| 1 | Liuwei Dihuang pill suppresses metastasis by regulating the wnt pathway and disrupting β-catenin/T cell factor interactions in a murine model of triple-negative breast cancer显示文摘OBJECTIVE:To investigate if the Liuwei Dihuang pill(LWDHP)can inhibit metastasis to the liver and lungs in mice bearing triple-negative breast cancer(TNBC),and the molecular mechanism underpinning this action.METHODS:Ninety-nine TNBC bearing-mice were distributed randomly to five groups:control(Con),paclitaxel(PTX),low-dose LWDHP(LLP,2.3 g·kg^-1·d^-1),middle-dose LWDHP(MLP,4.6 g·kg^-1·d^-1)and high-dose LWDHP(HLP,9.2 g·kg^-1·d^-1).The LWDHP were administered(p.o.)to the agonal stage.The morphology of BC cells was observed by hematoxylin&eosin staining.Expression of axin-2,β-catenin,T cell factor(TCF),cyclin-D1 and vascular endothelial growth factor(VEGF)was detected by western blotting or immunofluorescence.β-catenin/TCF-1 interaction was measured using a co-immunoprecipitation assay.RESULTS:After LWDHP treatment,metastasis of BC cells to the lungs and liver was inhibited,expression of axin-2 was increased,expression of TCF-1,β-catenin,cyclin-D1 and VEGF was decreased,andβ-catenin/TCF-1 interaction was disrupted.CONCLUSION:The LWDHP could inhibit metastasis of BC cells to the liver and lungs.The molecular mechanism underlying this action may be regulation of protein expression andβ-catenin/TCF-1 interactions in the Wnt pathway. | Zheng Lixiang Zheng Qing Yu Zhipeng Wang Jian Ren Xiaoying Gong Yan Yang Xue Hunag Ping Weng Meizhi Liu Hongning Liu Haizhou | 2019 | Journal of Traditional Chinese Medicine2019,39,6: | 9 |
| 2 | Status and development of high-power laser facilities at the NLHPLP显示文摘In this paper, we review the status of the multifunctional experimental platform at the National Laboratory of High Power Laser and Physics(NLHPLP). The platform, including the SG-II laser facility, SG-II 9th beam, SG-II upgrade(SG-II UP) facility, and SG-II 5 PW facility, is operational and available for interested scientists studying inertial confinement fusion(ICF) and a broad range of high-energy-density physics. These facilities can provide important experimental capabilities by combining different pulse widths of nanosecond, picosecond, and femtosecond scales. In addition, the SG-II UP facility, consisting of a single petawatt system and an eight-beam nanosecond system, is introduced including several laser technologies that have been developed to ensure the performance of the facility. | 3 | Analysis and construction status of SG-Ⅱ 5PW laser facility显示文摘We present a recent progress of the SG-II 5 PW facility, which designed a multi-petawatt ultrashort pulse laser based on optical parametric chirped-pulse amplification(OPCPA). The prior two optical parametric amplifiers have been accomplished and chirped pulses with an energy of 49.7 J and a full-width-at-half-maximum(FWHM) spectrum bandwidth of 85 nm have been achieved. In the PW-scale optical parametric amplification(OPA), with the pump pulse that has an energy of 118 J from the second harmonic generation of the SG-II 7 th beam, the pump-to-signal conversion efficiency is up to 41.9%, which to the best of our knowledge is the highest among all of the reported values for OPCPA systems. The compressed pulse is higher than 37 J in 21 fs(1.76 PW), and the focal spot is ~10 μm after the closed-loop corrections by the adaptive optics. Limited by the repetition of the pump laser, the SG-II 5 PW facility operates one shot per hour. | 4 | Thermostability of Subtilisin Nattokinase Obtained by Site-Directed Mutagenesis显示文摘To study the thermostability of Nattokinase(subtilisin NAT,NK),three double mutant plasmids(pET-28a-NKG61C/S98C,pET-28a-NKT22C/S87C,pET-28a-NKS24C/S87C)were constructed by site-directed mutagenesis.Target enzymes were detected using SDS-PAGE and disulfide bond formation was detected using Western blotting analysis.Thermostability was tested by rates of inactivation at certain temperature.The results showed that disulfide bond was not formed within two cysteines and the thermostability of three double mutants was not increased compared with the wild-type NK.The thermostability of NK performed in Ca2+was stronger than in ethylenediaminetetraacetic acid(EDTA).But when the temperature reached 62℃,the enzymes rapidly denatured and inactivated even in the presence of Ca2+.Although the thermostability of mutants was not increased,this study shows a tendency of improving thermostability of NK in protein engineering. | WENG Meizhi DENG Xiongwei WU Jieyuan ZOU Guolin | 2014 | Wuhan University Journal of Natural Sciences2014,19,3: | 6 |
| 5 | Design and experimental demonstration of a high conversion efficiency OPCPA pre-amplifier for petawatt laser facility显示文摘We present the design and experiment of a broadband optical parametric chirped-pulse amplifier(OPCPA) which provides high conversion efficiency and good beam quality at 808 nm wavelength. Using a three-dimensional spatial and temporal numerical model, several design considerations necessary to achieve high conversion efficiency, good beam quality and good output stability are discussed. To improve the conversion efficiency and broaden the amplified signal bandwidth simultaneously, the nonlinear crystal length and OPCPA parameters are analyzed and optimized with the concept of dissipating amplified idler between optical parametric amplification(OPA) of two crystals configuration.In the experiment, an amplifier consisting of two OPCPA stages of ‘L’ type configuration was demonstrated by using the optimized parameters. An amplified signal energy of 160 mJ was achieved with a total pump-to-signal efficiency of 35%(43% efficiency for the OPCPA stage 2). The output bandwidth of signal pulse reached 80 nm and the signal pulse was compressed to 24 fs. The energy stability reached 1.67% RMS at 3% pump energy variation. | 6 | Outline of Metallogenic Regularity of Bauxite Deposits in China显示文摘Among the abundant aluminum ore resources in China, bauxite is dominated, which is mainly distributed in 19 provinces and regions, including Shanxi, Henan, Guizhou and Guangxi. The major deposit type of bauxite is paleo-weathering crust sedimentary type, and the other one is the accumulation type. The main metallogenic period is the late Paleozoic Era followed by the Cenozoic Era. The metallogenic tectonic background is characterized by a cratonic environment. This paper summarizes the bauxite metallogenic regularity based on the characteristics of bauxite resources, bauxite deposit type, bauxite metallogenic belt and metallogenic series in China, and 15 bauxite metallogenic belts, 8 bauxite metallogenic series and 7 bauxite ore concentrated areas were identified in the study. Neofusicoccum parvum was isolated from these cankered tissues, with the identification based on morphology and an ITS-nr DNA sequence. In order to demonstrate how cultures of N. parvum could cause the expected symptoms, artificial infection, using these isolates and re-isolation of the pathogen, was used. Their structures, including the absolute configurations,were determined by spectroscopic analyses and ECD experiments. Meroterpenoids are special natural products that originate from mixed terpenoid-nonterpenoid pathway. As a member of meroterpenoid family, diisoprenyl-cyclohexene/ane-type meroterpenoids composed of two isoprenyl chains(C5 unit)and a cyclohexene/ane moiety(C6 unit), featuring diverse skeleton structures with wide range of bioactivities. In these reported diisoprenyl-cyclohexene/ane-type meroterpenoids, only three dimers were identified. | 9 | Contents Variation Analysis ofγ-Amino Butyric Acid in Semen sojae praeparatum Fermentation Using Online Pre-Column Derivatization-HPLC显示文摘This paper reported the contents variation analysis ofγ-amino butyric acid(GABA)in Semen sojae praeparatum(SSP)which is a famous traditional Chinese medicine.High performance liquid chromatography(HPLC)was used and GABA was derivatized by online pre-column derivatization with o-phthalaldehyde(OPA).To validate this method,the precision,stability,repeatability and recovery were discussed.In the concentration range from 0.0125 to 0.400 mg/m L,the calibration curve for GABA was linear and the regression equation was obtained with correlation coefficient(R2)of 0.9999.Relatively high levels of GABA exist in SSP and the content changes of GABA at different time points during the fermenting process were detected.At the'yellow cladding'stage,GABA level was very low or even undetectable;the'secondary fermentation'stage witnessed a rapid increase of GABA content to 1.39-5.52 mg/g,which remained stable after 18 days of'secondary fermentation'.This study demonstrated that GABA was generated at the'secondary fermentation'stage,revealing the significance and rationality of the'secondary fermentation'stage in the fermenting process of SSP.On the other hand,it suggested the downside of taking soy isoflavones as the only measurement in existing quality assessment and optimization approach for the fermenting process of SSP. | WENG Meizhi HE Jing REN Jiaxiu LIANG Yonghong CHEN Qingfeng SU Mingsheng WANG Liyuan XIE Xiaomei | 2021 | Wuhan University Journal of Natural Sciences2021,26,1: | 2 |

| 15 | Systematic study of spatiotemporal influences on temporal contrast in the focal region in large-aperture broadband ultrashort petawatt lasers显示文摘Temporal contrast is one of the crucial physical determinants which guarantee the successful performance of laser–matter interaction experiments. We generally reviewed the influences on the temporal contrast in three categories of noises based on the requirement by the physical mechanisms. The spatiotemporal influences on temporal contrast at the focal region of the chromatic aberration and propagation time difference introduced by large-aperture broadband spatial filters, which were spatiotemporally coupled with compression and focusing, were calculated and discussed with a practical case in SG-Ⅱ5 PW ultrashort petawatt laser. | 16 | Intersected functional zone of transcriptional regulators patterns stemness within stem cell niche of root apical meristem显示文摘Root stem cell niche(SCN)consists of a quiescent center(QC)and surrounding stem cells.Disrupted symplastic communication leads to loss of stemness in the whole SCN.Several SCN regulators were reported to move between cells for SCN main-tenance.However,single mutant of these regulators is insu?cient to abolish QC stemness despite the high differentiation rate in surrounding stem cells.To dis-sect the mechanism behind such distinct stemness in SCN,we combined the mis-expression strategy with pWOX5:icals3m system in which QC is symplastically isolated.We found the starch accumulation in QC could be synergistically repressed by WUSCHEL-RE-LATED HOMEOBOX 5(WOX5),SHORT-ROOT(SHR),SCARCROW(SCR),and PLETHORA(PLT).Like PLTs,other core regulators also exhibited dimorphicfunctions by inhibiting differentiation at a higher dose while promoting cell division at a low protein level.Being located in the center of the intersected ex-pression zones,QC cells receive the highest level of core regulators,forming the most robust stemness within SCN.WUSCHEL-RELATED HOMEOBOX 5 was su?cient to activate PLT1/2 expression,contributing to the QC-enriched PLTs.Our results provide ex-perimental evidence supporting the long-standing hypothesis that the combination of spatial ex-pression,synergistic function and dosage effect of core regulators result in spatially distinct stemness in SCN. | Meizhi Xu Xu Gu Nengsong Liang Xinxin Bian Hong Wang Yaxin Qin Limin Pi Shuang Wu | 2020 | Journal of Integrative Plant Biology2020,62,7: | 1 |
