1The EGFR-P38 MAPK axis up-regulates PD-L1 through miR-675-5p and down-regulates HLA-ABC via hexokinase-2 in hepatocellular carcinoma cells显示文摘Background:Immunotherapy has been shown to be a promising strategy against human cancers.A better understanding of the immune regulation in hepatocellular carcinoma(HCC)could help the development of immunotherapy against HCC.The epidermal growth factor receptor(EGFR)signaling is frequently activated in HCC and plays important roles in tumorigenesis.However,its role in HCC immunity is still largely unknown.This study aimed to investigate the impact of EGFR signaling on programmed death-ligand 1(PD-L1)and human leukocyte antigen class-I(HLA-I)expression in HCC cells and its underlying mechanisms.Methods:The expression of phosphorylated EGFR(p-EGFR),PD-L1,and HLAI(HLA-ABC)in HCC specimens was detected by immunohistochemistry,and their correlations were analyzed.PD-L1 and HLA-ABC expression in EGFRactivated HCC cells were detected by quantitative real-time PCR,Western blotting,and flow cytometry,and T cell-mediated lysis was performed to test the immunosuppressive effects of PD-L1 and HLA-ABC alterations in HCC cells.Furthermore,the underlying mechanisms of EGFR activation-induced PD-L1 up-regulation and HLA-ABC down-regulation were explored by animal experiments,luciferase reporter assay,and gene gain-and loss-of-function studies.Results:p-EGFR was positively correlated with PD-L1 and negatively correlated with HLA-ABC expression in HCCs.EGFR activation by its ligand EGF up-regulated PD-L1 and down-regulated HLA-ABC in HCC cells,which was functionally important and could be abolished by the EGFR inhibitor,gefitinib,both in vitro and in vivo.Mechanistically,enhanced P38 mitogenactivated protein kinase(MAPK)activation down-regulated microRNA-675-5p(miR-675-5p)and up-regulated glycolysis-related enzyme hexokinase 2(HK2);miR-675-5p down-regulation enhanced the stability of PD-L1 mRNA probably via the 3’-untranslated region(3’-UTR)of PD-L1 and thereby caused PD-L1 accumulation,and HK2 up-regulation enhanced aerobic glycolysis and mediated a decrease in HLA-ABC.Conclusions:The EGFR-P38 MAPK axis could up-regulate PD-L1 through miR-675-5p and down-regulate HLA-ABC via HK2 in HCC cells.Our study reveals a novel signaling network that may cause immune suppression in HCC and suggests that EGFR signaling can be targeted for HCC immunotherapy.Zongcai Liu Fen Ning Yanna Cai Huiying Sheng Ruidan Zheng Xi Yin Zhikun Lu Ling Su Xiaodan Chen Chunhua Zeng Haifang Wang Li Liu 2021Cancer Communications2021,41,1:5

4Farmland mercury contamination in the vicinity of an organic chemical factory in Guizhou,China显示文摘This study assesses the level of contamination of Hg in farmland soils along the irrigating channel down-stream from Guizhou Organic Chemical Factory (GOCF),where metallic mercury is used as a catalyst to produce acetic acid. The total input of mercury into the environment,as announced by GOCF,is 140 t in the past 30 years (1971-2000). Sampling sites were chosen based on the distance from the source of pollution—the chemical factory. A total of 39 samples were collected from the study area and analyzed for total mercury concentrations and methyl mercury concentrations. The characteristics of vertical and horizontal distributions of total mercury and methyl mer-cury in the study area (farmland) are described in this paper. Much attention was paid to the transformation of inor-ganic Hg into organic mercury species in soils as well. The results showed that the farmland has been heavily con-taminated by Hg. Land cultivation activity,land utilization style and distance from the pollution source could be the dominant factors controlling the distribution of THg and MeHg. 6DI-3-n-butylphthalide exerts neuroprotective effects by modulating hypoxia-inducible factor 1-alpha ubiquitination to attenuate oxidative stress-induced apoptosis显示文摘DI-3-n-butylphthalide is used to treat mild and moderate acute ischemic stroke.However,the precise underlying mechanism requires further investigation.In this study,we investigated the molecular mechanism of DI-3-n-butylphthalide action by various means.We used hydrogen peroxide to induce injury to PC12cells and RAW264.7 cells to mimic neuronal oxidative stress injury in stroke in vitro and examined the effects of DI-3-n-butylphthalide.We found that DI-3-nbutylphthalide pretreatment markedly inhibited the reduction in viability and reactive oxygen species production in PC12 cells caused by hydrogen peroxide and inhibited cell apoptosis.Furthermore,DI-3-n-butylphthalide pretreatment inhibited the expression of the pro-apoptotic genes Bax and Bnip3.DI-3-nbutylphthalide also promoted ubiquitination and degradation of hypoxia inducible factor 1α,the key transcription factor that regulates Bax and Bnip3 genes.These findings suggest that DI-3-n-butylphthalide exhibits a neuroprotective effect on stroke by promoting hypoxia inducible factor-1α ubiquitination and degradation and inhibiting cell apoptosis.Shuai Li Jingyuan Zhao Yan Xi Jiaqi Ren Yanna Zhu Yan Lu Deshi Dong 2023Neural Regeneration Research2023,18,11:3
7Resilient outage recovery of a distribution system: co-optimizing mobile power sources with network structure显示文摘Outage recovery is important for reducing the economic cost and improving the reliability of a distribution system(DS)in extreme weather and with equipment faults.Previous studies have separately considered network reconfigu-ration(NR)and dispatching mobile power sources(MPS)to restore the outage load.However,NR cannot deal with the scenario of an electrical island,while dispatching MPS results in a long power outage.In this paper,a resilient outage recovery method based on co-optimizing MPS and NR is proposed,where the DS and traffic network(TN)are considered simultaneously.In the DS,the switch action cost and power losses are minimized,and the access points of MPSs are changed by carrying out the NR process.In the TN,an MPS dispatching model with the objective of mini-mizing power outage time,routing and power generation cost is developed to optimize the MPSs’schedule.A solu-tion algorithm based on iteration and relaxation methods is proposed to simplify the solving process and obtain the optimal recovery strategy.Finally,numerical case studies on the IEEE 33 and 119-bus systems validate the proposed resilient outage recovery method.It is shown that the access point of MPS can be changed by NR to decrease the power outage time and dispatching cost of MPS.The results also show that the system operation cost can be reduced by considering power losses in the objective function.Chenchen Li Yanna Xi Yufan Lu Nian Liu Liudong Chen Li Ju Yibin Tao 2022Protection and Control of Modern Power Systems2022,7,1:2

9Transcriptional regulation of fleshy fruit texture显示文摘Fleshy fruit texture is a critically important quality characteristic of ripe fruit.Softening is an irreversible process which operates in most fleshy fruits during ripening which,together with changes in color and taste,contributes to improvements in mouthfeel and general attractiveness.Softening results mainly from the expression of genes encoding enzymes responsible for cell wall modifications but starch degradation and high levels of flavonoids can also contribute to texture change.Some fleshy fruit undergo lignification during development and post-harvest,which negatively affects eating quality.Excessive softening can also lead to physical damage and infection,particularly during transport and storage which causes severe supply chain losses.Many transcription factors(TFs)that regulate fruit texture by controlling the expression of genes involved in cell wall and starch metabolism have been characterized.Some TFs directly regulate cell wall targets,while others act as part of a broader regulatory program governing several aspects of the ripening process.In this review,we focus on advances in our understanding of the transcriptional regulatory mechanisms governing fruit textural change during fruit development,ripening and postharvest.Potential targets for breeding and future research directions for the control of texture and quality improvement are discussed.Yanna Shi Bai-Jun Li Guanqing Su Mengxue Zhang Donald Grierson Kun-Song Chen 2022Journal of Integrative Plant Biology2022,64,9:2
10Environmental factors affecting degradation of perfluorooctanoic acid(PFOA) by In2O3 nanoparticles显示文摘Nanophotocatalysts have shown great potential for degrading poly-and perfluorinated substances(PFAS).In light of the fact that most of these catalysts were studied in pure water,this study was designed to elucidate effects from common environmental factors on decomposing and defluorinating perfluorooctanoic acid(PFOA)by In2O3 nanoparticles.Results from this work demonstrated that among the seven parameters,pH,sulfate,chloride,H2O2,In2O3 dose,NOM and O2,the first four had statistically significant negative effects on PFOA degradation.Since PFOA is a strong acid,the best condition leading to the highest PFOA removal was identified for two pH ranges.When pH was between 4 and 8,the optimal condition was:pH=4.2;sulfate=5.00 mg/L;chloride=20.43 mg/L;H2O2=0 mmol/L.Under this condition,PFOA decomposition and defluorination were 55.22 and 23.56%,respectively.When pH was between 2 and 6,the optimal condition was:pH=2;sulfate=5.00 mg/L;chloride=27.31 mg/L;H2O2=0 mmol/L.With this condition,the modeled PFOA decomposition was 97.59%with a defluorination of approximately 100%.These predicted results were all confirmed by experimental data.Thus,In2O3 nanoparticles can be used for degrading PFOA in aqueous solutions.This approach works best when the target contaminated water contains low concentrations of NOM,sulfate and chloride and at a low pH.Weilan Zhang Harry Efstathiadis Lingyun Li Yanna Liang 2020Journal of Environmental Sciences2020,32,7:2

14Geochronology,Petrogenesis and Tectonic Setting of the Late Jurassic I-type Granites in the North Qinling Orogenic Belt,Central China显示文摘The North Qinling Orogenic Belt(NQOB)is a composite orogenic belt in central China.It started evolving during the Meso-Neoproterozoic period and underwent multiple stages of plate subduction and collision before entering intra-continental orogeny in the Late Triassic.The Meso-Cenozoic intra-continental orogeny and tectonic evolution had different responses in various terranes of the belt,with the tectonic evolution of the middle part of the belt being particularly controversial.The granites distributed in the Dayu and Kuyu areas in the middle part of the NQOB can provide an important window for revealing the geodynamic mechanisms of the NQOB.The main lithology of Dayu and Kuyu granites is biotite monzogranite,and the zircon U-Pb dating yielded intrusive ages of 151.3±3.4 Ma and 147.7±1.5 Ma,respectively.The dates suggest that the biotite monzogranite were formed at the end of the Late Jurassic.The whole-rock geochemistry analysis shows that the granites in the study areas are characterized by slightly high SiO_(2)(64.50-68.88 wt%)and high Al_(2)O_(3)(15.12-16.24 wt%)and Na_(2)O(3.55-3.80 wt%)contents.They are also enriched in light rare earth elements,large ion lithophile elements(e.g.,Ba,K,La,Pb and Sr),and depleted in high field strength elements(HFSEs)(e.g.,Ta,Nb,P and Ti).Additionally,the granites have weakly negative-slightly positive Eu anomalies(δEu=0.91-1.19).Zircon Lu-Hf isotopic analysis showedε_(Hf)(t)=-6.1--3.8,and the two-stage model age is T_(2DM(crust))=1.5-1.6 Ga.The granites in the study areas are analyzed as weak peraluminous high-K calc-alkaline I-type granites.They formed by partial melting of the thickened ancient lower crust,accompanied by the addition of minor mantle-derived materials.During magma ascent,they experienced fractional crystallization,with residual garnet and amphibole for a certain proportion in the magma source region.Comprehensive the geotectonic data suggest that the end of the Late Jurassic granite magmatism in the Dayu and Kuyu areas represents a compression-extension transition regime.It may have been a response to multiple tectonic mechanisms,such as the late Mesozoic intra-continental southward subduction of the North China Craton and the remote effect of the Paleo-Pacific Plate subduction.HUANG Yanna LI Dunpeng XIAO Aifang XU Songming 2023Acta Geologica Sinica(English Edition)2023,97,4:1

16Mutations in the miRNA165/166 binding site of the HB2 gene result in pleiotropic effects on morphological traits in wheat显示文摘Leaf,spike,stem,and root morphologies are key factors that determine crop growth,development,and productivity.Multiple genes that control these morphological traits have been identified in Arabidopsis,rice,maize,and other plant species.However,little is known about the genomic regions and genes associated with morphological traits in wheat.Here,we identified the ethyl methanesulfonate-derived mutant wheat line M133 that displays multiple morphological changes that include upward-curled leaves,paired spikelets,dwarfism,and delayed heading.Using bulked segregant RNA sequencing(BSR-seq)and a high-resolution genetic map,we identified TraesCS1D02G155200(HBD2)as a potential candidate gene.HB-D2 encodes a class III homeodomain-leucine zipper(HD-ZIP III)transcription factor,and the mutation was located in the miRNA165/166 complementary site,resulting in a resistant allele designated rHb-D2.The relative expression of rHb2 in the mutant plants was significantly higher(P<0.01)than in plants homozygous for the WT allele.Independent resistant mutations that disrupt the miRNA165/166 complementary sites in the A-(rHb-A2)and B-genome(rHb-B2)homoeologs showed similar phenotypic alterations,but the relative intensity of the effects was different.Transgenic plants expressing rHb-D2 gene driven by the maize UBIQUITIN(UBI)promoter showed similar phenotypes to the rHb-D2 mutant.These results confirmed that HB-D2 is the causal gene responsible for the mutant phenotypes.Finally,a survey of 1397 wheat accessions showed that the complementary sites for miRNA165/166 in all three HB2 homoeologs are highly conserved.Our results suggest that HB2 plays an important role in regulating growth and development in wheat.Dengji Jiang Lei Hua Chaozhong Zhang Hongna Li Zheng Wang Jian Li Guiping Wang Rui Song Tao Shen Hongyu Li Shengsheng Bai Yanna Liu Jian Wang Hao Li Jorge Dubcovsky Shisheng Chen 2023The Crop Journal2023,11,1:1
