1Up-Regulation of Trem2 Inhibits Hippocampal Neuronal Apoptosis and Alleviates Oxidative Stress in Epilepsy via the PI3K/Akt Pathway in Mice显示文摘Epilepsy is a chronic and severe neurological disorder that has negative effects on the autonomous activities of patients. Functionally, Trem2(triggering receptor expressed on myeloid cells-2) is an immunoglobulin receptor that affects neurological and psychiatric genetic diseases. Based on this rationale, we aimed to assess the potential role of Trem2 integration with the PI3 K/Akt pathway in epilepsy. We used microarray-based gene expression profiling to identify epilepsy-related differentially-expressed genes. In a mouse hippocampal neuron model of epilepsy, neurons were treated with lowMg^2+ extracellular fluid, and the protein and mRNA expression of Trem2 were determined. Using a gain-offunction approach with Trem2, neuronal apoptosis and its related factors were assessed by flow cytometry, RT-qPCR,and Western blot analysis. In a pilocarpine-induced epileptic mouse model, the malondialdehyde(MDA) and8-hydroxy-20-deoxyguanosine(8-OHdG) content and superoxide dismutase(SOD) and glutathione-peroxidase(GSH-Px) activity in the hippocampus were determined,and the protein expression of Trem2 was measured. In addition, the regulatory effect of Trem2 on the PI3 K/Akt pathway was analyzed by inhibiting this pathway in both the cell and mouse models of epilepsy. Trem2 was found to occupy a core position and was correlated with epilepsy.Trem2 was decreased in the hippocampus of epileptic miceand epileptic hippocampal neurons. Of crucial importance,overexpression of Trem2 activated the PI3 K/Akt pathway to inhibit neuronal apoptosis. Moreover, activation of the PI3 K/Akt pathway through over-expression of Trem2 alleviated oxidative stress, as shown by the increased expression of SOD and GSH-Px and the decreased expression of MDA and 8-OHdG. 2A novel gene delivery system targeting cells expressing VEGF receptors显示文摘Two ligand oligopeptides GV1 and GV2 were designed according to the putative binding region of VEGF to its receptors. GV1, GV2 and endosome releasing oligopeptide HA20 were conjugated with poly-L-lysine or protamine and the resulting conjugates could interact with DNA in a noncovalent bond to form a complex. Using pSV2-β-galactosidase as a reporter gene, it has been demonstrated that exogenous gene was transferred into bovine aortic arch-derived endothelial cells (ABAE) andhuman malignant melanoma cell lines (A375) in vitro. In vivo experiments, exogenous gene was transferred into tumor vascular endothelial cells and tumor cells of subcutaneously transplanted human colon cancer LOVO, human malignant melanoma A375 and human hepatoma graft in nude mice. This system could also target gene to intrahepatically transplanted human hepatoma injected via portal vein in nude mice. 4Effect of various crop rotations on rice yield and nitrogen use efficiency in paddy–upland systems in southeastern China显示文摘To evaluate the effects of various rotation systems on rice grain yield and N use efficiency, a paddy–upland cropping experiment(2013–2016) was conducted in southeastern China. The experiment was designed using six different rice––winter crop rotations: rice–fallow(RF),rice–wheat(RW), rice–potato with rice straw mulch(RP), rice–green manure(Chinese milk vetch; RC–G), rice–oilseed rape(RO), and rice–green manure crop(oilseed rape with fresh straw incorporated into soil at flowering; RO–G) and three N rates, N0(0 kg N ha-1), N1(142.5 kg N ha-1), and N2(202.5 kg N ha-1). Average rice yields in the RF(5.93 t ha-1) rotation were significantly lower than those in the rotations with winter crops(7.20–7.48 t ha-1)under the N0 treatment, suggesting that incorporation of straw might be more effective for increasing soil N than winter fallow. The rice yield differences among the rotations varied by year with the N input. In general, the grain yields in the RP and RO–G rotations –were respectively 11.6–28.5% and 14.80–37.19% higher than those in the RF in plots with N applied. Increasing the N rate may have tended to minimize the average yield gap between the RF and the other rotations; the yield gaps were 18.55%, 4.14%, and 0.23% in N0, N1, and N2, respectively. However, the N recovery efficiency in the RF was significantly lower than that in other rotations, except for 2015 under both N1 and N2 rates, a finding that implies a large amount of chemical N loss. No significant differences in nitrogen agronomic efficiency(NAE) and physiological efficiency(NPE) were found between the rotations with legume(RC–G) and non–legume(RO and RW) winter crops, a result that may be due partly to straw incorporation. For this reason, we concluded that the return of straw could reduce differences in N use efficiency between rotations with and without legume crops. The degree of synchrony between the crop N demand and the N supply was evaluated by comparison of nitrogen balance degree(NBD) values. The NBD values in the RP and RW were significantly lower than those in the other rotations under both N1 and N2 rates. Thus,in view of the higher grain yield in the RP compared to the RW under the N1 rate, the RP rotation might be a promising practice with comparable grain yield and greater N use efficiency under reduced N input relative to the other rotations. The primary yield components of the RF and RP were identified as number of panicles m-2 and numbers of kernels panicle-1, respectively. 8DDIT4 promotes gastric cancer proliferation and tumorigenesis through the p53 and MAPK pathways显示文摘Background:Gastric cancer(GC)is one of the most common malignancies worldwide,particularly in China.DNA damage-inducible transcript 4(DDIT4)is a mammalian target of rapamycin inhibitor and is induced by various cellular stresses;however,its critical role in GC remains poorly understood.The present study aimed to investigate the poten-tial relationship and the underlying mechanism between DDIT4 and GC development.Methods:We used western blotting,real-time polymerase chain reaction,and immunohistochemical or immunoflu-orescence to determine DDIT4 expression in GC cells and tissues.High-content screening,cell counting kit-8 assays,colony formation,and in vivo tumorigenesis assays were performed to evaluate cell proliferation.Flow cytometry was used to investigate cell apoptosis and cell cycle distribution.Results:DDIT4 was upregulated in GC cells and tissue.Furthermore,downregulating DDIT4 in GC cells inhibited proliferation both in vitro and in vivo and increased 5-fluorouracil-induced apoptosis and cell cycle arrest.In contrast,ectopic expression of DDIT4 in normal gastric epithelial cells promoted proliferation and attenuated chemosensitivity.Further analysis indicated that the mitogen-activated protein kinase and p53 signaling pathways were involved in the suppression of proliferation,and increased chemosensitivity upon DDIT4 downregulation.Conclusion:DDIT4 promotes GC proliferation and tumorigenesis,providing new insights into the role of DDIT4 in the tumorigenesis of human GC.Feng Du Lina Sun Yi Chu Tingyu Li Chao Lei Xin Wang Mingzuo Jiang Yali Min Yuanyuan Lu Xiaodi Zhao Yongzhan Nie Daiming Fan 2018Cancer Communications2018,38,1:10
9Novel SNP of EPAS1 gene associated with higher hemoglobin concentration revealed the hypoxia adaptation of yak(Bos grunniens)显示文摘Endothelial PAS domain protein 1 gene(EPAS1) is a key transcription factor that activates the expression of oxygen-regulated genes. In this study, in order to better understand the effects of EPAS1 gene on hematologic parameters in yak, we firstly quantified the tissue expression patterns for EPAS1 m RNA of yak, identified polymorphism in this gene and evaluated its association with hematologic parameters. Expression of EPAS1 m RNA was detected in all eight tissues(heart, liver, lung, spleen, pancreas, kidney, muscles and ovary). The expressions of EPAS1 in lung and pancreas were extremely higher than other tissues examined. Three novel single nucleotide polymorphisms(SNPs)(g.83052 C>T, g.83065 G>A and g.83067 C>A) within the EPAS1 were identified and genotyped in Pali(PL), Gannan(GN) and Tianzhu White(TZW) yak breeds. Significant higher frequencies of the AA and GA genotypes and A allele of the g.83065 G>A were observed in the PL and GN breeds than that in the TZW breed(P<0.01). Association analysis of the PL breed indicated that the g.83065 G>A polymorphism was significantly associated with hemoglobin(HGB) concentration in yaks(P<0.05). Individuals with genotype AA had significantly higher HGB concentration(P<0.05) than those with genotype GA and GG. 10PGE patterns of ores of Dajing Cu-polymetallic deposit in Linxi County, Inner Mongolia:Indicator to source of metallogenic elements显示文摘By comparing PGE patterns of sulfide ores with that of basic-ultrabasic dikes occurring in the Dajing Cu-polymetallic deposit, we discover that there are similar positive slopes between the basic-ultrabasic dikes and the black ores composed mainly of sphalerite and galena, reflecting affinity of their source. The PGE patterns of the yellow ores composed basically of chalcopyrite, arsenopyrite and pyrite are actually different. One is close to the pattern of the mantle 1herzolite, and others resemble the pattern of the basic rocks. However, the concentrations of Ru are higher in the yellow ores, reflecting that sulfur attains or approaches saturation in their parent magma. The PGE characteristics of ores and dikes show that the magmas derived from the deep of crust and the metallogenic elements were supplied mainly by growth material of the crust. 11Breaking the barriers: advances in acoustic functional materials显示文摘Acoustics is a classical field of study that has witnessed tremendous developments over the past 25 years.Driven by the novel acoustic effects underpinned by phononic crystals with periodic modulation of elastic building blocks in wavelength scale and acoustic metamaterials with localized resonant units in subwavelength scale, researchers in diverse disciplines of physics, mathematics, and engineering have pushed the boundary of possibilities beyond those long held as unbreakable limits. More recently, structure designs guided by the physics of graphene and topological electronic states of matter have further broadened the whole field of acoustic metamaterials by phenomena that reproduce the quantum effects classically. Use of active energy-gain components, directed by the parity–time reversal symmetry principle,has led to some previously unexpected wave characteristics. It is the intention of this review to trace historically these exciting developments, substantiated by brief accounts of the salient milestones. The latter can include, but are not limited to, zero/negative refraction, subwavelength imaging, sound cloaking, total sound absorption, metasurface and phase engineering, Dirac physics and topology-inspired acoustic engineering, non-Hermitian parity–time synthetic active metamaterials, and one-way propagation of sound waves. 12Simultaneous quantification of ginsenoside Rg1 and its metabolites by HPLC–MS/MS:Rg1 excretion in rat bile, urine and feces显示文摘Ginsenoside Rg1(Rg1), the major effective component of ginseng, has been shown to have multiple bioactivities, but low oral bioavailability. The aim of this study was to develop a simple, sensitive and rapid high performance liquid chromatography–tandem mass spectrometry(LC–MS/MS) method, which could be used to validate and quantify the concentrations of Rg1 and its metabolites in Sprague-Dawley rat bile,urine, and feces after oral administration(25 mg/kg). Calibration curves offered satisfactory linearity(r40.995)within the determined ranges. Both intra-day and inter-day variances were less than 15%, and the accuracy was within 80–120%. The excretion recoveries of Rg1, ginsenoside Rh1(Rh1), and protopanaxatriol(Ppt) in bile,urine, and feces combined were all greater than 70%. The fecal excretion recoveries of Rg1, Rh1, and Ppt were40.11%, 22.19%, and 22.88%, respectively, whereas 6.88% of Rg1 and 0.09% of Rh1 were excreted in bile.Urinary excretion accounted for only 0.04% of Rg1. 17A text-to-speech system with high intelligibility and naturalness for Chinese显示文摘A Chinese text-to-speech system, which is based on the time domain PitchSynchronous-Overlap-Add (PSOLA) method, with a Chinese syllable dictionary and a prosodicrule dictionaIy, can produce very clear and natural Chinese speech. Research work on naturalness of synthetic Chinese show that, when synthesizing Chinese, pitch, energy, syllable duration and coarticulation between syllables are main factors which affect the naturalness. Among them pitch and duration play the most important roles. The time domain PSOLA scheme provides a method to modify the pitch and duration of a speech segment in time domain, and this makes it possible to adjust the prosody of speech in word level and sentence level, when synthesizing Chinese using waveform concatenation technique. Acoustics analysis of news broadcast speech provides theoretical basis for building up prosodic rules in this system. 18Quasi-phase-matching-division multiplexing holography in a three-dimensional nonlinear photonic crystal显示文摘Nonlinear holography has recently emerged as a novel tool to reconstruct the encoded information at a new wavelength,which has important applications in optical display and optical encryption.However,this scheme still struggles with low conversion efficiency and ineffective multiplexing.In this work,we demonstrate a quasi-phasematching(QPM)-division multiplexing holography in a three-dimensional(3D)nonlinear photonic crystal(NPC).3D NPC works as a nonlinear hologram,in which multiple images are distributed into different Ewald spheres in reciprocal space.The reciprocal vectors locating in a given Ewald sphere are capable of fulfilling the complete QPM conditions for the high-efficiency reconstruction of the target image at the second-harmonic(SH)wave.One can easily switch the reconstructed SH images by changing the QPM condition.The multiplexing capacity is scalable with the period number of 3D NPC.Our work provides a promising strategy to achieve highly efficient nonlinear multiplexing holography for high-security and high-density storage of optical information.Pengcheng Chen Chaowei Wang Dunzhao Wei Yanlei Hu Xiaoyi Xu Jiawen Li Dong Wu Jianan Ma Shengyun Ji Leran Zhang Liqun Xu Tianxin Wang Chuan Xu Jiaru Chu Shining Zhu Min Xiao Yong Zhang 2021Light(Science & Applications)2021,10,8:5

20Effect of bevacizumab on the expression of fibrosis-related inflammatory mediators in ARPE-19 cells显示文摘AIM:To investigate the effect of anti-vascular epithelial growth factor(VEGF)agents on the expression of fibrosisrelated inflammatory mediators under normoxic and hypoxic conditions,and to further clarify the mechanism underlying fibrosis after anti-VEGF therapy. METHODS:Human retinal pigment epithelial(RPE)cells were incubated under normoxic and hypoxic conditions.For hypoxia treatment,CoCl_2 at 200μmol/L was added to the media. ARPE-19 cells were treated as following:1)control group:no treatment; 2)bevacizumab group:bevacizumab at 0.25 mg/mL was added to the media; 3)hypoxia group:CoCl_2 at 200 μmol/L was added to the media; 4)hypoxia+bevacizumab group:CoCl_2 at 200 μmol/L and bevacizumab at 0.25 mg/mL were added to the media.The expression of interleukin(IL)-1β,IL-6,IL-8 and tumor necrosis factor(TNF)-α were evaluated using real-time polymerase chain reaction(RT-PCR)and enzyme-linked immunosorbent assay(ELISA)at 6,12,24 and 48 h. RESULTS:Both m RNA and protein levels of IL-1β,IL-6 and IL-8 were statistically significantly higher in the bevacizumab group than in the control group at each time point,and TNF-α gene and protein expression was only significantly higher only at 24 and 48h(P<0.05). Under hypoxic conditions,bevacizumab significantly increased the expression of IL-1β,IL-6,IL-8 and TNF-α at 6,12,24 and 48h(P<0.05). IL-1β,IL-8 and TNF-α peaked at 24 h and IL-6 peaked at 12 h after the bevacizumab treatment under both normoxic and hypoxic conditions. CONCLUSION:Treatment of ARPE-19 cells with bevacizumab can significantly increase the expression of fibrosis-related inflammatory mediators under bothnormoxic and hypoxic conditions. Inflammatory factors might be involved in the process of fibrosis after antiVEGF therapy,and the up-regulation of inflammatory factors induced by anti-VEGF drugs might promote the fibrosis process.San-Jun Chu Zhao-Hua Zhang Min Wang Hai-Feng Xu 2017International Journal of Ophthalmology(English edition)2017,10,3:4
