| 1 | Sequential fate-switches in stem-like cells drive the tumorigenic trajectory from human neural stem cells to malignant glioma显示文摘Glioblastoma(GBM)is an incurable and highly heterogeneous brain tumor,originating from human neural stem/progenitor cells(hNSCs/hNPCs)years ahead of diagnosis.Despite extensive efforts to characterize hNSCs and end-stage GBM at bulk and single-cell levels,the de novo gliomagenic path from hNSCs is largely unknown due to technical difficulties in early-stage sampling and preclinical modeling.Here,we established two highly penetrant hNSC-derived malignant glioma models,which resemble the histopathology and transcriptional heterogeneity of human GBM.Integrating time-series analyses of whole-exome sequencing,bulk and single-cell RNA-seq,we reconstructed gliomagenic trajectories,and identified a persistent NSC-like population at all stages of tumorigenesis.Through trajectory analyses and lineage tracing,we showed that tumor progression is primarily driven by multi-step transcriptional reprogramming and fate-switches in the NSC-like cells,which sequentially generate malignant heterogeneity and induce tumor phenotype transitions.We further uncovered stage-specific oncogenic cascades,and among the candidate genes we functionally validated C1QL1 as a new glioma-promoting factor.Importantly,the neurogenic-to-gliogenic switch in NSC-like cells marks an early stage characterized by a burst of oncogenic alterations,during which transient AP-1 inhibition is sufficient to inhibit gliomagenesis.Together,our results reveal previously undercharacterized molecular dynamics and fate choices driving de novo gliomagenesis from hNSCs,and provide a blueprint for potential early-stage treatment/diagnosis for GBM. | Xiaofei Wang Ran Zhou Yanzhen Xiong Lingling Zhou Xiang Yan Manli Wang Fan Li Chuanxing Xie Yiming Zhang Zongyao Huang Chaoqiong Ding Kaidou Shi Weida Li Yu Liu Zhongwei Cao Zhen-Ning Zhang Shengtao Zhou Chong Chen Yan Zhang Lu Chen Yuan Wang | 2021 | Cell Research2021,31,6: | 8 |
| 2 | Disparity in reimbursement for tuberculosis care among different health insurance schemes: evidence from three counties in central China显示文摘Background:Health inequity is an important issue all around the world.The Chinese basic medical security system comprises three major insurance schemes,namely the Urban Employee Basic Medical Insurance(UEBMI),the Urban Resident Basic Medical Insurance(URBMI),and the New Cooperative Medical Scheme(NCMS).Little research has been conducted to look into the disparity in payments among the health insurance schemes in China.In this study,we aimed to evaluate the disparity in reimbursements for tuberculosis(TB)care among the abovementioned health insurance schemes.Methods:This study uses a World Health Organization(WHO)framework to analyze the disparities and equity relating to the three dimensions of health insurance:population coverage,the range of services covered,and the extent to which costs are covered.Each of the health insurance scheme’s policies were categorized and analyzed.An analysis of the claims database of all hospitalizations reimbursed from 2010 to 2012 in three counties of Yichang city(YC),which included 1506 discharges,was conducted to identify the differences in reimbursement rates and out-of-pocket(OOP)expenses among the health insurance schemes.Results:Tuberculosis patients had various inpatient expenses depending on which scheme they were covered by(TB patients covered by the NCMS have less inpatient expenses than those who were covered by the URBMI,who have less inpatient expenses than those covered by the UEBMI).We found a significant horizontal inequity of healthcare utilization among the lower socioeconomic groups.In terms of financial inequity,TB patients who earned less paid more.The NCMS provides modest financial protection,based on income.Overall,TB patients from lower socioeconomic groups were the most vulnerable.Conclusion:There are large disparities in reimbursement for TB care among the three health insurance schemes and this,in turn,hampers TB control.Reducing the gap in health outcomes between the three health insurance schemes in China should be a focus of TB care and control.Achieving equity through integrated policies that avoid discrimination is likely to be effective. | Yao Pan Shanquan Chen Manli Chen Pei Zhang Qian Long Li Xiang Henry Lucas | 2016 | Infectious Diseases of Poverty2016,5,1: | 7 |
| 3 | Conservation of Jiaohe ancient earthen site in China显示文摘Earthen sites,which are mainly made of earth,are cultural heritages with historical,artistic and scientific values.Many extremely valuable earthen sites have been preserved in the arid areas in China.The earthen site of Jiaohe Ancient City is one of the earliest National Protected Important Cultural Heritage Sites.The Jiaohe Ancient City site exhibits all kinds of deteriorations,which can be found in the earthen sites in arid environments in China.Through a case study of the Jiaohe ancient earthen site,we present in this paper the comprehensive conservation technologies,including the mud bricklaying and reparation,wooden rod anchorage,crack grouting,surface potassium silicate(PS) penetration consolidation,and suspended steel beam ceiling,etc.Results of this case study showed that better conservation effects could be achieved by selecting proper PS penetration and crack grouting processes based on the deterioration characteristics of the earthen sites.The technology of mud bricklaying and reparation was also an effective method for preventing the earthen body from collapsing.Compared with traditional conservation technologies,the suspended steel beam ceiling technology could effectively reduce the negative impacts to the original state of the earthen site.As for unstable cliffs,a new method using composite anchor rod of bamboo and steel with massive loose earth was applied.Deformation monitoring and temporary supports were critical and indispensable measures for the safe of site conservation projects.Through years of monitoring and practical operation at the Jiaohe ancient earthen site,deterioration at the site has been effectively controlled. | Zuixiong Li Xudong Wang Manli Sun Wenwu Chen Qinglin Guo Huyuan Zhang | 2011 | Journal of Rock Mechanics and Geotechnical Engineering2011,3,3: | 6 |

| 5 | Monitoring of microbial community structure and succession in the biohydrogen production reactor by denaturing gradient gel electrophoresis(DGGE)显示文摘To study the structure of microbial communities in the biological hydrogen produc- tion reactor and determine the ecological function of hydrogen producing bacteria, anaerobic sludge was obtained from the continuous stirred tank reactor (CSTR) in different periods of time, and the diversity and dynamics of microbial communities were investigated by denaturing gra- dient gel electrophoresis (DGGE). The results of DGGE demonstrated that an obvious shift of microbial population happened from the beginning of star-up to the 28th day, and the ethanol type fermentation was established. After 28 days the structure of microbial community became stable, and the climax community was formed. Comparative analysis of 16S rDNA sequences from reamplifying and sequencing the prominent bands indicated that the dominant population belonged to low G+C Gram-positive bacteria (Clostridium sp. and Ethanologenbacterium sp.), β- proteobacteria (Acidovorax sp.), γ-proteobacteria (Kluyvera sp.), Bacteroides (uncultured bacte- rium SJA-168), and Spirochaetes (uncultured eubacterium E1-K13), respectively. The hydrogen production rate increased obviously with the increase of Ethanologenbacterium sp., Clostridium sp. and uncultured Spirochaetes after 21 days, meanwhile the succession of ethanol type fer- mentation was formed. Throughout the succession the microbial diversity increased however it decreased after 21 days. Some types of Clostridium sp. Acidovorax sp., Kluyvera sp., and Bac- teroides were dominant populations during all periods of time. These special populations were essential for the construction of climax community. Hydrogen production efficiency was de- pendent on both hydrogen producing bacteria and other populations. | 6 | Inhibition of cyclooxygenase-2 activity in subchondral bone modifies a subtype of osteoarthritis显示文摘Osteoarthritis(OA) causes the destruction of joints. Its pathogenesis is still under investigation, and there is no effective diseasemodifying therapy. Here, we report that elevated cyclooxygenase-2(COX-2) expression in the osteocytes of subchondral bone causes both spontaneous OA and rheumatoid arthritis(RA). The knockout of COX-2 in osteocytes or treatment with a COX-2 inhibitor effectively rescues the structure of subchondral bone and attenuates cartilage degeneration in spontaneous OA(STR/Ort)mice and tumor necrosis factor-α transgenic RA mice. Thus, elevated COX-2 expression in subchondral bone induces both OAassociated and RA-associated joint cartilage degeneration. | 7 | Bamboo charcoal enhances cellulase and urease activities during chicken manure composting:Roles of the bacterial community and metabolic functions显示文摘Microbial enzymes are crucial for material biotransformation during the composting process.In this study,we investigated the effects of adding bamboo charcoal(BC)(i.e.,at 5%,10%,and 20%corresponding to BC5,BC10,and BC20,respectively)on the enzyme activity levels during chicken manure composting.The results showed that BC10 could increase the cellulose and urease activities by 56%and 96%,respectively.The bacterial community structure in BC10 differed from those in the other treatments,and Luteivirga,Lactobacillus,Paenalcaligenes,Ulvibacter,Bacillus,Facklamia,Pelagibacterium,Sporosarcina,Cellvibrio,and Corynebacterium had the most important roles in composting.Compared with other treatments,BC10 significantly enhanced the average rates of degradation of carbohydrates(Dxylose(40%)andα-D-lactose(44%))and amino acids(L-arginine(16%),L-asparagine(14%),and L-threonine(52%)).We also explored the associations among the bacterial community and their metabolic functions with the changes in the activities of enzymes.Network analysis demonstrated that BC10 altered the co-occurrence patterns of the bacterial communities,where Ulvibacter and class Bacilli were the keystone bacterial taxa with high capacities for degrading carbon source,and they were related to increases in the activities of cellulase and urease,respectively.The results obtained in this study may help to further enhance the efficiency of composting. | Yanan Yin Chao Yang Jingrui Tang Jie Gu Haichao Li Manli Duan Xiaochang Wang Rong Chen | 2021 | Journal of Environmental Sciences2021,33,10: | 4 |

| 9 | Development of EST-SSR primers and their practicability test for Laminaria显示文摘Laminaria (Saccharina) is one of the economic important seaweeds.Through sequence analysis of 4 099 ESTs from Laminaria (971 were generated from our own cDNA laboratories and 3 128 were downloaded from updated EST databases) with SSRIT software,two hundred and fifty-four SSRs in 206 ESTs were found.From the 254 SSRs,sixty-three SSR primer-pairs were designed. In order to test their practicability, the 63 primer-pairs were tested in commonly used SSR reaction conditions using 12 Laminara DNA samples as templates. The results show that 23 SSR primer-pairs gave good amplification patterns on most (more than 80%) of the 12 Laminara DNA templates. | 11 | Improving Baculovirus Transduction of Mammalian Cells by Incorporation of Thogotovirus Glycoproteins显示文摘Baculovirus can transduce a wide range of mammalian cells and is considered a promising gene therapy vector. However,the low transduction efficiency of baculovirus into many mammalian cells limits its practical application. Co-expressing heterologous viral glycoproteins(GPs), such as vesicular stomatitis virus G protein(VSV G), with baculovirus native envelope protein GP64 is one of the feasible strategies for improving virus transduction. Tick-borne thogotoviruses infect mammals and their GPs share sequence/structure homology and common evolutionary origins with baculovirus GP64.Herein, we tested whether thogotovirus GPs could facilitate the entry of the prototype baculovirus Autographa californica multiple multiple nucleopolyhedrovirus(AcMNPV) into mammalian cells. The gp genes of two thogotoviruses, Thogoto virus and Dhori virus, were inserted into the AcMNPV genome. Both GPs were properly expressed and incorporated into the envelope of the recombinant AcMNPVs. The transduction rates of recombinant AcMNPVs expressing the two thogotovirus GPs increased for approximately 4–12 fold compared to the wild type AcMNPV in six of the 12 tested mammalian cell lines. It seemed that thogotovirus GPs provide the recombinant AcMNPVs with different cell tropisms and showed better performance in several mammalian cells compared to VSV G incorporated AcMNPV. Further studies showed that the improved transduction was a result of augmented virus-endosome fusion and endosome escaping, rather than increased cell binding or internalization. We found the AcMNPV envelope protein GP64-mediated fusion was enhanced by the thogotovirus GPs at relatively higher p H conditions. | 12 | Three dimensional distribution of surfactant in microspheres revealed by synchrotron radiation X-ray microcomputed tomography显示文摘This study investigated the formulation mechanism of microspheres via internal surfactant distribution. Eudragit L100 based microspheres loaded with bovine serum albumin were prepared by solid in oil in oil emulsion solvent evaporation method using acetone and liquid paraffin system containing sucrose stearate as a surfactant. The fabricated microspheres were evaluated for encapsulation efficiency, particle size, production yield, and in vitro release characteristics. The internal structures of microspheres were characterized using synchrotron radiation X-ray microcomputed tomography(SR-μCT). The enhanced contrast made the sucrose stearate distinguished from Eudragit to have its three dimensional(3D) distribution. Results indicated that the content and concentration determined the state of sucrose stearate and had significant influences on the release kinetics of protein. The dispersity of sucrose stearate was the primary factor that controlled the structure of the microspheres and further affected the encapsulation efficiency, effective drug loading, as well as in vitro release behavior. In conclusion, the 3D internal distribution of surfactant in microspheres and its effects on protein release behaviors have been revealed for the first time.
