5Comprehensive Marine Observing Experiment Based on High-Altitude Large Unmanned Aerial Vehicle(South China Sea Experiment2020 of the “Petrel Project”)显示文摘In collaboration with 12 other institutions, the Meteorological Observation Center of the China Meteorological Administration undertook a comprehensive marine observation experiment in the South China Sea using the Yilong-10 high-altitude large unmanned aerial vehicle(UAV). The Yilong-10 UAV carried a self-developed dropsonde system and a millimeter-wave cloud radar system. In addition, a solar-powered unmanned surface vessel and two drifting buoys were used. The experiment was further supported by an intelligent, reciprocating horizontal drifting radiosonde system that was deployed from the Sansha Meteorological Observing Station, with the intent of producing a stereoscopic observation over the South China Sea. Comprehensive three-dimensional observations were collected using the system from 31 July to2 August, 2020. This information was used to investigate the formation and development processes of Typhoon Sinlaku(2020). The data contain measurements of 21 oceanic and meteorological parameters acquired by the five devices, along with video footage from the UAV. The data proved very helpful in determining the actual location and intensity of Typhoon Sinlaku(2020). The experiment demonstrates the feasibility of using a high-altitude, large UAV to fill in the gaps between operational meteorological observations of marine areas and typhoons near China, and marks a milestone for the use of such data for analyzing the structure and impact of a typhoon in the South China Sea. 6Climatology of Cloud-base Height from Long-term Radiosonde Measurements in China显示文摘Clouds are critical to the global radiation budget and hydrological cycle, but knowledge is still poor concerning the observed climatology of cloud-base height(CBH) in China. Based on fine-resolution sounding observations from the China Radiosonde Network(CRN), the method used to estimate CBH was modified, and uncertainty analyses indicated that the CBH is good enough. The accuracy of CBH estimation is verified by the comparison between the sounding-derived CBHs and those estimated from the micro-pulse lidar and millimeter-wave cloud radar. As such, the CBH climatology was compiled for the period 2006–16. Overall, the CBH exhibits large geographic variability across China, at both 0800 Local Standard Time(LST) and 2000 LST, irrespective of season. In addition, the summertime cloud base tends to be elevated to higher altitudes in dry regions [i.e., Inner Mongolia and the North China Plain(NCP)]. By comparison, the Tibetan Plateau(TP),Pearl River Delta(PRD) and Sichuan Basin(SCB) have relatively low CBHs(< 2.4 km above ground level). In terms of seasonality, the CBH reaches its maximum in summer and minimum in winter. A low cloud base tends to occur frequently(> 70%) over the TP, PRD and SCB. In contrast, at most sites over the Yangtze River Delta(YRD) and the NCP, about half the cloud belongs to the high-cloud category. The CBH does not exhibit marked diurnal variation in summer, throughout all CRN sites, probably due to the persistent cloud coverage caused by the East Asia Summer Monsson. 7Production of transgenic pigs over-expressing theantiviral gene Mx1显示文摘The myxovirus resistance gene (Mx1) has a broad spectrum of antiviral activities. It is therefore an interestingcandidate gene to improve disease resistance in farm animals. In this study, we report the use of somatic cellnuclear transfer (SCNT) to produce transgenic pigs over-expressing the Mx1 gene. These transgenic pigs expressapproximately 15–25 times more Mx1 mRNA than non-transgenic pigs, and the protein level of Mx1 was alsomarkedly enhanced. We challenged fibroblast cells isolated from the ear skin of transgenic and control pigs withinfluenza A virus and classical swine fever virus (CFSV). Indirect immunofluorescence assay (IFA) revealed a profounddecrease of influenza A proliferation in Mx1 transgenic cells. Growth kinetics showed an approximately 10-foldreduction of viral copies in the transgenic cells compared to non-transgenic controls. Additionally, we found thatthe Mx1 transgenic cells were more resistant to CSFV infection in comparison to non-transgenic cells. 8Efficient and high-fidelity base editor with expanded PAM compatibility for cytidine dinucleotide显示文摘Cytidine base editor(CBE),which is composed of a cytidine deaminase fused to Cas9 nickase,has been widely used to induce C-to-T conversions in a wide range of organisms.However,the targeting scope of current CBEs is largely restricted to protospacer adjacent motif(PAM)sequences containing G,T,or A bases.In this study,we developed a new base editor termed“nNme2-CBE”with excellent PAM compatibility for cytidine dinucleotide,significantly expanding the genome-targeting scope of CBEs.Using nNme2-CBE,targeted editing efficiencies of 29.0%-55.0%and 17.3%—52.5%were generated in human cells and rabbit embryos,respectively.In contrast to conventional nSp-CBE,the nNme2-CBE is a natural high-fidelity base editing platform with minimal DNA off-targeting detected in vivo.Significantly increased efficiency in GC context and precision were determined by combining nNme2Cas9 with rationally engineered cytidine deaminases.In addition,the Founder rabbits with accurate single-base substitutions at Fgf5 gene loci were successfully generated by using the nNme2-CBE system.These novel nNme2-CBEs with expanded PAM compatibility and high fidelity will expand the base editing toolset for efficient gene modification and therapeutic applications.Zhiquan Liu Siyu Chen Yingqi Jia Huanhuan Shan Mao Chen Yuning Song Liangxue Lai Zhanjun Li 2021Science China(Life Sciences)2021,64,8:2
9The effect of iron on the sound velocities ofδ-AlOOH up to 135 GPa显示文摘δ-(Al,Fe)OOH is considered to be one of the most important hydrous phases on Earth,remaining stable under the extreme conditions throughout the mantle.The behavior ofδ-(Al,Fe)OOH at high pressure is essential to understanding the deep water cycle.δ-(Al_(0.956)Fe_(0.044))OOH crystals synthesized at 21 GPa and 1473 K were investigated by high-pressure Brillouin light scattering spectroscopy and synchrotron X-ray diffraction up to 135.4 GPa in diamond anvil cells.The incorporation of 5 mol%FeOOH increases the unit-cell volume ofδ-AlOOH by~1%and decreases the shear-wave velocity(VS)by~5%at 20–135 GPa.In particular,the compressional(V_(P))and shear(VS)wave velocities ofδ-(Al_(0.956)Fe_(0.044))OOH are 7%–16%and 10%–24%greater than all the major minerals in the mantle transition zone including wadsleyite,ringwoodite,and majorite.The distinctly high sound velocities ofδ-(Al_(0.956)Fe_(0.044))OOH at 20–25 GPa may contribute to the seismic anomalies observed at~560–680 km depths in the cold and stagnant slab beneath Izu-Bonin and/or Korea.Furthermore,the VS ofδ-(Al_(0.956)Fe_(0.044))OOH is about 10%and 4%–12%lower than iron-bearing bridgmanite Mg_(0.96)Fe_(0.05)Si_(0.99O3)and ferropericlase(Mg_(0.92)Fe_(0.08))O,respectively,under the lowermost mantle conditions,which might partially contribute to the large low-shear-velocity provinces and ultralow velocity zones at the bottom of the lower mantle.Xiaowan Su Chaoshuai Zhao Chaojia Lv Yukai Zhuang Nilesh Salke Liangxu Xu Hu Tang Huiyang Gou Xiaohui Yu Qiang Sun Jin Liu 2021Geoscience Frontiers2021,12,2:1

11Reprogramming mature terminally differentiated adipocytes to induced pluripotent stem cells显示文摘Mature adipocytes are terminally differentiated somatic cells. Here, we report the successful generation of induced pluripotent stem(i PS) cells from mouse mature adipocytes by forced expression of six transcription factors(Oct4, Sox2, c-Myc, Klf4, Rarc, and Lrh1) with a piggy Bac transposon-based strategy. The resulting i PS cells were pluripotent as evidenced by the fact that they stained positive for alkaline phosphatase, expressed high levels of key pluripotency markers including Oct4, Nanog, and SSEA1,and remained pluripotent on a 2i media. In vitro differentiation of the i PS cells showed that the cell derivatives of all three germ layers could be readily obtained through formation of embryoid bodies. Most importantly, these adipocytederived i PS cells were capable of producing chimera with high frequencies when reintroduced into early-stage embryos and transmitted through the germ line. 20Three-dimensional-printed collagen/chitosan/secretome derived from HUCMSCs scaffolds for efficient neural network reconstruction in canines with traumatic brain injury显示文摘The secretome secreted by stem cells and bioactive material has emerged as a promising therapeutic choice for traumatic brain injury(TBI).We aimed to determine the effect of 3D-printed collagen/chitosan/secretome derived from human umbilical cord blood mesenchymal stem cells scaffolds(3D-CC-ST)on the injured tissue regeneration process.3D-CC-ST was performed using 3D printing technology at a low temperature(20C),and the physical properties and degeneration rate were measured.The utilization of low temperature contributed to a higher cytocompatibility of fabricating porous 3D architectures that provide a homogeneous distribution of cells.Immediately after the establishment of the canine TBI model,3D-CC-ST and 3D-CC(3D-printed collagen/chitosan scaffolds)were implanted into the cavity of TBI.Following implantation of scaffolds,neurological examination and motor evoked potential detection were performed to analyze locomotor function recovery.Histological and immunofluorescence staining were performed to evaluate neuro-regeneration.The group treated with 3D-CC-ST had good performance of behavior functions.Implanting 3D-CC-ST significantly reduced the cavity area,facilitated the regeneration of nerve fibers and vessel reconstruction,and promoted endogenous neuronal differentiation and synapse formation after TBI.The implantation of 3D-CC-ST also markedly reduced cell apoptosis and regulated the level of systemic inflammatory factors after TBI.Xiaoyin Liu Guijun Zhang Pan Wei Lin Zhong Yaxing Chen Jianyong Zhang Xuyi Chen Liangxue Zhou 2022Regenerative Biomaterials2022,9,1:0
