| 1 | Whole-Genome Resequencing of a Worldwide Collection of Rapeseed Accessions Reveals the Genetic Basis of Ecotype Divergence显示文摘Rapeseed (Brassica napus),an important oilseed crop,has adapted to diverse climate zones and latitudes by forming three main ecotype groups,namely winter,semiwinter,and spring types. However,genetic variations underlying the divergence of these ecotypes are largely unknown. Here,we report the global pattern of genetic polymorphisms in rapeseed determined by resequencing a worldwide collection of 991 germplasm accessions.A total of 5.56 and 5.53 million singlenucleotide polymorphisms (SNPs)as Well as 1.86 and 1.92 million InDels were identified by mapping reads to the reference genomes of 'Darmor-bzh'and 'Tapidor,'respectively.We generated a map of allelic drift paths that shows splits and mixtures of the main populations,and revealed an asymmetric evolution of the two subgenomes of B.napus by calculating the genetic diversity and linkage disequilibrium parameters.Selective-sweep analysis revealed genetic changes in genes orthologous to those regulating various aspects of plant development and response to stresses.A genome-wide association study identified SNPs in the promoter regions of FLOWERING LOCUS T and FLOWERING LOCUS C orthologs that corresponded to the different rapeseed ecotype groups. | 2 | Neuroprotective effects of total saponins from Rubus parvifolius L. on cerebral ischemia/reperfusion injury in rats显示文摘This study examines the neuroprotective effects and mechanisms of action of total saponins from Rubus parvifolius L. (TSRP) on focal cerebral ischemia and reperfusion injury in rats. Focal cerebral ischemia and reperfusion injury was performed in rats using the suture method. The results indicate that intragastric injection of TSRP, at 5, 10 and 20 mg/kg, could decrease neurological impairment, reduce cerebral infarct volume, diminish pathological changes, and significantly inhibit the apoptosis of neurons surrounding the ischemic area. In addition, TSRP upregulated the expression of the anti-apoptotic factor Bcl-2, at the protein and mRNA levels, and it downregulated the expression of the pro-apoptotic factor Bax, at the protein and mRNA levels. | 3 | The dichotomous and incomplete adaptive immunity in COVID-19 patients with different disease severity显示文摘The adaptive immunity that protects patients from coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is not well characterized.In particular,the asymptomatic patients have been found to induce weak and transient SARS-CoV-2 antibody responses,but the underlying mechanisms remain unknown;meanwhile,the protective immunity that guide the recovery of these asymptomatic patients is elusive.Here,we characterized SARS-CoV-2-specific B-cell and T-cell responses in 10 asymptomatic patients and 64 patients with other disease severity(mild,n=10,moderate,n=32,severe,n=12)and found that asymptomatic or mild symptomatic patients failed to mount virus-specific germinal center(GC)B cell responses that result in robust and prolonged humoral immunity,assessed by GC response indicators including follicular helper T(TFH)cell and memory B cell responses as well as serum CXCL13 levels.Alternatively,these patients mounted potent virus-specific TH1 and CD8+T cell responses.In sharp contrast,patients of moderate or severe disease induced vigorous virus-specific GC B cell responses and associated TFH responses;however,the virus-specific TH1 and CD8+T cells were minimally induced in these patients.These results,therefore,uncovered the protective immunity in asymptomatic patients and also revealed the strikingly dichotomous and incomplete humoral and cellular immune responses in COVID-19 patients with different disease severity,providing important insights into rational design of effective COVID-19 vaccines. | Leiqiong Gao Jing Zhou Sen Yang Lisha Wang Xiangyu Chen Yang Yang Ren Li Zhiwei Pan Jing Zhao Zhirong Li Qizhao Huang Jianfang Tang Li Hu Pinghuang Liu Guozhong Zhang Yaokai Chen Lilin Ye | 2021 | Signal Transduction and Targeted Therapy2021,6,4: | 2 |

| 9 | Healing Effect and Underlying Mechanism of Silver Nanoparticles Prepared Using Carboxymethyl Chitosan under Solar Irradiation显示文摘In our previous study,silver nanoparticles were prepared using AgNO_(3) and carboxymethyl chitosan(CMCS)which is commercially available with solar irradiation.In this study,the efficacy and safety of silver nanoparticles prepared by this method were evaluated for healing wounds in rats with diabetes.We also attempted to determine the underlying mechanism and influencing factors of the silver nanomaterial preparation method.Compared with growth factors,silver nanoparticles exhibit better healing effects for rats with diabetes.No remnant silver ions were detected in the major organs of these rats after the application of silver nanoparticles.Silver nanoparticles prepared using CMCS are less toxic than those prepared from the conventional method,promote the proliferation of skin fibroblasts,and are promising as a topical medication for infected wounds.An obvious breakage process of the particles is observed during the growth of nanocrystalline silver in CMCS.In this study,we also attempted to determine whether this method is suitable for synthesizing silver nanoparticles using N-trimethyl chitosan chloride and sodium alginate were used in this particular experiment.The characteristic UV-vis absorbance peak of silver nanoparticles was found only in the reaction mixture containing N-trimethyl chitosan chloride.Our study demonstrates that free radicals are the key factor in this silver nanoparticle preparation method. | Yi Long Chunling Zhang Lan Chen Tietao Di Lu Chen Lisha Tang Kaizhong Luo Yu Yan Pingzhen Tong Jie Xiang | 2021 | Journal of Renewable Materials2021,9,6: | 0 |
| 10 | Literature and patent analysis of the cloning and identification of human functional genes in China显示文摘The Human Genome Project was launched at the end of the 1980s.Since then,the cloning and identification of functional genes has been a major focus of research across the world.In China too,the potentially profound impact of such studies on the life sciences and on human health was realized,and relevant studies were initiated in the 1990s.To advance China's involvement in the Human Genome Project,in the mid-1990s,Committee of Experts in Biology from National High Technology Research and Development Program of China(863 Program) proposed the 'two 1%' goal.This goal envisaged China contributing 1% of the total sequencing work,and cloning and identifying 1% of the total human functional genes.Over the past 20 years,tremendous achievement has been accomplished by Chinese scientists.It is well known that scientists in China finished the 1% of sequencing work of the Human Genome Project,whereas,there is no comprehensive report about 'whether China had finished cloning and identifying 1% of human functional genes'.In the present study,the GenBank database at the National Center of Biotechnology Information,the PubMed search tool,and the patent database of the State Intellectual Property Office,China,were used to retrieve entries based on two screening standards:(i) Were the newly cloned and identified genes first reported by Chinese scientists?(ii) Were the Chinese scientists awarded the gene sequence patent? | 18 | In-situ synthesis of high thermal stability and salt resistance carbon dots for injection pressure reduction and enhanced oil recovery显示文摘Carbon dots(CDs)show great potential as a new type of oil-displacing agent for unconventional oil and gas development.However,the instability and easy aggregation epitomize the challenges that accompany the application of CDs in high temperature and high salinity(HT/HS)reservoirs.In this research,novel benzene sulfonate-modified carbon dots(BS-CDs)with remarkable thermal stability and salt resistance were fabricated through an in-situ electrochemical exfoliation method.Molecular simulation verifies that the introduction of benzene sulfonate groups substantially strengthens the electrostatic repulsion between BS-CDs,leading to outstanding dispersibility and stability even at a temperature of 100℃ and salinity of 14×10^(4) mg/L.Core flooding tests show that 0.05 wt.%BS-CDs nanofluid can significantly reduce the water injection pressure by 50.00%and enhanced oil recovery(EOR)to 68.39%under HT/HS conditions.According to the atomic force microscopy(AFM)scanning results,the adhesion force between the core(after BS-CDs treatment)and oil decreased by 11.94 times,indicating that the hydrophilicity of the core surface was increased.In addition,the distribution of the adhesion force curve is more concentrated,which means that the micro-scale wettability of the core changes from oil-wet to more homogeneous water-wet.This study provides a feasible way for the development and application of good thermal stability and salt resistance CDs in unconventional reservoir development. | Yining Wu Lisha Tang Dayu Liu Demin Kong Liu Kai Mengjiao Cao Qingshan Zhao | 2023 | Nano Research2023,16,10: | 0 |
