| 2 | Orlistat induces ferroptosis-like cell death of lung cancer cells显示文摘Aberrant de novo lipid synthesis is involved in the progression and treatment resistance of many types of cancers,including lung cancer;however,targeting the lipogenetic pathways for cancer therapy remains an unmet clinical need.In this study,we tested the anticancer activity of orlistat,an FDA-approved anti-obesity drug,in human and mouse cancer cells in vitro and in vivo,and we found that orlistat,as a single agent,inhibited the proliferation and viabilities of lung cancer cells and induced ferroptosis-like cell death in vitro.Mechanistically,we found that orlistat reduced the expression of GPX4,a central ferroptosis regulator,and induced lipid peroxidation.In addition,we systemically analyzed the genome-wide gene expression changes affected by orlistat treatment using RNA-seq and identified FAF2,a molecule regulating the lipid droplet homeostasis,as a novel target of orlistat.Moreover,in a mouse xenograft model,orlistat significantly inhibited tumor growth and reduced the tumor volumes compared with vehicle control(P<0.05).Our study showed a novel mechanism of the anticancer activity of orlistat and provided the rationale for repurposing this drug for the treatment of lung cancer and other types of cancer. | Wenjing Zhou Jing Zhang Mingkun Yan Jin Wu Shuo Lian Kang Sun Baiqing Li Jia Ma Jun Xia Chaoqun Lian | 2021 | Frontiers of Medicine2021,15,6: | 1 |
| 3 | Potential Secretory Transporters and Biosynthetic Precursors of Biological Nitrification Inhibitor 1,9-Decanediol in Rice as Revealed by Transcriptome and Metabolome Analyses显示文摘Biological nitrification inhibitors(BNIs)are released from plant roots and inhibit the nitrification activity of microorganisms in soils,reducing NO_(3)^(‒)leaching and N2O emissions,and increasing nitrogenuse efficiency(NUE).Several recent studies have focused on the identification of new BNIs,yet little is known about the genetic loci that govern their biosynthesis and secretion.We applied a combined transcriptomic and metabolomic analysis to investigate possible biosynthetic pathways and transporters involved in the biosynthesis and release of BNI 1,9-decanediol(1,9-D),which was previously identified in rice root exudates.Our results linked four fatty acids,icosapentaenoic acid,linoleate,norlinolenic acid,and polyhydroxy-α,ω-divarboxylic acid,with 1,9-D biosynthesis and three transporter families,namely the ATP-binding cassette protein family,the multidrug and toxic compound extrusion family,and the major facilitator superfamily,with 1,9-D release from roots into the soil medium.Our finding provided candidates for further work on the genes implicated in the biosynthesis and secretion of 1,9-D and pinpoint genetic loci for crop breeding to improve NUE by enhancing 1,9-D secretion,with the potential to reduce NO_(3)^(‒)leaching and N2O emissions from agricultural soils. | DI Dongwei MA Mingkun ZHANG Xiaoyang LU Yufang Herbert J.KRONZUCKER SHI Weiming | 2024 | Rice science2024,31,1: | 0 |

| 6 | Tribological properties of PTFE-based fabric composites at cryogenic temperature显示文摘Fabric composites are widely employed in self-lubricating bearing liners as solid lubrication materials.Although the tribological behaviors of fabric composites have been extensively studied,the cryogenic tribological properties and mechanisms have been scarcely reported and are largely unclear to instruct material design for aerospace and other high-tech applications.Herein,the tribological properties of polytetrafluoroethylene(PTFE)-based hybrid-fabric composites were investigated at cryogenic and ambient temperatures in the form of pin-on-disk friction under heavy loads.The results suggest that the friction coefficients of the hybrid-fabric composites obviously increase with a decrease in wear when the temperature drops from 25 to−150°C.Moreover,thermoplastic polyetherimide(PEI),as an adhesive for fabric composites,has better cryogenic lubrication performance than thermosetting phenol formaldehyde(PF)resin,which can be attributed to the flexible chemical structure of PEI.The excellent lubrication performance of hybrid-fabric composites is attributed to the transfer film formed by PTFE fibers on the surface of fabrics. | Mingkun XU Zidan WANG Lihe GUO Liming TAO Tianbao MA Tingmei WANG Qihua WANG | 2024 | Friction2024,12,2: | 0 |
| 7 | Genomic Perspectives on the Emerging SARS-CoV-2 Omicron Variant显示文摘A new variant of concern for SARS-CoV-2,Omicron(B.1.1.529),was designated by the World Health Organization on November 26,2021.This study analyzed the viral genome sequencing data of 108 samples collected from patients infected with Omicron.First,we found that the enrichment efficiency of viral nucleic acids was reduced due to mutations in the region where the primers anneal to.Second,the Omicron variant possesses an excessive number of mutations compared to other variants circulating at the same time(median:62 vs.45),especially in the Spike gene.Mutations in the Spike gene confer alterations in 32 amino acid residues,more than those observed in other SARS-CoV-2 variants.Moreover,a large number of nonsynonymous mutations occur in the codons for the amino acid residues located on the surface of the Spike protein,which could potentially affect the replication,infectivity,and antigenicity of SARS-CoV-2.Third,there are 53 mutations between the Omicron variant and its closest sequences available in public databases.Many of these mutations were rarely observed in public databases and had a low mutation rate.In addition,the linkage disequilibrium between these mutations was low,with a limited number of mutations concurrently observed in the same genome,suggesting that the Omicron variant would be in a different evolutionary branch from the currently prevalent variants.To improve our ability to detect and track the source of new variants rapidly,it is imperative to further strengthen genomic surveillance and data sharing globally in a timely manner. | Wentai Ma Jing Yang Haoyi Fu Chao Su Caixia Yu Qihui Wang Ana Tereza Ribeiro de Vasconcelos Georgii A.Bazykin Yiming Bao Mingkun Li | 2022 | Genomics, Proteomics & Bioinformatics2022,20,1: | 0 |
