| 1 | Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. | 2 | RDX crystals with high sphericity prepared by resonance acoustic mixing assisted solvent etching technology显示文摘In order to obtain high-quality spherical RDX crystal particles,the RDX crystals were suspended in a mixed solvent of cyclohexanone and cyclohexane,subsequently a solvent etching study was carried out under the action of vibration/acoustic flow coupled flow field,which generated by resonance acoustic mixing.The effects of solvent ratio,temperature,acceleration and experiment time on morphology as well as particle size of RDX crystals were studied.Not only were the morphology,particle size distribution and crystal form of RDX crystals determined,but also the thermal decomposition performance and mechanical sensitivity of spherical RDX were examined and discussed.Results indicated that under the process of solvent/non-solvent volume ratio at 1:2,temperature of 40℃,acceleration of 40 g and experiment time of 4 h,α-type RDX crystal with sphericity of 0.92 can be obtained.Furthermore,the median particle size(D_(50))of spherical RDX crystals is 215.8 μm with a unimodal particle size distribution(size span 1.34).For one thing,the thermal decomposition peak temperature of spherical RDX is about 2.5℃ higher than that of raw RDX,and apparent activation energy reaches 444.68 kJ/mol.For another thing,impact sensitivity and friction sensitivity of spherical RDX are 18.18% and 33.33% lower than that of raw RDX,respectively.It demonstrates that safety of spherical RDX under thermal,impact and friction stimuli has been improved. | Dongjie Liao Qian Liu Chunyan Li Ning Liu Mingchang Wang Chongwei An | 2023 | Defence Technology(防务技术)2023,28,10: | 1 |

| 6 | Azobenzene quaternary ammonium salt for photo-controlled and reusable disinfection without drug resistance显示文摘The abuse of antibiotics causes severe bacterial resistance, and the shortage of antibiotics has created a global public health crisis. This situation has prompted people to develop new antibacterial agents independent of traditional antibiotics. Here, we created a series of photosensitive azobenzene-quaternary ammonium salt smart antibacterial agents by connecting azobenzene with amines with different chain lengths to improve the antibacterial selectivity of quaternary ammonium salt(QAS) and prevent the accumulation of active QAS in the environment. After trans-cis isomerization, the solubility of the title compound(compound 4) increased and the antibacterial property enhanced. The experimental results suggested that the antibacterial effect of compound 4 was significantly enhanced after 365 nm light irradiation, and it had photosensitive intelligent antibacterial activity and could be reused. Notably, we did not obtain any mutants of Staphylococcus aureus or Escherichia coli resistant to compound 4. In general,compound 4 has the advantages of high yield, photo-controllable antibacterial properties, reusability, and does not induce bacterial resistance. | 8 | Cross-linked lipoic acid nanocapsules serve as H_(2)O_(2) amplifier to strengthen the H_(2)O_(2)-sensitive prodrug activation显示文摘The H_(2)O_(2) -triggered prodrug activation represents a hot topic while the low H_(2)O_(2) level of cancer cells hindered the complete activation of parent drugs and resulted in the poor therapeutic effect. Here, we developed an H_(2)O_(2) amplifier fabricated by crosslinked lipoic acid nanocapsules(cLANCs), which would selectively elevate the intracellular H_(2)O_(2) level of cancer cells 3.4-fold higher than that of untreated cancer cells so as to speed up the activation of parent drug from the loaded prodrugs(Pro-5-FU). The cytotoxicity showed that the killing effect of Pro-5-FU loaded in cLANCs(Pro-5-FU@cLANCs) arrived up to 1.6-time higher than that of Pro-5-FU alone against A549 cells, and was even close to free 5-FU. The in vivo anticancer evaluation gave the tumor inhibition rate(TIR) and survival rate(SR) of Pro-5-FU@cLANCs against A549 tumor-bearing nude mice up to 73.1% and 80%,respectively, much higher than those of Pro-5-FU(TIR: 41.2%, SR: 20%) or free 5-FU(TIR: 50.6%, SR: 0%). | 9 | Characterization of Nucleation Behavior in Temperature-Induced BCC-to-HCP Phase Transformation for High Entropy Alloy显示文摘Phase transformation is one of the essential topics in the studies on high entropy alloys(HEAs).However,characterization of the nucleation behavior in the phase transformation for HEAs is still challenging through experimental methods.In the present work,HfNbTaTiZr HEA was chosen as the representative material,and molecular dynamics/Monte Carlo(MD/MC)simulations were performed to investigate the nucleation behavior in temperature-induced BCC-to-HCP transformation for this HEA system.The results indicate that Nb–Ta,Ti–Zr,Hf–Zr and Hf–Ti atom pairs are preferred in the BCC solid solution of HfNbTaTiZr HEA and Hf–Ti–Zr-rich atom cluster with chemical short range order acts as the nucleation site for HCP phase.The nucleation process follows the non-classical two-step nucleation model:BCC-like structure with severe lattice distortion forms first and then HCP structure nucleates from the BCC-like structure.Moreover,at low temperature,the BCC-to-HCP nucleation hardly occurs,and the BCC solid solution is stabilized.The present work provides more atomic details of the nucleation behavior in temperature-induced BCC-to-HCP phase transformation for HEA,and can help in deep understanding of the phase stability for HEAs. | Xiusong Huang Lehua Liu Weibing Liao Jianjun Huang Huibin Sun Chunyan Yu | 2021 | Acta Metallurgica Sinica(English Letters)2021,34,11: | 0 |
