1Microbiome and ileum transcriptome revealed the boosting effects of selenium yeast on egg production in aged laying hens显示文摘The declines in laying performance during the late productionperiod have adverse effects on the length of theproduction cycle. Improving the nutrition of laying hens is a crucialmeasure to reverse this declination. Thisstudy investigatedthe effect of seleniumyeast (SY)on egg production, ileal gene expressionandmicrobiota, aswell as elucidating their associations in aged laying hens. A total of 375 Jinghong laying hens at 76weeks oldwere randomly assigned into 5 dietary treatments, which included a selenium-deficient basal diet based oncorn-soybean meal, and dietary supplementation of SY at 0.15, 0.30 and 0.45 mg/kg, and sodium selenite at0.45mg/kg.The results showed that SYamelioratedthe depressionin aged layingperformance inthe 0.30mg/kg group (P < 0.01). Selenium yeast significantly increased ileum selenium concentration (P < 0.05), and SYgroups had higher seleniumdeposition efficiency than the sodiumselenite group. Functional enrichment andShort Time-series Expression Miner (STEM) analysis indicated that SY activated metabolic progress (e.g.,glycerolipidmetabolism, glycerophospholipidmetabolism, andfattyacidmetabolism),immune responseandoxidative stress response. Four hub genes including thioredoxin reductase 1 (TXNRD1), dihydrolipoamidedehydrogenase (DLD), integrin linked kinase (ILK) and leucine zipper tumor suppressor 2 (LZTS2) wereinvolved in intestinal metabolismwhich was closely associated with selenium deposition/status. Moreover,the relative abundance of Veillonella, Turicibacter and Lactobacillus was significantly increased, but the relativeabundance of Stenotrophomonas was significantly decreased by SY supplementation. Multi-omics dataintegration and Canonical correspondence analysis (CCA) showed that both the ileumselenium content andthe laying rate were highly correlated with pathways and bacteria enriched in metabolism and immuneresponse. Meanwhile, the “switched on” gene prostate stem cell antigen (PSCA) had a positive relationshipwith Veillonella and a negative relationship with the opportunistic pathogens Stenotrophomonas. 2Single-cell transcriptomic landscape of human blood cells显示文摘High throughput single-cell RNA-seq has been successfully implemented to dissect the cellular and molecular features underlying hematopoiesis.However,an elaborate and comprehensive transcriptome reference of the whole blood system is lacking.Here,we profiled the transcriptomes of 7551 human blood cells representing 32 immunophenotypic cell types,including hematopoietic stem cells,progenitors and mature blood cells derived from 21 healthy donors.With high sequencing depth and coverage,we constructed a single-cell transcriptional atlas of blood cells(ABC) on the basis of both protein-coding genes and long noncoding RNAs(lncRNAs),and showed a high consistence between them.Notably,putative lncRNAs and transcription factors regulating hematopoietic cell differentiation were identified.While common transcription factor regulatory networks were activated in neutrophils and monocytes,lymphoid cells dramatically changed their regulatory networks during differentiation.Furthermore,we showed a subset of nucleated erythrocytes actively expressing immune signals,suggesting the existence of erythroid precursors with immune functions.Finally,a web portal offering transcriptome browsing and blood cell type prediction has been established.Thus,our work provides a transcriptional map of human blood cells at single-cell resolution,thereby offering a comprehensive reference for the exploration of physiological and pathological hematopoiesis.Xiaowei Xie Mengyao Liu Yawen Zhang Bingrui Wang Caiying Zhu Chenchen Wang Qing Li Yingying Huo Jiaojiao Guo Changlu Xu Linping Hu Aiming Pang Shihui Ma Una Wang Wenbin Cao Shulian Chen Qiuling Li Sudong Zhang Xueying Zhao Wen Zhou Hongbo Luo Guoguang Zheng Erlie Jiang Sizhou Feng Lixiang Chen Lihong Shi Hui Cheng Sha Hao Ping Zhu Tao Cheng 2021National Science Review2021,8,3:3
3Environmental taxation and regional inequality in China显示文摘In order to combat environmental pollution, China enacted the Environmental Protection Tax Law in early 2018. Yet the impacts of the environmental tax on individual regions with different socioeconomic statuses, which are crucial for social justice and public acceptance, remain unclear. Based on a MultiRegional Input-Output(MRIO) table and a nationally regulated tax payment calculation method, this study analyzes the distributional impacts of an environmental tax based upon province’s consumption from both inter-provincial and rural-urban aspects. The national tax revenue based on the current levy mechanism is estimated to be only one seventh of the economic loss from premature mortality caused by ambient particulate matter(PM2.5). The taxation may slightly alleviate urban-rural inequality but may not be helpful with reducing inter-provincial inequality. We further analyze two alternative levy mechanisms. 4Effects of Integrated Soil-Crop System Management on Soil Organic Carbon Characteristics in a Primosol in Northeast China显示文摘A synchronous increase in crop productivity, nutrient use efficiency, and soil carbon(C) sequestration is important from the point of view of food security and environmental protection. In recent years, integrated soil-crop system management(ISSM), which uses crop models and advanced nutrient management to redesign cropping systems, has been successfully demonstrated to achieve both high crop productivity and high nutrient use efficiency in China, but the effects of ISSM on soil organic C(SOC) characteristics remain unknown. In this study, the effects of current farmers' practice(FP), high-yielding practice(HY), which maximizes yields without considering costs, and ISSM on the content and chemical composition of SOC were studied in a 4-year(2009–2013) field plot experiment with maize(Zea mays L.) monoculture in an Alluvic Primosol in Northeast China. The ISSM resulted in higher soil total organic C(TOC), water-soluble organic C, easily-oxidizable organic C, particulate organic C, and humic acid C compared with HY and FP in the region. The SOC contents in aggregate size fractions generally followed a similar pattern to TOC. Compared with FP,HY decreased the mean weight diameter, geometric mean diameter, percentage of > 0.25-mm water-stable aggregates, and the stability ratio of water-stable aggregates, and increased the structure-deterioration rate and index of unstable aggregates. The opposite trend was observed between ISSM and HY. Solid-state ^(13)C nuclear magnetic resonance spectra of bulk soil showed that ISSM had higher O-alkyl C and aliphatic C/aromatic C ratio, but lower aromatic C, carbonyl C, and alkyl C/O-alkyl C and hydrophobic C/hydrophilic C ratios than HY and FP. 6Multivalent Aptamer-modified DNA Origami as Drug Delivery System for Targeted Cancer Therapy显示文摘In spite of great development in nanoparticle-based drug delivery systems(DDSs)for improved therapeutic efficacy,it remains challenging for effective delivery of chemotherapeutic drugs to targeted tumor cells.In this work,we report a triangle DNA origami as targeted DDS for cancer therapy.DNA origami shows excellent biocompatibility and stability in cell culture medium for 24 h.In addition,the DNA origami structures conjugated with multivalent aptamers enable for efficient delivery of anticancer drug doxorubicin(Dox)into targeted cancer cell due to their targeting function,reducing side effects associated with nonspecific distribution.Moreover,we also demonstrated that the multivalent aptamer-modified DNA origami loading Dox exhibits prominent therapeutic efficacy in vitro.Accordingly,this work provides a good paradigm for the development of DNA origami nanostructure-based targeted DDS for cancer therapy.CAO Mengyao SUN Yueyang XIAO Mingshu LI Li LIU Xiaohui JIN Hong PEI Hao 2020Chemical Research in Chinese Universities2020,36,2:1
7Dietary emodin alleviates lipopolysaccharide-induced intestinal mucosal barrier injury by regulating gut microbiota in piglets显示文摘This study was to determine the effects of dietary emodin(ED)on the intestinal mucosal barrier,nuclear factor kappa-B(NF-κB)pathways,and gut microbial flora in lipopolysaccharide(LPS)-induced piglets.Twenty-four weaned piglets were chosen and 4 treatments were created by randomly distributing piglets into CON,ED,LPS,and ED_LPS groups.Experiments were done in a 2×2 factorial arrangement and maintained for 21 d.Dietary treatment(a basal diet or 300 mg/kg ED)and immunological challenge(LPS or sterile saline)were 2 major factors.Intraperitoneal injections of LPS or sterilized saline were given to piglets on d 21.Six hours after the LPS challenge,all piglets were euthanized for sample collection and analysis.The results showed that piglets of the ED_LPS group had higher(P<0.05)villus height to crypt depth ratio(VCR),and lower(P<0.05)plasma D-lactate and diamine oxidase(DAO)than the LPS group.Furthermore,ED inhibited(P<0.05)the decrease of glutathione peroxidase(GSH-Px)and catalase(CAT)activities and increase of malonaldehyde level(P<0.05)in jejunal mucosa induced by LPS.The mRNA levels of pro-inflammatory cytokine genes(IL-6,IL-1β,and TNF-a)were significantly reduced(P<0.05),and the mRNA levels of antioxidant enzyme genes(GPX-1,SOD2 and CAT),as well as protein and mRNA levels of tight junction proteins(occludin,claudin-1,and ZO-1),were also significantly increased(P<0.05)by ED addition in LPS-induced piglets.Meanwhile,ED supplementation significantly decreased the LPS-induced protein levels of cyclooxygenase-2 and phosphorylation levels of NF-κB p65and IκBa in jejunal mucosa.Emodin had a significant effect on the composition of gut microbial flora at various taxonomic positions as indicated by 16S RNA sequencing.The acetic acid,isobutyric acid,valeric acid,and isovaleric acid concentrations in the cecum were also increased by ED addition in pigs(P<0.05).Furthermore,the correlation analysis revealed that some intestinal microbiota had a potential relationship with jejunal VCR,plasma D-lactate and DAO,jejunal mucosa GSH-Px and CAT activity,and cecal short-chain fatty acid concentration.These data suggest that ED is effective in alleviating LPSinduced intestinal mucosal barrier injury by modulating gut microbiota in piglets.Wenjuan Xun Mengyao Ji Zhonghua Ma Tanjie Deng Wen Yang Guanyu Hou Liguang Shi Ting Cao 2023Animal Nutrition2023,,3:0
8A NIR fluorescent probe for imaging thiophenol in the living system and revealing thiophenol-induced oxidative stress显示文摘Thiophenol(PhSH)is an important raw material for organic synthesis,while its high toxicity to organisms makes it an environmental pollutant.Therefore,it is crucial to accurately detect PhSH and explore its metabolic process in the living system.Herein,a near-infrared(NIR)fluorescent probe TEM-FB was developed for sensing PhSH with a turn-on fluorescent signal at 719nm and a large Stokes shift(198 nm)based on generating the intramolecular charge transfer(ICT)process.TEM-FB shows high specificity and significant sensitivity towards PhSH(detection limit:10 nmol/L)via the aromatic nucleophilic substitution mechanism.Furthermore,it was successfully applied to image PhSH in multiple cell lines and in zebrafish.Notably,we revealed the oxidative stress process caused by PhSH and demonstrated that the hydrogen peroxide(H_(2)O_(2))in cells would alleviate the poisonousness from exogenous PhSH for the first time.This work provides a promising bioimaging tool for monitoring PhSH in living systems and visualizing the process of oxidative stress induced by PhSH.Zesi Wang Jiao Li Jiao Chen Zifeng Cao Hui Li Yaopeng Cao Quanquan Li Mengyao She Ping Liu Shengyong Zhang Jianli Li 2023Chinese Chemical Letters2023,34,8:0
9Impacts of different surface features on soil detachment in the subtropical region显示文摘Near-surface features have a great influence on runoff and detachment processes by overland flow,but the contributions are still unclear on steep slopes with yellow soil in subtropical humid regions.Field scouring experiments were conducted to investigate how near-surface features affect hydraulic parameters and detachment rate by overland flow.Five treatments and a baseline(disturbed rootless bare cropland)were designed to identify the contributions from stem-leaves,litter,biological soil crusts(BSCs),root systems and non-disturbance,respectively.The results showed that(1)the values of velocity,Reynolds number and Froude number for vegetated slopes were significantly lower than that of baseline,and stem-leaves made a greater contribution(average of 47.30%)to reducing kinetic energy than the other features;(2)the total contribution rate of grassland was 99.38%,and of this total,1.19,1.44,2.49,49.79 and 44.47%reductions were attributed to the stem-leaves,litter,BSCs,root systems and non-disturbance,respectively.Root system and non-disturbance dominated the detachment rate reduction;(3)with increasing flow rate or slope gradient,the total contribution rate remained between 98%and 100%;and(4)for each treatment,the relationship between detachment rate and hydraulic parameters remained constant,and the stream power was the best predictor to detachment rate.The study results are helpful in evaluating the effects of near-surface features on erosion control and providing reference for government decision-makers to choose appropriate soil conservation and management practices.Qianhong Ma Keli Zhang Zihao Cao Zhicheng Yang Mengyao Wei Zaike Gu 2021International Soil and Water Conservation Research2021,9,4:0

11Effects of two vectors on the expression of the NbNAC1 transcription factor and preparation of its polyclonal antibody显示文摘The NAC(NAM,ATAF,and CUC)superfamily is one of the largest plant-specific families containing transcription factors.An increasing number of studies suggest that NAC1 is involved in plants response to different biotic and abiotic stimulis.Nicotiana benthamiana is a widely used system for evaluating plant-pathogen interactions.In order to study the biochemical function of NbNAC1,NbNAC1 protein and antibody are essential.Therefore,we focused on developing a prokaryotic expression system for producing the Nicotiana benthamiana NbNAC1 protein of in Escherichia coli and the preparation of its polyclonal antibody.Firstly,we constructed two different molecular weight prokaryotic expression vectors:pGE vector with GST tag(pGEX4T-1–NbNAC1)and pET expression vector with His tag(pET28a-NbNAC1).The NbNAC1 protein can be successfully expressed in both vectors.The His-tagged NbNAC1 proteins are insoluble,while the GST-tagged NbNAC1 proteins are partially soluble.We then successfully purified and enriched both proteins.The His-tagged NbNAC1 was chosen to immunize rabbits owing to an unknown protein accompanying the GST-tagged NbNAC1.The anti-NbNAC1 polyclonal antibody had good specificity and could be used in subsequent protein-related studies.FENG ZHU QINQIN ZHANG YANGKAI ZHOU QIPING ZHANG MENGYAO CAO ZHAOLIN JI 2022BIOCELL2022,46,9:0
12Detecting gravitational wave with an interferometric seismometer array on lunar nearside显示文摘Our Moon is by far the easiest natural, extraterrestrial celestial object for us to reach, and many space agencies, in particular the China National Space Administration, have publicized plans for manned missions there. Such high-profile projects must yield commensurate scientific outputs to justify their cost to the taxpayers. An important criterion for the selection of candidate experiments is the extent to which the Moon plays an integral and irreplaceable role, as opposed to offering only marginal advantages over Earth-orbit, space-based alternatives. In this study, we propose an experiment that excels in this regard, which considers the Moon as part of the experimental apparatus. Specifically, the Moon acts as a Weber bar-style resonant mass that responds to deci-Hertz gravitational waves(GWs). GW-excited lunar vibrations can be picked up by an array of(prefabricated on Earth and easy to transport to the Moon) small laser interferometric seismometers yielding GW detection. Such a setup would plug a gap in the GW frequency range already covered by the existing or underdevelopment detectors, and it can inform us on important astrophysics topics, such as the progenitors of Type Ia supernovae(SNe Ia) or intermediate mass black holes.
