| 1 | A LuxR family transcriptional regulator AniF promotes the production of anisomycin and its derivatives in Streptomyces hygrospinosus var.beijingensis显示文摘The protein synthesis inhibitor anisomycin features a unique benzylpyrrolidine system and exhibits potent selective activity against pathogenic protozoa and fungi.It is one of the important effective components in Agricultural Antibiotic120,which has been widely used as naturally-originated agents for treatment of crop decay in China.The chemical synthesis of anisomycin has recently been reported,but the complex process with low productivity made the biosynthesis still to be a vital mainstay in efforts.The biosynthetic gene cluster(BGC)of anisomycin in Streptomyces hygrospinosus var.beijingensis has been identified in our previous work,while poor understanding of the regulatory mechanism limited the yield enhancement via regulation engineering of S.hygrospinosus var.beijingensis.In this study here,we characterized AniF as an indispensable LuxR family transcriptional regulator for the activation of anisomycin biosynthesis.The genetic manipulations of aniF and the real-time quantitative PCR(RT-qPCR)revealed that it positively regulated the transcription of the anisomycin BGC.Moreover,the overexpression of aniF contributed to the improvement of the production of anisomycin and its derivatives.Dissection of the mechanism underlying the function of AniF revealed that it directly activated the transcription of the genes aniR-G involved in anisomycin biosynthesis.Especially,one AniF-binding site in the promoter region of aniR was identified by DNase I footprinting assay and an inverted repeat sequence(5′-GGGC-3′)composed of two 4-nt half sites in the protected region was found.Taken together,our systematic study confirmed the positive regulatory role of AniF and might facilitate the future construction of engineering strains with high productivity of anisomycin and its derivatives. | Jufang Shen Lingxin Kong Yan Li Xiaoqing Zheng Qing Wang Weinan Yang Zixin Deng Delin You | 2019 | Synthetic and Systems Biotechnology2019,4,1: | 10 |

| 3 | Quantitative investigation on collapse margin of steel high-rise buildings subjected to extremely severe earthquakes显示文摘Reponses of structures subjected to severe earthquakes sometimes significantly surpass what was considered in the design.It is important to investigate the failure mechanism and collapse margin of structures beyond design,especially for high-rise buildings.In this study,steel high-rise buildings using either square concrete-filled-tube(CFT) columns or steel tube columns are designed.A detailed three-dimensional(3 D) structural model is developed to analyze the seismic behavior of a steel high-rise towards a complete collapse.The effectiveness is verified by both component tests and a full-scale shaking table test.The collapse margin,which is defined as the ratio of PGA between the collapse level to the design major earthquake level(Level 2),is quantified by a series of numerical simulations using incremental dynamic analyses(IDA).The baseline building using CFT columns collapsed with a weak first story mechanism and presented a collapse margin ranging from 10 to 20.The significant variation in the collapse margin was caused by the different characteristics of the input ground motions.The building using equivalent steel columns collapsed earlier due to the significant shortening of the locally buckled columns,exhibiting only 57% of the collapse margin of the baseline building.The influence of reducing the height of the first story was quite significant.The shortened first story not only enlarged the collapse margin by 20%,but also changed the collapse mode. | Lin Xuchuan Mikiko Kato Zhang Lingxin Masayoshi Nakashima | 2018 | Earthquake Engineering and Engineering Vibration2018,17,3: | 9 |

| 5 | Characterization of the positive SARP family regulator PieR for improving piericidin A1 production in Streptomyces piomogeues var.Hangzhouwanensis显示文摘Piericidin A1,a member ofɑ-pyridone antibiotic,exhibits various biological activities such as antimicrobial,antifungal,and antitumor properties and possesses potent respiration-inhibitory activity against insects due to its competitive binding capacity to mitochondrial complex I.The biosynthetic pathway of piericidin A1 has been reported in Streptomyces piomogeues var.Hangzhouwanensis,while the regulatory mechanism remains poorly understood.In this study,a Streptomyces antibiotic regulatory protein(SARP)family transcriptional regulator PieR was characterized.Genetic disruption and complementation manipulations revealed that PieR positively regulated the production of piericidin A1.Moreover,the overexpression of pieR contributed to the improvement of piericidin A1 productivity.The real-time quantitative PCR(RT-qPCR)was carried out and the data showed that pieR stimulated the transcription of all the biosynthesis-related genes for piericidin A1.In order to explore the regulatory mechanism,electrophoresis mobility shift assays(EMSA)and DNase I footprinting experiments have been conducted.A protected region covering 50 nucleotides within the upstream region of pieR was identified and two 5-nt direct repeat sequences(5′-CCGGA-3′)in the protected region were found.These findings,taken together,set stage for transcriptional control engineering in the view of optimizing piericidin A1 production and thus provide a viable potent route for the construction of strains with high productivity. | Yan Li Lingxin Kong Jufang Shen Qing Wang Qian Liu Weinan Yang Zixin Deng Delin You | 2019 | Synthetic and Systems Biotechnology2019,4,1: | 4 |

| 10 | In Situ Electrochemical Mn(Ⅲ)/Mn(Ⅳ) Generation of Mn(Ⅱ)O Electrocatalysts for High-Performance Oxygen Reduction显示文摘Among various earth-abundant and noble metal-free catalysts for oxygen reduction reaction(ORR),manganese-based oxides are promising candidates owing to the rich variety of manganese valence.Herein,an extremely facile method for the synthesis of cubic and orthorhombic phase coexisting Mn(Ⅱ)O electrocatalyst as an efficient ORR catalyst was explored.The obtained MnO electrocatalyst with oxygen vacancies shows a significantly elevated ORR catalytic activity with a half-wave potential(E1/2) of as high as 0.895 V,in comparison with that of commercial Pt/C(E1/2=0.877 V).More impressively,the MnO electrocatalyst exhibits a marked activity enhancement after test under a constant applied potential for 1000 s thanks to the in situ generation and stable presence of high-valence manganese species(Mn^3+ and Mn^4+) during the electrochemical process,initiating a synergetic catalytic effect with oxygen vacancies,which is proved to largely accelerate the adsorption and reduction of O_2 molecules favoring the ORR activity elevation.Such an excellent ORR catalytic performance of this MnO electrocatalyst is applied in Zn-air battery,which shows an extra-high peak power density of 63.2 mW cm^-2 in comparison with that(47.4 m W cm^-2) of commercial Pt/C under identical test conditions. | Han Tian Liming Zeng Yifan Huang Zhonghua Ma Ge Meng Lingxin Peng Chang Chen Xiangzhi Cui Jianlin Shi | 2020 | Nano-Micro Letters2020,12,11: | 2 |
| 11 | Multimarker Detection of MAGE-1,MAGE-3 and AFP mRNAs by a Real-time Quantitative PCR Assay:a Possible Predictor of Hematogenous Micrometastasis of Hepatocellular Carcinoma显示文摘OBJECTIVE To explore the relationship between multimarker detection of MAGE-1,MAGE-3 and AFP mRNAs in the peripheral blood of patients with hepatocellular carcinoma and micrometastasis using a realtime quantitative-PCR(real-time Q-PCR)assay. METHODS Peripheral blood samples were obtained from control subjects and 86 patients with hepatocellular carcinoma (HCC).Real-time Q-PCR was used to detect MAGE-1,MAGE-3, and AFP mRNAs in the blood cells. RESULTS In 86 tumor specimens,the positivity for MAGE-1, MAGE-3,and AFP genes was respectively 34.9%(30/86),60.5% (52/86)and 69.8%(60/86).All specimens expressed at least one marker.MAGE-1,MAGE-3,and AFP transcripts were detected respectively in 12(14.0%),18(20.1%)and 29(33.7%)of the 86 blood specimens from hepatocellular carcinoma patients,while 45 specimens(52.3%)were positive for at least one marker.In addition,MAGE-1,MAGE-3 and AFP gene transcripts were not detected in any peripheral blood specimens from 25 chronic liver disease patients and 28 normal healthy volunteers.The positive rate correlated with the TNM clinical stages,extrahepatic metastasis and portal vein carcinothrombosis(P<0.05).No correlation was found between tumor size,tumor number, differentiation,serum a-fetoprotein(AFP)and the positive rate.
