| 2 | Calcium signaling and T-type calcium channels in cancer cell cycling显示文摘Regulation of intracellular calcium is an important signaling mechanism for cell proliferation in both normal and cancerous cells. In normal epithelial cells, free calcium concentration is essential for cells to enter and accomplish the S phase and the M phase of the cell cycle. In contrast, cancerous cells can pass these phases of the cell cycle with much lower cytoplasmic free calcium concentrations, indicating an alternative mechanism has developed for fulfilling the intracellular calcium requirement for an increased rate of DNA synthesis and mitosis of fast replicating cancerous cells. The detailed mechanism underlying the altered calcium loading pathway remains unclear; however, there is a growing body of evidence that suggests the T-type Ca2+ channel is abnormally expressed in cancerous cells and that blockade of these channels may reduce cell proliferation in addition to inducing apoptosis. Recent studies also show that the expression of T-type Ca2+ channels in breast cancer cells is proliferation state dependent, i.e. the channels are expressed at higher levels during the fast-replication period, and once the cells are in a non-proliferation state, expression of this channel isminimal. Therefore, selectively blocking calcium entry into cancerous cells may be a valuable approach for preventing tumor growth. | 3 | Observations of the Karimata Strait througflow from December 2007 to November 2008显示文摘In order to quantitatively estimate the volume and property transports between the South China Sea and Indonesian Seas via the Karimata Strait, two trawl-resistant bottom mounts, with ADCPs embedded, were deployed in the strait to measure the velocity profile as part of the South China Sea-Indonesian Seas transport/exchange (SITE) program. A pair of surface and bottom acoustic modems was employed to transfer the measured velocity without recovering the mooring. The advantage and problems of the instruments in this field work are reported and discussed. The field observations confirm the existence of the South China Sea branch of Indonesian throughflow via the Karimata Strait with a stronger southward flow in boreal winter and weaker southward bottom flow in boreal summer, beneath the upper layer northward (reversal) flow. The estimate of the averaged volume, heat and freshwater transports from December 2007 to March 2008 (winter) is (-2.7 ± 1.1) × 10 6 m3/s, (-0.30 ± 0.11) PW, (-0.18 ± 0.07) × 106m3/s and from May to September 2008 (summer) is (1.2 ± 0.6) × 106m3/s, (0.14 ± 0.03) PW, (0.12 ± 0.04) × 106m3/s and for the entire record from December 2007 to October 2008 is (-0.5 ± 1.9) × 10 6 m3/s, (-0.05 ± 0.22) PW, (-0.01 ± 0.15) × 106m3/s (negative/positive represents southward/northward transport), respectively. The existence of southward bottom flow in boreal summer implies that the downward sea surface slope from north to south as found by Fang et al. | 6 | EXPERIMENTAL AND NUMERICAL RESEARCH ON BULLDOZER WORKING PROCESS显示文摘A simulative analysis coupled with experiment on behaviors of a soil bed cut by a model bulldozer blade is carried out using the finite element/distinct element method(FE/DEM) facility built in the ELFEN package. Before simulation,the soil specimens are examined through uniaxial tensile/compression,triaxial compression and direct shear tests to obtain model characteristics and relevant parameters,then soil cutting experiments are carried out via a mini-soil bin system with a soil bed of 60/120 mm in width and 10 mm in depth cut by a 1/9 scale model bulldozer blade moving with the velocity of 10 mm/s. The soil constitutive model includes the tensile elastic model for tensile breakage and the compressive elastoplastic relationship with Mohr-Coulomb criterion. The cutting length in simulation is set as 1/4 of that in the experiment divided into 1 869 triangular elements. The comparison between the simulated results and experimental ones shows that the used model is capable of analyzing soil dynamic behaviors qualitatively,and the predicted fracturing profiles in general conform to the experiment. | 7 | Centrifugal model test on a riverine landslide in the Three Gorges Reservoir induced by rainfall and water level fluctuation显示文摘Frequent soil landslide events are recorded in the Three Gorges Reservoir area,China,making it necessary to investigate the failure mode of such riverside landslides.Geotechnical centrifugal test is considered to be the most realistic laboratory model,which can reconstruct the required geo-stress.In this study,the Liangshuijing landslide in the Three Gorgers Reservoir area is selected for a scaled centrifugal model experiment,and a water pump system is employed to retain the rainfall condition.Using the techniques of digital photography and pore water pressure transducers,water level fluctuation is controlled,and multi-physical data are thus obtained,including the pore water pressure,earth pressure,surface displacement and deep displacement.The analysis results indicate that:Three stages were set in the test(waterflooding stage,rainfall stage and drainage stage).Seven transverse cracks with wide of 1–5 mm appeared during the model test,of which 3 cracks at the toe landslide were caused by reservoir water fluctuation,and the cracks at the middle and rear part were caused by rainfall.During rainfall process,the maximum displacement of landslide model reaches 3 cm.And the maximum deformation of the model exceeds 12 cm at the drainage stage.The failure process of the slope model can be divided into four stages:microcracks appearance and propagation stage,thrust-type failure stage,retrogressive failure stage,and holistic failure stage.When the thrust-type zone caused by rainfall was connected or even overlapped with the retrogressive failure zone caused by the drainage,the landslide would start,which displayed a typical composite failure pattern.The failure mode and deformation mechanism under the coupling actions of water level fluctuation and rainfall are revealed in the model test,which could appropriately guide for the analysis and evaluation of riverside landslides. | Fasheng Miao Yiping Wu Ákos Török Linwei Li Yang Xue | 2022 | Geoscience Frontiers2022，13，3： | 3 |
