1First dark matter search results from the PandaX-Ⅰ experiment显示文摘We report on the first dark-matter(DM)search results from PandaX-I,a low threshold dual-phase xenon experiment operating at the China JinPing Underground Laboratory.In the 37-kg liquid xenon target with 17.4 live-days of exposure,no DM particle candidate event was found.This result sets a stringent limit for low-mass DM particles and disfavors the interpretation of previously-reported positive experimental results.The minimum upper limit,3.7×10-44cm2,for the spin-independent isoscalar DM-particle-nucleon scattering cross section is obtained at a DM-particle mass of 49 GeV/c2at 90%confidence level.XIAO MengJiao XIAO Xiang ZHAO Li CAO XiGuang CHEN Xun CHEN YunHua CUI XiangYi FANG DeQing FU ChangBo GIBONI Karl L. 3Abnormal Effective Connectivity in the Brain is Involved in Auditory Verbal Hallucinations in Schizophrenia显示文摘Information flow among auditory and language processing-related regions implicated in the pathophysiology of auditory verbal hallucinations(AVHs) in schizophrenia(SZ) remains unclear. In this study, we used stochastic dynamic causal modeling(s DCM) to quantify connections among the left dorsolateral prefrontal cortex(inner speech monitoring), auditory cortex(auditory processing), hippocampus(memory retrieval), thalamus(information filtering), and Broca's area(language production) in 17 first-episode drug-na?¨ve SZ patients with AVHs, 15 without AVHs, and 19 healthy controls using resting-state functional magnetic resonance imaging.Finally, we performed receiver operating characteristic(ROC) analysis and correlation analysis between image measures and symptoms. s DCM revealed an increasedsensitivity of auditory cortex to its thalamic afferents and a decrease in hippocampal sensitivity to auditory inputs in SZ patients with AVHs. The area under the ROC curve showed the diagnostic value of these two connections to distinguish SZ patients with AVHs from those without AVHs. Furthermore, we found a positive correlation between the strength of the connectivity from Broca's area to the auditory cortex and the severity of AVHs. These findings demonstrate, for the first time, augmented AVHspecific excitatory afferents from the thalamus to the auditory cortex in SZ patients, resulting in auditory perception without external auditory stimuli. Our results provide insights into the neural mechanisms underlying AVHs in SZ. 5Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). 6An UPLC-MS/MS method for simultaneous determination of multiple constituents in Guizhi Fuling capsule with ultrafast positive/negative ionization switching显示文摘Guizhi Fuling capsule(GFC), a traditional Chinese medicine(TCM) with effects of promoting blood circulation and dissipating blood stasis, has been widely used in the clinic. Because of the complex matrix and various chemical structure types, quality control of GFC remains great challenge. In the present study, an ultra performance liquid chromatography hybrid triple-quadrupole mass spectrometry(UPLC-QQQ MS) method with ultrafast positive/negative ionization switching was developed for simultaneous determination of 18 bioactive components in GFC, including methyl gallate, ethyl gallate, oxypaeoniflorin, benzoic acid, albiflorin, paeonolide, paeoniflorin, 1, 2, 3, 4, 6-pentagalloylglucose, mudanpioside C, benzoyloxypaeoniflorin, benzoylpaeoniflorin, pachymic acid, amygdalin, cinnamaldehyde, paeonol, cinnamic acid, 4-hydroxybenzoic acid, and gallic acid. Separation was performed on an Agilent Zorbax Extend–C18 column(2.1 mm × 50 mm, 1.8 μm), using a gradient elution with acetonitrile and water containing 0.1% formic acid. Cholic acid was selected as the internal standard. This newly developed method was fully validated for linearity, precision, accuracy, and stability, and then applied to quality assessment of GFC. Finally, the batch-to-batch reproducibility of GFC samples was evaluated by the cosine ration and Euclidean distance method, which showed high quality consistency. 9Characterization of edible bird’s nest by peptide fingerprinting with principal component analysis显示文摘OBJECTIVES:Proteins are the major component and play a key role in nutritious and therapeutic functions of edible bird’s nest(EBN);however,limited studies have been conducted on the protein due to difficulties in extraction,isolation as well as identification.This study aimed to provide comprehensive information for the quality evaluation of EBN peptides,which would be a valuable reference for further study on EBN proteins.METHODS:Here,we developed a quality control method using high performance liquid chromatography(HPLC)peptide fingerprints deriving from EBN being digested with simulated gastric fluid.The characteristic peptide peaks were collected and identified by LC-MS/MS.RESULTS:The characteristic peptide peaks,corresponding to the protein fragments of acidic mammalian chitinase-like,lysyl oxidase,and Mucin-5AC-like,were identified and quantified.Interestingly,the principal component analysis indicated that the fingerprints were able to discriminate colour of EBN(white/red)and production sites(cave/house)of White EBN on the same weight basis.As proposed by the model developed in this study,Muc-5AC-like and AMCaselike proteins were the markers with the highest discriminative power.CONCLUSIONS:The overall findings suggest that HPLC peptide fingerprints were able to clearly demonstrate peptide profile differences between genuine and adulterated EBN samples;and classify EBN samples by its color and production site.In addition,the protein identification results suggested that Muc-5AC-like protein was the major protein in EBN.Chun-Fai Wong Gallant Kar-Lun Chan Ming-Lu Zhang Ping Yao Huang-Quan Lin Tina Ting-Xia Dong Geng Li Xiao-Ping Lai Karl Wah-Keung Tsim 2017Food Quality and Safety2017,1,1:2

19Isolation of prostrate turfgrass mutants via screening of dwarf phenotype and characterization of a perennial ryegrass prostrate mutant显示文摘Prostrate turf varieties are desirable because of their increased low mowing tolerance,heat resistance,traffic resistance and ground coverage compared with upright varieties.Mutation breeding may provide a powerful tool to create prostrate varieties,but there are no simple,straightforward methods to screen for such mutants.Elucidation of the molecular basis of the major‘green revolution’traits,dwarfism and semi-dwarfism,guided us to design a simple strategy for isolating dwarf mutants of perennial ryegrass(Lolium perenne L.).We have shown that gamma-ray-mediated dominant dwarf mutants can be easily screened for at the three-leaf stage.About 10%of dwarf mutant lines also displayed a prostrate phenotype at mature stages(>10 tillers).One prostrate line,Lowboy I,has been characterized in detail.Lowboy I had significantly shorter canopy,leaf blade and internode lengths compared with wild type.Lowboy I also exhibited greater tolerance to low mowing stress than wild type.Exogenous gibberellic acid(GA)restored Lowboy I to a wild-type phenotype,indicating that the dwarf and prostrate phenotypes were both due to GA deficiency.We further showed that phenotypes of Lowboy I were dominant and stably inherited through sexual reproduction.Prostrate turfgrass mutants are difficult to screen for because the phenotype is not observed at young seedling stages,therefore our method represents a simple strategy for easily isolating prostrate mutants.Furthermore,Lowboy I may provide an outstanding germplasm for breeding novel prostrate perennial ryegrass cultivars.Junmei Chen Chandra Thammina Wei Li Hao Yu Huseyin Yer Rania El-Tanbouly Manon Marron Lorenzo Katin-Grazzini Yongqin Chen John Inguagiato Richard J.McAvoy Karl Guillard Xian Zhang Yi Li 2016Horticulture Research2016,3,1:1
20Development and internal validation of the Comprehensive ALPPS Preoperative Risk Assessment(CAPRA)score:is the patient suitable for Associating Liver Partition and Portal vein ligation for Staged hepatectomy(ALPPS)?显示文摘Background:Preoperative patient selection in Associating Liver Partition and Portal vein ligation for Staged hepatectomy(ALPPS)is not always reliable with currently available scores,particularly in patients with primary liver tumor.This study aims to(I)to determine whether comorbidities and patients characteristics are a risk factor in ALPPS and(II)to create a score predicting 90-day mortality preoperatively.Methods:Thirteen high-volume centers participated in this retrospective multicentric study.A risk analysis based on patient characteristics,underlying disease and procedure type was performed to identify risk factors and model the Comprehensive ALPPS Preoperative Risk Assessment(CAPRA)score.A nonparametric receiver operating characteristic analysis was performed to estimate the predictive ability of our score against the Charlson Comorbidity Index(CCI),the age-adjusted CCI(aCCI),the ALPPS risk score before Stage 1(ALPPS-RS1)and Stage 2(ALPPS-RS2).The model was internally validated applying bootstrapping.Results:A total of 451 patients were included.Mortality was 14.4%.The CAPRA score is calculated based on the following formula:(0.1×age)−(2×BSA)+1(in the presence of primary liver tumor)+1(in the presence of severe cardiovascular disease)+2(in the presence of moderate or severe diabetes)+2(in the presence of renal disease)+2(if classic ALPPS is planned).The predictive ability was 0.837 for the CAPRA score,0.443 for CCI,0.519 for aCCI,0.693 for ALPPS-RS1 and 0.807 for ALPPS-RS2.After 1,000 cycles of bootstrapping the C statistic was 0.793.The accuracy plot revealed a cut-off for optimal prediction of postoperative mortality of 4.70.Conclusions:Comorbidities play an important role in ALPPS and should be carefully considered when planning the procedure.By assessing the patient’s preoperative condition in relation to ALPPS,the CAPRA score has a very good ability to predict postoperative mortality.Ivan Capobianco Karl J.Oldhafer Mohammed-Hossein Fard-Aghaie Ricardo Robles-Campos Roberto Brusadin Henrik Petrowsky Michael Linecker Arianeb Mehrabi Katrin Hoffmann Jun Li Asmus Heumann Roberto Hernandez-Alejandro Mauro Enrique Tun-Abraham Elio Jovine Matteo Serenari Bergthor Bjornsson Per Sandström Ruslan Alikhanov Mikhail Efanov Paolo Muiesan Andrea Schlegel Thomas M.van Gulik Pim B.Olthof Gregor Alexander Stavrou Lina Maria Serna-Higuita Alfred Königsrainer Silvio Nadalin 2022Hepatobiliary Surgery and Nutrition2022,11,1:1
