| 1 | Neuroprotective effect of pretreatment with ganoderma lucidum in cerebral ischemia/reperfusion injury in rat hippocampus显示文摘Ganoderma lucidum is a traditional Chinese medicine,which has been shown to have both anti-oxidative and anti-inflammatory effects,and noticeably decreases both the infarct area and neuronal apoptosis of the ischemic cortex.This study aimed to investigate the protective effects and mechanisms of pretreatment with ganoderma lucidum(by intragastric administration)in cerebral ischemia/reperfusion injury in rats.Our results showed that pretreatment with ganoder-ma lucidum for 3 and 7 days reduced neuronal loss in the hippocampus,diminished the content of malondialdehyde in the hippocampus and serum,decreased the levels of tumor necrosis fac-tor-αand interleukin-8 in the hippocampus,and increased the activity of superoxide dismutase in the hippocampus and serum.These results suggest that pretreatment with ganoderma lucidum was protective against cerebral ischemia/reperfusion injury through its anti-oxidative and an-ti-inflammatory actions. | Wangxin Zhang Qiuling Zhang Wen Deng Yalu Li Guoqing Xing Xianjun Shi Yifeng Du | 2014 | Neural Regeneration Research2014,9,15: | 6 |
| 2 | Machine learning aided design of perovskite oxide materials for photocatalytic water splitting显示文摘Suffering from the inefficient traditional trial-and-error methods and the huge searching space filled by millions of candidates, discovering new perovskite visible photocatalysts with higher hydrogen production rate(RH_(2)) still remains a challenge in the field of photocatalytic water splitting(PWS). Herein, we established structural-property models targeted to RH_(2) and the proper bandgap(Eg) via machine learning(ML) technology to accelerate the discovery of efficient perovskite photocatalysts for PWS. The Pearson correlation coefficients(R) of leave-one-out cross validation(LOOCV) were adopted to compare the performances of different algorithms including gradient boosting regression(GBR), support vector regression(SVR), backpropagation artificial neural network(BPANN), and random forest(RF). It was found that the BPANN model showed the highest R values from LOOCV and testing data of 0.9897 and 0.9740 for RH_(2),while the GBR model had the best values of 0.9290 and 0.9207 for Eg. Furtherly, 14 potential PWS perovskite candidates were screened out from 30,000 ABO3-type perovskite structures under the criteria of structural stability, Eg, conduction band energy, valence band energy and RH_(2). The average RH_(2) of these14 perovskites is 6.4% higher than the highest value in the training data set. | 3 | Protocatechuic aldehyde protects cardiomycoytes against ischemic injury via regulation of nuclear pyruvate kinase M2显示文摘Rescuing cells from stress damage emerges a potential therapeutic strategy to combat myocardial infarction.Protocatechuic aldehyde(PCA)is a major phenolic acid in Chinese herb Danshen(Salvia miltiorrhiza root).This study investigated whether PCA regulated nuclear pyruvate kinase isoform M2(PKM2)function to protect cardiomyocytes.In rats subjected to isoprenaline,PCA attenuated heart injury and protected cardiomyocytes from apoptosis.Through DARTS and CETSA assays,we identified that PCA bound and promoted PKM2 nuclear translocation in cardiomyocytes exposed to oxygen/glucose deprivation(OGD).In the nucleus,PCA increased the binding of PKM2 to β-Catenin via preserving PKM2 acetylation,and the complex,in cooperation with T-cell factor 4(TCF4),was required for transcriptional induction of genes encoding anti-apoptotic proteins,contributing to rescuing cardiomyocyte survival.In addition,PCA ameliorated mitochondrial dysfunction and prevented mitochondrial apoptosis dependent on PKM2.Consistently,PCA increased the binding of PKM2 to β-Catenin,improved heart contractive function,normalized heart structure and attenuated oxidative damage in mice subjected to artery ligation,but the protective effects were lost in Pkm2-deficient heart.Together,we showed that PCA regulated nuclear PKM2 function to rescue cardiomyocyte survival via β-Catenin/TCF4 signaling cascade,suggesting the potential of pharmacological intervention of PKM2 shuttle to protect the heart. | Xunxun Wu Lian Liu Qiuling Zheng Haiping Hao Hui Ye Ping Li Hua Yang | 2021 | Acta Pharmaceutica Sinica B2021,11,11: | 4 |

| 6 | Formation mechanism of yolk-shell LaMnO3 microspheres prepared by P123-template and oxidation of NO显示文摘The yolk-shell LaMnO3 perovskite microspheres were fabricated by a novel,simple and mild soft template approach.A series of template-P123 concentrations(0-6.12 mmol·L^-1)were employed to optimize the most complete spheres.When the concentration of P123 is 3.0 mmol·L^-1,the obtained yolk-shell microspheres with a diameter of 200-700 nm were constructed by nanoparticles.The possible formation mechanism of the yolk-shell microspheres was revealed step by step via XRD,SEM,TEM,EDS and HRTEM.Molecules of P123 were suitably mixed with solvents for double shelled vesicles through self-assembly,which interacted with metal complexes to form P123-metal vesicles.After the removal of P123 and citric acid by calcination at 700 ℃,the yolk-shell LaMnO3 microspheres with through-channels were obtained.Through-channels on the surface were due to citric acid and the solid core was attributed to the shrink of inner vesicles.Prepared yolk-shell microsphere samples possessed a larger surface area and a higher maximum NO conversion value of 78% at 314 ℃ for NO oxidation,compared with samples without the yolk-shell structure. | Lihui WU Qiuling JIANG Li WANG Ying WANG Mengxue WANG | 2019 | Frontiers of Materials Science2019,13,1: | 3 |

The product has an hourglass or half-hourglass morphology with a narrow size distribution of 150-200 nm. Major endothermal peak of decomposition was found at 270°C by differential thermal analysis. X-ray diffraction identified the product as bayerite. | 9 | GM130 regulates pulmonary surfactant protein secretion in alveolar typeⅡcells显示文摘Pulmonary surfactant is a lipid-protein complex secreted by alveolar typeⅡepithelial cells and is essential for the maintenance of the delicate structure of mammalian alveoli to promote efficient gas exchange across the air-liquid barrier.The Golgi apparatus plays an important role in pulmonary surfactant modification and secretory trafficking.However,the physiological function of the Golgi apparatus in the transport of pulmonary surfactants is unclear.In the present study,deletion of GM130,which encodes for a matrix protein of the cis-Golgi cisternae,was shown to induce the disruption of the Golgi structure leading to impaired secretion of lung surfactant proteins and lipids.Specifically,the results of in vitro and in vivo analysis indicated that the loss of GM130 resulted in trapping of Sftpa in the endoplasmic reticulum,Sftpb and Sftpc accumulation in the Golgi apparatus,and an increase in the compensatory secretion of Sftpd.Moreover,global and epithelial-specific GM130 knockout in mice resulted in an enlargement of alveolar airspace and an increase in alveolar epithelial autophagy;however,surfactant repletion partially rescued the enlarged airspace defects in GM130-deficient mice.Therefore,our results demonstrate that GM130 and the mammalian Golgi apparatus play a critical role in the control of surfactant protein secretion in pulmonary epithelial cells. | Qianqian Pang Chunyi Liu Yulong Qiao Jian Zhao Sin Man Lam Mei Mei Guanghou Shui Shilai Bao Qiuling Li | 2022 | Science China(Life Sciences)2022,65,1: | 2 |
| 10 | Transgene expression and differentiation of baculovirus-transduced adipose-derived stem cells from dystrophin-utrophin double knock-out mouse显示文摘In this study, recombinant baculovirus carrying the microdystrophin and β-catenin genes was used to infect adipose-derived stem cells from a dystrophin-utrophin double knock-out mouse. Results showed that, after baculovirus transgene infection, microdystrophin and β-catenin genes were effectively expressed in adipose-derived stem cells from the dystrophin-utrophin double knock-out mouse. Furthermore, this transgenic expression promoted adipose-derived stem cell differentiation into muscle cells, but inhibited adipogenic differentiation. In addition, protein expression related to the microdystrophin and Wnt/β-catenin signaling pathway was upregulated. | 11 | Clinical Benefit of Epidermal Growth Factor Receptor-Tyrosine Kinase Inhibitors Plus Radiotherapy for Epidermal Growth Factor Receptor-Mutated Metastatic Non-Small Cell Lung Cancer: A Retrospective Analysis on Real World Data显示文摘Objective: To investigate the benefit of epidermal growth factor receptor( EGFR) tyrosine kinase inhibitors( TKIs)with radiotherapy in patients with EGFR mutation-positive metastatic non-small cell lung cancer( NSCLC),compared with TKIs alone.Methods: A total of 103 patients with stage Ⅳ EGFR-mutated NSCLC treated from February 2015 to May 2017 at Sichuan Cancer Hospital were analyzed retrospectively. Fifty patients were treated with EGFR-TKIs( gefitinib or erlotinib) plus radiotherapy( the TKI +RT group) and 53 patients received EGFR-TKIs alone( the TKI group). Tumor response,survival and toxicities were compared between the two groups. Results: Median follow-up time was 11. 7 months( 2. 8-36. 3 months). The overall response rate( ORR) and disease control rate( DCR) in the TKI + RT group vs the TKI group were 62% vs 37. 7%( P = 0. 014) and 88% vs 75. 5%( P =0. 101),respectively. The median progression-free survival( PFS) and median overall survival( OS) in the TKI + RT group were superior to those of the TKI group( 18. 87 months vs 12. 80 months,P = 0. 035 and 23. 10 months vs 18. 30 months,P = 0. 011). OS rates in the TKI + RT group and the TKI group were 56. 0% vs 35. 8% at year 1( P = 0. 04) and 16. 0% vs 3. 8% at year 2( P =0. 036). | 13 | The combined effects of lysozyme and ascorbic acid on microstructure and properties of zein-based films显示文摘Edible zein-based films containing lysozyme(LY) and ascorbic acid(AA) were developed in the presence of polyethylene glycol 400(PEG 400), the combined effects of LY and AA on the microstructure, mechanical properties and release properties of developed zein films were investigated in detail. The results of microstructure characterization indicated that zein-based films became compact and smooth, and LY aggregates were well distributed in the zein matrix because of the simultaneous addition of LY and AA. The results of mechanical tests showed that because of the synergistic effects of LY and AA on zein film, elongation at break of zein-based film could be up to 138%, which was 34.5 times higher than that of zein control film. LY release tests showed that when the concentration of AA was less than 3.1 mg·cm^(-2), the release rate of LY significantly decreased by 33.7%, and the total release increased by 80.6%. While the release profiles of AA showed that the release rate and total release of AA from the films containing LY increased by approximately 68.9% and 61.7% than the films without LY. | 20 | Dihydrotanshinone I preconditions myocardium against ischemic injury via PKM2 glutathionylation sensitive to ROS显示文摘Ischemic preconditioning(IPC)is a potential intervention known to protect the heart against ischemia/reperfusion injury,but its role in the no-reflow phenomenon that follows reperfusion is unclear.Dihydrotanshinone I(DT)is a natural compound and this study illustrates its role in cardiac ischemic injury from the aspect of IPC.Pretreatment with DT induced modest ROS production and protected cardiomyocytes against oxygen and glucose deprivation(OGD),but the protection was prevented by a ROS scavenger.In addition,DT administration protected the heart against isoprenaline challenge.Mechanistically,PKM2 reacted to transient ROS via oxidization at Cys423/Cys424,leading to glutathionylation and nuclear translocation in dimer form.In the nucleus,PKM2 served as a co-factor to promote HIF-1a-dependent gene induction,contributing to adaptive responses.In mice subjected to permanent coronary ligation,cardiac-specific knockdown of Pkm2 blocked DT-mediated preconditioning protection,which was rescued by overexpression of wild-type Pkm2,rather than Cys423/424-mutated Pkm2.In conclusion,PKM2 is sensitive to oxidation,and subsequent glutathionylation promotes its nuclear translocation.Although IPC has been viewed as a protective means against reperfusion injury,our study reveals its potential role in protection of the heart from no-reflow ischemia. | Xunxun Wu Lian Liu Qiuling Zheng Hui Ye Hua Yang Haiping Hao Ping Li | 2023 | Acta Pharmaceutica Sinica B2023,13,1: | 1 |
