1ALK,the Key Gene for Gelatinization Temperature,is a Modifier Gene for Gel Consistency in Rice显示文摘Gelatinization temperature(GT) is an important parameter in evaluating the cooking and eating quality of rice.Indeed,the phenotype,biochemistry and inheritance of GT have been widely studied in recent times.Previous map-based cloning revealed that GT was controlled by ALK gene,which encodes a putative soluble starch synthase II-3.Complementation vector and RNAi vector were constructed and transformed into Nipponbare mediated by Agrobacterium.Phenotypic and molecular analyses of transgenic lines provided direct evidence for ALK as a key gene for GT.Meanwhile,amylose content,gel consistency and pasting properties were also affected in transgenic lines.Two of four nonsynonymous single nucleotide polymorphisms in coding sequence of ALK were identified as essential for GT.Based on the single nucleotide polymorphisms(SNPs),two new sets of SNP markers combined with one cleaved amplified polymorphic sequence marker were developed for application in rice quality breeding.Zhenyu Gao Dali Zeng Fangmin Cheng Zhixi Tian Longbiao Guo Yan Su Meixian Yan Hua Jiang Guojun Dong Yuchen Huang Bin Han Jiayang Li Qian Qian 2011Journal of Integrative Plant Biology2011,53,9:13
2Hysteresis in sap flow and its controlling mechanisms for a deciduous broad-leaved tree species in a humid karst region显示文摘The critical zone(CZ) represents the intersection of the biosphere with the atmosphere, hydrosphere and lithosphere.Understanding the hydrological processes and human impact factors on the CZ is fundamental to sustainable water resources management for agroforestry. Transpiration(T) is an important component of terrestrial evapotranspiration(ET), and understanding the time lag(TL) between vegetation transpiration and meteorological factors can improve our knowledge of the mechanisms of vegetation adaptability to a changing environment. However, the controlling factors on the TL remain poorly understood. Therefore, the objective of this study is identifying the temporal dynamics of key controlling factors on the TL, using a typical deciduous broad-leaved tree species(Zenia insigins Chun) of CZ in subtropical humid karst regions. This species is used as an example to explore the characteristics of the TL between SF(sap flow) and hydro-meteorological forcing. Sap flow in these 6 trees was monitored using the thermal dissipation probes(TDP). Results showed that:(1) the peak of diurnal sap flow generally lagged behind PAR but preceded Ta(air temperature), RH(relative humidity) and VPD(vapor pressure deficit), with the mean TL of-67.4 min(PAR), 90.5 min(Ta), 91.6 min(RH) and 92.9 min(VPD), respectively;(2) TL had no significant relationships with the daily mean meteorological factors and soil moisture, but was highly(R^2>0.66) correlated to CRs(changing rates of meteorological factors) in the morning;(3) At seasonal scale, the sap flow rate and TL both were controlled by the seasonality of precipitation and temperature. Overall, the seasonality of the TL was caused by plants' high water loss and strongly active physiological response in hot seasons, leading to close stomata earlier than in cold seasons;(4) The reason why CRs proposed can explain the TL better than mean values of metrological factors is that the CRs considered the distribution and change processes of metrological factors in the daytime. 3Identification and functional analysis of the MOC1 interacting protein 1显示文摘Rice tillering is one of the most important agronomic traits that determine grain yields.Our previous study has demonstrated that the MONOCULM1 (MOC1) gene is a key component that controls the formation of rice tiller buds.To further elucidate the molecular mechanism of MOC1 involved in the regulation of rice tillering,we performed a yeast-two-hybrid screening to identify MOC1 interacting proteins (MIPs).Here we reported that MIP1 interacted with MOC1 both in vitro and in vivo.The overexpression of MIP1 resulted in enhanced tillering and reduced plant height.In-depth characterization of the context of MIP1 and MOC1 would further our understanding of molecular regulatory mechanisms of rice tillering.Fengli Sun Weiping Zhang Guosheng Xiong Meixian Yan Qian Qian Jiayang Li Yonghong Wang 2010Journal of Genetics and Genomics2010,37,1:9

5Abnormal Adipokines Associated with Various Types of Obesity in Chinese Children and Adolescents显示文摘Objective To explore the role of adipokines including insulin, resistin, leptin, adiponectin, acylation stimulating protein (ASP) and complement C3 (C3) in various types of obesity (peripheral obesity, abdominal obesity and mixed obesity) in Chinese children and adolescents, and their relationships with body size and pubertal development. Methods Children and adolescents (n=3 508) aged 6 to 18 years, with 1 788 boys and 1 720 girls were assessed for body mass index, waist circumference, pubertal development, blood insulin, resistin, leptin, adiponectin, ASP and C3 levels. Three types of obesity [peripheral obesity (n=43), abdominal obesity (n=473), mixed obesity (n=1 187)] and non‐obese control (n=1 805) were defined with combined use of Chinese body mass index and waist circumference criteria. Results Serum resistin, leptin and adiponectin levels were higher in girls than those in boys (all P<0.01). Insulin and leptin increased and adiponectin decreased across five Tanner stages in both girls and boys (all P<0.001), while ASP changed only in girls (P<0.001) and C3 only in boys (P<0.001). Insulin, leptin and ASP were higher, but adiponectin was lower in all three types of obesity vs. the non‐obese control (all P<0.05). The greatest abnormalities of all six adipokines were found in the mixed obesity group. With inclusion of body mass index and waist circumference in simultaneous regression analyses, both body size indices were independently and significantly correlated with insulin, leptin and adiponectin after age and gender adjustment. Compared with waist circumference, the body mass index was stronger in interpreting insulin, leptin, adiponectin and ASP levels, whereas it was weaker in explaining variance of plasma C3. Conclusions Obese children have a worse metabolic profile with high insulin, resistin, leptin, ASP and C3, and low adiponectin levels. The adipokine profile in mixed obesity is worse than that in peripheral or abdominal obesity. 6In vitro comparison of the hemocompatibility of diamond-like carbon and carbon nitride coatings with different atomic percentages of N显示文摘Carbon nitride (CNx) and diamond-like carbon (DLC) coatings were prepared by dc magnetron sputtering at room temperature. Different partial pressures of N2 were used to synthesize CNx to evaluate the relationship between the atomic percentage of nitrogen and hemocompatibility. Auger electron spectroscopy and atomic force microscopy indicated atomic percentages of N of 0.12 and 0.22 and that the CNx coatings were smooth. An in vitro study of the hemocompatibility of the coatings revealed that both CNx coatings had better anticoagulant properties and lower platelet adhesion than DLC. Compared with CN0.12, the CN0.22 coating showed longer dynamic clotting time (about 42 min), static clotting time (23.6 min) and recalcification time (45.6 s), as well as lower platelet adhesion (102 cells μm 2), aggregation, and activation. 8Identification and characterization of Mini1, a gene regulating rice shoot development显示文摘The aerial parts of higher plants are generated from the shoot apical meristem(SAM). In this study, we isolated a small rice(Oryza sativa L.) mutant that showed premature termination of shoot development and was named mini rice 1(mini1). The mutant was first isolated from a japonica cultivar Zhonghua11(ZH11) subjected to ethyl methanesulfonate(EMS)treatment. With bulked segregant analysis(BSA) and map-based cloning method, Mini1 gene was finally fine-mapped to an interval of 48.6 kb on chromosome 9. Sequence analyses revealed a single base substitution from G to A was found in the region, which resulted in an amino acid change from Gly to Asp.The candidate gene Os09g0363900 was predicted to encode a putative adhesion of calyx edges protein ACE(putative HOTHEAD precursor) and genetic complementation experiment confirmed the identity of Mini1. Os09g0363900 contains glucose-methanol-choline(GMC) oxidoreductase and NAD(P)-binding Rossmann-like domain, and exhibits high similarity to Arabidopsis HOTHEAD(HTH). 9Enhanced catalysis of ultrasmall Au-MoS_2 clusters against reactive oxygen species for radiation protection显示文摘Ionizing radiation produces excessive reactive oxygen species(ROS) which impose detrimental effects on biological systems. Thus, it is important to explore clinically safe and efficacious radioprotection agents to scavenge ROS and reduce the risks of radiotherapy. Recently, emerging catalytic nanomaterials such as sulfide nanomaterials have shown capability of clearing ROS in vivo by unique electron transfers between atoms, but their catalytic activities are yet suboptimal. As such, there is an unmet need to improve catalytic properties for stronger antioxidant activities and radiation protection. Herein, we prepared ultrasmall Au-MoS_2 clusters(~2.5 nm) and they showed enhanced catalytic properties via gold intercalation facilitating increased active sites and synergistic effects. Electrocatalysis results revealed that the catalytic activity of Au-MoS_2 towards H2O_2 was superior to ultrasmall MoS_2 without Au. As a result, we found that improving the electrocatalytic property of Au-MoS_2 can effectively enhance corresponding antioxidant activities and radioprotection effects in vivo. In addition, Au-MoS_2 also showed significant radioprotection in vitro and dramatically reduced the excess of radiation-induced adverse ROS. 11C-dots assisted synthesis of gold nanoparticles as labels to catalyze copper deposition for ultrasensitive electrochemical sensing of proteins显示文摘We report an ultrasensitive protocol for electrochemical sensing using the hydroxyl-rich C-dots assisted synthesis of gold nanoparticles(C-dots@AuNP) as labels with copper depositon reaction. The C-dots catalyzing copper deposition reaction was implemented for the first time. We constructed a sandwich-type immunosensor on the chitosan modified glassy carbon electrode(GCE) by glutaraldehyde(GA) crosslinking, with C-dots@AuNP as biolabels. Copper was deposited on the catalytic surfaces of second antibody-conjugated C-dots@AuNP nanoparticles through CuSO_4-ascorbic acid reduction, because both C-dots and AuNPs could strongly catalyze the CuSO_4 and ascorbic acid to form Cu particles, which amplified the detection signal. Then the corresponding antigen was quantified based on simultaneous chemical-dissolution/cathodic-preconcentration of copper for insitu analysis using anodic stripping square wave voltammetry(ASSWV) directly on the modified electrode. Under optimized conditions, these electrodes were employed for sandwich-type immunoanaly sis, pushing the lower limits of detection(LODs)down to the fg mL^(-1) level for human immunoglobulin G(IgG) and cardiac troponin I(cTnI), a cardiac biomarker.
