1Adiponectin:a versatile player of innate immunity显示文摘Adiponectin acts as a key regulator of the innate immune system and plays a major role in the progression of inflammation and metabolic disorders.Macrophages and monocytes are representative components of the innate immune system,and their proliferation,plasticity,and polarization are a key component of metabolic adaption.Innate-like lymphocytes such as group 2 innate lymphoid cells(ILC2s),natural killer T(NKT)cells,and gamma delta T(gd T)cells are also members of the innate immune system and play important roles in the development of obesity and its related diseases.Adiponectin senses metabolic stress and modulates metabolic adaption by targeting the innate immune system under physiological and pathological conditions.Defining the mechanisms underlying the role of adiponectin in regulating innate immunity is crucial to adiponectin-based therapeutic intervention.Yan Luo Meilian Liu 2016Journal of Molecular Cell Biology2016,8,2:6
2The novel ER stress inducer Sec C triggers apoptosis by sulfating ER cysteine residues and degrading YAP via ER stress in pancreatic cancer cells显示文摘Pancreatic adenocarcinoma(PAAD) is one of the most lethal malignancies. Although gemcitabine(GEM) is a standard treatment for PAAD, resistance limits its application and therapy. Secoemestrin C(Sec C) is a natural compound from the endophytic fungus Emericella, and its anticancer activity has not been investigated since it was isolated. Our research is the first to indicate that Sec C is a broad-spectrum anticancer agent and could exhibit potently similar anticancer activity both in GEM-resistant and GEM-sensitive PAAD cells. Interestingly, Sec C exerted a rapid growth-inhibiting effect(80% death at 6 h), which might be beneficial for patients who need rapid tumor shrinkage before surgery. Liquid chromatography/mass spectrometry and N-acetyl-L-cysteine(NAC) reverse assays show that Sec C sulfates cysteines to disrupt disulfide-bonds formation in endoplasmic reticulum(ER) proteins to cause protein misfolding, leading to ER stress and disorder of lipid biosynthesis. Microarray data and subsequent assays show that ER stress-mediated ER-associated degradation(ERAD) ubiquitinates anddownregulates YAP to enhance ER stress via destruction complex(YAP-Axin-GSK-βTr CP), which also elucidates a unique degrading style for YAP. 4Rheb promotes brown fat thermogenesis by Notch-dependent activation of the PKA signaling pathway显示文摘Increasing brown and beige fat thermogenesis have an anti-obesity effect and thus great metabolic benefits.However,the molecular mechanisms regulating brown and beige fat thermogenesis remain to be further elucidated.We recently found that fat-specific knockout of Rheb promoted beige fat thermogenesis.In the current study,we show that Rheb has distinct effects on thermogenic gene expression in brown and beige fat.Fat-specific knockout of Rheb decreased protein kinase A(PKA)activity and thermogenic gene expression in brown adipose tissue of high-fat diet-fed mice.On the other hand,overexpression of Rheb activated PKA and increased uncoupling protein 1 expression in brown adipocytes.Mechanistically,Rheb overexpression in brown adipocytes increased Notch expression,leading to disassociation of the regulatory subunit from the catalytic subunit of PKA and subsequent PKA activation.Our study demonstrates that Rheb,by selectively modulating thermogenic gene expression in brown and beige adipose tissues,plays an important role in regulating energy homeostasis.Wen Meng Xiuci Liang Ting Xiao Jing Wang Jie Wen Hairong Luo Jianhui Teng Yanquan Fei Qinghai Zhang Bilian Liu Fang Hu juli Bai Meilian Liu Zhiguang Zhou Feng Liu 2019Journal of Molecular Cell Biology2019,11,9:2
5Using multi-source data to assess livability in Hong Kong at the community-based level:A combined subjective-objective approach显示文摘With the emergence of new types of data(e.g.social media data)and cutting-edge computer technology(e.g.Natural Language Processing),the shortcomings of traditional methods(subjective and objective ways)for de-tecting urban livability can be overcome by an integrated approach.This study aims to develop a comprehensive approach to measure urban livability based on statistic data,geo-data(e.g.points of interest),questionnaires survey,and social media data(Instagram),from both objective and subjective angles.Hong Kong,as a city with a high level of urbanization and contrasting urban environments,is chosen as the study area in this research.Through this study,the question“which area of Hong Kong is more suitable for living”is answered by the visu-alization of GIS-based analysis.Also,the correlation between livability scores and individuals’sentiment scores are explored.Specifically,the results show that central areas of Hong Kong with a higher level of urbanization are relatively more livable than suburban regions.However,through sentiment analysis,individuals who post Instagram in suburban areas of Hong Kong usually express more positive content and happier emotion than those who post Instagram in central urban areas.The study could offer useful information for the policy action of authorities as well as the residential location choices of citizens.Jianxiao LIU Han BI Meilian Wang 2020Geography and Sustainability2020,1,4:2

9Acute and Sub-Acute Toxicological Evaluations of Bioactive Alkaloidal Extract from Melodinus henryi and Their Main Chemical Constituents显示文摘Melodinus henryi is a good source of terpenoid indole alkaloids,and traditionally used as a folk medicine in the treatment of meningitis and fracture.In order to further exploit their potential uses,its anti-inflammatory and immunosuppressive activi-ties,safety evaluations and chemical profiles have been illustrated.Compared to the crude methanol extract from M.henryi and its non-alkaloidal fraction,the total alkaloidal fraction(MHTA)had the strongest anti-inflammatory and immunosup-pressive activities.In the acute oral toxicity assay,the half lethal dose(LD50)of MHTA was more than 2000 mg/kg.The sub-acute toxicity assay for consecutive 28 days exhibited MHTA at a lower concentrations of less than 500 mg/kg might be regarded as safe,and might damage spleen,liver,kidney,and heart when the dose is higher than 1000 mg/kg.In addition,a phytochemical investigation on MHTA led to the isolation of 15 monoterpenoid indole alkaloids.Thus,in regard with the potent side effects of MHTA,it should be used with caution in the development of phytomedicine.Meilian Yang Yudan Wang Zhifeng Fan Qingwang Xue Guy Sedar Singor Njateng Yaping Liu Jianxin Cao Tianrui Zhao Guiguang Cheng 2020Natural Products and Bioprospecting2020,10,4:1

11A Case of Congenital Syringocystadenoma Papilliferum显示文摘Syringocystadenoma papilliferum(SCAP)is a rare benign skin tumor that develops from the apocrine or eccrine glands.Here,we report the case of a 2-year-old girl who presented with an asymptomatic,slowly growing lesion on her scalp.On physical examination,a 2 cm×1 cm red papillary plaque with a crusted surface was noted on her scalp.Histo­pathological examination revealed papillomatous expansion of the epidermis and cystic invaginations extending from the epidermis down to the deep dermis.The cystic structures were lined by papillae with two layers of columnar epithelium,which showed decapitation secretion.Based on the above clinicopathological findings,a diagnosis of congenital scalp SCAP was made.Liansheng ZHONG Meilian LIU Xingqiang GAO 2021Chinese Journal Of Plastic and Reconstructive Surgery2021,3,2:0

13Effects of plastic film mulching on flax(Linum usitatissimum L.) root distribution and water use efficiency显示文摘To evaluate effects of plastic film mulching on soil moisture, majority root distribution and water use efficiency (WUE), experiment was designed in flax field of Gansu,China. Three treatments including flat drill without mulching (CK), whole field soil plastic film mulched which covered with soil and bunch seeded (T1), and ridges mulched with plastic film and row-seeding in furrows (T2) were tested. Results indicated that mulching practice increased soil water storage compared to CK treatment in general. In both topsoil (0-30 cm) and underlying soil (30-100 cm) layer, T1 treatment had significantly greater (P<0.05) water storage than that in CK and T2 treatment. T1 treatment increased most significantly of soil water storage in 0-100 cm soil layer after rain, and also maintained high soil moisture at flax mature stage. Compared to CK, T1 and T2 produced obviously larger (P<0.05) taproot diameter and greater dry weight at both juvenile growth stage and first flowering stage, but had shorter lateral root length at all three stages. Consequently, seed yields and WUE with mulching treatment were increased by 22.3%-38.8% for T1 and 35.4%-56.2% for T2 compared to CK respectively. 14Effect of dehydroepiandrosterone on insulin action and development of insulin-induced resistance in C_2C_(12) muscle cells显示文摘Dehydroepiandrosterone (DHEA), a precursor of androgens and estrogens, has been demonstrated to have effect of preventing insulin resistance and development of diabetes mellitus. Administration of testosterone appears to induce a marked insulin resistance. How these two hormones affect insulin resistance through regulation of sensitivity of tissues to insulin deserves further studies. Here, the effects of DHEA and testosterone on response to insulin in C2C12 muscle cells are analyzed. After 24 h of DHEA (10-6 mol/L) treatment, C2C12 cells showed an increased insulin- stimulated glucose uptake and enhanced activities of glycogen synthase (GS), phosphofructokinase (PFK) and pyruvate dehydrogenase (PDH), whereas testosterone gave the opposite effects. Incubation of C2C12 cells with high-dose insulin (5×10-7 mol/L) for 24 hours decreased their sensitivity to insulin and led to a state of resistance as assessed on insulin-stimulated glucose uptake and activities of GS, PFK and PDH. Addition of DHEA to insulin-resistant C2C12 cells could reverse the response of these cells to high-dose insulin, but testosterone could further impair insulin sensitivity in insulin-resistant C2C12 cells. 15Comprehensive bioinformatics analysis of CYB561 expression in breast cancer:Link between prognosis and immune infiltration显示文摘Background:Cytochrome b561(CYB561)plays a critical role in neuroendocrine function,cardiovascular regulation,and tumor growth;however,the prognostic value of CYB561 in patients with breast cancer and the relationship between CYB561 expression and immune infiltration in breast cancer remain unclear.Methods:The mRNA expression and clinical data of patients with breast cancer were obtained from The Cancer Genome Atlas database.Functional enrichment analysis was used to explore underlying biological functions associated with CYB561.The methylation status of CYB561 was analyzed using the MethSurv database.The enrichment score of immune cell infiltration for CYB561 in breast cancer was calculated using single-sample gene set enrichment analysis.The prognostic value of CYB561 was evaluated using Kaplan-Meier method and Cox regression analysis.Based on the results of the multivariate Cox analysis,a nomogram was constructed to predict the effect of CYB561 expression on overall survival(OS).Results:The results showed that CYB561 was highly expressed in breast cancer tissues.Hypomethylation of CYB561 is associated with an unfavorable prognosis.In multivariate Cox regression analysis,CYB561 was an independent prognostic factor for OS.Functional enrichment analysis indicated that estrogen signaling pathway,inflammatory response,KRAS signaling pathway,epithelial-mesenchymal transition,leukocyte migration,and regulation of lymphocyte activation were strongly enriched in the low CYB561 expression group.Additionally,CYB561 expression was negatively correlated with immune infiltration of B cells,plasmacytoid dendritic cells,dendritic cells,and neutrophils.Conclusion:CYB561 may serve as a potential biomarker for breast cancer diagnosis and prognosis.XI YANG HUIXIAN WU CHAO XIONG BO ZHAO MEILIAN LIU JIE QIN MEI DENG 2023BIOCELL2023,47,5:0
