| 1 | Management of cytokine release syndrome related to CAR-T cell therapy显示文摘Chimeric antigen receptor T (CAR-T) cell therapy is a novel cellular immunotherapy that is widely used to treat hematological malignancies, including acute leukemia, lymphoma, and multiple myeloma. Despite its remarkable clinical effects, this therapy has side effects that cannot be underestimated. Cytokine release syndrome (CRS) is one of the most clinically important and potentially life-threatening toxicities. This syndrome is a systemic immune storm that involves the mass cytokines releasing by activated immune cells. This phenomenon causes multisystem damages and sometimes even death. In this study, we reported the management of a patient with recurrent and refractory multiple myeloma and three patients with acute lymphocytic leukemia who suffered CRS during CAR-T treatment. The early application of tocilizumab, an anti-IL-6 receptor antibody, according to toxicity grading and clinical manifestation is recommended especially for patients who suffer continuous hyperpyrexia, hypotensive shock, acute respiratory failure, and whose CRS toxicities deteriorated rapidly. Moreover, low doses of dexamethasone (5-10 mg/day) were used for refractory CRS not responding to tocilizumab. | 2 | Generation of B Cell-Deficient Pigs by Highly Efficient CRISPR/Cas9-Mediated Gene Targeting显示文摘Generating B cell-deficient mutant is the first step to produce human antibody repertoires in large animal models. In this study, we applied the clustered regularly interspaced short palindromic repeat(CRISPR)/CRISPR-associated(Cas) system to target the JHregion of the pig Ig M heavy chain gene which is crucial for B cell development and differentiation. Transfection of Ig M-targeting Cas9 plasmid in primary porcine fetal fibroblasts(PFFs) enabled inducing gene knock out(KO) in up to 53.3% of colonies analyzed, a quarter of which harbored biallelic modification, which was much higher than that of the traditional homologous recombination(HR). With the aid of somatic cell nuclear transfer(SCNT) technology, three piglets with the biallelic Ig M heavy chain gene mutation were produced. The piglets showed no antibody-producing B cells which indicated that the biallelic mutation of the Ig M heavy chain gene effectively knocked out the function of the Ig M and resulted in a B cell-deficient phenotype. | 3 | Unusual high-density and saline aqueous inclusions in ultrahigh pressure metamor- phic rocks from Sulu terrane in eastern China显示文摘Primary high-density fluid inclusions were identified in garnet from ultrahigh pressure eclogite in the southern part of the Sulu terrane. They occur isolatedly or in cluster together with relatively low-density two-phase inclu-sions. The eutectic temperature of the inclusions is as low as ≤ -52℃. A bubble was nucleated in a liquid inclusion dur-ing the specific stage of cyclic cooling-heating runs, and the liquid-gas homogenization temperature was measured to be ≤-12.5℃. The composition of the inclusions modeled by the system CaCl2-NaCl-H2O, yields the fluid density of 1.27g/cm3 that corresponds to a pressure of ca. 2.4 GPa at thetemperature of peak eclogite-facies metamorphism, close tothe ultrahigh pressure metamorphic conditions. | 4 | A telomere-to-telomere gap-free reference genome of watermelon and its mutation library provide important resources for gene discovery and breeding显示文摘Watermelon,Citrullus lanatus,is the world's third largest fruit crop.Reference genomes with gaps and a narrow genetic base hinder functional genomics and genetic improvement of watermelon.Here,we report the assembly of a telomere-to-telomere gap-free genome of the elite watermelon inbred line G42 by incorporating high-coverage and accurate long-read sequencing data with multiple assembly strategies.All 11 chromosomes have been assembled into single-contig pseudomolecules without gaps,representing the highest completeness and assembly quality to date.The G42 reference genome is 369321829 bp in length and contains 24205 predicted protein-coding genes,with all 22 telomeres and 11 centromeres characterized.Furthermore,we established a pollen-EMS mutagenesis protocol and obtained over 200000M1 seeds from G42.In a sampling pool,48 monogenic phenotypic mutations,selected from 223M1and 78 M2 mutants with morphological changes,were confirmed.The average mutation density was 1 SNP/1.69Mband1 indel/4.55 Mb per M1 plant and 1SNP/1.08Mb and 1 indel/6.25 Mb per M2 plant.Taking advantage of the gap-free G42 genome,8039 mutations from 32 plants sampled from M1 and M2 families were identified with 100%accuracy,whereas only 25% of the randomly selected mutations identified using the 97103v2 reference genome could be confirmed.Using this library and the gap-free genome,two genes responsible for elongated fruit shape and male sterility(CiMs1)were identified,both caused by a single basechange from G to A.The validated gap-free genome and its EMS mutation library provide invaluable resources for functional genomics and genetic improvement of watermelon. | Yun Deng Shoucheng Liu Yilin Zhang Jingsheng Tan Xiaopeng Li Xiao Chu Binghua Xu Yao Tian Yudong Sun Bosheng Li YunbiXu Xing Wang Deng Hang He Xingping Zhang | 2022 | Molecular Plant2022,15,8: | 9 |
| 5 | Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes,downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. | 6 | Satellite-observed trends in the Arctic sea ice concentration for the period 1979–2016显示文摘Arctic sea ice cover has decreased dramatically over the last three decades. This study quanti?es the sea ice concentration(SIC) trends in the Arctic Ocean over the period of 1979–2016 and analyzes their spatial and temporal variations. During each month the SIC trends are negative over the Arctic Ocean, wherein the largest(smallest) rate of decline found in September(March) is-0.48%/a(-0.10%/a).The summer(-0.42%/a) and autumn(-0.31%/a) seasons show faster decrease rates than those of winter(-0.12%/a) and spring(-0.20%/a) seasons. Regional variability is large in the annual SIC trend. The largest SIC trends are observed for the Kara(-0.60%/a) and Barents Seas(-0.54%/a), followed by the Chukchi Sea(-0.48%/a), East Siberian Sea(-0.43%/a), Laptev Sea(-0.38%/a), and Beaufort Sea(-0.36%/a). The annual SIC trend for the whole Arctic Ocean is-0.26%/a over the same period. Furthermore, the in?uences and feedbacks between the SIC and three climate indexes and three climatic parameters, including the Arctic Oscillation(AO), North Atlantic Oscillation(NAO), Dipole anomaly(DA), sea surface temperature(SST), surface air temperature(SAT), and surface wind(SW), are investigated. Statistically, sea ice provides memory for the Arctic climate system so that changes in SIC driven by the climate indices(AO, NAO and DA) can be felt during the ensuing seasons. Positive SST trends can cause greater SIC reductions, which is observed in the Greenland and Barents Seas during the autumn and winter. In contrast, the removal of sea ice(i.e., loss of the insulating layer) likely contributes to a colder sea surface(i.e., decreased SST), as is observed in northern Barents Sea. Decreasing SIC trends can lead to an in-phase enhancement of SAT, while SAT variations seem to have a lagged in?uence on SIC trends. | 7 | Aneuploidy of chromosome 8 in circulating tumor cells correlates with prognosis in patients with advanced gastric cancer显示文摘Objective: Previous work indicated that aneuploidy of chromosome 8 in circulating tumor cells(CTCs)correlated with therapeutic efficacy for advanced gastric cancer(AGC) patients. In this follow-up study performed on the same population of AGC patients, we investigated whether and how aneuploidy of chromosome 8 in CTCs correlates with patients' clinical prognosis.Methods: The prospective study was performed on 31 patients with newly diagnosed AGC. Previously established integrated subtraction enrichment(SE) and immunostaining-fluorescence in situ hybridization(i FISH)platform was applied to identify, enumerate and characterize CTCs. Quantification of CTCs and analysis of their aneuploidy of chromosome 8 were performed on patients before and after therapy.Results: CTCs were measured in 93.5% of AGC patients, and two CTC subtypes with diverse threshold values were identified, multiploid CTCs with the threshold of ≥2 per 7.5 m L and multiploid plus triploid CTCs with the threshold of ≥4, which were found to significantly correlate with poor progression-free survival(PFS) and overall survival(OS). In particular, patients with ≥10% increased multiploid CTCs after an initial 6 weeks of therapy had poor PFS and OS, whereas improved PFS and OS were observed on those who had ≥10% decreased multiploid CTCs. After adjusting for clinically significant factors, ≥10% increased post-therapy multiploid CTCs was the only independent predictor of PFS and OS.Conclusions: Aneuploidy of CTCs correlates with prognosis of AGC patients. | 9 | Motility-based label-free detection of parasites in bodily fluids using holographic speckle analysis and deep learning显示文摘Parasitic infections constitute a major global public health issue.Existing screening methods that are based on manual microscopic examination often struggle to provide sufficient volumetric throughput and sensitivity to facilitate early diagnosis.Here,we demonstrate a motility-based label-free computational imaging platform to rapidly detect motile parasites in optically dense bodily fluids by utilizing the locomotion of the parasites as a specific biomarker and endogenous contrast mechanism.Based on this principle,a cost-effective and mobile instrument,which rapidly screens~3.2 mL of fluid sample in three dimensions,was built to automatically detect and count motile microorganisms using their holographic time-lapse speckle patterns.We demonstrate the capabilities of our platform by detecting trypanosomes,which are motile protozoan parasites,with various species that cause deadly diseases affecting millions of people worldwide.Using a holographic speckle analysis algorithm combined with deep learningbased classification,we demonstrate sensitive and label-free detection of trypanosomes within spiked whole blood and artificial cerebrospinal fluid(CSF)samples,achieving a limit of detection of ten trypanosomes per mL of whole blood(~five-fold better than the current state-of-the-art parasitological method)and three trypanosomes per mL of CSF.We further demonstrate that this platform can be applied to detect other motile parasites by imaging Trichomonas vaginalis,the causative agent of trichomoniasis,which affects 275 million people worldwide.With its costeffective,portable design and rapid screening time,this unique platform has the potential to be applied for sensitive and timely diagnosis of neglected tropical diseases caused by motile parasites and other parasitic infections in resource-limited regions. | Yibo Zhang Hatice Ceylan Koydemir Michelle M.Shimogawa Sener Yalcin Alexander Guziak Tairan Liu Ilker Oguz Yujia Huang Bijie Bai Yilin Luo Yi Luo Zhensong Wei Hongda Wang Vittorio Bianco Bohan Zhang Rohan Nadkarni Kent Hill Aydogan Ozcan | 2018 | Light(Science & Applications)2018,7,1: | 6 |
| 10 | Optimization of a new animal glue binder system cured by CO_2 for use in foundry显示文摘A new sand binder system cured by CO2 was prepared based on the animal bone glue.To overcome the disadvantages of animal glue such as agglomeration at room temperature,high energy consumption and low efficiency,an alkaline decomposition process was selected,and certain modifier was used to modify the performance of the animal glue binder.For the alkaline decomposition,NaOH was used as the catalyst with an addition of 4wt.% to the animal glue.A modifier was determined through the orthogonal experiment with a weight ratio of glycerin:glycol:dextrin:animal glue = 9:16:15:100,and the optimal modification reaction should be performed at 75 ℃ with a reaction time of 90 min.Ca(OH)2 was used as a promoter;the optimal CO2 gas flux blowing into the sand was 0.7 m3.h-1 for a duration of 60s under the experimental conditions.Results show that an original strength above 0.7 MPa and a final strength about 4.2 MPa can be achieved,which could meet the requirement of rapid moulding and core-making for foundry.The new binder was characterized and analyzed by means of IR,and the modification and CO2-cured mechanisms of this animal glue binder were also discussed. | Liu Weihua Zhang Yilin Li Yingmin Liu Xiuling | 2012 | China Foundry2012,9,4: | 5 |

| 12 | Correlation-enhanced electron-phonon coupling for accurate evaluation of the superconducting transition temperature in bulk FeSe显示文摘It has been widely recognized that,based on standard density functional theory calculations of the electron-phonon coupling,the superconducting transition temperature(T_(c))in bulk FeSe is exceptionally low(almost 0 K)within the Bardeen-Cooper-Schrieffer formalism.Yet the experimentally observed T_(c)is much higher(∼10 K),and the underlying physical origin remains to be fully explored,especially at the quantitative level.Here we present the first accurate determination of T_(c)in FeSe where the correlation-enhanced electron-phonon coupling is treated within first-principles dynamical mean-field theory.Our studies treat both the multiple electronic bands across the Fermi level and phononic bands,and reveal that all the optical phonon modes are effectively coupled with the conduction electrons,including the important contributions of a single breathing mode as established by previ-ous experiments.Accordingly,each of those phonon modes contributes pronouncedly to the electron pairing,and the resultant T_(c)is drastically enhanced to the experimentally observed range.The approach developed here should be broadly applicable to other superconducting systems where correlation-enhanced electron-phonon coupling plays an important role. | Wenjun Ding Yilin Wang Tong Wei Jiaqing Gao Ping Cui Zhenyu Zhang | 2022 | Science China(Physics,Mechanics & Astronomy)2022,65,6: | 5 |
| 13 | Preliminary Investigation of the Reversible 4D Printing of a Dual-Layer Component显示文摘The rapid development of additive manufacturing and advances in shape memory materials have fueled the progress of four-dimensional (4D) printing. With increasing improvements in design, reversible 4D printing or two-way 4D printing has been proven to be feasible. This technology will fully eliminate the need for human interference, as the programming is completely driven by external stimuli, which allows 4D-printed parts to be actuated in multiple cycles. This study proposes a new reversible 4D print- ing actuation method. The swelling of an elastomer and heat are used in the programming stage, and heat is used in the recovery stage. The main focus of this study is on the self-actuated programming step. To attain control over the bending, a simple predictive model has been developed to study the degree of cur- vature. The parameters, temperature, and elastomer thickness have also been studied in order to gain a better understanding of how well the model predicts the curvature. | 15 | An implantable microelectrode array for simultaneous L-glutamate and electrophysiological recordings in vivo显示文摘L-glutamate,the most common excitatory neurotransmitter in the mammalian central nervous system(CNS),is associated with a wide range of neurological diseases.Because neurons in CNS communicate with each other both electrically and chemically,dualmode(electric and chemical)analytical techniques with high spatiotemporal resolution are required to better understand glutamate function in vivo.In the present study,a silicon-based implantable microelectrode array(MEA)composed of both platinum electrochemical and electrophysiological microelectrodes was fabricated using micro-electromechanical system.In the MEA probe,the electrophysiological electrodes have a low impedance of 0.018 MΩat 1 kHz,and the electrochemical electrodes show a sensitivity of 56 pAμM^(−1) to glutamate and have a detection limit of 0.5μM.The MEA probe was used to monitor extracellular glutamate levels,spikes and local field potentials(LFPs)in the striatum of anaesthetised rats.To explore the potential of the MEA probe,the rats were administered to KCl via intraperitoneal injection.K+significantly increases extracellular glutamate levels,LFP low-beta range(12–18 Hz)power and spike firing rates with a similar temporal profile,indicating that the MEA probe is capable of detecting dual-mode neuronal signals.It was concluded that the MEA probe can help reveal mechanisms of neural physiology and pathology in vivo. | Wenjing Wei Yilin Song Li Wang Song Zhang Jinping Luo Shengwei Xu Xinxia Cai | 2015 | Microsystems & Nanoengineering2015,1,1: | 4 |

| 17 | PG-CODE:Latent Dirichlet Allocation Embedded Policy Knowledge Graph for Government Department Coordination显示文摘Government policy-group integration and policy-chain inference are significant to the execution of strategies in current Chinese society.Specifically,the coordination of hierarchical policies implemented among government departments is one of the key challenges to rural revitalization.In recent years,various well-established quantitative methods have been proposed to evaluate policy coordination,but the majority of these relied on manual analysis,which can lead to subjective results.Thus,in this paper,a novel approach called“policy knowledge graph for the coordination among the government departments”(PG-CODE)is proposed,which incorporates topic modeling into policy knowledge graphs.Similar to a knowledge graph,a policy knowledge graph uses a graph-structured data model to integrate policy discourse.With latent Dirichlet allocation embedding,a policy knowledge graph could capture the underlying topics of the policies.Furthermore,coordination strength and topic diffusion among hierarchical departments could be inferred from the PG-CODE,as it can provide a better representation of coordination within the policy space.We implemented and evaluated the PG-CODE in the field of rural innovation and entrepreneurship policy,and the results effectively demonstrate improved coordination among departments. | Yilin Kang Renwei Ou Yi Zhang Hongling Li Shasha Tian | 2022 | Tsinghua Science and Technology2022,27,4: | 3 |

Aptamers have proved to be one kind of novel functional molecules in life science and chemistry. After being labeled by signaling groups, the aptamer probe can conveniently transfer the characteristics of aptamer-target recognition to a form of high-sensitive signal, and the high-affinity, high-specificity measurements of metal ion, organic molecules, nucleic acid, proteins, or cells become possible. | 19 | 2D organic semiconductors, the future of green nanotechnology显示文摘The discovery of 2D organic semiconductors of atomically thin structures has attracted great attention due to their emerging optical, electronic, optoelectronic and mechatronic properties. Recent progress in such organic nanostructures has opened new opportunities for engineering material properties in many ways, such as, 0D/1D/2D nanoparticles hybridization, strain engineering, atomic doping etc. Moreover, 2D organic nanostructures exhibit a unique feature of bio–functionality and are highly sensitive to bio-analytes. Such peculiar behavior in 2D organics can be utilized to design highly-efficient bio-sensors. Also, a bio-molecular integrated electronic/optoelectronic device with enhanced performance can be attained. Furthermore, the bio-degradable, biocompatible, biometabolizable, non-toxic behaviour and natural origin of organic nanomaterials can address the current ecological concerns of increasing inorganic material based electronic waste. This review highlights the benefits of 2D organic semiconductors. Considering the importance of strategic techniques for growing thin 2D organic layers,this review summarizes progress towards this direction. The possible challenges for long-time stability and future research directions in 2D organic nano electronics/optoelectronics are also discussed. | 20 | Two haplotype-resolved,gap-free genome assemblies for Actinidia latifolia and Actinidia chinensis shed light on the regulatory mechanisms of vitamin C and sucrose metabolism in kiwifruit显示文摘Kiwifruit is a recently domesticated horticultural fruit crop with substantial economic and nutritional value,especially because of the high content of vitamin C in its fruit.In this study,we de novo assembled two telomere-to-telomere kiwifruit genomes from Actinidia chinensis var.‘Donghong’(DH)and Actinidia latifolia‘Kuoye’(KY),with total lengths of 608327852 and 640561626 bp for 29 chromosomes,respectively.With a burst of structural variants involving inversion,translocations,and duplications within 8.39 million years,the metabolite content of DH and KY exhibited differences in saccharides,lignans,and vitamins.A regulatory ERF098 transcription factor family has expanded in KY and Actinidia eriantha,both of which have ultra-high vitamin C content.With each assembly phased into two complete haplotypes,we identified allelic variations between two sets of haplotypes,leading to protein sequence variations in 26494 and 27773 gene loci and allele-specific expression of 4687 and 12238 homozygous gene pairs.Synchronized metabolome and transcriptome changes during DH fruit development revealed the same dynamic patterns in expression levels and metabolite contents;free fatty acids and flavonols accumulated in the early stages,but sugar substances and amino acids accumulated in the late stages.The AcSWEET9b gene that exhibits allelic dominance was further identified to positively correlate with high sucrose content in fruit.Compared with wild varieties and other Actinidia species,AcSWEET9b promoters were selected in red-flesh kiwifruits that have increased fruit sucrose content,providing a possible explanation on why red-flesh kiwifruits are sweeter.Collectively,these two gap-free kiwifruit genomes provide a valuable genetic resource for investigating domestication mechanisms and genome-based breeding of kiwifruit. | Xue Han Yilin Zhang Qiong Zhang Ni Ma Xiaoying Liu Wenjing Tao Zhiying Lou Caihong Zhong Xing Wang Deng Dawei Li Hang He | 2023 | Molecular Plant2023,16,2: | 3 |
