| 1 | Roles of Fas signaling pathway in vitamin E succinateinduced apoptosis in human gastric cancer SGC-7901 cells显示文摘AIM: To investigate the roles of Fas signaling pathway in vitamin E succinate-induced apoptosis in human gastric cancer SGC-7901 cells.METHODS: Human gastric cancer SGC-7901 cells were treated with VES at 5, 10, 20 mg@L-1, succinic acid and vitamin E as vehicle control and condition media only as untreated (UT) control. Apoptotic morphology was observed by DAPI staining. Western blot analysis was applied to measure the expression of Fas, FADD and caspase-8 proteins. After the cells were transiently transfected with Fas and FADD antisense oligonucleotides, respectively, caspase-8 activity was determined by flurometric method.RESULTS: The morphologically apoptotic changes were observed after VES treatment by DAPI staining. 23.7 % and 89.6 % apoptosis occurred after 24 h and 48 h of 20 mg@L-1 VES treatment, respectively. The protein levels of Fas, FADD and caspase-8 were evidently increased in a dose-dependent manner after 24 h of VES treatment. The blockage of Fas by transfection with Fas antisense oligonucleotides obviously inhibited the expression of FADD protein. | 3 | Individualized prediction of perineural invasion in colorectal cancer: development and validation of a radiomics prediction model显示文摘Objective: To develop and validate a radiomics prediction model for individualized prediction of perineural invasion(PNI) in colorectal cancer(CRC).Methods: After computed tomography(CT) radiomics features extraction, a radiomics signature was constructed in derivation cohort(346 CRC patients). A prediction model was developed to integrate the radiomics signature and clinical candidate predictors [age, sex, tumor location, and carcinoembryonic antigen(CEA) level]. Apparent prediction performance was assessed. After internal validation, independent temporal validation(separate from the cohort used to build the model) was then conducted in 217 CRC patients. The final model was converted to an easy-to-use nomogram.Results: The developed radiomics nomogram that integrated the radiomics signature and CEA level showed good calibration and discrimination performance [Harrell's concordance index(c-index): 0.817; 95% confidence interval(95% CI): 0.811–0.823]. Application of the nomogram in validation cohort gave a comparable calibration and discrimination(c-index: 0.803; 95% CI: 0.794–0.812).Conclusions: Integrating the radiomics signature and CEA level into a radiomics prediction model enables easy and effective risk assessment of PNI in CRC. | 4 | Improving Plant Genome Editing with High-Fidelity xCas9 and Non-canonical PAM-Targeting Cas9-NG显示文摘Two recently engineered SpCas9 variants, namely xCas9 and Cas9-NG, show promising potential in improving targeting specificity and broadening the targeting range. In this study, we evaluated these Cas9 variants in the model and crop plant, rice. We first tested xCas9-3.7, the most effective xCas9 variant in mammalian cells, for targeted mutagenesis at 16 possible NGN PAM (protospacer adjacent motif) combinations in duplicates. xCas9 exhibited nearly equivalent editing efficiency to wild-type Cas9 (Cas9-WT) at most canonical NGG PAM sites tested, whereas it showed limited activity at non-canonical NGH (H = A, C, T) PAM sites. High editing efficiency of xCas9 at NGG PAMs was further demonstrated with C to T base editing by both rAPOBECI and PmCDAI cytidine deaminases. With mismatched sgRNAs, we found that xCas9 had improved targeting specificity over the Cas9-WT. Furthermore, we tested two Cas9-NG variants, Cas9-NGv1 and Cas9-NG, for targeting NGN PAMs. Both Cas9-NG variants showed higher editing efficiency at most non-canonical NG PAM sites tested, and enabled much more efficient editing than xCas9 at AT-rich PAM sites such as GAT, GAA, and CAA. Nevertheless, we found that Cas9-NG variants showed significant reduced activity at the canonical NGG PAM sites. In stable transgenic rice lines, we demonstrated that Cas9-NG had much higher editing efficiency than Cas9-NGv1 and xCas9 at NG PAM sites. To expand the base-editing scope, we developed an efficient C to T base-editing system by making fusion of Cas9-NG nickase (D10A version), PmCDAI, and UGI. | 8 | Failure mechanism and control technology of water-immersed roadway in high-stress and soft rock in a deep mine显示文摘Aiming at soft rock ground support issues under conditions of high stress and long-term water immersion, the ground failure mechanism is revealed by taking the deep-water sumps of Jiulong Mine as the engineering background and employing field investigation, tests of rock structure, mechanical properties and mineral composition. The main factors leading to the surrounding rock failure include the high and complex stress state of the water sumps, high-clay content and water-weakened rock, and the unreasonable support design. In this paper, the broken and fractured rock mass near roadway opening is considered as ground small-structure, and deep stable rock mass as ground large-structure. A support technology focusing on cutting off the water, strengthening the small structure of the rock and transferring the large structure of the rock is proposed. The proposed support technology of interconnecting the large and small structures, based on high-strength bolts, high-stiffness shotcrete layer plugging water,strengthening the small structure with deep-hole grouting and shallow-hole grouting, highpretensioned cables tensioned twice to make the large and small structures bearing the pressure evenly,channel-steel and high-pretensioned cables are used to control floor heave. | 9 | Apatinib, a selective VEGFR2 inhibitor, improves the delivery of chemotherapeutic agents to tumors by normalizing tumor vessels in LoVo colon cancer xenograft mice显示文摘Tumor vascular normalization has been proposed as a therapeutic strategy for malignant neoplasms, which can also interpret the synergistic effect of anti-angiogenesis agents combined with chemotherapy. Apatinib (Apa), a highly selective VEGFR2 inhibitor, attracts much attentions due to its encouraging anticancer activity, especially in the clinical trials of combined treatment. In this study, we investigated whether Apa could promote vascular normalization in tumor in a certain time window. Mice bearing LoVo colon cancer xenograft were orally administrated Apa (150 mg kg^-1 per day) for 5, 7, 10, or 12 days. Apa significantly inhibited tumor growth and decreased the microvessel density. Using multi-photon microscopy and electron microscopy, we found that Apa improved tumor vessel morphology by pruning distorted vessel branches and decreased the gap between endothelial cells after a 7-day treatment. Furthermore, Apa decreased vessel leakage and in creased pericyte coverage on vascular en dothelial cells, suggesting that tumor vessels were more mature and integrated. The intratumoral distribution of adriamycin (ADR) in Apa group was improved from day 7 to 10 without change in plasma drug concen tration. Tumor blood perfusion was also in creased in this window, and the expression of hypoxia induced factor 1a was downregulated, suggesting the effect of Apa on alleviating tumor hypoxic micro-e nvironme nt. In conclusi on, Apa may improve the effective perfusi on of tumor vessels and in crease the intratumoral distribution of ADR in a certain time window via normalizing tumor vessels. | 10 | Mechanisms and pharmacokinetic/pharmacodynamic profiles underlying the low nephrotoxicity and ototoxicity of etimicin显示文摘Etimicin(ETM),a fourth-generation aminoglycosides(AGs),is now widely clinically used in China due to its high efficacy and low toxicity.However,the mechanisms underlying its low nephrotoxicity and ototoxicity remain unclear.In the present study we compared the antibacterial and toxicity profiles of etimicin,gentamicin(GM,a second-generation AG),and amikacin(AMK,a third-generation AG),and investigated their pharmacokinetic properties in the toxicity target organs(kidney and inner ear)and subcellular compartments.We first demonstrated that ETM exhibited superior antibacterial activities against clinical isolates to GM and AMK,and it exerted minimal nephrotoxicity and ototoxicity in rats following multi-dose administration.Then,we conducted pharmacokinetic studies in rats,showed that the three AGs accumulated in the kidney and inner ear with ETM being distributed to a lesser degree in the two toxicity target organs as compared with GM and AMK high-dose groups.Furthermore,we conducted in vitro experiments in NRK-52E rat renal tubular epithelial cells and HEI-OC1 cochlear hair cells,and revealed that all the three AGs were distributed predominantly in the mitochondria with ETM showing minimal accumulation;they not only directly inhibited the activity of mitochondrial complexesⅣandⅤbut also inhibited mitochondrial function and its related PGC-1α-NRF1-TFAM pathway;ETM caused minimal damage to the mitochondrial complex and mitochondrial biogenesis.Our results demonstrate that the minimal otonephrotoxicity of ETM results from its lesser accumulation in mitochondria of target cells and subsequently lesser inhibition of mitochondrial function.These results provide a new strategy for discovering novel AGs with high efficacy and low toxicity. | Lan Yao Jing-wei Zhang Bin Chen Ming-min Cai Dong Feng Qi-zhi Wang Xin-yu Wang Jian-guo Sun Yi-wen Zheng Guang-ji Wang Fang Zhou | 2020 | Acta Pharmacologica Sinica2020,41,6: | 10 |

| 12 | Transcriptional Regulation of miR528 by OsSPL9 Orchestrates Antiviral Response in Rice显示文摘Many microRNAs (miRNAs) are critical regulators of plant antiviral defense.However,little is known about how these miRNAs respond to virus invasion at the transcriptional level.We previously show that defense against Rice stripe virus (RSV) invasion entailed a reduction of miR528 accumulation in rice,alleviating miR528-mediated degradation of L-Ascorbate Oxidase (AO) mRNA and bolstering the antiviral activity of AO.Here we show that the miR528-A0 defense module is regulated by the transcription factor SPL9.SPL9 displayed high-affinity binding to specific motifs within the promoter region of miR528 and activated the expression of miR528 gene in vivo.Loss-of-function mutations in SPL9 caused a significant reduction in miR528 accumulation but a substantial increase of AO mRNA,resulting in enhanced plant resistance to RSV.Conversely,transgenic overexpression of SPL9 stimulated the expression of miR528 gene,hence lowering the level of AO mRNA and compromising rice defense against RSV.Importantly,gain in RSV susceptibility did not occur when SPL9 was overexpressed in mir528 loss-of-function mutants,or in transgenic rice expressing a miR528-resistant AO.Taken together,the finding of SPL9-mediated transcriptional activation of miR528 expression adds a new regulatory layer to the miR528-A0 antiviral defense pathway. | Shengze Yao Zhirui Yang Rongxin Yang Yu Huang Ge Guo Xiangyue Kong Ying Lan Tong Zhou He Wang Wenming Wang Xiaofeng Cao Jianguo Wu Yi Li | 2019 | Molecular Plant2019,12,8: | 10 |

| 15 | Incidence and risk factors of tuberculosis among the elderly population in China:a prospective cohort study显示文摘Background:China is facing challenges of the shifting presentation of tuberculosis(TB)from younger to elderly due to an ageing population,longer life expectancy and reactivation disease.However,the burden of elderly TB and influence factors are not yet clear.To fill the gap,we generated a cohort study to measure the magnitude of TB incidence and associated factors among the elderly population aged 65 years and above in China.Methods:In this cohort established in 2013 through a prevalence survey conducted in selected sites,a total of 34076 elderlies without TB were enrolled into two-year follow-up.We used both active and passive case findings to find out all TB patients among them.The person-year(PY)incidence rates for both bacteriologically positive TB and active TB were calculated.Cox proportional regression model was performed to test effect of risk factors,and the population attributable fraction(PAF)of each risk factor contributing to incident TB among elderlies was calculated.Results:Over the two-year follow-up period,a total of 215 incident active TB were identified,62 of which were bacteriologically positive.The incidence rates for active TB and bacteriologically positive TB were 481.8 per 100000 PY(95%CI:417.4–546.2 per 100000 PY)and 138.9 per 100000 PY(95%CI:104.4–173.5 per 100000 PY),respectively.Incident cases detected by active case finding were significantly higher(P<0.001).Male,non-Han nationality,previously treated TB,ex/current smoker and body mass index(BMI)<18.5 presented as independent predictors for developing TB disease.For developing bacteriologically positive TB,the biggest contribution was from self-reported ex or current smoker(18.06%).And,for developing active TB,the biggest contribution was from non-Han nationality(35.40%),followed by male(26.80%)and age at 75 years and above(10.85%).Conclusions:Ageing population in China had a high TB incidence rate and risk to develop TB disease,implying that National TB Program(NTP)needs to prioritize for elderly.Active case finding should be applied capture more active TB cases among this particular population,especially for male,non-Han nationality,and those with identified risk factors. | Jun Cheng Yan-Ni Sun Can-You Zhang Yan-Ling Yu Li-Hong Tang Hong Peng Ying Peng Yu-Xia Yao Shuang-Yi Hou Jian-Wei Li Jin-Ming Zhao Lan Xia Lin Xu Yin-Yin Xia Fei Zhao Li-Xia Wang Hui Zhang | 2020 | Infectious Diseases of Poverty2020,9,1: | 10 |
| 16 | Design and test of operation parameters for rice air broadcasting by unmanned aerial vehicle显示文摘Considering the difficulty of broadcasting in the small plots and complex terrain in South China,this research aimed to explore a new efficient broadcasting way and figure out advisable operation parameters by using a hollow 12-axis,rotor-wing Unmanned Aerial Vehicle(UAV)which is typically made up of four groups of solid support structure,with each consisting of three axes and two rotor wings.A 3.7 L reverse pyramid-shape seed hopper with a 60 mm×13 mm rectangular outlet at the bottom was designed to realize self-gravity seeding.Rice seed firstly directly falls on the rotating disc driven by direct-current dynamo before being sown.The disc was located 120 mm above the ground,with a diameter of 350 mm.Under constant flight conditions,parameters of the on-board broadcasting devices(dropping speed referring to speed for the outlet and the broadcasting speed for the disc’s rotation speed)determine the uniformity of air broadcasting.The FUTABA T8FG transmitter and receiver system were employed as the remote control device.When the UAV flies at 3 m/s and 2 m above the ground with an expected seeding of 180 grain/m^(2),the RD(Right Down)knob marked 29 and LD(Left Down)knob marked-24 with a disc rotation speed of 900 r/min,the broadcasting lasts for 15 seconds with a 25%opening(1.95 cm^(2))of rectangular outlet.In order to verify the feasibility of air broadcasting parameters,the project team carried out a field broadcasting test by using a hollow multi-rotor UAV at Zhongluotan Test Base in Guangzhou in July,2014.In the square field plots of 0.09 hm^(2),flying path of air broadcasting operation was designed.The collected sampling data of broadcasting quantity per unit area after the test showed that there were,on average,187.4 grains/m^(2)of the five sampling points with a standard deviation of 22.77,and a coefficient of variation of 12.15%which was far smaller than that of artificial broadcasting.The average yield of field broadcasted by UAV is 7705.5 kg/hm^(2)in 2014,implying that rice air broadcasting by UAV is feasible. | Li Jiyu Lan Yubin Zhou Zhiyan Zeng Shan Huang Cong Yao Weixiang Zhang Yang Zhu Qiuyang | 2016 | International Journal of Agricultural and Biological Engineering2016,9,5: | 10 |

| 19 | Graphitic carbon nitride/biochar composite synthesized by a facile ball-milling method for the adsorption and photocatalytic degradation of enrofloxacin显示文摘In order to enhance the removal performance of graphitic carbon nitride(g-C_(3)N_(4)) on organic pollutant, a simultaneous process of adsorption and photocatalysis was achieved via the compounding of biochar and g-C_(3)N_(4). In this study, g-C_(3)N_(4) was obtained by a condensation reaction of melamine at 550 °C. Then the g-C_(3)N_(4)/biochar composites were synthesized by ball milling biochar and g-C_(3)N_(4) together, which was considered as a simple, economical, and green strategy. The characterization of resulting g-C_(3)N_(4)/biochar suggested that biochar and g-C_(3)N_(4) achieved effective linkage. The adsorption and photocatalytic performance of the composites were evaluated with enrofloxacin(EFA) as a model pollutant. The result showed that all the g-C_(3)N_(4)/biochar composites displayed higher adsorption and photocatalytic performance to EFA than that of pure g-C_(3)N_(4). The 50% g-C_(3)N_(4)/biochar performed best and removed 45.2% and 81.1% of EFA(10 mg/L) under darkness and light with a dosage of 1 mg/mL, while g-C_(3)N_(4) were 19.0% and 27.3%, respectively. Besides, 50% g-C_(3)N_(4)/biochar showed the highest total organic carbon(TOC) removal efficiency(65.9%). Radical trapping experiments suggested that superoxide radical( ·O_(2)^(-)) and hole(h~+) were the main active species in the photocatalytic process.
