1Marsdenia tenacissima extract induces G_0/G_1 cell cycle arrest in human esophageal carcinoma cells by inhibiting mitogen-activated protein kinase(MAPK) signaling pathway显示文摘Marsdenia tenacissima extract(MTE, trade name: Xiao-Ai-Ping injection) is an extract of a single Chinese plant medicine. It has been used for the treatment of cancer in China for decades, especially for esophageal cancer and other cancers in the digestive tract. In the present study, the potential mechanism for MTE's activity in esophageal cancer was explored. The effects of MTE on the proliferation of human esophageal cancer cells(KYSE150 and Eca-109) were investigated by the MTT assay, the Brd U(bromodeoxyuridine) incorporation immunofluorescence assay, and flow cytometric analysis. MTE inhibited cell proliferation through inducing G0/G1 cell cycle arrest in KYSE150 and Eca-109. Western blot analysis was employed to determine protein levels in the MTE treated cells. Compared with the control cells, the expression levels of the cell cycle regulatory proteins cyclin D1/D2/D3, cyclin E1, CDK2/4/6(CDK: cyclin dependent kinase), and p-Rb were decreased significantly in the cells treated with MTE at 40 mg·m L-1. In addition, MTE had an inhibitory effect on the MAPK(mitogen-activated protein kinase) signal transduction pathway, including ERK(extracellular signal-regulated kinase), JNK(c-Jun N-terminal kinase), and p38 MAPK. Moreover, MTE showed little additional effects on the regulation of cyclin D1/D3, CDK4/6, and p-Rb when the ERK pathway was already inhibited by the specific ERK inhibitor U0126. 2Mechanism of exosomal microRNA-224 in development of hepatocellular carcinoma and its diagnostic and prognostic value显示文摘BACKGROUND Exosomes contain proteins, lipids, and biological molecules such as DNA and RNA. Nucleic acids in exosomes are a group of molecules that can act as biomarkers. Currently, there are many reports on exosomal microRNAs, which are ideal biomarkers for the early diagnosis of cancer. However, there are few reports on the role of exosomal microRNAs in the diagnosis and prognosis of hepatocellular carcinoma(HCC).AIM To understand the mechanism of exosomal microRNA-224(miR-224) in the development of HCC and evaluate its diagnostic and prognostic value.METHODS Cell culture and transfection of exosomal miRNA-224, real-time quantitative PCR, luciferase reporter assay, and other methods were used to find new biomarkers related to the development of HCC that can be used to diagnose HCC and predict HCC prognosis.RESULTSBy targeting glycine N-methyltransferase, incubating exosomes with miR-224 mimic resulted in a significant increase in cell proliferation compared to that of the control group, while incubation with the miR-224 inhibitor significantly reduced cell proliferation. The same results were obtained for the cell invasion assay. 3Relationship between gene expression and the accumulation of catechin during spring and autumn in tea plants (Camellia sinensis L.)显示文摘The tea plant(Camellia sinensis L.)is an important commercial crop with remarkably high catechin concentrations.Tea is popular worldwide given the plant's health benefits.Catechins are the main astringent substance in tea and are synthesized mainly via the phenylpropanoid pathway.In this study,eight cultivars of tea plants harvested both in spring and autumn were used to investigate differences in catechin concentrations by using high-performance liquid chromatography.The expression levels of genes associated with catechin biosynthesis were investigated using reverse transcription-quantitative polymerase chain reaction.The results indicated that the total catechin(TC)concentrations were significantly higher in tea plants harvested in autumn than in those harvested in spring,based on higher concentrations of epigallocatechin(EGC)in autumn tea(P<0.01).The expression of the genes phenylalanine ammonia-lyase(PAL),flavanone 3-hydroxylase(F3H),flavonoid 3′,5′-hydroxylase(F3′5′H),dihydroflavonol 4-reductase(DFR),and anthocyanidin synthase(ANS)is closely related to the TC content of tea plants in both spring and autumn.Positive correlations between PAL,cinnamate 4-hydroxylase(C4H),F3H,and DFR expression and EGC accumulation in autumn tea were identified,with correlation coefficients of 0.710,0.763,0.884,and 0.707,respectively.A negative correlation between ANS expression level and EGC concentrations in tea plants harvested in spring was noted(r=−0.732).Additionally,negative correlations between F3H and ANS expression levels and the catechin content were identified in spring tea,whereas the correlations were positive in autumn tea.Significant differences in the F3H and ANS expression levels between spring and autumn tea indicate that F3H and ANS are potentially key genes affecting catechin accumulation in tea plants.Min Liu Heng-lu Tian Jian-Hua Wu Ren-Rong Cang Run-Xian Wang Xiao-Hua Qi Qiang Xu Xue-Hao Chen 2015Horticulture Research2015,2,1:10
4Knockdown of HMGB1 improves apoptosis and suppresses proliferation and invasion of glioma cells显示文摘Background: Effective methods for managing patients with solitary pulmonary nodules(SPNs) depend critically on the predictive probability of malignancy.Methods: Between July 2009 and June 2011, data on gender, age, cancer history, tumor familial history, smoking status, tumor location, nodule size, spiculation, calcification, the tumor border, and the final pathological diagnosis were collected retrospectively from 154 surgical patients with an SPN measuring 3-30 mm. Each final diagnosis was compared with the probability calculated by three predicted models—the Mayo, VA, and Peking University(PU) models. The accuracy of each model was assessed using area under the receiver operating characteristics(ROC) and calibration curves.Results: The area under the ROC curve of the PU model [0.800; 95% confidence interval(CI): 0.708-0.891] was higher than that of the Mayo model(0.753; 95% CI: 0.650-0.857) or VA model(0.728; 95% CI: 0.623-0.833); however, this finding was not statistically significant. To varying degrees, calibration curves showed that all three models overestimated malignancy.Conclusions: The three predicted models have similar accuracy for prediction of SPN malignancy, although the accuracy is not sufficient. For Chinese patients, the PU model may has greater predictive power.Background: Here, we introduced our short experience on the application of a new CUSA Excel ultrasonic aspiration system, which was provided by Integra Lifesciences corporation, in skull base meningiomas resection.Methods: Ten patients with anterior, middle skull base and sphenoid ridge meningioma were operated using the CUSA Excel ultrasonic aspiration system at the Neurosurgery Department of Shanghai Huashan Hospital from August 2014 to October 2014. There were six male and four female patients, aged from 38 to 61 years old(the mean age was 48.5 years old). Five cases with tumor located at anterior skull base, three cases with tumor on middle skull base, and two cases with tumor on sphenoid ridge.Results: All the patents received total resection of meningiomas with the help of this new tool, and the critical brain vessels and nerves were preserved during operations. All the patients recovered well after operation.Conclusions: This new CUSA Excel ultrasonic aspiration system has the advantage of preserving vital brain arteries and cranial nerves during skull base meningioma resection, which is very important for skull base tumor operations. This key step would ensure a well prognosis for patients. We hope the neurosurgeons would benefit from this kind of technique.Background: The purposes of this study were to explore the effects of high mobility group protein box 1(HMGB1) gene on the growth, proliferation, apoptosis, invasion, and metastasis of glioma cells, with an attempt to provide potential therapeutic targets for the treatment of glioma. Methods: The expressions of HMGB1 in glioma cells(U251, U-87 MG and LN-18) and one control cell line(SVG p12) were detected by real time PCR and Western blotting, respectively. Then, the effects of HMGB1 on the biological behaviors of glioma cells were detected: the expression of HMGB1 in human glioma cell lines U251 and U-87 MG were suppressed using RNAi technique, then the influences of HMGB1 on the viability, cycle, apoptosis, and invasion abilities of U251 and U-87 MG cells were analyzed using in a Transwell invasion chamber. Also, the effects of HMGB1 on the expressions of cyclin D1, Bax, Bcl-2, and MMP 9 were detected. Results: As shown by real-time PCR and Western blotting, the expression of HMGB1 significantly increased in glioma cells(U251, U-87 MG, and LN-18) in comparison with the control cell line(SVG p12); the vitality, proliferation and invasive capabilities of U251 and U-87 MG cells in the HMGB1 siR NA-transfected group were significantly lower than those in the blank control group and negative control(NC) siR NA group(P<0.05) but showed no significant difference between the blank control group and NC siR NA group. The percentage of apoptotic U251 and U-87 MG cells was significantly higher in the HMGB1 siR NA-transfected group than in the blank control group and NC siR NA group(P<0.05) but was similar between the latter two groups. The HMGB1 siR NA-transfected group had significantly lower expression levels of Cyclin D1, Bcl-2, and MMP-9 protein in U251 and U-87 MG cells and significantly higher expression of Bax protein than in the blank control group and NC siR NA group(P<0.05); the expression profiles of cyclin D1, Bax, Bcl-2, and MMP 9 showed no significant change in both blank control group and NC siR NA group. Conclusions: HMGB1 gene may promote the proliferation and migration of glioma cells and suppress its effects of apoptosis. Inhibition of the expression of HMGB1 gene can suppress the proliferation and migration of glioma cells and promote their apoptosis. 5Viral and Bacterial Etiology of Acute Febrile Respiratory Syndrome among Patients in Qinghai, China显示文摘Objective This study was conducted to investigate the viral and bacterial etiology and epidemiology of patients with acute febrile respiratory syndrome(AFRS) in Qinghai using a commercial routine multiplex-ligation-nucleic acid amplification test(NAT)-based assay. Methods A total of 445 nasopharyngeal swabs specimens from patients with AFRS were analyzed using the RespiFinderSmart22 kit(PathoFinder BV, Netherlands) and the LightCycler 480 real-time PCR system. Results Among the 225(225/445, 51%) positive specimens, 329 positive pathogens were detected, including 298(90.58%) viruses and 31(9%) bacteria. The most commonly detected pathogens were influenza virus(IFV;37.39%;123/329), adenovirus(AdV;17.02%;56/329), human coronaviruses(HCoVs;10.94%;36/329), rhinovirus/enterovirus(RV/EV;10.03%;33/329), parainfluenza viruses(PIVs;8.51%;28/329), and Mycoplasma pneumoniae(M. pneu;8.51%;28/329), respectively. Among the co-infected cases(17.53%;78/445), IFV/AdV and IFV/M. pneu were the most common co-infections. Most of the respiratory viruses were detected in summer and fall. Conclusion In our study, IFV-A was the most common respiratory pathogen among 22 detected pathogens, followed by AdV, HCoV, RV/EV, PIV, and M. pneu. Bacteria appeared less frequently than viruses, and co-infection was the most common phenomenon among viral pathogens. Pathogens were distributed among different age groups and respiratory viruses were generally active in July, September, and November. 6Complementation in the composition of steel slag and red mud for preparation of novel ceramics显示文摘A method for preparing novel ceramics was developed in this study. Different ratios red muds were added to steel slags to optimize the preparation of novel ceramics by a traditional ceramic preparation process. The sintering mechanism, microstructure, and performance were studied by X-ray diffraction techniques, scanning electron microscopy, and combined experiments of linear shrinkage, water absorption, and flexural strength. The results confirmed that red mud can reduce the volumetric instabilities through the complementarity of red mud and ferroalloy slag. The crystal phases in the ceramics are all pyroxene group minerals, including diopside ferrian, augite, and diopside. 7Casting structure of pure aluminum by electric pulse modification at different superheated temperatures显示文摘Electric pulse modification (EPM) is a novel technique that reduces grain size by altering the structure of a melt. It was investigated that the response of the casting structure of high pure aluminum to EPM in different superheated melts. The results indi- cate that the grain refining effect of a given pulse electric field holds an optimal temperature range, moreover, a lower or higher su- perheated temperature will both disadvantage the improvements of casting structure. 8Approximate Iteration Detection and Precoding in Massive MIMO显示文摘Massive multiple-input multiple-output provides improved energy efficiency and spectral efficiency in 5 G. However it requires large-scale matrix computation with tremendous complexity, especially for data detection and precoding. Recently, many detection and precoding methods were proposed using approximate iteration methods, which meet the demand of precision with low complexity. In this paper, we compare these approximate iteration methods in precision and complexity, and then improve these methods with iteration refinement at the cost of little complexity and no extra hardware resource. By derivation, our proposal is a combination of three approximate iteration methods in essence and provides remarkable precision improvement on desired vectors. The results show that our proposal provides 27%-83% normalized mean-squared error improvement of the detection symbol vector and precoding symbol vector. Moreover, we find the bit-error rate is mainly controlled by soft-input soft-output Viterbi decoding when using approximate iteration methods. 10Correlation analysis between the chemical contents and bioactivity for the quality control of Alismatis Rhizoma显示文摘In order to clarify regions of production and to discriminate processing methods, quantitative and qualitative analyses for saccharides and terpenes in 35 batches of Alismatis Rhizoma were performed.Methodologies included HPLC–PDA, HPLC–VWD and UHPLC–MSn, combined with principal component analysis(PCA) and partial least squares regression techniques(PLSR). The inhibitory effects of triterpenes and Alismatis Rhizoma extracts on lipase activity were evaluated in vitro. PLSR analysis revealed significant positive correlations(R2= 0.5795) between the contents of triterpenes 10, 14, 15, 18 and 22 and the inhibitory effects of Alismatis Rhizoma. The present study establishes an effective method for simultaneous determination of multiple components, and identifies key bioactive triterpenes. 11Effectiveness of flesh-moistening paste in psoriasis vulgaris patients with symptom pattern of blood stasis:a randomized and parallel-controlled trial显示文摘OBJECTIVE:To evaluate the efficacy and safety of flesh-moistening paste for treating psoriasis vulgaris in patients with blood stasis pattern in terms of Traditional Chinese Medicine(TCM).METHODS:Eudipleural rashes on both the left and right side of the same patients with psoriasis vulgaris,diagnosed via TCM as blood stasis pattern,were selected as the targeted skin lesions.A randomized,double-blind,parallel-controlled,multicenter controlled trial was conducted.The targeted skin lesions were categorized into either the treatment or control group.The treatment group used the flesh-moistening paste;the control group used a placebo.Both the paste and the placebo were topically applied twice daily for eight weeks.The patients were examined biweekly to evaluate the effects.The two groups were compared in terms of the psoriasis area and severity index(PASI)of the targeted skin lesions,which is scored according to erythema,desquamation,infiltration,area,pruritus,and improvement of skin barrier function.Adverse events were recorded during the study period.SPSS 21.0 was used to analyze the data.RESULTS:Fifty-six patients were enrolled between February 2016 and October 2017.Two were complicated by cardio-cerebrovascular disease and were excluded;thus,54 outpatients were finally enrolled in the study.Four dropped out during the study period:three failed to complete their follow-up visits for unknown reasons,and one exited due to an adverse event.The final trial comprised 50 of the 56 originally selected patients,with a 92.6%completion rate.After 8 weeks of treatment,the targeted skin lesion scores differed significantly(P<0.05).The PASI scores of the targeted skin lesions differed significantly beginning at week 6(P<0.05).The treatment group presented better results than those of the control group.Only one patient had an adverse reaction associated with the treatment.Improvements in skin barrier function differed significantly(P<0.05).CONCLUSION:The flesh-moistening paste demonstrated a reliable curative effect and safety for treating psoriasis vulgaris in patients with blood stasis patterns.The topical paste improved the barrier function of the skin lesions.Guo Xinwei Lang Na Li Guanru Li Ping Qu Jianhua Zhou Dongmei Wang Ping Cang Zhang Deng BingXu Lou Weihai Xu Jingna Lan Haibing Liu Yumin Zhang Weishu Sun Liyun 2020Journal of Traditional Chinese Medicine2020,40,6:4

13Preparation of hydro-sodalite from fly ash using a hydrothermal method with a submolten salt system and study of the phase transition process显示文摘Hydro-sodalites are zeolitic materials with a wide variety of applications.Fly ash is an abundant industrial solid waste,rich in silicon and aluminum,from which hydro-sodalite can be synthesized.However,traditional hydrothermal synthesis methods are complex and cannot produce high-purity products.Therefore,there is a demand for processing routes to obtain high-purity hydro-sodalites.In the present study,high-purity hydro-sodalite(90.2 wt%)was prepared from fly ash by applying a hydrothermal method to a submolten salt system.Samples were characterized by powder X-ray diffraction(XRD),scanning electron microscopy(SEM),thermogravimetry and differential thermal analysis(TG–DTA),and Fourier transform infrared spectroscopy(FTIR)to confirm and quantify conversion of the raw material into the product phase.Purity of the samples prepared with an H2O/Na OH mass ratio of 1.5 and an H2O/fly ash mass ratio of 10 was calculated and the conversion process of the product phase was studied.Crystallinity of the product was influenced more by the Na OH concentration,less by the H2O/fly ash mass ratio.The main reaction process of the system is that the Si O ions produced by dissolution of the vitreous body in the fly ash and Na+ions in the solution reacted on the destroyed mullite skeleton to produce hydro-sodalite.This processing route could help mitigate processing difficulties,while producing high-purity hydro-sodalite from fly ash.Yan-bing Zong Cheng-yu Zhao Wen-hui Chen Zhao-bo Liu Da-qiang Cang 2020International Journal of Minerals,Metallurgy and Materials2020,27,1:3
14A Quantitative Process-Based Inventory Study on Material Embodied Carbon Emissions of Residential, Office, and Commercial Buildings in China显示文摘Studies on building carbon emissions focus mainly on the materialization phase of life cycle, as carbon emissions in this stage is intensive and high. This paper proposes a simplified model to calculate embodied carbon emissions in building design stage by conducting a process-based inventory analysis of carbon emissions from materials used in 129 residential buildings, 41 office buildings, and 21 commercial buildings during materialization phase. The results indicate that average carbon emissions per unit area from building materials used in residential buildings, office buildings, and commercial buildings are 514.66 kgCO2 e/m2, 533.69 kg CO2 e/m2 and 494.19 kgCO2 e/m2, respectively. Besides, ten kinds of building materials(namely, steel, commercial concrete, wall building materials, mortar, copper core cables, architectural ceramics, PVC pipes, thermal insulation materials, doors and windows, and water paint) constitute 99% of total carbon emissions in all three types of buildings. These materials are major carbon emissions sources in materialization phase. 15Effect of Traditional Chinese Medicine plus narrow-band medium-wave ultraviolet B radiation on moderate-to-severe psoriasis vulgaris in a case series显示文摘OBJECTIVE: To investigate the efficacy and safety of Traditional Chinese Medicine (TCM) therapy combined with ultraviolet B light therapy in the treatment of moderate-to-severe psoriasis. METHODS: Patients with moderate-to-severe psoriasis (skin lesion area > 10% of the body surface area) for 2 consecutive years were treated with TCM (oral and external use of herbal medicines, acupuncture, and herbal bathing) and narrow-band medium-wave ultraviolet B light treatment for 12 weeks. The treatment effect was evaluated based on the Psoriasis Area Severity Index (PASI), the achievement of a 50% reduction in the PASI (PASI50), the achievement of a 75% reduction in the PASI (PASI75), pruritus score, Dermatology Life Quality Index, and safety. RESULTS: A total of 95 outpatients were enrolled, and 92 subjects (96.8%) completed the 12-week treatment course. At baseline, the average proportion of the body surface area covered by skin lesions was 12.4%, and the average PASI was 17.7. All patients had previously been treated with conventional medicine (89.1% of patients received ultraviolet light treatment, 50.0% received glucocorticoids, and 21.7% received acitretin). After the 12-week treatment course, 22 patients (23.9%) achieved PASI75, and 43 (46.7%) achieved PASI50. The post-treatment pruritus score and Dermatology Life Quality Index of all treated patients were significantly lower than the respective baseline values (P < 0.0001). 16Variation in individual biomass decreases faster than mean biomass with increasing density of bamboo stands显示文摘The total biomass of a stand is an indicator of stand productivity and is closely related to the density of plants. According to the self-thinning law, mean individual biomass follows a negative power law with plant density. If the variance of individual biomass is constant, we can expect increased stand productivity with increasing plant density. However, Taylor's power law(TPL) that relates the variance and the mean of many biological measures(e.g. bilateral areal differences of a leaf, plant biomass atdifferent times, developmental rates at different temperatures, population densities on different spatial or temporal scales), affects the estimate of stand productivity when it is defined as the total biomass of large plants in a stand.Because the variance of individual biomass decreases faster than mean individual biomass, differences in individual biomass decline with increasing density, leading to more homogeneous timbers of greater economic value. We tested whether TPL in plant biomass holds for different species and whether the variance of individual biomass changes faster than the mean with increasing stand density.The height, ground diameter and fresh weight of 50 bamboo species were measured in 50 stands ranging from 1 m by 1 m to 30 m by 30 m to ensure more than 150 bamboos in every stand. We separately examined TPL in height,ground diameter, and weight, and found that TPL holds for all three biological measures, with the relationship strongest for weight. Using analysis of covariance to compare the regression slopes of logarithmic mean and variance against the logarithm of density, we found that the variance in individual biomass declined faster than the mean with increasing density. This suggests that dense planting reduced mean individual biomass but homogenized individual biomass. Thus, there exists a trade-off between effective stand productivity and stand density for optimal forest management. Sparse planting leads to large variation in individual biomass, whereas dense planting reduces mean individual biomass. 18Two-dimensional Electrophoresis Analysis of Proteins in Response to Cold Stress in Extremely Cold-resistant Winter Wheat Dongnongdongmai 1 Tillering Nodes显示文摘The overwintering survival ratio of the cultivar Dongnongdongmai 1 with strong cold-resistance in paramos of Heilongjiang Province in China are over 85%. The tillering nodes are the most important organs for overwintering survival of winter wheat, because there are more substances associated with cold resistance in tillering nodes than those in leaves and roots. Proteins in the tillering nodes of the cold-resistant cultivar Dongnongdongmai 1 grown under field conditions with or without any lowtemperature stress were analyzed by 2-dimensional electrophoresis and identified by mass spectrometry. In the range of pH 4-7, the expression of 37 proteins showed obvious difference (±more than two fold) in the proteomic maps of cold-stressed and non-stressed tillering nodes, including a new protein spot. All proteins exhibiting the difference in expression were identified using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry, followed by a database search for protein identification and function prediction. Five groups of proteins were confirmed, namely stress-related proteins (22%), metabolism-associated proteins (35%), and signaling molecules (24%), cell wall-binding proteins (5%), unclear proteins (14%).
