| 1 | Chemical characteristics of haze particles in Xi'an during Chinese Spring Festival: Impact of fireworks burning显示文摘Fireworks burning releases massive fine particles and gaseous pollutants, significantly deteriorating air quality during Chinese Lunar New Year(LNY) period. To investigate the impact of the fireworks burning on the atmospheric aerosol chemistry, 1-hr time resolution of PM_(2.5) samples in Xi'an during the winter of 2016 including the LNY were collected and detected for inorganic ions, acidity and liquid water content(LWC) of the fine aerosols. PM_(2.5) during the LNY was 167 ± 87 μg/m^3, two times higher than the China National Ambient Air Quality Standard(75 μg/m^3). K^+(28 wt.% of the total ion mass) was the most abundant ion in the LNY period, followed by SO_4^(2-)(25 wt.%) and Cl-(18 wt.%). In contrast, NO_3^-(34 wt.%) was the most abundant species in the haze periods(hourly PM32-2.5> 75 μg/m), followed by SO_4(29.2 wt.%) and NH_4^+(16.3 wt.%), while SO_4^(2-)(35 wt.%) was the most abundant species in the clean periods(hourly PM_(2.5)< 75 μg/m^3), followed by NO_3^-(23.1 wt.%) and NH_4^+(11 wt.%). Being different from the acidic nature in the non-LNY periods, aerosol in the LNY period presented an alkaline nature with a pH value of 7.8 ± 1.3. LWC during the LNY period showed a robust linear correlation with K_2SO_4 and KCl, suggesting that aerosol hygroscopicity was dominated by inorganic salts derived from fireworks burning. | 2 | WUSCHEL-related homeobox1(WOX1)regulates vein patterning and leaf size in Cucumis sativus显示文摘In plants,WUSCHEL-related homeobox1(WOX1)homologs promote lamina mediolateral outgrowth.However,the downstream components linking WOX1 and lamina development remain unclear.In this study,we revealed the roles of WOX1 in palmate leaf expansion in cucumber(Cucumis sativus).A cucumber mango fruit(mf)mutant,resulting from truncation of a WOX1-type protein(CsWOX1),displayed abnormal lamina growth and defects in the development of secondary and smaller veins.CsWOX1 was expressed in the middle mesophyll and leaf margins and rescued defects of the Arabidopsis wox1 prs double mutant.Transcriptomic analysis revealed that genes involved in auxin polar transport and auxin response were highly associated with leaf development.Analysis of the cucumber mf rl(round leaf)double mutant revealed that CsWOX1 functioned in vein development via PINOID(CsPID1)-controlled auxin transport.Overexpression of CsWOX1 in cucumber(CsWOX1-OE)affected vein patterning and produced‘butterfly-shaped’leaves.CsWOX1 physically interacted with CsTCP4a,which may account for the abnormal lamina development in the mf mutant line and the smaller leaves in the CsWOX1-OE plants.Our findings demonstrated that CsWOX1 regulates cucumber leaf vein development by modulating auxin polar transport;moreover,CsWOX1 regulates leaf size by controlling CIN-TCP genes. | Hu Wang Huanhuan Niu Chuang Li Guoyan Shen Xiaofeng Liu Yiqun Weng Tao Wu Zheng Li | 2020 | Horticulture Research2020,7,1: | 6 |

| 4 | Elevated CO2 differentially affects tobacco and rice defense against lepidopteran larvae via the jasmonic acid signaling pathway显示文摘Atmospheric CO_2 levels are rapidly increasing due to human activities. However, the effects of elevated CO_2(ECO_2) on plant defense against insects and the underlying mechanisms remain poorly understood. Here we show that ECO_2 increased the photosynthetic rates and the biomass of tobacco and rice plants, and the chewing lepidopteran insects Spodoptera litura and Mythimna separata gained less and more mass on tobacco and rice plants, respectively. Consistently, under ECO_2, the levels of jasmonic acid(JA), the main phytohormone controlling plant defense against these lepidopteran insects, as well as the main defense-related metabolites, were increased and decreased in insectdamaged tobacco and rice plants. Importantly, bioassaysand quantification of defense-related metabolites in tobacco and rice silenced in JA biosynthesis and perception indicate that ECO_2 changes plant resistance mainly by affecting the JA pathway. We further demonstrate that the defensive metabolites, but not total N or protein, are the main factors contributing to the altered defense levels under ECO_2. | 5 | Targeting a novel inducible GPX4 alternative isoform to alleviate ferroptosis and treat metabolic-associated fatty liver disease显示文摘Metabolic-associated fatty liver disease(MAFLD),which is previously known as non-alcoholic fatty liver disease(NAFLD),represents a major health concern worldwide with limited therapy.Here,we provide evidence that ferroptosis,a novel form of regulated cell death characterized by iron-driven lipid peroxidation,was comprehensively activated in liver tissues from MAFLD patients.The canonical-GPX4(cGPX4),which is the most important negative controller of ferroptosis,is downregulated at protein but not mRNA level.Interestingly,a non-canonical GPX4 transcript-variant is induced(inducible-GPX4,iGPX4)in MAFLD condition.The high fat-fructose/sucrose diet(HFFD)and methionine/choline-deficient diet(MCD)-induced MAFLD pathologies,including hepatocellular ballooning,steatohepatitis andfibrosis,were attenuated and aggravated,respectively,in cGPX4-and iGPX4-knockin mice.cGPX4 and iGPX4 isoforms also displayed opposing effects on oxidative stress and ferroptosis in hepatocytes.Knockdown of iGPX4 by siRNA alleviated lipid stress,ferroptosis and cell injury.Mechanistically,the triggered iGPX4 interacts with cGPX4 to facilitate the transformation of cGPX4 from enzymatic-active monomer to enzymatic-inactive oligomers upon lipid stress,and thus promotes ferroptosis.Co-immunoprecipitation and nano LC–MS/MS analyses confirmed the interaction between iGPX4 and cGPX4.Our results reveal a detrimental role of non-canonical GPX4 isoform in ferroptosis,and indicate selectively targeting iGPX4 may be a promising therapeutic strategy for MAFLD. | Jie Tong Dongjie Li Hongbo Meng Diyang Sun Xiuting Lan Min Ni Jiawei Ma Feiyan Zeng Sijia Sun Jiangtao Fu Guoqiang Li Qingxin Ji Guoyan Zhang Qirui Shen Yuanyuan Wang Jiahui Zhu Yi Zhao Xujie Wang Yi Liu Shenxi Ouyang Chunquan Sheng Fuming Shen Pei Wang | 2022 | Acta Pharmaceutica Sinica B2022,12,9: | 3 |

The local controller first selects candidate substrate nodes for each virtual node in the domain. Then the global controller abstracts substrate network topology based on the candidate nodes and boundary nodes of each domain, and applies Particle Swarm Optimization Algorithm on it to divide virtual network requests. Each local controller then embeds the virtual nodes of the divided single-domain virtual network requests in the domain, and cooperates with other local controllers to embed the inter-domain virtual links. | 13 | Sensitive detection of DNA methyltransferase activity based on supercharged fluorescent protein and template-free DNA polymerization显示文摘DNA methylation, catalyzed by DNA methyltransferases(MTases), is a key component of genetic regulation, and DNA MTases have been regarded as potential targets in anticancer therapy. Herein, based on our previously developed DNA-mediated supercharged green fluorescent protein(Sc GFP)/graphene oxide(GO) interaction, coupled with methylation-initiated template-free DNA polymerization, we propose a novel fluorescence assay strategy for sensitive detection of DNA MTase activity. A hairpin DNA with a methylation-sensitive site and an amino-modified 3′-terminal(DNA-1) was designed and worked as a starting molecule. In the presence of DNA MTase, methylation-sensitive restriction endonuclease, and terminal deoxynucleotidyl transferase(Td T), DNA-1 can be sequentially methylated, cleaved, and further elongated. The resulting long DNA fragments quickly bind with Sc GFP and form the Sc GFP/DNA nanocomplex. Such nanocomplex can effectively protect Sc GFP from being adsorbed and quenched by GO. Without the methylation-initiated DNA polymerization, the fluorescence of Sc GFP will be quenched by GO. Thus, the DNA MTase activity, which is proportional to the amount of DNA polymerization products, can be measured by reading the fluorescence of Sc GFP/GO. The method was successfully used to detect the activity of DNA adenine methylation(Dam) MTase with a wide linear range(0.1–100 U/m L) and a low detection limit of 0.1 U/m L. In addition, the method showed high selectivity and the potential to be applied in a complex sample. | 19 | Screening of Extraction Methods for Glycoproteins from Jellyfish (Rhopilema esculentum) Oral-Arms by High Performance Liquid Chromatography显示文摘In order to select an optimum extraction method for the target glycoprotein (TGP) from jellyfish (Rhopilema esculentum) oralarms, a high performance liquid chromatography (HPLC)–assay for the determination of the TGP was developed. Purified target glycoprotein was taken as a standard glycoprotein. The results showed that the calibration curves for peak area plotted against concentration for TGP were linear (r = 0.9984, y = 4.5895x+47.601) over concentrations ranging from 50 to 400 mgL-1. The mean extraction recovery was 97.84% (CV2.60%). The fractions containing TGP were isolated from jellyfish (R. esculentum) oral-arms by four extraction methods: 1) water extraction (WE), 2) phosphate buffer solution (PBS) extraction (PE), 3) ultrasound-assisted water extraction (UA-WE), 4) ultrasound-assisted PBS extraction (UA-PE). The lyophilized extract was dissolved in Milli-Q water and analyzed directly on a short TSK-GEL G4000PWXL (7.8 mm×300 mm) column.
