Robust MVDR beamforming based on covariance matrix reconstruction显示文摘The minimum variance distortionless response(MVDR) beamformer has better resolution and much better interference rejection capability than the data-independent beamformers. However, the former is more sensitive to errors, such as the steering vector errors caused by direction of arrival mismatch, imprecise array calibration or any other possible factors, especially in the case of high signal-to-noise ratio(SNR).A new robust MVDR beamformer against the general steering vector errors is proposed in this paper. This method is based on the reconstruction of the covariance matrix which aims to reduce the power of the signal of interest(SOI) in the covariance matrix. The eigenanalysis is used in the process of removing the SOI component and modifying the residual covariance matrix.Simulation results show that the proposed method has excellent signal-to-interference-plus-noise ratio(SINR) performance under the mismatch condition.MU PengCheng LI Dan YIN QinYe GUO Wei 2013Science China(Information Sciences)2013,56,4:11
Interferon-c and interleukin-4 detected in serum and saliva from patients with oral lichen planus显示文摘Our previous salivary study had demonstrated an apparent T helper 2(Th2)-predominance in saliva of oral lichen planus(OLP) patients and suggested a potential of salivary interleukin-4(IL-4) as a biomarker for monitoring disease severity. To further determine the consistency of Th1/Th2 bias of OLP, this study investigated the expression profile of interferon-c(IFN-c) and IL-4 in serum and the relationship of the serum levels of these cytokines with their saliva partners. Sixty ethnic Chinese patients with OLP(40 of the erythematous/ulcerative form and 20 of the reticular form) were recruited for this study, with 40 age–sex-matched healthy volunteers as control group. IFN-c and IL-4 levels in serum and paired saliva samples were screened by enzyme-linked immunosorbent assay. OLP patient showed a low-level IFN-c but high-level IL-4 expression profile in both serum and saliva, with a lower IFN-c/IL-4 ratio. Serum IL-4 level in the erythematous/ulcerative group was significantly higher than that in the reticular group. Serum levels of IFN-c and IL-4 were significantly and positively correlated with their saliva partners. These results provided more evidence for Th2 cytokinepredominant immune imbalance in OLP, as well as the potential of IL-4 as the biomarker for monitoring severity of OLP.Wen-Zhao Liu Ming-Jing He Long Long Dong-Liang Mu Ming-Shu Xu Xue Xing Xin Zeng Ga Liao Hong-Xia Dan Qian-Ming Chen 2014International Journal of Oral Science2014,6,1:10
A multi-channel collagen scaffold loaded with neural stem cells for the repair of spinal cord injury显示文摘Collagen scaffolds possess a three-dimensional porous structure that provides sufficient space for cell growth and proliferation, the passage of nutrients and oxygen, and the discharge of metabolites. In this study,a porous collagen scaffold with axially-aligned luminal conduits was prepared. In vitro biocompatibility analysis of the collagen scaffold revealed that it enhances the activity of neural stem cells and promotes cell extension, without affecting cell differentiation. The collagen scaffold loaded with neural stem cells improved the hindlimb motor function in the rat model of T8 complete transection and promoted nerve regeneration. The collagen scaffold was completely degraded in vivo within 5 weeks of implantation, exhibiting good biodegradability.Rectal temperature,C-reactive protein expression and CD68 staining demonstrated that rats with spinal cord injury that underwent implantation of the collagen scaffold had no notable inflammatory reaction. These findings suggest that this novel collagen scaffold is a good carrier for neural stem cell transplantation, thereby enhancing spinal cord repair following injury. This study was approved by the Animal Ethics Committee of Nanjing Drum Tower Hospital(the Affiliated Hospital of Nanjing University Medical School),China(approval No.2019AE02005)on June 15,2019.Shuo Liu Yuan-Yuan Xie Liu-Di Wang Chen-Xu Tai Dong Chen Dan Mu Yan-Yan Cui Bin Wang 2021Neural Regeneration Research2021,16,11:6
Comparison of Histological, Microbiological, and Molecular Methods in Diagnosis of Patients with TBLN Having Different Anti-TB Treatment Background显示文摘Objective The influence of anti-tuberculosis(TB) treatment history on tuberculous lymphadenitis(TBLN) diagnosis is unclear. Therefore, this study aims to evaluate the diagnostic methods, including histology, microbiology, and molecular tests, used for TBLN. Methods In this study, suspected patients with TBLN and having different anti-TB treatment background were enrolled. All the samples were tested simultaneously by histology, Ziehl-Neelsen(ZN) staining, mycobacterial culture(culture), Xpert MTB/RIF(xpert), real-time PCR, and high-resolution melting curve PCR(HRM). Thereafter, the performance of these methods on samples with different anti-TB treatment background was assessed. Results In our study, 89 patients were prospectively included 82 patients with TBLN and 7 with other diseases. The overall sensitivities of Xpert, real-time PCR, histology, ZN staining, and culture were 86.6%, 69.5%, 58.5%, 43.9%, and 22.0%, respectively. The anti-TB treatment history revealed dramatic influences on the sensitivity of culture(P < 0.0001). In fact, the treatment that lasted over 3 months also influenced the sensitivity of Xpert(P < 0.05). However, the treatment history did not affect the performance of remaining tests(P > 0.05). For rifampicin drug susceptibility test(DST), the anti-TB treatment showed only significant influence on the success rate of culture DST(P = 0.001), but not on those of Xpert and HRM tests(P > 0.05). Conclusion Other tests as well as culture should be considered for patients with TBLN having retreatment history or over 1-month treatment to avoid false negative results.CHE Nan Ying HUANG Shao Jun MA Yan HAN Yi LIU Zi Chen ZHANG Chen MU Jing ZHAO Dan QU Yang ZHANG Hai Qing LIU Zhi Dong XU Shao Fa 2017Biomedical and Environmental Sciences2017,30,6:6
Expression Patterns of Cytochrome P450 Aromatase Genes During Ovary Development and Their Responses to Temperature Stress in Female Yellow Catfish (Pelteobagrus fulvidraco)显示文摘Cytochrome P450 aromatase(P450arom)plays a pivotal role in ovary development. In this study,we used semi-quanti-tative reverse transcription PCR(RT-PCR)to analyze spatiotemporal expressions of two P450arom genes(CYP19A and CYP19B)and their responses to temperature stress in female yellow catfish(Pelteobagrus fulvidraco).Tissue distribution pattern of CYP19 showed that CYP19B was abundantly expressed in fish brain and ovary(brain > ovary),but weakly in intestines, whereas CYP19A was exclusively expressed in ovary.Semi-quantitative RT-PCR analyses showed high transcript abundance of both CYP19A and CYP19B in the ovarian reproductive cycle, corresponding with serum estradiol-17β(E2)levels.Increases in aromatases, serum E2 and testosterone(T)levels in fish exposed to higher temperature indicated stimulation of ovarian maturation and recrudescence by heat stress in stages Ⅱ and Ⅴ during the ovarian cycle, whereas associated decreases in stage Ⅲ suggested vitellogenesis inhibition by heat stress.Gene expression of CYP19 was closely related to levels of serum E2.Results demonstrated CYP19 played a crucial role in the reproductive cycle of female yellow catfish.Different temperature stress affected CYP19 gene expression in the fish ovarian re-productive cycle.Associated P450arom genes could be useful for studying physiological aspects of yellow catfish.LIU Miao WEN Haishen HE Feng LI Jifang SHI Dan HU Jian ZHANG Yuanqing MA Ruiqin MU Weijie QI Baoxia 2011Journal of Ocean University of China2011,10,4:5
Poly-PR in C9ORF72-Related Amyotrophic Lateral Sclerosis/Frontotemporal Dementia Causes Neurotoxicity by Clathrin-Dependent Endocytosis显示文摘GGGGCC repeat expansions in the C9 ORF72 gene are the most common cause of amyotrophic lateral sclerosis and frontotemporal dementia(c9 ALS/FTD). It has been reported that hexanucleotide repeat expansions in C9 ORF72 produce five dipeptide repeat(DPR) proteins by an unconventional repeat-associated non-ATG(RAN)translation. Within the five DPR proteins, poly-PR and poly-GR that contain arginine are more toxic than the other DPRs(poly-GA, poly-GP, and poly-PA). Here, we demonstrated that poly-PR peptides transferred into cells by endocytosis in a clathrin-dependent manner, leading to endoplasmic reticulum stress and cell death. In SH-SY5 Y cells and primary cortical neurons, poly-PR activated JUN amino-terminal kinase(JNK) and increased the levels of p53 and Bax. The uptake of poly-PR peptides by cells was significantly inhibited by knockdown of clathrin or by chlorpromazine, an inhibitor that blocks clathrin-mediated endocytosis. Inhibition of clathrin-dependent endocytosis by chlorpromazine significantly blocked the transfer of poly-PR peptides into cells, and attenuated poly-PRinduced JNK activation and cell death. Our data revealed that the uptake of poly-PR undergoes clathrin-dependentendocytosis and blockade of this process prevents the toxic effects of synthetic poly-PR peptides.Rui Wang Xingyun Xu Zongbing Hao Shun Zhang Dan Wu Hongyang Sun Chenchen Mu Haigang Ren Guanghui Wang 2019Neuroscience Bulletin2019,35,5:4
Dynamic changes in the systemic immune responses of spinal cord injury model mice显示文摘Intraspinal inflammatory and immune responses are considered to play central roles in the pathological development of spinal cord injury. This study aimed to decipher the dynamics of systemic immune responses, initiated by spinal cord injury. The spinal cord in mice was completely transected at T8.Changes in the in vivo inflammatory response, between the acute and subacute stages, were observed.A rapid decrease in C-reactive protein levels, circulating leukocytes and lymphocytes,spleen-derived CD4~+interferon-γ+T-helper cells, and inflammatory cytokines, and a marked increase in neutrophils, monocytes, and CD4~+CD25~+FOXP3~+regulatory T-cells were observed during the acute phase. These systemic immune alterations were gradually restored to basal levels during the sub-acute phase.During the acute phase of spinal cord injury, systemic immune cells and factors showed significant inhibition;however, this inhibition was transient, and the indicators of these serious disorders gradually returned to baseline levels during the subacute phase. All experiments were performed in accordance with the institutional animal care guidelines, approved by the Institutional Animal Care and Use Committee of Experimental Animal Center of Drum Tower Hospital,China(approval No.2019 AE01040)on June 25,2019.Tian-Yun Gao Fei-Fei Huang Yuan-Yuan Xie Wen-Qing Wang Liu-Di Wang Dan Mu Yi Cui Bin Wang 2021Neural Regeneration Research2021,16,2:3
Numerical study of M_2 internal tide generation and propagation in the Luzon Strait显示文摘Based on the z-coordinate ocean model HAMSOM,we introduced the internal-tide viscosity term and applied the model to numerically investigate the M2 internal tide generation and propagation in the Luzon Strait (LS).The results show that (1) in the upper 250 m depth,at the thermocline, the maximum amplitude of the generated internal tides in the LS can reach 40 m;(2) the major internal tides are generated to the northwest of Itbayat Island, the southwest of Batan Island and the northwest of the Babuyan Islands;(3) during the propagation the baroclinic energy scattering and reflection is obvious, which exists under the effect of the specific topography in the South China Sea (SCS);(4) the westward-propagating internal tides are divided into two branches entering the SCS.While passing through 118 E,the major branch is divided into two branches again. The strongest internal tides in the LS are generated to the northwest of Itbayat Island and propagate northeastward to the Pacific. However,to the east of 122 E,most of the internal tides propagate southeastward to the Pacific as a beam.LI Huan SONG Dan CHEN Xueen QIAN Hongbao MU Lin SONG Jun 2011Acta Oceanologica Sinica2011,30,5:3

Structure, retrogradation and digestibility of waxy corn starch modified by a GtfC enzyme from Geobacillus sp.12AMOR1显示文摘To lower the retrogradation and digestibility of waxy corn starch for different food applications,a novel thermostable GtfC type 4,6-α-glucanotransferase without N-and C-terminals(GsGtfC)from Geobacillus sp.12AMOR1 was used.Waxy corn starch of 50 g/L was incubated with GsGtfC of 40-100 U/g substrate at 65℃and pH 5.5 for 1 h.Its molecular weight, iodine affinity,XRD crystallinity, and FTIR ratio of heights of bands at 1047 and 1022 cm^(-1) decreased, but ratio of DP<6 to DP≥25 branches and degree of branching increased.GsGtfC cleavedα-1,4-glycosidic bonds and inducedα-1,6-branching points to produce reuteran-likes polymers, which is different from Exiguobacterium sibiricum GtfC enzyme cleavingα-1,4-glycosidic bonds and synthesizing consecutiveα-1,6-glycosidic bonds to produce isomalto/malto-oligosaccharides.GsGtfC modified waxy corn starch had significantly lower DSC retrogradation enthalpies during the storage at 4℃for 3-14 days and significantly lower released glucose during the incubation with mammalian mucosalα-glucosidase at 37℃for 10-360 min.GsGtfC at 100 U/g substrate increased slowly digestible portion from 11.07%to 24.11%.Xiaolei Li Yong Wang Siyu Mu Xiangqing Ji Cheng Zeng Dingkuan Yang Liyuan Dai Cuicui Duan Dan Li 2022Food Bioscience2022,46,2:2
Tracing the impact of stack configuration on interface resistances in reverse electrodialysis by in situ electrochemical impedance spectroscopy显示文摘Reverse electrodialysis(RED)is an emerging membrane-based technology for the production of renewable energy from mixing waters with different salinities.Herein, the impact of the stack configuration on the Ohmic and non-Ohmic resistances as well as the performance of RED were systematically studied by using in situ electrochemical impedance spectroscopy(EIS).Three different parameters(membrane type, number of cell pairs and spacer design)were controlled. The Ohmic and non-Ohmic resistances were evaluated for RED stacks equipped with two types of commercial membranes(Type I and Type II)supplied by Fujifilm Manufacturing Europe B.V:Type I Fuji membranes displayed higher Ohmic and non-Ohmic resistances than Type II membranes, which was mainly attributed to the difference in fixed charge density. The output power of the stack was observed to decrease with the increasing number of cell pairs mainly due to the increase in ionic shortcut currents. With the reduction in spacer thickness from 750 to 200μm,the permselectivity of membranes in the stack decreased from 0.86 to 0.79 whereas the energy efificiency losses increased from 31%to 49%.Overall,the output of the present study provides a basis for understanding the impact of stack design on internal losses during the scaling-up of RED.Wenjuan Zhang Bo Han Ramato Ashu Tufa Chuyang Tang Xunuo Liu Ge Zhang Jing Chang Rui Zhang Rong Mu Caihong Liu Dan Song Junjing Li Jun Ma Yufeng Zhang 2022Frontiers of Environmental Science & Engineering2022,16,4:1

Analysis of cyclin E co-expression genes reveals nuclear transcription factor Y subunit alpha is an oncogene in gastric cancer显示文摘Objective: To explore genes potentially co-expressed with cyclin E in gastric cancer and discover possible targets for gastric cancer treatment. Methods: The Cancer Genome Atlas (TCGA) stomach adenocarcinoma sequencing data were used to predict genes co-expressed with cyclin E. Co-expression genes predicted by cBioPortal online analysis with Pearson correlation coefficient ≥0.4 were analyzed by gene ontology (GO) enrichment annotation using the PANTHER online platform (Ver. 7). Interactions between proteins encoded by these genes were analyzed using the STRING online platform (Ver. 10.5) and Cytoscape software (Ver. 3.5.1). Genes displaying a high degree of connection were analyzed by transcription factor enrichment prediction using FunRich software (Ver. 3). The significant transcription factor and cyclin E expression levels and their impact on gastric cancer progression were analyzed by Western blotting and Kaplan-Meier survival curve analysis. Results: After filtering the co-expression gene prediction results, 78 predicted genes that included 73 protein coding genes and 5 non-coding genes with Pearson correlation coefficient ≥0.4 were selected. The expressions of the genes were considered to be correlated with cyclin E expression. Among the 78 genes co-expressed with cyclin E, 19 genes at the central of the regulatory network associated with cyclin E were discovered. Nuclear transcription factor Y subunit alpha (NF-YA) was identified as a significant transcription factor associated with cyclin E co-expressing genes. Analysis of specimen donors’ clinical records revealed that high expression of NF-YA tended to be associated with increased cyclin E expression. The expression of both was associated with progression of gastric cancer. Western blotting results showed that compared with normal tissues, NF-YA and cyclin E were highly expressed in tumor tissues (P < 0.001). Survival curve analysis clearly demonstrated relatively poor overall survival of gastric cancer patients with high cyclin E or high NF-YA expression level, compared to patients with low cyclin E or NF-YA expression (P < 0.05). Conclusions: NF-YA may promote gastric cancer progression by increasing the transcription of cyclin E and other cell cycle regulatory genes. NF-YA might be a potential therapeutically useful prognostic factor for gastric cancer.Liang-Yu Bie Dan Li Yu Mu Sheng Wang Bei-Bei Chen Hui-Fang Lyu Li-Li Han Cai-Yun Nie Chang-Cheng Yang Lin Wang Chuan-Chuan Ren Wei-Jie Zhang Ping Guo Feng Shi Qing-Xia Fan Liu-Xing Wang Xiao-Bing Chen Su-Xia Luo 2019Chronic Diseases and Translational Medicine2019,5,1:1
