State of the Art and Prospects in Metal-Organic Framework-Derived Microwave Absorption Materials显示文摘Microwave has been widely used in many fields, including communication, medical treatment and military industry;however, the corresponding generated radiations have been novel hazardous sources of pollution threating human’s daily life. Therefore, designing high-performance microwave absorption materials(MAMs)has become an indispensable requirement.Recently,metal-organic frameworks(MOFs)have been considered as one of the most ideal precursor candidates of MAMs because of their tunable structure, high porosity and large specific surface area.Usually,MOF-derived MAMs exhibit excellent electrical conductivity, good magnetism and sufficient defects and interfaces, providing obvious merits in both impedance matching and microwave loss.In this review, the recent research progresses on MOF-derived MAMs were profoundly reviewed, including the categories of MOFs and MOF composites precursors, design principles, preparation methods and the relationship between mechanisms of microwave absorption and microstructures of MAMs.Finally, the current challenges and prospects for future opportunities of MOF-derived MAMs are also discussed.Shuning Ren Haojie Yu Li Wang Zhikun Huang Tengfei Lin Yudi Huang Jian Yang Yichuan Hong Jinyi Liu 2022Nano-Micro Letters2022,14,4:6
A triple-RBD-based mucosal vaccine provides broad protectionagainst SARS-CoV-2 variants of concern显示文摘The rapid mutation and spread of SARS-CoV-2 variants urge the development of effective mucosal vaccines to provide broadspectrum protection against the initial infection and thereby curb the transmission potential. Here,we designed a chimeric tripleRBD immunogen,3Ro-NC,harboring one Delta RBD and two Omicron RBDs within a novel protein scaffold.3Ro-NC elicits potent and broad RBD-specific neutralizing immunity against SARS-CoV-2 variants of concern.Notably, intranasal immunization with 3RoNC plus the mucosal adjuvant KFD(3Ro-NC+KFDi.n)elicits coordinated mucosal IgA and higher neutralizing antibody specificity(closer antigenic distance)against the Omicron variant. In Omicron-challenged human ACE2 transgenic mice,3Ro-NC+KFDi.n immunization significantly reduces the tissue pathology in the lung and lowers the viral RNA copy numbers in both the lung(85.7-fold)and the nasal turbinate(13.6-fold).Nasal virologic control is highly correlated with RBD-specific secretory IgA antibodies. Our data show that 3Ro-NC plus KFD is a promising mucosal vaccine candidate for protection against SARS-CoV-2 Omicron infection, pathology and transmission potential.Jingyi Yang Mei-Qin Liu Lin Liu Xian Li Mengxin Xu Haofeng Lin Shuning Liu Yunqi Hu Bei Li Bowen Liu Min Li Ying Sun Yao-Qing Chen Zheng-Li Shi Huimin Yan 2022Cellular & Molecular Immunology2022,19,11:3
Glycerolipid Profiling of Yellow Sarson Seeds Using Ultra High Performance Liquid Chromatography Coupled to Triple Time-of-Flight Mass Spectrometry显示文摘Yellow sarson(Brassica rapa ssp. trillocularis) is an important rapeseed-mustard species of Brassica rapa due to its high seed oil content. Glycerolipids and fatty acid composition affect seed germination and determine the quality of seed oil. To date, no information is available on the composition of individual glycerolipids in this species. Therefore, in this study the glycerolipid profiling of yellow sarson seeds was performed using ultra high performance liquid chromatography coupled to triple time-of-flight mass spectrometry(UPLC-Triple-TOF-MS).A fast and efficient chromatographic separation of glycerolipids was accomplished based on an UPLCTMBEH C8 column within 22 min.In ESI positive ion mode, TOF-MS scan-information dependent acquisition-product ion scan was carried out to acquire both high resolution MS and MS/MS information from one injection. According to MS/MS spectra, predominant fragmentation patterns of glycerolipids were elucidated in detail. Based on retention time, accurate mass, isotopic distribution, and fragmentation patterns, the composition of 144 glycerolipids and fatty acids were finally identified in yellow sarson seeds, including 77 triacylglycerols, 32 diacylglycerols, 18sulfoquinovosyl-diacylglycerols, 5 monogalactosyl-diaclyglycerols, and 12 digalactosyl-diacylglycerols. Of them, the most abundant glycerolipids in yellow sarson seeds were triacylglycerols, the major storage form of seed oil in plants. In addition, diacylglycerols were found as a minor component of glycerolipids. The lowest amounts of glycerolipids detected in seeds were glycosyl-acylglycerols. The results revealed the composition and relative content of glycerolipids in yellow sarson seeds, which will provide a more comprehensive assessment of the quality of seed oil and also help to select functional cultivars with higher beneficial glycerolipids. This profiling method has the advantages of high throughput, high sensitivity and good accuracy, which will provide a reliable analysis platform for glycerolipids metabolism in plants.ZHENG Shuning ZHANG Yanguo LIU Guangyang LV Jun LIU Zhongxiao XU Xiaomin LI Lingyun XU Donghui 2017Horticultural Plant Journal2017,3,4:2
Rapid isolation and immune profiling of SARS-CoV-2 specific memory B cell in convalescent COVID-19 patients via LIBRA-seq显示文摘B cell response plays a critical role against SARS-CoV-2 infection. However, little is known about the diversity and frequency of the paired SARS-CoV-2 antigen-specific BCR repertoire after SARS-CoV-2 infection. Here,we performed single-cell RNA sequencing and VDJ sequencing using the memory and plasma B cells isolated from five convalescent COVID-19 patients, and analyzed the spectrum and transcriptional heterogeneity of antibody immune responses.Via linking BCR to antigen specificity through sequencing(LIBRA-seq),we identified a distinct activated memory B cell subgroup(CD11c^(high) CD95^(high))had a higher proportion of SARS-CoV-2 antigen-labeled cells compared with memory B cells. Our results revealed the diversity of paired BCR repertoire and the non-stochastic pairing of SARS-CoV-2 antigen-specific immunoglobulin heavy and light chains after SARS-CoV-2 infection. The public antibody clonotypes were shared by distinct convalescent individuals. Moreover, several antibodies isolated by LIBRA-seq showed high binding affinity against SARS-CoV-2 receptor-binding domain(RBD)or nucleoprotein(NP)via ELISA assay. Two RBD-reactive antibodies C14646P3S and C2767P3S isolated by LIBRA-seq exhibited high neutralizing activities against both pseudotyped and authentic SARS-CoV-2 viruses in vitro. Our study provides fundamental insights into B cell response following SARS-CoV-2 infection at the single-cell level.Bing He Shuning Liu Yuanyuan Wang Mengxin Xu Wei Cai Jia Liu Wendi Bai Shupei Ye Yong Ma Hengrui Hu Huicui Meng Tao Sun Yanling Li Huanle Luo Mang Shi Xiangjun Du Wenjing Zhao Shoudeng Chen Jingyi Yang Haipeng Zhu Yusheng Jie Yuedong Yang Deyin Guo Qiao Wang Yuwen Liu Huimin Yan Manli Wang Yao-Qing Chen 2021Signal Transduction and Targeted Therapy2021,6,6:2
An oral second-generation proteasome inhibitor oprozomib significantly inhibits lung cancer in a p53 independent manner in vitro显示文摘The destruction of proteins via the ubiquitin-proteasome system is amulti-step,complex process involving polyubiquitination of substrate proteins, follow ed by proteolytic degradation by the m acrom olecular 26S proteasome complex.Inhibitors of the proteasome prom ote the accum ulation of proteins that are deleterious to cell survival and are prom ising anticancer agents.Oprozomib(OPZ),an oral second-generation proteasome inhibitor, has been shown to inhibit the grow th of several cancers in preclinical and clinical trials, including m ultiple m yelom a and head and neck cancers, but its effects on lung cancer has not yet been determ ined.In this study,we evaluated the inhibitory effects of OPZ on lung cancer cell lines in vitro. The results showed that OPZ significantly suppressed cell proliferation and strongly induced apoptosis in both tested lung cancer cells independent of p53 expression.OPZ was able to cause obvious caspase 3 and PARP cleavages and stabilize p53 and its transcriptional targets p21,PUMA,and Noxa. Moreover,OPZ was capable of sensitizing lung cancer cells to the conventional chem otherapeutic drug cisplatin. Our study provides preclinical data and sheds light on the potential applications of proteasome inhibitor OPZ in lung cancer treatment.Hongge Zhu Tianhai Wang Zhou Xin Yiyi Zhan Guoming Gu Xiaoqin Li Xiuli Wang Shune Yang Chunling Liu 2019Acta Biochimica et Biophysica Sinica2019,51,10:2
VAV2 is required for DNA repair and implicated in cancer radiotherapy resistance显示文摘Radiotherapy remains the mainstay for treatment of various types of human cancer;however, the clinical efficacy is often limited by radioresistance,in which the underlying mechanism is largely unknown. Here, using esophageal squamous cell carcinoma(ESCC)as a model,we demonstrate that guanine nucleotide exchange factor 2(VAV2),which is overexpressed in most human cancers, plays an important role in primary and secondary radioresistance. We have discovered for the first time that VAV2 is required for the Ku70/Ku80 complex formation and participates in non-homologous end joining repair of DNA damages caused by ionizing radiation.Weiling Liu Chuanwang Miao Shaosen Zhang Yachen Liu Xiangjie Niu Yiyi Xi Wenjia Guo Jiahui Chu Ai Lin Hongjin Liu Xinyu Yang Xinjie Chen Ce Zhong Yuling Ma Yuqian Wang Shihao Zhu Shuning Liu Wen Tan Dongxin Lin Chen Wu 2021Signal Transduction and Targeted Therapy2021,6,9:1
Exome sequencing identifies rare mutations of LDLR and QTRT1 conferring risk for early-onset coronary artery disease in Chinese显示文摘Despite the advances made over the past decades, the known loci for coronary artery disease(CAD)still only explain<20%of the genetic variation in risk[1,2].The known loci with the strongest effects usually confer a 20%-37%increased CAD risk and the most loci modulate risk by≤10%[1].While most of the current data are from European populations, the use of trans-ethnic analyses could be helpful to identify additional loci.Genetic inheritance may impose a high burden on early-onset coronary artery disease(EOCAD).Kang Yao Yuxiang Dai Juan Shen Yi Wang Huanjie Yang Runda Wu Qijun Liao Hongyi Wu Xiaodong Fang Shalaimaiti Shali Lili Xu Meng Hao Chenhao Lin Zhonghan Sun Yilian Liu Mengxin Li Zhen Wang Qiang Gao Shuning Zhang Chenguang Li Wei Gao Lei Ge Yunzeng Zou Aijun Sun Juying Qian Li Jin Shangyu Hong Yan Zheng Junbo Ge 2022National Science Review2022,9,8:1
Enhancing single-molecule conductance of platinum (Ⅱ) complexes through synergistic aromaticity-assisted structural asymmetry显示文摘Seeking the strategies of designing highly conductive molecular structures is one of the core researches in molecular electronics. As asymmetric structure has manifested feasible properties in comprehensive fields, we introduce the structures of asymmetric platinum(Ⅱ) complexes into the charge transport study at single-molecule scale for the first time. The single-molecule conductance measurement results reveal that, in platinum(Ⅱ)-aryloligoynyl structures, the conductance of asymmetrically coordinated complexes is obviously higher than that of the symmetric isomers with the same molecular length, while the conductance is almost identical in symmetric and asymmetric platinum(Ⅱ)-oligoynyl complexes. Theoretical study uncovers that, upon connecting to the oligoynyl structure, the aromatic group effectively extends the π-system of the whole conductive backbone and gathers the HOMO population mainly on the longer oligoynyl ligand, which reduces the energy barrier in electron transport and enhances the conductance through HOMO energy lifting. This result provides feasible strategy for achieving high conductive molecular devices.Ping Duan Junyang Liu Jin-Yun Wang Kai Qu Shuning Cai Fei Wang Lichuan Chen Xiaoyan Huang Ruihao Li Jia Shi Qian-Chong Zhang Wenjing Hong Zhong-Ning Chen 2020Science China Chemistry2020,63,4:0
Monitoring NAD(H) and NADP(H) dynamics during organismal development with genetically encoded fluorescent biosensors显示文摘Cell metabolism plays vital roles in organismal development, but it has been much less studied than transcriptional and epigenetic control of developmental programs. The difficulty might be largely attributed to the lack of in situ metabolite assays.Genetically encoded fluorescent sensors are powerful tools for noninvasive metabolic monitoring in living cells and in vivo by highly spatiotemporal visualization.Among all living organisms, the NAD(H)and NADP(H)pools are essential for maintaining redox homeostasis and for modulating cellular metabolism. Here,we introduce NAD(H)and NADP(H)biosensors, present example assays in developing organisms, and describe promising prospects for how sensors contribute to developmental biology research.Ting Li Yejun Zou Shuning Liu Yi Yang Zhuo Zhang Yuzheng Zhao 2022Cell Regeneration2022,11,1:0
High BRCA1 expression is an independent prognostic biomarker in LUAD and correlates with immune infiltration显示文摘Non-small-cell lung cancer(NSCLC)is the primary histopathological subtype of lung cancer and accounts for around 82%of all pulmonary neoplasms[1].Breast cancer susceptibility gene 1(BRCA1)is a tumor suppressor gene that regulates cellular responses to stress through DNA damage repair and is frequently mutated in breast and ovarian cancers[2].Furthermore,previous studies have increasingly shown that the expression of BRCA1 and the relevant poly(ADP-ribose)polymerase inhibitors is related to the prognosis and therapeutic response of other tumors, including lung cancer[3,4].However,previous findings on the prognostic value of BRCA1 for NSCLC have been inconsistent or conflicting, which may be attributed to the small number of patients from a single center or disparity in the ratio of patients with lung adenocarcinoma(LUAD)versus squamous cell carcinoma(LUSC)[5].The aim of this study was to determine the prognostic value of BRCA1 in NSCLC and its association with immune infiltration.Fengzhu Guo Cong Li Shuning Liu Zhijun Li Jingtong Zhai Zhiwu Wang Binghe Xu 2023Cancer Innovation2023,2,2:0

Network pharmacology assessment of Qingkailing injection(清开灵注射液)treatment of cholestatic hepatitis显示文摘OBJECTIVE:To investigate the targets and mechanisms of action of Qingkailing injection(清开灵注射液,QKL)in the treatment of cholestatic hepatitis.METHODS:A network pharmacology method was implemented using drug and disease databases to target QKL and cholestasis hepatitis, respectively. The functional protein association network STRING database was used to construct a protein-protein interaction network using R language and the Bioconductor toolkit. The org.Hs.eg.db and cluster Profiler packages were used for gene ontology and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis, which explored biological functions and pathways of potential targets.Targets were then visualized using Cytoscape 3.6.0 software.RESULTS:We screened 121 compounds in QKL and identified 112 targets for the treatment of cholestatic hepatitis.QKL played a role in the treatment of cholestatic hepatitis through 305 biology process terms,15 cellular component and 29 molecular function terms. The mechanism of QKL action was mainly related to tumor necrosis factor,mitogen-activated protein kinase, and PI3 K-Akt signaling pathways.CONCLUSION:The treatment of cholestatic hepatitis by QKL involved multiple targets, biological functions, and signaling pathways that are closely associated with the disease.LIAN Yajun WANG Qingguo MU Jie LIU Haixia XU Tian FAN Shuning TANG Feifei FENG Tianyi XU Wenxiu JIN Na CHENG Fafeng WANG Xueqian 2021Journal of Traditional Chinese Medicine2021,41,1:0
Effect of Na^(+)and Sr^(2+)Substitution on the Formation of the Oxygen Vacancies in Y_(3)Al_(2)Ga_(3)O_(12):Ce^(3+)Phosphor显示文摘The low-valence cations Na^(+)and Sr^(2+)were selected as the co-dopants to increase the vacancies concentration in the Y_(2.982)Ce_(0.018)Al_(2)Ga_(3)O_(12)phosphor.The successful incorporation of Na^(+)and Sr^(2+)was confirmed by the X-Ray Difiraction(XRD)results. All the samples show 5d-4f green persistent luminescence of Ce^(3+)after 450 nm excitation. The decay curves demonstrate that the persistent luminescence is efiectively enhanced with Na+and Sr2+doping.The thermoluminescence glow curves also show not only does the trap concentration increase, but also the distribution of trap depths is broadened. In addition, the air-and H_(2)/Ar-annealing treatments were conducted on every as-made sample. The experimental results prove that the increased traps after the Na^(+)/Sr^(2+)doping are mainly attributed to the oxygen vacancies, and the traps have a continuous and broad distribution of trap depths. We hope this work could give new inspiration for designing a high-performance persistent phosphor.YANG Ting JIANG Hongyi DONG Ye GAO Shuai LIU Shuning 2023Journal of Wuhan University of Technology(Materials Science)2023,38,5:0
Ultra-lightweight ceramic scaffolds with simultaneous improvement of pore interconnectivity and mechanical strength显示文摘The high porosity and interconnectivity of scaffolds are critical for nutrient transmission in bone tis-sue engineering but usually lead to poor mechanical properties.Herein,a novel method that combines acid etching(AE)with selective laser sintering(SLS)and reaction bonding(RB)of Al particles is pro-posed to realize highly improved porosity, interconnectivity, mechanical strength, and in vitro bioactivity in 3D Al_(2)O_(3) scaffolds.By controlling the oxidation and etching behaviors of Al particles,a tunable hol-low spherical feature can be obtained, which brings about the distinction in compressive response and fracture path. The prevention of microcrack propagation on the in situ formed hollow spheres results in unique near elastic buckling rather than traditional brittle fracture, allowing an unparalleled compressive strength of 3.72±0.17 MPa at a high porosity of 87.7%±0.4%and pore interconnectivity of 94.7%±0.4%.Furthermore,scaffolds with an optimized pore structure and superhydrophilic surface show excellent cell proliferation and adhesion properties. Our findings offer a promising strategy for the coexistence of out-standing mechanical and biological properties, with great potential for tissue engineering applications.Ye Dong Annan Chen Ting Yang Shuai Gao Shuning Liu Hongyi Jiang Yusheng Shi Chenglong Hu 2023Journal of Materials Science & Technology2023,,6:0
A Piecewise Linear Programming Algorithm for Sparse Signal Reconstruction显示文摘In order to recover a signal from its compressive measurements, the compressed sensing theory seeks the sparsest signal that agrees with the measurements, which is actually an l_0 norm minimization problem. In this paper, we equivalently transform the l_0 norm minimization into a concave continuous piecewise linear programming, and propose an optimization algorithm based on a modified interior point method. Numerical experiments demonstrate that our algorithm improves the sufficient number of measurements, relaxes the restrictions of the sensing matrix to some extent, and performs robustly in the noisy scenarios.Kuangyu Liu Xiangming Xi Zhiming Xu Shuning Wang 2017Tsinghua Science and Technology2017,22,1:0
Method of Hill Tunneling via Weighted Simplex Centroid for Continuous Piecewise Linear Programming显示文摘This paper works on a heuristic algorithm with determinacy for the global optimization of Continuous PieceWise Linear(CPWL) programming. The widely applied CPWL programming can be equivalently transformed into D.C. programming and concave optimization over a polyhedron. Considering that the super-level sets of concave piecewise linear functions are polyhedra, we propose the Hill Tunneling via Weighted Simplex Centroid(HTWSC) algorithm, which can escape a local optimum to reach the other side of its contour surface by cutting across the super-level set. The searching path for hill tunneling is established via the weighted centroid of a constructed simplex. In the numerical experiments, different weighting methods are studied first, and the best is chosen for the proposed HTWSC algorithm. Then, the HTWSC algorithm is compared with the hill detouring method and the software CPLEX for the equivalent mixed integer programming, with results indicating its superior performance in terms of numerical efficiency and the global search capability.Zhiming Xu Yu Bai Kuangyu Liu Shuning Wang 2019Tsinghua Science and Technology2019,24,3:0

A neutralizing-protective supersite of human monoclonal antibodies for yellow fever virus显示文摘The yellow fever virus(YFV)is a life-threatening human pathogen.Owing to the lack of available therapeutics,non-vaccinated individuals are at risk. Here,we isolated eight human monoclonal antibodies that neutralize YFV infection.Five recognized overlapping epitopes and exhibited potent neutralizing activity.Two(YD6 and YD73)were ultra-potent and conferred complete protection against the lethal challenge of YFV as both prophylactics and therapeutics in a mouse model.Crystal structures revealed that YD6 engaged the YFV envelope protein in both pre-and post-fusion states, suggesting viral inhibition by a“double-lock”mechanism.The recognition determinants for YD6 and YD73 are clustered at the premembrane(prM)-binding site.Notably, antibodies targeting this site were present in minute traces in YFV-infected individuals but contributed significantly to neutralization, suggesting a vulnerable supersite of YFV.We provide two promising candidates for immunotherapy against YFV,and the supersite represents an ideal target for epitope-based vaccine design.Yan Li Zhihai Chen Lili Wu Lianpan Dai Jianxun Qi Yan Chai Shihua Li Qihui Wang Zhou Tong Sufang Ma Xiaomin Duan Shuning Ren Rui Song Mifang Liang Wenjun Liu Jinghua Yan George F.Gao 2022The Innovation2022,3,6:0
The impact of venetoclax based regimens in the preemptive of measurable residual disease in acute myeloid leukemia显示文摘To The Editor:The role of measurable residual disease(MRD)in prognosis and treatment in acute myeloid leukemia(AML)is evolving.Studies have demonstrated the correlation between MRD and risks of relapse in adult AML:persistently positive MRD after induction is associated with a high risk of relapse,1,2 and these patients should consider allogeneic transplantation(allo-Hematopoietic Stem Cell Transplantation(HSCT))and clinical trial, even in favorable-risk groups. However, because of the financial issue or lack of suitable transplant donors, many of the patients could not receive allo-HSCT,so how to prolong the relapse-free survival of these patients remains a challenge.Platzbecker et al treated MRD-positive patients with azacytidine(AZA),and found pre-emptive therapy with AZA can prevent or substantially delay hematological relapse in MRD-positive patients with MDS(myelodysplastic syndrome)or AML who are at a high risk of relapse.Qiuyun Fang Xiaoyuan Gong Yan Li Benfa Gong Yuntao Liu Kaiqi Liu Guangji Zhang Shuning Wei Dong Lin Bingcheng Liu Ying Wang Hui Wei Yingchang Mi Jianxiang Wang 2022Blood Science2022,4,1:0
