| 1 | Analysis of HSV-I ICP22 effects on HCMV major immediate-early promoter structure显示文摘The human cytomegalovirus (HCMV) major immediate-early (MIE) promoter has strong transcriptional promoting capability. Its cis-acting regulatory elements form a special structure in this region that is repeated multiple times; the biological significance of these elements and their different compositions in the transcriptional promoting process remain unclear. Our results demonstrate that the HSV-I MIE protein ICP22 can generate strong repression of many viral and cellular promoters and enhancers. We further studied the transcriptional effects of ICP22 on structural elements and mutations in various HCMV MIE promoters by using a CAT assay. In spite of different transcriptional effects of all the ele- ments in the presence of ICP22, the transcriptional efficiencies exhibited by mutations generated by different compositions and an entire HCMV promoter, are not the simple sum of the functions of these elements. Furthermore, the transcriptional activities of specific sequences were not affected by the presence of ICP22. | 5 | Shen Qi Wan attenuates renal interstitial fibrosis through upregulating AQP1显示文摘Renal interstitial fibrosis(RIF)is the crucial pathway in chronic kidney disease(CKD)leading to the end-stage renal failure.However,the underlying mechanism of Shen Qi Wan(SQW)on RIF is not fully understood.In the current study,we investigated the role of Aquaporin 1(AQP1)in SQW on tubular epithelial-to-mesenchymal transition(EMT).A RIF mouse model induced by adenine and a TGF-β1-stimulated HK-2 cell model were etablished to explore the involvement of AQP 1 in the protective effect of SQW on EMT in vitro and in vivo.Subsequently,the molecular mechanism of SQW on EMT was explored in HK-2 cells with AQP1 knockdown.The results indicated that SQW alleviated kidney injury and renal collagen deposition in the kidneys of mice induced by adenine,increased the protein expression of E-cadherin and AQP1 expression,and decreased the expression of vimentin andα-smooth muscle actin(α-SMA).Similarly,treatmement with SQW-containing serum significantly halted EMT process in TGF-β1 stimulated HK-2 cells.The expression of snail and slug was significantly upregulated in HK-2 cells after knockdown of AQP1.AQP1 knockdown also increased the mRNA expression of vimentin andα-SMA,and decreased the expression of E-cadherin.The protein expression of vimentin increased,while the expression of E-cadherin and CK-18 significantly decreased after AQP1 knockdown in HK-2 cells.These results revealed that AQP1 knockdown promoted EMT.Furthermore,AQP1 knockdown abolished the protective effect of SQW-containing serum on EMT in HK-2 cells.In sum,SQW attentuates EMT process in RIF through upregulation of the expression of AQP1. | LIN Yiyou WEI Jiale ZHANG Yehui HUANG Junhao WANG Sichen LUO Qihan YU Hongxia JI Liting ZHOU Xiaojie LI Changyu | 2023 | Chinese Journal of Natural Medicines2023,21,5: | 1 |

| 7 | XAS:Cross-API scripting attacks in social ecosystems显示文摘With the rapid development of online social networks, various Web application programming interfaces(APIs) on social platforms are released to share profitable social data with all kinds of third-party online services. However, it also brings new risks to social networks once Web APIs are insecurely designed,implemented, and invoked. The focused topic in this paper is security analysis of a new type of cross-site scripting(XSS) which is based on Web APIs in new complicated social ecosystems which consist of social networks,third-party apps, and other online services. In this paper, we refer to Web API-based XSS as cross-API scripting(XAS). For the first time, we take typical XAS attacks in diversified context as cases to demonstrate the new exploiting opportunities and threats in social ecosystems. Also, we design a tool to identify the design and implementation flaws of Web APIs in 11 popular social networks. We discover several security flaws of API via our experiment. According to the results, we conclude causes of XAS flaws in depth. We also examined 143Web-based apps and verified the prevalence of XAS flaws.
