3Targeting lactate dehydrogenase A (LDHA) exerts antileukemic effects on T-cell acute lymphoblastic leukemia显示文摘Background:T-cell acute lymphoblastic leukemia(T-ALL)is an uncommon and aggressive subtype of acute lymphoblastic leukemia(ALL).In the serum of T-ALL patients,the activity of lactate dehydrogenase A(LDHA)is increased.We proposed that targeting LDHA may be a potential strategy to improve T-ALL outcomes.The current study was conducted to investigate the antileukemic effect of LDHA gene-targeting treatment on T-ALL and the underlying molecular mechanism.Methods:Primary T-ALL cell lines Jurkat and DU528 were treated with the LDH inhibitor oxamate.MTT,colony formation,apoptosis,and cell cycle assays were performed to investigate the effects of oxamate on T-ALL cells.Quantitative real-time PCR(qPCR)and Western blotting analyses were applied to determine the related signaling pathways.A mitochondrial reactive oxygen species(ROS)assay was performed to evaluate ROS production after T-ALL cells were treated with oxamate.A T-ALL transgenic zebrafish model with LDHA gene knockdown was established using CRISPR/Cas9 gene-editing technology,and then TUNEL,Western blotting,and T-ALL tumor progression analyses were conducted to investigate the effects of LDHA gene knockdown on T-ALL transgenic zebrafish.Results:Oxamate significantly inhibited proliferation and induced apoptosis of Jurkat and DU528 cells.It also arrested Jurkat and DU528 cells in G0/G1 phase and stimulated ROS production(all P<0.001).Blocking LDHA significantly decreased the gene and protein expression of c-Myc,as well as the levels of phosphorylated serine/threonine kinase(AKT)and glycogen synthase kinase 3 beta(GSK-3β)in the phosphatidylinositol 3′-kinase(PI3K)signaling pathway.LDHA gene knockdown delayed disease progression and down-regulated c-Myc mRNA and protein expression in T-ALL transgenic zebrafish.Conclusion:Targeting LDHA exerted an antileukemic effect on T-ALL,representing a potential strategy for T-ALL treatment.Haizhi Yu Yafei Yin Yifang Yi Zhao Cheng Wenyong Kuang Ruijuan Li Haiying Zhong Yajuan Cui Lingli Yuan Fanjie Gong Zhihua Wang Heng Li Hongling Peng Guangsen Zhang 2020Cancer Communications2020,40,10:2

8Optimization of Fermentation Process for Hyaluronic Acid Production by Streptococcus zooepidemicus显示文摘Hyaluronic acid(HA) is a high molecular weight glycosaminoglycan consisting of alternating D-glucuronic acid and N-acetylglucosamine and plays extremely important roles in many biological processes.In this study,we optimized fermentation process for the production of HA by Streptococcus zooepidemicus ATCC35246,including fermentation broth composition and various fermentation parameters.The experimental results showed that the optimal fermentation broth composition was:glucose 45 g/L,yeast extract 10 g/L,tryptone 12 g/L,KH_2PO_42 g/L,K_2HPO_4·3H_2O 2 g/L,MgSO_4 ·7H_2O2 g/L,and(NH_4)_2SO_40.4g/L.The optimal parameters involved in fermentation was:liquid volume 20%,pH 6.0,rotation speed 180 r/min,fermentation temperature 35 °C,fermentation duration 18 h,CTAB concentration 25 mg/L.Under the optimized conditions,the yield of HA was 0.305 g/L,which was dramatically improved by 43.87%compared to that of 0.212 g/L before optimization.Jing YUAN Zhiping ZHAO Baoqu WANG Lingli WU Li ZHANG 2015Agricultural Biotechnology2015,4,5:1
9Acetylation of ezrin regulates membrane–cytoskeleton interaction underlying CCL18-elicited cell migration显示文摘Ezrin,a membrane–cytoskeleton linker protein,plays an essential role in cell polarity establishment,cell migration,and division.Recent studies show that ezrin phosphorylation regulates breast cancer metastasis by promoting cancer cell survivor and promotes intrahepatic metastasis via cell migration.However,it was less characterized whether there are additional post-translational modifications and/or post-translational crosstalks on ezrin underlying context-dependent breast cancer cell migration and invasion.Here we show that ezrin is acetylated by p300/CBP-associated factor(PCAF)in breast cancer cells in response to CCL18 stimulation.Ezrin physically interacts with PCAF and is a cognate substrate of PCAF.The acetylation site of ezrin was mapped by mass spectrometric analyses,and dynamic acetylation of ezrin is essential for CCL18-induced breast cancer cell migration and invasion.Mechanistically,the acetylation reduced the lipid-binding activity of ezrin to ensure a robust and dynamic cycling between the plasma membrane and cytosol in response to CCL18 stimulation.Biochemical analyses show that ezrin acetylation prevents the phosphorylation of Thr567.Using atomic force microscopic measurements,our study revealed that acetylation of ezrin induced its unfolding into a dominant structure,which prevents ezrin phosphorylation at Thr567.Thus,these results present a previously undefined mechanism by which CCL18-elicited crosstalks between the acetylation and phosphorylation on ezrin control breast cancer cell migration and invasion.This suggests that targeting PCAF signaling could be a potential therapeutic strategy for combating hyperactive ezrin-driven cancer progression.Xiaoyu Song Wanjuan Wang Haowei Wang Xiao Yuan Fengrui Yang Lingli Zhao McKay Mullen Shihao Du Najdat Zohbi Saravanakumar Muthusamy Yalei Cao Jiying Jiang Peng Xia Ping He Mingrui Ding Nerimah Emmett Mingming Ma Quan Wu Hadiyah-Nicole Green Xia Ding Dongmei Wang Fengsong Wang Xing Liu 2020Journal of Molecular Cell Biology2020,12,6:1

12Dorsal root ganglion progenitors differentiate to gamma-aminobutyric acid-and choline acetyltransferase-positive neurons显示文摘This study examined the isolation and differentiation of dorsal root ganglion progenitor cells for therapeutic use in neurodegenerative diseases. Rat embryonic dorsal root ganglia progenitors were isolated and purified using the differential adhesion method combined with cytosine arabinoside treatment. After culture in serum-free medium supplemented with B27, basic fibroblast growth factor and epidermal growth factor, these cells remained viable and survived for more than 18 months in vitro. Most cells differentiated to neurons that were immunoreactive for gamma-aminobutyric acid and choline acetyltransferase as detected by immunohistochemical staining. In addition, nerve growth factor and neurotrophic tyrosine kinase receptor expression were also observed in dorsal root ganglion progenitors and differentiated cells. 16Amino acid variation analysis of surface spike glycoprotein at 614 in SARS-CoV-2 strains显示文摘As severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)continues to disperse globally with worrisome speed,identifying amino acid variations in the virus could help to understand the characteristics of it.Here,we studied 489 SARS-CoV-2 genomes obtained from 32 countries from the Nextstrain database and performed phylogenetic tree analysis by clade,country,and genotype of the surface spike glycoprotein(S protein)at site 614.We found that virus strains from China's Mainland were mostly distributed in Clade B and Clade undefined in the phylogenetic tree,with very few found in Clade A.In contrast,Clades A2(one case)and A2a(112 cases)predominantly contained strains from European regions.Moreover,Clades A2 and A2a differed significantly from those of China's Mainland in age of infected population(P Z 0.0071,mean age 40.24 to 46.66),although such differences did not exist between the US and China's Mainland.Further analysis demonstrated that the variation of the Sprotein at site 614(QHD43416.1:p.614D>G)was a characteristic of stains in Clades A2 and A2a.Importantly,this variation was predicted to have neutral or benign effects on the function of the S protein.In addition,global quality estimates and 3D protein structures tended to be different between the two S proteins.In summary,we identified different genomic epidemiology among SARS-CoV-2 strains in different clades,especially in an amino acid variation of the S protein at 614,revealing potential viral genome divergence in SARS-CoV-2 strains.Canhui Cao Liang Huang Kui Liu Ke Ma Yuan Tian Yu Qin Haiyin Sun Wencheng Ding Lingli Gui Peng Wu 2020Genes & Diseases2020,7,4:0

18Effects of Polygonatum odoratum Polysaccharide on Reproductive Hormones in Male Rats Fed a High-fat Diet显示文摘[Objectives]This study was conducted to investigate the effects of Polygonatum odoratum polysaccharide(POP)on organ relative weights and reproductive hormone levels in male rats fed a high-fat diet.[Methods]Thirty healthy male Sprague-Dawley(SD)rats were randomly divided into two groups according to their body weight:10 in normal control group(Group NC,n=10)and 20 in experimental group(n=20).The rats in experimental group were fed a high-fat diet for eight weeks before they were further randomly divided into two groups:high fat group(Group HF)and high fat+400 mg/(kg·d)POP group(Group HF+POP).In Group HF+POP,the rats were administered with POP for another six weeks,before their blood plasma was collected,and the relative weights of their testis and epididymis were calculated.The plasma levels of testosterone(T),estrogen(E2),follicle-stimulating hormone(FSH),cortisol(C)and luteinizing hormone(LH)were measured by radioimmunoassay,and the plasma levels of sex hormone-binding globulin(SHBG)and insulin-like growth factor-1(IGF-1)were determined by enzyme-linked immunosorbent assay.[Results]Compared with Group HF,POP could effectively inhibit rat obesity caused by high-fat diets,increase the relative weights of their testis and epididymis,plasma levels of LH,E2,FSH,T,SHBG and IGF-1,and reduce the plasma level of E2.[Conclusions]Polygonatum odoratum polysaccharide(POP)is able to effectively regulate the level of reproductive hormones in high-fat diet fed rats,and helps to protect their reproductive function.Yixin YAN Gang ZHANG Lingli BAI Xin TANG Mengjun YUAN Yihong HU Yan WANG Chenzhong JIN 2023Agricultural Biotechnology2023,12,2:0
19A 36 Mb terminal deletion of chromosome 2BL is responsible for a wheat semi-dwarf mutation显示文摘Reduced plant height is one of the most important traits related to lodging resistance and crop yield. The use of reduced height genes has been one of the main features in breeding modern high-yielding wheat varieties with less lodging. A spontaneous dwarf mutant DD399 was identified in a high yielding, gibberellic acid(GA)-insensitive, lodging-resistant variety Nongda 399(ND399). Significant differences in upper internode lengths between mutant DD399 and wild type ND399 were caused by reduced cell elongation. The plant height of ND399 × DD399 F_(1) hybrids was intermediate between the parents, indicating incomplete dominance or a dose–response effect of a reduced height gene. Plant height showed continuous distribution in the F_(2) population, and segregation distortion was observed among the 2292 F_(2:3) progenies. The reduced height mutation was characterized by Illumina 90 K iSelect SNP genotyping and bulked segregant RNA-Seq(BSR-Seq) analysis of the segregating population. A concentrated cluster of polymorphic SNPs associated with the reduced height phenotype was detected in the distal region of chromosome arm 2 BL.
