2Chinese progress in geomorphometry显示文摘Geomorphometry,the science of digital terrain analysis(DTA),is an important focus of research in both geomorphology and geographical information science(GIS).Given that 70% of China is mountainous,geomorphological research is popular among Chinese scholars,and the development of GIS over the last 30 years has led to significant advances in geomorphometric research.In this paper,we review Chinese progress in geomorphometry based on the published literature.There are three major areas of progress:digital terrain modelling methods,DTA methods,and applications of digital terrain models(DTMs).First,traditional vector-and raster-based terrain modelling methods,including the assessment of uncertainty,have received widespread attention.New terrain modelling methods such as unified raster and vector,high-fidelity,and real-time dynamic geographical scene modelling have also attracted research attention and are now a major focus of digital terrain modelling research.Second,in addition to the popular DTA methods based on topographical derivatives,geomorphological features,and hydrological factors extracted from DTMs,DTA methods have been extended to include analyses of the structure of underlying strata,ocean surface features and even socioeconomic spatial structures.Third,DTMs have been applied to fields including global climate change,analysis of various typical regions,lunar surface and other related fields.Clearly,Chinese scholars have made significant progress in geomorphometry.Chinese scholars have had the greatest international impact in areas including high-fidelity digital terrain modelling and DTM-based regional geomorphological analysis,particularly in the Loess Plateau and the Tibetan Plateau regions.LV Guonian XIONG Liyang CHEN Min TANG Guoan SHENG Yehua LIU Xuejun SONG Zhiyao LU Yuqi YU Zhaoyuan ZHANG Ka WANG Meizhen 2017Journal of Geographical Sciences2017,27,11:14
3Generation of obese rat model by transcription activator-like effector nucleases targeting the leptin receptor gene显示文摘The laboratory rat is a valuable mammalian model organism for basic research and drug discovery. Here we demonstrate an efficient methodology by applying transcription activator-like effector nucleases(TALENs) technology to generate Leptin receptor(Lepr) knockout rats on the Sprague Dawley(SD) genetic background. Through direct injection of in vitro transcribed m RNA of TALEN pairs into SD rat zygotes, somatic mutations were induced in two of three resulting pups. One of the founders carrying bi-allelic mutation exhibited early onset of obesity and infertility. The other founder carried a chimeric mutation which was efficiently transmitted to the progenies. Through phenotyping of the resulting three lines of rats bearing distinct mutations in the Lepr locus, we found that the strains with a frame-shifted or premature stop codon mutation led to obesity and metabolic disorders. However, no obvious defect was observed in a strain with an in-frame 57 bp deletion in the extracellular domain of Lepr. This suggests the deleted amino acids do not significantly affect Lepr structure and function. This is the first report of generating the Lepr mutant obese rat model in SD strain through a reverse genetic approach. 5Identification of m RNA-like non-coding RNAs and validation of a mighty one named MAR in Panax ginseng显示文摘Increasing evidence suggests that long non-coding RNAs(lnc RNAs) play significant roles in plants.However,little is known about lnc RNAs in Panax ginseng C.A.Meyer,an economically significant medicinal plant species.A total of3,688 m RNA-like non-coding RNAs(mlnc RNAs),a class of lnc RNAs,were identified in P.ginseng.Approximately 40% of the identified mlnc RNAs were processed into small RNAs,implying their regulatory roles via small RNA-mediated mechanisms.Eleven mi RNA-generating mlnc RNAs also produced si RNAs,suggesting the coordinated production of mi RNAs and si RNAs in P.ginseng.The mlnc RNA-derived small RNAs might be 21-,22-,or 24-nt phased and could be generated from both or only one strand of mlnc RNAs,or from super long hairpin structures.A full-length mlnc RNA,termed MAR(multiple-function-associated mlnc RNA),was cloned.It generated the most abundant si RNAs.The MAR si RNAs were Researchpredominantly 24-nt and some of them were distributed in a phased pattern.A total of 228 targets were predicted for 71 MAR si RNAs.Degradome sequencing validated 68 predicted targets involved in diverse metabolic pathways,suggesting the significance of MAR in P.ginseng.Consistently,MAR was detected in all tissues analyzed and responded to methyl jasmonate(Me JA) treatment.It sheds light on the function of mlncRNAs in plants.Meizhen Wang Bin Wu Chao Chen Shanfa Lu 2015Journal of Integrative Plant Biology2015,57,3:2
6Predictive models for characterizing the atomization process in pyrolysis of methyl ricinoleate显示文摘Pyrolysis of methyl ricinoleate(MR)can produce undecylenic acid methyl ester and heptanal which are important chemicals.Atomization feeding favors the heat exchange in the pyrolysis process and hence increases the product yield.Herein,predictive models to characterize the atomization process were developed.The effect of spray distance on Sauter mean diameter(SMD)of atomized MR droplets was examined,with the optimal spray distance to be 40-50 mm.Temperature mainly affected the physical properties of feedstock,with smaller droplet size obtained at increasing temperature.In addition,pressure had significant influence on SMD and higher pressure resulted in smaller atomized droplets.Then,a model for SMD prediction,combining temperature,pressure,spray distance,and structural parameters of nozzle,was developed through dimensionless analysis.The results showed that SMD was a power function of Reynolds number(Re),Ohnesorge number(Oh),and the ratio of spray distance to diameter of swirl chamber in the nozzle(H/dsc),with the exponents of-1.6618,-1.3205 and 0.1038,respectively.The experimental measured SMD was in good agreement with the calculated values,with the error within±15%.Moreover,the droplet size distribution was studied by establishing the relationship between the standard deviation of droplet size and SMD.This study could provide reference to the regulation and optimization of the atomization process in MR pyrolysis.Xiaoning Mao Qinglong Xie Ying Duan Shangzhi Yu Xiaojiang Liang Zhenyu Wu Meizhen Lu Yong Nie 2020Chinese Journal of Chemical Engineering2020,28,4:1

8Long-term correction of hemophilia B through CRISPR/Cas9 induced homology-independent targeted integration显示文摘CRISPR/Cas9-mediated site-specific insertion of exogenous genes holds potential for clinical applications.However,it is still infeasible because homologous recombination(HR)is inefficient,especially for nondividing cells.To overcome the challenge,we report that a homology-independent targeted integration(HITI)strategy is used for permanent integration of high-specificity-activity Factor IX variant(F9 Padua,R338L)at the albumin(Alb)locus in a novel hemophilia B(HB)rat model.The knock-in efficiency reaches 3.66%,as determined by droplet digital PCR(dd PCR).The clotting time is reduced to a normal level four weeks after treatment,and the circulating factor IX(FIX)level is gradually increased up to 52%of the normal level over nine months even after partial hepatectomy,demonstrating the amelioration of hemophilia.Through primer-extension-mediated sequencing(PEM-seq),no significant off-target effect is detected.This study not only provides a novel model for HB but also identifies a promising therapeutic approach for rare inherited diseases.Xi Chen Xuran Niu Yang Liu Rui Zheng Lei Yang Jian Lu Shuming Yin Yu Wei Jiahao Pan Ahmed Sayed Xueyun Ma Meizhen Liu Fengxiang Jing Mingyao Liu Jiazhi Hu Liren Wang Dali Li 2022Journal of Genetics and Genomics2022,49,12:0
