| 1 | Analysis of genetic architecture and favorable allele usage of agronomic traits in a large collection of Chinese rice accessions显示文摘Genotyping and phenotyping large natural populations provide opportunities for population genomic analysis and genome-wide association studies(GWAS). Several rice populations have been re-sequenced in the past decade;however, many major Chinese rice cultivars were not included in these studies. Here, we report large-scale genomic and phenotypic datasets for a collection mainly comprised of 1,275 rice accessions of widely planted cultivars and parental hybrid rice lines from China. The population was divided into three indica/Xian and three japonica/Geng phylogenetic subgroups that correlate strongly with their geographic or breeding origins. We acquired a total of 146 phenotypic datasets for 29 agronomic traits under multi-environments for different subpopulations. With GWAS, we identified a total of 143 significant association loci, including three newly identified candidate genes or alleles that control heading date or amylose content. Our genotypic analysis of agronomically important genes in the population revealed that many favorable alleles are underused in elite accessions, suggesting they may be used to provide improvements in future breeding efforts. | 2 | Characterization of a Novel Anti-DR5 Monoclonal Antibody WD1 with the Potential to Induce Tumor Cell Apoptosis显示文摘TNF-related apoptosis-inducing ligand (TRAIL) is a TNF family member capable of inducing apoptosis. Death receptor 5 (DR 5) is a key receptor of TRAIL and plays an important role in TRAIL-induced apoptosis. To prepare monoclonal antibodies (mAbs) against DR5,cDNA encoding soluble DR5 (sDR5) was firstly amplified by reverse transcriptase-polymerase chain reaction (RT-PCR) with specific primers,and then inserted into a prokaryotic expression vector pET-30a. The recombinant plasmid was expressed in Escherichia coli strain BL21 (DE3),and sDR5 was purified by nickel affinity chromatography. As an antigen,sDR5 was used to immunize mice. Hybridomas secreting antibodies against sDR5 were identified. One positive clone was selected to produce antibody,WD1. ELISA and immunofluorescence demonstrated that WD1 could bind recombinant sDR5 and membrane-bound DR5 (mDR5) on Jurkat and Molt-4 cells. ATPLite assays showed that Jurkat and Molt-4 cells were sensitive to the antibody in a dose dependent manner. The Annexin V/PI assays and Giemsa's staining both showed that WD1 could induce Jurkat cell apoptosis efficiently. Transient transfection of 293T cells and indirect immunofluorescence assay demonstrated that mAb (WD1) couldn't cross-react with DR4. Our findings indicated that the novel antibody,WD1 could act as a direct agonist,bind DR5 characteristically,and initiate efficient apoptotic signaling and tumor regression. | 5 | Effects of Bacterial-Feeding Nematodes and Glucose on Phenanthrene Removal by Pseudomonas putida显示文摘Bacterial-feeding nematodes can promote the bacterial activity through feeding.Bacterial abundance and their activity affect the degradation of polycyclic aromatic hydrocarbons(PAH) such as phenanthrene.The effects of bacterial-feeding nematodes,bacteria,and their interactions on the degradation of phenanthrene with or without glucose were studied through a microcosm experiment.The results showed that up to 57.0%of phenanthrene in mineral medium contaminated with phenanthrene was degraded in the control with bacteria alone and bacteria with the presence of nematodes and/or glucose increased the degradation of phenanthrene by 25.6%to 36.6%.Although both nematode and bacteria abundance decreased gradually,catechol 2,3-dioxygenase(C230) activity increased during the incubation period.Compared with bacteria alone,the presence of nematodes significantly increased C230 activity as well as the abundance of bacteria;this effect was more pronounced when glucose was present.The results imply that nematodes might promote the removal of phenanthrene from medium by stimulating bacteria and C230 activities. | JING Yongping LIYan LIU Zhaohui ZHANG Yingpeng LIU Ping SUN Ming Jiafa LUO | 2017 | Pedosphere2017,27,1: | 2 |

| 13 | Construction, photoelectric response and phase transition for new hybrid double perovskites showing narrow band gaps显示文摘To explore the lead-free key scientific issue in perovskite, double perovskite based on Ag Bi and Cu Bi was naturally selected as a competitive candidate due to its fascinating functional features, such as self-powered circularly polarized light detection, X-ray detection, photoluminescence and so on. However, the most challenging point is to simulate the structure and function of traditional lead-based perovskite in new double perovskite. At the same time, there are few suitable double perovskite systems with optical and electrical potential. The above two points greatly limit the competitiveness of double perovskite. In order to solve this problem, firstly, by analyzing and comparing previous studies,we used 2,2-dimethylpropan-1-aminium(abbreviated as 2,2-DPA) as the organic template to assemble materials. Solid-to-solid phase transition materials(2,2-DPA)3Bi2I91 and(2,2-DPA)3Pb2I72 were constructed. Along the path of lead-free and two-dimensional maintenance, we successfully synthesized(2,2-DPA)4AgBiI8.H_(2)O 3 and(2,2-DPA)_(4)CuBiI8.H_(2)O 4. As two typical semiconductors, 3 and 4 with narrower optical band gaps of 1.98 and 1.76 e V show obvious photo-response when the xenon lamp with intensity of 20 m W/cm^(2)is on or off, implying that they may be applied to light-harvesting and light-detecting devices. By referring to the phase transition mechanism of 1 and 2, 3 may be caused by ordered-disordered transition of the organic part, which was proven to be the first solid-to-solid phase transition material with <100>-oriented layered double perovskites with n = 1 by systematic characterization methods after dehydration for all we know. | 14 | Multi-Omics-Guided Discovery of Omicsynins Produced by Streptomyces sp.1647:Pseudo-Tetrapeptides Active Against Influenza A Viruses and Coronavirus HCoV-229E显示文摘Many microorganisms have mechanisms that protect cells against attack from viruses.The fermentation components of Streptomyces sp.1647 exhibit potent anti-influenza A virus(IAV)activity.This strain was isolated from soil in southern China in the 1970s,but the chemical nature of its antiviral substance(s)has remained unknown until now.We used an integrated multi-omics strategy to identify the antiviral agents from this streptomycete.The antibiotics and Secondary Metabolite Analysis Shell(antiSMASH)analysis of its genome sequence revealed 38 biosynthetic gene clusters(BGCs)for secondary metabolites,and the target BGCs possibly responsible for the production of antiviral components were narrowed down to three BGCs by bioactivity-guided comparative transcriptomics analysis.Through bioinformatics analysis and genetic manipulation of the regulators and a biosynthetic gene,cluster 36 was identified as the BGC responsible for the biosynthesis of the antiviral compounds.Bioactivity-based molecular networking analysis of mass spectrometric data from different recombinant strains illustrated that the antiviral compounds were a class of structural analogues.Finally,18 pseudo-tetrapeptides with an internal ureido linkage,omicsynins A1–A6,B1–B6,and C1–C6,were identified and/or isolated from fermentation broth.Among them,11 compounds(omicsynins A1,A2,A6,B1–B3,B5,B6,C1,C2,and C6)are new compounds.Omicsynins B1–B4 exhibited potent antiviral activity against IAV with the 50%inhibitory concentration(IC_(50))of approximately 1μmol·L^(-1)and a selectivity index(SI)ranging from 100 to 300.Omicsynins B1–B4 also showed significant antiviral activity against human coronavirus HCoV-229E.By integrating multi-omics data,we discovered a number of novel antiviral pseudo-tetrapeptides produced by Streptomyces sp.1647,indicating that the secondary metabolites of microorganisms are a valuable source of novel antivirals. | Hongmin Sun Xingxing Li Minghua Chen Ming Zhong Yihua Li Kun Wang Yu Du Xin Zhen Rongmei Gao Yexiang Wu Yuanyuan Shi Liyan Yu Yongsheng Che Yuhuan Li Jian-Dong Jiang Bin Hong Shuyi Si | 2022 | Engineering2022,,9: | 0 |
