| 1 | A Simplified PCR-SSP Method for HLA-A2 Subtype in a Population of Wuhan,China显示文摘HLA-A2 is the most frequent HLA-A allele in all ethnic populations, and an important restriction element for peptide presentation to T cells in infectious disease and cancer. However, the HLA-A2 supertype consisting of up to 75 subtypes, mutation studies and analyses using cytotoxic T lymphocytes suggest the functional relevance of subtype-specific differences in HLA-A2 molecules for peptide binding and T-cell recognition. Therefore, it is necessary for T-cell response study to discriminate the HLA-A2 subtypes and to understand the profile of HLA-A2 allelic distribution in a given population. In this study, we developed a simple, robust approach based on the nested polymerase chain reaction using sequence-specific primers (PCR-SSP) to discriminate 17 HLA-A2 subtypes which cover the most HLA-A2 alleles (> 99% allele frequency) reported in Chinese, using 15 combinations of 19 allelic specific primers. In the first round of PCR, 3 combinations of 5 primers were used to determine whether the tested sample was HLA-A2 positive, meanwhile the subtypes of HLA-A*0209 and HLA-A*0215N were determined for the variant position of these two subtypes is in exon 4 instead of exon 2, 3. Samples of HLA-A2 positive were subtyped in the second round of PCR, using PCR products of the first round as templates. This strategy was applied to test the samples of 78 random HLA-A2 positive individuals for their HLA-A2 subtypes. Those samples were screened for HLA-A2 positive by the first round PCR-SSP from 154 healthy blood donors in Wuhan, China. The subtyping results were verified by using flow cytometric analysis (FCM) with HLA-A2 specific monoclonal antibody BB7.2 and DNA sequencing. The typing results of the samples show 50.7% random individuals in the population carry HLA-A2, HLA-A*0201 ranks the first (allele frequency = 15.5%), followed by A*0207 (5.8%), A*0206 (4.7%), A*0203 (2.6%), A*0210 (0.7%), and these 5 alleles account for 99.0% HLA-A2 subtypes of allele frequency. | 5 | A Novel Model of Predicting Archie's Cementation Factor from Nuclear Magnetic Resonance(NMR) Logs in Low Permeability Reservoirs显示文摘The resistivity experimental measurements of core samples drilled from low permeability reservoirs of Ordos Basin,Northwest China,illustrate that the cementation factors are not agminate,but vary from 1.335 to 1.749.This leads to a challenge for the estimation of water and hydrocarbon saturation.Based on the analysis of Purcell equation and assumption that rock resistivity is determined by the parallel connection of numerous capillary resistances,a theoretical expression of cementation factor in terms of porosity and permeability is established.Then,cementation factor can be calculated if the parameters of porosity and permeability are determined.In the field application,porosity can be easily obtained by conventional logs.However,it is a tough challenge to estimate permeability due to the strong heterogeneity of low permeability reservoirs.Thus,the Schlumberger Doll Research(SDR)model derived from NMR logs has been proposed to estimate permeability.Based on the analysis of the theoretical expressions of cementation factor and SDR model,a novel cementation factor prediction model,which is relevant to porosity and logarithmic mean of NMR T2spectrum(T2lm),is derived.The advantage of this model is that all the input information can be acquired from NMR logs accurately.In order to confirm the credibility of the novel model,the resistivity and corresponding laboratory NMR measurements of 27 core samples are conducted.The credibility of the model is confirmed by comparing the predicted cementation factors with the core analyzed results.The absolute errors for all core samples are lower than 0.071.Once this model is extended to field application,the accuracy of water and hydrocarbon saturation estimation will be significantly improved. | Liang Wang Zhiqiang Mao Yujiang Shi Qin'e Tao Yumei Cheng Yong Song | 2014 | Journal of Earth Science2014,25,1: | 4 |
| 6 | Differently sized cuckoos pose different threats to hosts显示文摘Hole-nesting tits Parus spp.have been classified as'unsuitable'hosts for cuckoo parasitism because cuckoos cannot enter a cavity if the entrance is too small.However,Chinese tits could re-ject alien eggs and egg ejection rate increased with the local diversity of parasitic cuckoo species.Antiparasitic behavior among Chinese tits may have evolved due to greater size variation among sympatric cuckoo species.This raises the question of whether differently sized parasitic cuckoos pose different threats to Chinese tits.A green-backed tit Parus monticolus population that is sym-patric with Asian emerald cuckoo Chrysococcyx maculatus(eme-cuckoo,small-sized parasite)and common cuckoo Cuculus canorus(com-cuckoo,large-sized parasite),and a cinereous tit P.cinereus population that is only sympatric with com-cuckoo were chosen as study organisms.We observed behavioral response and recorded alarm calls of the 2 tit species to eme-cuckoo,com-cuckoo,chipmunk Tamias sibiricus(a nest predator)and dove Streptopelia orientalis(a harm-less control),and subsequently played back alarm calls to conspecific incubating females.In dummy experiments,both tit species performed intense response behavior to chipmunk,but rarely responded strongly to the 3 avian species.In playback experiments,both tit species responded strongly to conspecific chipmunk alarm calls,but rarely responded to dove alarm calls.The inten-sity of response of incubating female green-backed tits to eme-cuckoo and com-cuckoo alarm calls were similar to that of chipmunk alarm calls,while the intensity to eme-cuckoo alarm calls was higher than the intensity to dove alarm calls which was similar to that of com-cuckoo alarm calls.In contrast,few female cinereous tits responded to eme-cuckoo and com-cuckoo alarm calls.These findings indicated that the threat level of eme-cuckoo was slightly greater than that of com-cuckoo for sympatric green-backed tits,but not for allopatric cinereous tits. | Jiangping YU Mingju E Wei SUN Wei LIANG Haitao WANG Anders Pape MΦLLER | 2020 | Current Zoology2020,66,3: | 4 |
| 7 | Development and calibration of the Moon-based EUV camera for Chang'e-3显示文摘The process of development and calibration for the first Moon-based extreme ultraviolet(EUV) camera to observe Earth's plasmasphere is introduced and the design, test and calibration results are presented. The EUV camera is composed of a multilayer film mirror, a thin film filter, a photon-counting imaging detector, a mechanism that can adjust the direction in two dimensions, a protective cover, an electronic unit and a thermal control unit. The center wavelength of the EUV camera is 30.2 nm with a bandwidth of 4.6 nm. The field of view is 14.7° with an angular resolution of 0.08°, and the sensitivity of the camera is 0.11 count s-1Rayleigh-1. | 8 | Outcomes of Medicare beneficiaries hospitalised with transient ischaemic attack and stratification using the ABCD^(2) score显示文摘Background Long-term outcomes for Medicare beneficiaries hospitalised with transient ischaemic attack(TIA)and role of ABCD^(2) score in identifying high-risk individuals are not studied.Methods We identified 40825 Medicare beneficiaries hospitalised from 2011 to 2014 for a TIA to a Get With The Guidelines(GWTG)-Stroke hospital and classified them using ABCD^(2)s of mortality and rehospitalisation(all-cause,ischaemic stroke,haemorrhagic stroke,myocardial infarction,and gastrointestinal and intracranial haemorrhage)for high-risk versus low-risk groups adjusted for patient and hospital characteristics.Results Of the 40825 patients,35118(86%)were high risk(ABCD^(2)≥4)and 5707(14%)were low risk(ABCD^(2)=0-3).Overall rate of mortality during 1-year follow-up after hospital discharge for the index TIA was 11.7%,44.3% were rehospitalised for any reason and 3.6%were readmitted due to stroke.Patients with ABCD^(2) score≥4 had higher mortality at 1 year than not(adjusted HR 1.18,95%CI 1.07 to 1.30).Adjusted risks for ischaemic stroke,all-cause readmission and mortality/all-cause readmission at 1 year were also significantly higher for patients with ABCD^(2) score≥4 vs 0-3.In contrast,haemorrhagic stroke,myocardial infarction,gastrointestinal bleeding and intracranial haemorrhage risk were not significantly different by ABCD^(2) score.Conclusions This study validates the use of ABCD^(2) score for long-term risk assessment after TIA in patients aged 65 years and older.Attentive efforts for community-based follow-up care after TIA are needed for ongoing prevention in Medicare beneficiaries who were hospitalised for TIA. | Shreyansh Shah Li Liang Durgesh Bhandary Saga Johansson Eric E Smith Deepak L Bhatt Gregg C Fonarow Naeem D Khan Eric Peterson Janet Prvu Bettger | 2021 | Stroke & Vascular Neurology2021,6,2: | 3 |

| 15 | Effects of Slow-release Fertilizers and Sugarcane-specific Fertilizers on Yield and Quality of Sugarcane显示文摘The effects of slow-release fertilizers and sugarcane-specific fertilizers on the growth and quality of sugarcane were studied to provide a theoretical basis for cultivation of sugarcane with lower costs but higher efficiency. Field experiments were carried out in two major sugarcane areas in Guangxi and three fertilization treatments were studied: single application of compound fertilizers( treatment I),compound fertilizers + slow-release fertilizers( treatment II) and sugarcane-specific base fertilizers + sugarcane-specific topdressing( treatment III). Effects of equal fertilization conditions of treatment I,II and III on growth,yield and sugar of sugarcane were studied. The three fertilization treatments had little effects on emergence,tillering,and effective stems of sugarcane,but compared with the treatment of compound fertilizer( treatment I) with a ratio of N,P,and K of 1∶1∶1,treatment II using slow-release fertilizers as topdressing had better growth,higher plant height and stem diameter,so the yield was higher. Treatment III designed sugarcane-specific fertilizers with proper ratio according to fertilizer demands of sugarcane. Besides,the treatment III sugarcane-specific base fertilizers,containing certain amount of organic matters,could promote the sugar accumulation of sugarcane. Therefore,the sugarcane yield of treatment III was higher than that of treatment II. In conclusion,slow-release fertilizers and sugarcane-specific fertilizers can significantly increase sugarcane yield,especially sugarcane-specific fertilizers.
