrival times to coastal provinces in several tsunami scenarios is almost the same.According to the tsunami hazard analysis results,Yalongwan Beach and eight other bathing beaches are at the highest hazard level.A high-resolution numerical calculation model was established to analyze the tsunami physical characteristics for the high-risk bathing beaches.To explore mitigating effects of a tsunami disaster,this study simulated tsunami propagation with the addition of seawalls.The experimental results show that the tsunami prevention seawalls constructed in an appropriate shallow water location have some effect on reducing tsunami hazard.Seawalls separated by a certain distance work even better.The analysis results can provide a scientific reference for subsequent preventive measures such as facility construction and evacuation. | Jingming Hou Wei Lu Tingting Fan Peitao Wang | 2022 | Acta Oceanologica Sinica2022,41,12: | 0 |

| 9 | Pre-Earthquake Ionospheric Anomalies of the Wenchuan Earthquake Studied with DEMETER Satellite显示文摘The pre-earthquake ionospheric anomalies in Wenchuan,China(21°-41°N,93°-113°E)are studied and analyzed using the summer nighttime data from 2005 to 2008 measured by DEMETER(Detection of Electro-Magnetic Emission Transmitted from Earthquake Regions)satellite detectors ICE(Internet Communications Engine),IAP(In Application Programming),and ISL(Interior Switching Link).In this paper,we take the 12 May 2008 Wenchuan earthquake as an example,use the spatial gridding method to construct the background field over the epicenter,analyze the background characteristics of very low frequency(VLF)electric field components,low-energy particle parameters,and plasma parameters,and define the perturbation intensity index of each parameter before the earthquake to extract each parameter anomaly in both space and time dimensions.The results show that the background values of some ionospheric parameters in the Wenchuan area are related to spatial distribution.Moreover,anomalous enhancement of low-frequency electric field power spectral density,H+concentration,He+concentration and ion concentration with different intensities and anomalous weakening of ion temperature were extracted in the fifteen days before the Wenchuan earthquake.After filtering the data to exclude external interference,such as solar activity,this paper concludes that there is some connection between these anomalies and the Wenchuan earthquake. | LU Jingming HU Yaogai JIANG Chunhua ZHAO Zhengyu ZHANG Yuannong | 2023 | Wuhan University Journal of Natural Sciences2023,28,5: | 0 |
