2Effects of warming and clipping on plant and soil properties of an alpine meadow in the Qinghai-Tibetan Plateau, China显示文摘Climate warming and livestock grazing are known to have great influences on alpine ecosystems like those of the Qinghai-Tibetan Plateau(QTP) in China. However, it is lacking of studies on the effects of warming and grazing on plant and soil properties in these alpine ecosystems. In this study, we reported the related research from manipulative experiment in 2010–2012 in the QTP. The aim of this study was to investigate the individual and combined effects of warming and clipping on plant and soil properties in the alpine meadow ecosystem. Infrared radiators were used to simulate climate warming starting in July 2010, while clipping was performed once in October 2011 to simulate the local livestock grazing. The experiment was designed as a randomized block consisting of five replications and four treatments: control(CK), warming(W), clipping(C) and warming+clipping combination(WC). The plant and soil properties were investigated in the growing season of the alpine meadow in 2012. The results showed that W and WC treatments significantly decreased relative humidity at 20-cm height above ground as well as significantly increases air temperature at the same height, surface temperature, and soil temperature at the depth of 0–30 cm. However, the C treatment did not significantly decrease soil moisture and soil temperature at the depth of 0–60 cm. Relative to CK, vegetation height and species number increased significantly in W and WC treatment, respectively, while vegetation aboveground biomass decreased significantly in C treatment in the early growing season. However, vegetation cover, species diversity, belowground biomass and soil properties at the depth of 0–30 cm did not differ significantly in W, C and WC treatments. Soil moisture increased at the depth of 40–100 cm in W and WC treatments, while belowground biomass, soil activated carbon, organic carbon and total nitrogen increased in the 30–50 cm soil layer in W, C and WC treatments. Although the initial responses of plant and soil properties to experimental warming and clipping were slow and weak, the drought induced by the downward shift of soil moisture in the upper soil layers may induce plant belowground biomass to transfer to the deeper soil layers. This movement would modify the distributions of soil activated carbon, organic carbon and total nitrogen. 3Ursolic acid reduces hepatocellular apoptosis and alleviates alcohol-induced liver injury via irreversible inhibition of CASP3 in vivo显示文摘Alcoholic liver disease (ALD) is one of the pathogenic factors of chronic liver disease with the highest clinical morbidity worldwide. Ursolic acid (UA), a pentacyclic terpenoid carboxylic acid, has shown many health benefits including antioxidative, anti- inflammatory, anticancer, and hepatoprotective activities. We previously found that UA was metabolized in vivo into epoxy- modified UA containing an epoxy electrophilic group and had the potential to react with nucleophilic groups. In this study we prepared an alkynyl-modified UA (AM-UA) probe for tracing and capturing the target protein of UA from liver in mice, then investigated the mode by which UA bound to its target in vivo. By conducting proteome identification and bioinformatics analysis, we identified caspase-3 (CASP3) as the primary target protein of UA associated with liver protection. Molecule docking analysis showed that the epoxy group of the UA metabolite reacted with Cys-163 of CASP3, forming a covalent bond with CASP3. The binding mode of the UA metabolites (UA, CM-UA, and EM-UA) was verified by biochemical evaluation, demonstrating that the epoxy group produced by metabolism played an important role in the inhibition of CASP3. In alcohol-treated HepG2 cells, pretreatment with the UA metabolite (10 μM) irreversibly inhibited CASP3 activities, and subsequently decreased the cleavage of PARP and cell apoptosis. Finally, pre-administration of UA (20–80 mg· kg−1 per day, ig, for 1 week) dose-dependently alleviated alcohol-induced liver injury in mice mainly via the inhibition of CASP3. 8Key Techniques and Applications of Adaptive Growth Method for Stiffener Layout Design of Plates and Shells显示文摘The application of the adaptive growth method is limited because several key techniques during the design process need manual intervention of designers.Key techniques of the method including the ground structure construction and seed selection are studied,so as to make it possible to improve the effectiveness and applicability of the adaptive growth method in stiffener layout design optimization of plates and shells.Three schemes of ground structures,which are comprised by different shell elements and beam elements,are proposed.It is found that the main stiffener layouts resulted from different ground structures are almost the same,but the ground structure comprised by 8-nodes shell elements and both 3-nodes and 2-nodes beam elements can result in clearest stiffener layout,and has good adaptability and low computational cost.An automatic seed selection approach is proposed,which is based on such selection rules that the seeds should be positioned on where the structural strain energy is great for the minimum compliance problem,and satisfy the dispersancy requirement.The adaptive growth method with the suggested key techniques is integrated into an ANSYS-based program,which provides a design tool for the stiffener layout design optimization of plates and shells.Typical design examples,including plate and shell structures to achieve minimum compliance and maximum bulking stability are illustrated.In addition,as a practical mechanical structural design example,the stiffener layout of an inlet structure for a large-scale electrostatic precipitator is also demonstrated.The design results show that the adaptive growth method integrated with the suggested key techniques can effectively and flexibly deal with stiffener layout design problem for plates and shells with complex geometrical shape and loading conditions to achieve various design objectives,thus it provides a new solution method for engineering structural topology design optimization.DING Xiaohong JI Xuerong MA Man HOU Jianyun 2013Chinese Journal of Mechanical Engineering2013,26,6:2
9Clinical study on the related markers of blood coagulation in the patients with ANFH after SARS显示文摘The aim of this research was to investigate the blood coagulation function in the patients with avascular necrosis of the femoral head(ANFH)after severe acute respiratory syndrome(SARS).The expression of CD31,CD61,CD62p,CD63 and PAC-1 on platelet membrane was measured respectively by flowcytometry,and the plasma prothrombin time(PT),activated partial thromboplastin time(APTT),thrombin time(TT)and fibrinogen(Fbg)were mea-sured by blood clotting instrument in 26 patients with ANFH after SARS and in 17 healthy adults.The expression of CD31,CD61,CD 62p,CD63 and PAC-1 on platelet membrane in 26 patients was all lower than that in 17 healthy subjects(P<0.01).The levels of PT,APTT,TT and Fbg in 26 patients were all normal.There is no significant difference(P>0.05)in those markers between patients and 17 healthy adults.The blood may not be in hypercoagulable state in patients with ANFH after SARS.WU Lianhua GAO Chunjin WANG Guozhong YANG Lin HOU Xiaomin GE Huan XIA Chengqing QI Man 2007Frontiers of Medicine2007,1,4:2
10Pt nanocrystallines/TiO_2 with thickness-controlled carbon layers:Preparation and activities in CO oxidation显示文摘In this work, a series of Pt nanocrystallines(Pt NCs) supported on TiO_2 substrate with controlled thickness of carbon layers(C-Pt/TiO_2) were synthesized. Well-dispersed Pt NCs were facilely synthesized at room temperature by a photo-reduction process in lytropic liquid crystal(LCs). Surface tuning of the carbon layers on Pt/TiO_2 catalysts was achieved by varying the calcination atmospheres(in argon, air, and oxygen) and characterized by XPS and HRTEM. The influence of the coated carbon layers on the catalytic activity of catalysts is investigated by CO oxidation reaction which presented the following ranks: C-Pt/TiO_2-O_2> C-Pt/TiO_2-Air > C-Pt/TiO_2-Ar. It is found that the carbon layer coating can stabilize the Pt NCs and enable them anti-sintering at high temperature. 13Aspirin triggers ferroptosis in hepatocellular carcinoma cells through restricting NF-κB p65-activated SLC7A11 transcription显示文摘A number of studies have shown that aspirin,as commonly prescribed drug,prevents the development of hepatocellular carcinoma(HCC).Ferroptosis as a dynamic tumor suppressor plays a vital role in hepatocarcinogenesis.In this study we investigated whether aspirin affected ferroptosis in liver cancer cells.RNA-seq analysis revealed that aspirin up-regulated 4 ferroptosis-related drivers and down-regulated 5 ferroptosis-related suppressors in aspirin-treated HepG2 cells.Treatment with aspirin(4 mM)induced remarkable ferroptosis in HepG2 and Huh7 cells,which was enhanced by the ferroptosis inducer erastin(10μM).We demonstrated that NF-κB p65 restricted ferroptosis in HepG2 and Huh7 cells through directly binding to the core region of SLC7A11 promoter and activating the transcription of ferroptosis inhibitor SLC7A11,whereas aspirin induced ferroptosis through inhibiting NF-κB p65-activated SLC7A11 transcription.Overexpression of p65 rescued HepG2 and Huh7 cells from aspirin-induced ferroptosis.HCC patients with high expression levels of SLC7A11 and p65 presented lower survival rate.Functionally,NF-κB p65 blocked the aspirin-induced ferroptosis in vitro and in vivo,which was attenuated by erastin.We conclude that aspirin triggers ferroptosis by restricting NF-κB-activated SLC7A11 transcription to suppress the growth of HCC.These results provide a new insight into the mechanism by which aspirin regulates ferroptosis in hepatocarcinogenesis.A combination of aspirin and ferroptosis inducer may provide a potential strategy for the treatment of HCC in clinic.Yu-fei Wang Jin-yan Feng Li-na Zhao Man Zhao Xian-fu Wei Yu Geng Hong-feng Yuan Chun-yu Hou Hui-hui Zhang Guo-wen Wang Guang Yang Xiao-dong Zhang 2023Acta Pharmacologica Sinica2023,44,8:1

16LIFU-responsive nanomedicine enables acoustic droplet vaporization-induced apoptosis of macrophages for stabilizing vulnerable atherosclerotic plaques显示文摘Due to the high risk of tearing and rupture,vulnerable atherosclerotic plaques would induce serious cardiovascular and cerebrovascular diseases.Despite the available clinical methods can evaluate the vulnerability of plaques and specifically treat vulnerable plaques before a cardiovascular event,but the efficiency is still low and undesirable.Herein,we rationally design and engineer the low-intensity focused ultrasound(LIFU)-responsive FPD@CD nanomedicine for the highly efficient treatment of vulnerable plaques by facilely loading phase transition agent perfluorohexane(PFH)into biocompatible PLGA-PEG-PLGA nanoparticles(PPP NPs)and then attaching dextran sulphate(DS)onto the surface of PPP NPs for targeting delivery.DS,as a typical macrophages-targeted molecule,can achieve the precise vaporization of NPs and subsequently controllable apoptosis of RAW 264.7 macrophages as induced by acoustic droplet vaporization(ADV)effect.In addition,the introduction of DiR and Fe3O4 endows nanomedicine with near-infrared fluorescence(NIRF)and magnetic resonance(MR)imaging capabilities.The engineered FPD@CD nanomedicine that uses macrophages as therapeutic targets achieve the conspicuous therapeutic effect of shrinking vulnerable plaques based on in vivo and in vitro evaluation outcomes.A reduction of 49.4%of vascular stenosis degree in gross pathology specimens were achieved throughout the treatment period.This specific,efficient and biosafe treatment modality potentiates the biomedical application in patients with cardiovascular and cerebrovascular diseases based on the relief of the plaque rupture concerns.Jingxin Hou Jun Zhou Meiqi Chang Guangcheng Bao Jie Xu Man Ye Yixin Zhong Shuling Liu Junrui Wang Wei Zhang Haitao Ran Zhigang Wang Yu Chen Dajing Guo 2022Bioactive Materials2022,7,10:1
