1Microstructure, biodegradation, antibacterial and mechanical properties of ZK60-Cu alloys prepared by selective laser melting technique显示文摘Magnesium(Mg) alloys are receiving increasing attention for body implants owing to their good biocompatibility and biodegradability. However, they often suffer from bacterial infections on account of their insufficient antibacterial ability. In this study, ZK60-x Cu(x = 0, 0.2, 0.4, 0.6 and 0.8 wt%) alloys were prepared by selective laser melting(SLM) with alloying copper(Cu) to enhance their antibacterial ability.Results showed that ZK60-Cu alloys exhibited strong antibacterial ability due to combination of release of Cu ions and alkaline environment which could kill bacteria by destroying cellular membrane structure,denaturing enzymes and inhibiting deoxyribonucleic acid(DNA) replication. In addition, their compressive strength increased due to grain refinement and uniformly dispersing of short-bar shaped MgZnCu phases. Moreover, ZK60-Cu alloys also exhibited good cytocompatibility. 2MicroTom Metabolic Network: Rewiring Tomato Metabolic Regulatory Network throughout the Growth Cycle显示文摘Tomato(Solanum lycopersicum)is a major horticultural crop worldwide and has emerged as a preeminent model for metabolic research.Although many research efforts have focused on the analysis of metabolite differences between varieties and species,the dynamics of metabolic changes during the tomato growth cycle and the regulatory networks that underlie these changes are poorly understood.In this study,we integrated high-resolution spatio-temporal metabolome and transcriptome data to systematically explore the metabolic landscape across 20 major tomato tissues and growth stages.In the resulting MicroTom Metabolic Network,the 540 detected metabolites and their co-expressed genes could be divided into 10 distinct clusters based on their biological functions.Using this dataset,we constructed a global map of the major metabolic changes that occur throughout the tomato growth cycle and dissected the underlying regulatory network.In addition to verifying previously well-established regulatory networks for important metabolites,we identified novel transcription factors that regulate the biosynthesis of important secondary metabolites such as steroidal glycoalkaloids and flavonoids.Our findings provide insights into spatiotemporal changes in tomato metabolism and generate a valuable resource for the study of metabolic regulatory processes in model plants.Yan Li Yang Chen Lu Zhou Shengjie You Heng Deng Ya Chen Saleh Alseekh Yong Yuan Rao Fu Zixin Zhang Dan Su Alisdair R.Fernie Mondher Bouzayen Tao Ma Mingchun Liu Yang Zhang 2020Molecular Plant2020,13,8:10
3Two Types of Granites in the Western Yangtze Block and Their Implications for Regional Tectonic Evolution: Constraints from Geochemistry and Isotopic Data显示文摘In the western Yangtze Block, widespread Mesoproterozoic to Neoproterozoic rocks are the key to understanding the Precambrian tectonic-magmatic evolution of the region. However, their petrogenesis and tectonic setting are still controversial. In this paper, zircon U-Pb ages, Sm-Nd isotopic and whole-rock geochemical data are reported from selected fresh samples in the southern Dechang county, southwestern China, in order to constrain their emplacement age and magma source, as well as their petrogenesis and tectonic setting. They are mainly composed of biotite monzogranite, monzonitic granite, biotite granodiorites, and quartz diorite. Two ages of 1055 ± 43 Ma and 837.6 ± 3.8 Ma were obtained through zircon U-Pb dating by LA-ICP-MS and LA-MC-ICP-MS, respectively. According to their major element compositions, the Grenville-age granites are peraluminous calc-alkaline series calcic S-type granite. In contrast, the mid-Neoproterozoic granites are metaluminous calc-alkaline series alkalic I-type granite. Furthermore, the S-type granites are enriched in LREEs relative to HREEs with(La/Yb)_N ratios of 3.85–18.56 and underwent major fractionation with strongly negative Eu anomalies(Eu/Eu~* = 0.38–0.66). In the MORB-normalized trace element variation diagram, all the samples are enriched in Ce and large ion lithophile elements such as Rb, Th, and K, and depleted in high field strength elements such as Nb, and Ti, with negative Sr and Ti anomalies. The I-type granites are enriched in LREEs with slight negative Eu anomalies(Eu/Eu~* = 0.83–0.93). They are characterized by the enrichment of highly incompatible elements(such as K, Rb, Ba, Th) and LREEs, relative to MORB. Neodymium isotopic data show that the S-type granites display ^(143)Nd/(^(144) Nd) values of 0.51241–0.51256, and have ε_(Nd)(t = 1055 Ma) values of(-3.29) to(-3.81). Calculated t_(DM) ages yield values from 1.87 to 1.91 Ga with the t_(DM).2 stg ages of 1.86 to 1.9 Ga. The I-type granites have ^(143)Nd/(^(144) Nd) ratios between 0.51192 and 0.51195, corresponding to initial ε_(Nd)(t = 837 Ma) values of 1.22 to 5.63. Calculated t_(DM) ages yield values from 1.0 to 1.38 Ga and the t_(DM).2 stg ages yield values from 0.99 to 1.06 Ga. The S-type granites are distinguished as syn-collision granite, whereas the I-type granites were formed as arc magmas according to the Rb-(Yb+Ta) and R_1-R_2 tectonic discrimination diagrams. To conclude, there are two types of spatially associated granite, the Mesoproterozoic S-type granite which were derived from remelting of upper crustal mudstone and/or clastics and resulted from the convergence of two continental plates, and the mid-Neoproterozoic I-type granite which formed in continental arc and resulted from mantle-derived magma mixed crust material, in the western Yangtze Block. Furthermore, we suggest that collision between the Yangtze and Cathaysia blocks occurred at about 1055 Ma, and caused the Stype granite. 4Designing a high Si reduced activation ferritic/martensitic steel for nuclear power generation by using Calphad method显示文摘A high Si reduced activation ferritic/martensitic(RAFM) steel for nuclear structure application is successfully designed by using Calphad method. The main designed chemical composition is C 0.18–0.22%, Cr10.0–10.5%, W 1.0–1.5%, Si 1.0–1.3%, V+Ta 0.30–0.45%, and Fe in balance. High Si design brings excellent corrosion resistance, while low activation is advantageous in the nuclear waste processing. The experimental results indicate that the newly designed high Si RAFM steel had full martensitic structure and uniformly distributed fine second phase particles, and exhibited excellent mechanical properties and corrosion resistance. 7Characteristics of fat metabolism in skeletal muscle of rats after hypobaric hypoxic acclimation显示文摘Objective: To investigate the characteristics of fat metabolism in rat skeletal muscle after hypobaric hypoxia acclimation. Methods: Sprague-Dawley rats were divided into 3 groups randomly: control group (H0), hypoxic 5-day group (H5), and hypoxic 15-day group (H15). Animals of H5 and 15 groups were exposed to hypobaric hypoxia chamber simulating 5 000 m high altitude for 5 d or 15 d respectively, 23 h per day. H0 group stayed outside of chamber. The level of fatty acid oxidation and uptake, and glucose oxidation were examined, and the level of non-esterified fatty acids (NEFA), ATP and phosphocreatine (PCr) were also assayed in rat skeletal muscles. Results: The contents of ATP and PCr in H5 group were lower than those in H0 and H15 groups (P<0.05), while there was no significant difference between H0 and H15. Compared with H0, the blood NEFA level in all hypoxia groups was increased significantly (P<0.05). The muscle NEFA level in H15 group was greatly higher than that in H0 and H5 groups. The rates of fatty acid oxidation and uptake in H15 group were significantly higher than those in H0 and H5 groups (P<0.05), and the rate of glucose oxidation in all hypoxia groups was significantly decreased than that in H0 group (P<0.05). 19Pathogen infection-responsive nanoplatform targeting macrophage endoplasmic reticulum for treating life-threatening systemic infection显示文摘Systemic infections caused by life-threatening pathogens represent one of the main factors leading to clinical death.In this study,we developed a pathogen infection-responsive and macrophage endoplasmic reticulum-targeting nanoplatform to alleviate systemic infections.The nanoplatform is composed of large-pore mesoporous silica nanoparticles(MSNs)grafted by an endoplasmic reticulum-targeting peptide,and a pathogen infection-responsive cap containing the reactive oxygen speciescleavable boronobenzyl acid linker and bovine serum albumin.The capped MSNs exhibited the capacity to high-efficiently load the antimicrobial peptide melittin,and to rapidly release the cargo triggered by H_(2)O_(2) or the pathogen-macrophage interaction system,but had no obvious toxicity to macrophages.During the interaction with pathogenic Candida albicans cells and macrophages,the melittin-loading nanoplatform MSNE+MEL+TPB strongly inhibited pathogen growth,survived macrophages,and suppressed endoplasmic reticulum stress together with pro-inflammatory cytokine secretion.In a systemic infection model,the nanoplatform efficiently prevented kidney dysfunction,alleviated inflammatory symptoms,and protected the mice from death.This study developed a macrophage organelle-targeting nanoplatform for treatment of life-threatening systemic infections.Yan Zhao Shuo Liu Zhishang Shi Hangqi Zhu Mingchun Li Qilin Yu 2022Nano Research2022,15,7:0
20RIN enhances plant disease resistance via root exudate-mediated assembly of diseasesuppressive rhizosphere microbiota显示文摘The RIPENING-INHIBITOR(RIN)transcriptional factor is a key regulator governing fruit ripening.While RIN also affects other physiological processes,its potential roles in triggering interactions with the rhizosphere microbiome and plant health are unknown.Here we show that RIN affects microbiome-mediated disease resistance via root exudation,leading to recruitment of microbiota that suppress the soil-borne,phytopathogenic Ralstonia solanacearum bacterium.Compared with the wild-type(WT)plant,RIN mutants had different root exudate profiles,which were associated with distinct changes in microbiome composition and diversity.Specifically,the relative abundances of antibiosis-associated genes and pathogensuppressing Actinobacteria(Streptomyces)were clearly lower in the rhizosphere of rin mutants.The composition,diversity,and suppressiveness of rin plant microbiomes could be restored by the application of 3-hydroxyflavone and riboflavin,which were exuded in much lower concentrations by the rin mutant.Interestingly,RIN-mediated effects on root exudates,Actinobacteria,and disease suppression were evident from the seedling stage,indicating that RIN plays a dual role in the early assembly of diseasesuppressive microbiota and late fruit development.Collectively,our work suggests that,while plant disease resistance is a complex trait driven by interactions between the plant,rhizosphere microbiome,and the pathogen,it can be indirectly manipulated using'prebiotic'compounds that promote the recruitment of disease-suppressive microbiota.Keming Yang Ruixin Fu Haichao Feng Gaofei Jiang Omri Finkel Tianyu Sun Mingchun Liu Baowen Huang Shan Li Xiaofang Wang Tianjie Yang Yikui Wang Shimei Wang Yangchun Xu Qirong Shen Ville-Petri Friman Alexandre Jousset Zhong Wei 2023Molecular Plant2023,16,9:0
