1Interaction between the gut microbiome and mucosal immune system显示文摘The gut microbiota, the largest symbiotic ecosystem with the host, has been shown to play important roles in maintaining intestinal homeostasis. Dysbiosis of the gut microbiome is caused by the imbalance between the commensal and pathogenic microbiomes. The commensal microbiome regulates the maturation of the mucosal immune system, while the pathogenic microbiome causes immunity dysfunction, resulting in disease development.The gut mucosal immune system, which consists of lymph nodes, lamina propria and epithelial cells, constitutes a protective barrier for the integrity of the intestinal tract. The composition of the gut microbiota is under the surveillance of the normal mucosal immune system. Inflammation, which is caused by abnormal immune responses,influences the balance of the gut microbiome, resulting in intestinal diseases. 4Type I IFN augments IL-27-dependent TRIM25 expression to inhibit HBV replication显示文摘Hepatitis B virus(HBV)can cause chronic hepatitis B,which may lead to cirrhosis and liver cancer.Type I interferon(IFN)is an approved drug for the treatment of chronic hepatitis B.However,the fundamental mechanisms of antiviral action by type I IFN and the downstream signaling pathway are unclear.TRIM25 is an IFN-stimulated gene(ISG)that has an important role in RIG-I ubiquitination and activation.Whether TRIM25 is induced in liver cells by type I IFN to mediate anti-HBV function remains unclear.Here we report that interleukin-27(IL-27)has a critical role in IFN-induced TRIM25 upregulation.TRIM25 induction requires both STAT1 and STAT3.In TRIM25 knockout HepG2 cells,type I IFN production was consistently attenuated and HBV replication was increased,whereas overexpression of TRIM25 in HepG2 cells resulted in elevated IFN production and reduced HBV replication.More interestingly,we found that TRIM25 expression was downregulated in HBV patients and the addition of serum samples from HBV patients could inhibit TRIM25 expression in HepG2 cells,suggesting that HBV might have involved a mechanism to inhibit antiviral ISG expression and induce IFN resistance.Collectively,our results demonstrate that type I IFN-induced TRIM25 is an important factor in inhibiting HBV replication,and the IFN-IL-27-TRIM25 axis may represent a new target for treating HBV infection.Guangyun Tan Qingfei Xiao Hongxiao Song Feng Ma Fengchao Xu Di Peng Na Li Xiaosong Wang Junqi Niu Pujun Gao F Xiao-Feng Qin Genhong Cheng 2018Cellular & Molecular Immunology2018,15,3:13
5Local administration of icariin contributes to peripheral nerve regeneration and functional recovery显示文摘Our previous study showed that systemic administration of the traditional Chinese medicine Epimedium extract promotes peripheral nerve regeneration. Here, we sought to explore the therapeutic effects of local administration of icariin, a major component of Epimedium extract, on peripheral nerve regeneration. A poly(lactic-co-glycolic acid) biological conduit sleeve was used to bridge a 5 mm right sciatic nerve defect in rats, and physiological saline, nerve growth factor, icariin suspension, or nerve growth factor-releasing microsphere suspension was injected into the defect. Twelve weeks later, sciatic nerve conduction velocity and the number of myelinated fibers were notably greater in the rats treated with icariin suspension or nerve growth factor-releasing microspheres than those that had received nerve growth factor or physiological saline. The effects of icariin suspension were similar to those of nerve growth factor-releasing microspheres. 6A Low-Complexity PTS Based on Greedy and Genetic Algorithm for OFDM Systems显示文摘Partial transmit sequence(PTS) is one of effective technique to reduce high Peak-to-average power ratio(PAPR) in Orthogonal frequency division multiplexing(OFDM) system. However, the complexity of Original PTS(O-PTS) increases exponentially with the number of sub-blocks. To reduce the computational complexity while still offering a lower PAPR, a new PTS method is proposed to search for suboptimal rotating vectors in this paper. In the proposed method, the candidate rotation vectors are generated based on greedy and genetic algorithm.We also combine the proposed method and the superimposed training sequence method to get a further PAPR reduction. 7Programmed cell death, antioxidant response and oxidative stress in wheat f lag leaves induced by chemical hybridization agent SQ-1显示文摘Male sterility induced by a chemical hybridization agent(CHA) is an important tool for utilizing crop heterosis. Leaves, especially the flag leaves, as CHA initial recipients play a decisive role in inducing male sterility. To investigate effects of different treatment times of CHA-SQ-1 used, morphological, biochemical and physiological responses of wheat flag leaves were detected in this study. CHA induced programmed cell death(PCD) as shown in terminal deoxynucleotidyl transferase-mediated d UTP nick end-labelling(TUNEL) and DNA laddering analysis. In the early phase, CHA-SQ-1 triggered organelle changes and PCD in wheat leaves accompanied by excess production of reactive oxygen species( O^-._2and H_2O_2) and down-regulation of the activities of superoxide dismutase(SOD), catalase(CAT) and guaiacol peroxidase(POD). Meanwhile, leaf cell DNAs showed ladder-like patterns on agarose gel, indicating that CHA-SQ-1 led to the activation of the responsible endonuclease. The oxidative stress assays showed that lipid peroxidation was strongly activated and photosynthesis was obviously inhibited in SQ-1-induced leaves. However, CHA contents in wheat leaves gradually reduced along with the time CHA-SQ-1 applied. Young flags returned to an oxidative/antioxidative balance and ultimately developed into mature green leaves. 8A novel ASBT inhibitor, IMB17-15, repressed nonalcoholic fatty liver disease development in high-fat diet-fed Syrian golden hamsters显示文摘The manipulation of bile acid (BA) homeostasis by blocking the ileal apical Na+-dependent bile salt transporter (ASBT/SLC10A2) may have therapeutic effects in nonalcoholic fatty liver disease. We developed a novel ASBT inhibitor, an N-(3,4-o-dichlorophenyl)?2-(3-trifluoromethoxy) benzamide derivative referred to as IMB17–15, and investigated its therapeutic effects and the molecular mechanisms underlying the effects. Syrian golden hamsters were challenged with high-fat diet (HFD) to induce NAFLD and were subsequently administered 400 mg/kg IMB17–15 by gavage daily for 21 days. Serum, liver, and fecal samples were collected for further analysis. Plasma concentration-time profiles of IMB17–15 were also constructed. The human hepatocyte cell line HL-7702 was treated with Oleic acid (OA) with or without IMB17–15. Western blotting and real-time PCR were used to study the molecular mechanisms of IMB17–15. We found that IMB17–15 inhibited ASBT and subsequently suppressed ileal farnesoid X receptor (FXR) and FXR-activated fibroblast growth factor15/19 (FGF15/19) expression, which reduced the hepatic phosphorylated extracellular regulated protein kinase (ERK) and c-Jun N-terminal kinase (JNK) levels and upregulated the cholesterol 7α-hydroxylase (CYP7A1) activity. Additionally, IMB17–15 stimulated adenosine monophosphate (AMP)-activated protein kinase (AMPKα) phosphorylation and enhanced peroxisome proliferator activated receptor α(PPARα) expression and thus promoted triglyceride (TG) oxidation and high-density lipoprotein cholesterol (HDL-c) metabolism through an ASBT-independent mechanism. In conclusion, a novel ASBT inhibitor known as IMB17–15 protected hamsters against HFD-induced NFALD by manipulating BA and lipid homeostasis. 9Protective effect of hydroxychloroquine on rheumatoid arthritis-associated atherosclerosis显示文摘Background: Patients with rheumatoid arthritis (RA) have an increased risk for cardiovascular disease. We examined the effect of gut microbiota in a mouse model of RA that develops atherosclerosis. Methods: We created three groups of K/BxN female mice that were positive for the anti‐glucose‐6‐phosphate isomerase (GPI) antibody: control diet (CD), high fat diet (HFD), and HFD with hydroxychloroquine (HFD + HCQ). Serological tests were used to detect the serum levels of total cholesterol (TCHO), low‐density lipoprotein cholesterol (LDL‐C), triglyceride (TG), high‐density lipoprotein cholesterol (HDL‐C), anti‐ GPI antibody titers, and serum cytokines. Atherosclerotic plaque was determined by histological analysis, and gut microbiota were determined by 16sV4 sequencing. Results: Relative to mice given the CD, those receiving the HFD had increased serum levels of LDL‐C, TCHO, and TG, decreased serum levels of HDL‐C, increased atherosclerotic lesions in the aortic root, and altered gut microbiota. Addition of HCQ to HFD decreased the serum levels of LDL‐C, TCHO, and TG, increased serum levels of HDL‐C, and decreased the atherosclerotic lesions in the aortic root. Mice receiving HFD + HCQ also had the greatest bacterial diversity among the three experimental groups. Moreover, HCQ treatment significantly increased the abundance of Akkermansia and Parabacteroides, and decreased the abundance of Clostridium sensu stricto cluster 1, and therefore may be responsible for the reduced RA‐associated atherosclerosis and dyslipidemia. 10Role of autophagy in acute myeloid leukemia therapy显示文摘Despite its dual role in determining cell fate in a wide array of solid cancer cell lines, autophagy has been robustly shown to suppress or kill acute myeloid leukemia cells via degradation of the oncogenic fusion protein that drives leukemogenesis. However, autophagy also induces the demise of acute leukemia cells that do not express the known fusion protein, though the molecular mechanism remains elusive. 11Comparative genomic and transcriptomic analyses uncover the molecular basis of high nitrogen-use efficiency in the wheat cultivar Kenong 9204显示文摘Studying the regulatory mechanisms that drive nitrogen-use efficiency(NUE)in crops is important for sustainable agriculture and environmental protection.In this study,we generated a high-quality genome assembly for the high-NUE wheat cultivar Kenong 9204 and systematically analyzed genes related to nitrogen uptake and metabolism.By comparative analyses,we found that the high-affinity nitrate transporter gene family had expanded in Triticeae.Further studies showed that subsequent functional differentiation endowed the expanded family members with saline inducibility,providing a genetic basis for improving the adaptability of wheat to nitrogen deficiency in various habitats.To explore the genetic and molecular mechanisms of high NUE,we compared genomic and transcriptomic data from the high-NUE cultivar Kenong 9204(KN9204)and the low-NUE cultivar Jing 411 and quantified their nitrogen accumulation under high-and low-nitrogen conditions.Compared with Jing 411,KN9204 absorbed significantly more nitrogen at the reproductive stage after shooting and accumulated it in the shoots and seeds.Transcriptome data analysis revealed that nitrogen deficiency clearly suppressed the expression of genes related to cell division in the young spike of Jing 411,whereas this suppression of gene expression was much lower in KN9204.In addition,KN9204 maintained relatively high expression of NPF genes for a longer time than Jing 411 during seed maturity.Physiological and transcriptome data revealed that KN9204 was more tolerant of nitrogen deficiency than Jing 411,especially at the reproductive stage.The high NUE of KN9204 is an integrated effect controlled at different levels.Taken together,our data provide new insights into the molecular mechanisms of NUE and important gene resources for improving wheat cultivars with a higher NUE trait.Xiaoli Shi Fa Cui Xinyin Han Yilin He Long Zhao Na Zhang Hao Zhang Haidong Zhu Zhexin Liu Bin Ma Shusong Zheng Wei Zhang Jiajia Liu Xiaoli Fan Yaoqi Si Shuiquan Tian Jianqing Niu Huilan Wu Xuemei Liu Zhuo Chen Deyuan Meng Xiaoyan Wang Liqiang Song Lijing Sun Jie Han Hui Zhao Jun Ji Zhiguo Wang Xiaoyu He Ruilin Li Xuebin Chi Chengzhi Liang Beifang Niu Jun Xiao Junming Li Hong-Qing Ling 2022Molecular Plant2022,15,9:6
12Intraoperative single administration of neutrophil peptide 1 accelerates the early functional recovery of peripheral nerves after crush injury显示文摘Neutrophil peptide 1 belongs to a family of peptides involved in innate immunity. Continuous intramuscular injection of neutrophil peptide 1 can promote the regeneration of peripheral nerves, but clinical application in this manner is not convenient. To this end, the effects of a single intraoperative administration of neutrophil peptide 1 on peripheral nerve regeneration were experimentally observed. A rat model of sciatic nerve crush injury was established using the clamp method. After model establishment, a normal saline group and a neutrophil peptide 1 group were injected with a single dose of normal saline or 10 μg/mL neutrophil peptide 1, respectively. A sham group, without sciatic nerve crush was also prepared as a control. Sciatic nerve function tests, neuroelectrophysiological tests, and hematoxylin-eosin staining showed that the nerve conduction velocity, sciatic functional index, and tibialis anterior muscle fiber cross-sectional area were better in the neutrophil peptide 1 group than in the normal saline group at 4 weeks after surgery. At 4 and 8 weeks after surgery, there were no differences in the wet weight of the tibialis anterior muscle between the neutrophil peptide 1 and saline groups. Histological staining of the sciatic nerve showed no significant differences in the number of myelinated nerve fibers or the axon cross-sectional area between the neutrophil peptide 1 and normal saline groups. The above data confirmed that a single dose of neutrophil peptide 1 during surgery can promote the recovery of neurological function 4 weeks after sciatic nerve injury. 13Plasma spray of biofunctional(Mg, Sr)-substituted hydroxyapatite coatings for titanium alloy implants显示文摘Plasma-sprayed hydroxyapatite (HA) coatings have been widely utilized in load-bearing titanium alloy implants. In this study, Mg, Sr co-substituted HA ((Mg,Sr)-HA) nano-scale powders have been synthesized, which are further used to prepare (Mg,Sr)-HA coatings on Ti-6A1-4V alloys in order to improve the biological functions. The average size of (Mg,Sr)-HA nano particles is ~75nm. The average bonding strength for (Mg,Sr)-HA coating and samples after heat treatment at 500℃ or 600℃ for 3h are 26.17±2.11 MPa, 36.07±4.48 MPa and 37.07 ±2.95 MPa, respectively. There is a significantly increase of bonding strength likely due to low residual stress after heated treatment.MC3T3-E1 cells show a high proliferation rate when cultured with (Mg,Sr)-HA coating extract compared to the normal culture medium, which also exhibit large extension and depositi on of extracellular matrices when adhered on the coating surfaces. 14Pantoprazole ameliorates liver fibrosis and suppresses hepatic stellate cell activation in bile duct ligation rats by promoting YAP degradation显示文摘Liver fibrosis is one of the most severe pathologic consequences of chronic liver diseases,and effective therapeutic strategies are urgently needed.Proton pump inhibitors(PPIs)are H+/K+-ATPase inhibitors and currently used to treat acid-related diseases such as gastric ulcers,which have shown other therapeutic effects in addition to inhibiting acid secretion.However,few studies have focused on PPIs from the perspective of inhibiting hepatic fibrosis.In the present study,we investigated the effects of pantoprazole(PPZ),a PPI,against liver fibrosis in a bile duct ligation(BDL)rat model,human hepatic stellate cell(HSC)line LX-2 and mouse primary HSCs(pHSCs),and explored the potential mechanisms underlying the effects of PPZ in vitro and in vivo.In BDL rats,administration of PPZ(150 mg·kg^(−1)·d^(−1),i.p.for 14 d)significantly attenuated liver histopathological injury,collagen accumulation,and inflammatory responses,and suppressed fibrogenesis-associated gene expression including Col1a1,Acta2,Tgfβ1,and Mmp-2.In LX-2 cells and mouse pHSCs,PPZ(100–300μM)dose-dependently suppressed the levels of fibrogenic markers.We conducted transcriptome analysis and subsequent validation in PPZ-treated LX-2 cells,and revealed that PPZ inhibited the expression of Yes-associated protein(YAP)and its downstream targets such as CTGF,ID1,survivin,CYR61,and GLI2.Using YAP overexpression and silencing,we demonstrated that PPZ downregulated hepatic fibrogenic gene expression via YAP.Furthermore,we showed that PPZ promoted the proteasome-dependent degradation and ubiquitination of YAP,thus inhibiting HSC activation.Additionally,we showed that PPZ destabilized YAP by disrupting the interaction between a deubiquitinating enzyme OTUB2 and YAP,and subsequently blocked the progression of hepatic fibrosis.Zhen-ning Lu Wei-xiao Niu Na Zhang Mao-xu Ge Yun-yang Bao Yu Ren Xiu-li Guo Hong-wei He 2021Acta Pharmacologica Sinica2021,42,11:5

16Osteoclasts are not a source of SLIT3显示文摘The axon guidance cue SLIT3 was identified as an osteoanabolic agent in two recent reports. However, these reports conflict in their nomination of osteoblasts versus osteoclasts as the key producers of skeletal SLIT3 and additionally offer conflicting data on the effects of SLIT3 on osteoclastogenesis. Here, aiming to address this discrepancy, we found no observable SLIT3 expression during human or mouse osteoclastogenesis and the only modest SLIT3-mediated effects on osteoclast differentiation. Conditional deletion of SLIT3 in cathepsin K(CTSK)-positive cells, including osteoclasts, had no effect on the number of osteoclast progenitors, in vitro osteoclast differentiation, overall bone mass, or bone resorption/formation parameters. Similar results were observed with the deletion of SLIT3 in Lys M-positive cells, including osteoclast lineage cells. Consistent with this finding, bone marrow chimeras made from Slit3-/-donors that lacked SLIT3 expression at all stages of osteoclast development displayed normal bone mass relative to controls. 19Extraction of Hemicellulose from Acacia Wood via Autohydrolysis and Ethanol Precipitation显示文摘Isolation and utilization of hemicellulose are important processes in the pulp mill-based biorefinery. Considering the potential end use of the hemicellulose,this study investigated the concentration of monomeric and oligomeric sugars in the hydrolysate derived from autohydrolysis of acacia wood and the obtainment of these sugars via ethanol precipitation. The rate of generation of monomers and oligomers increased dramatically with an increase of the temperature from 160℃ to 180℃. The maximum content of oligomers and total sugars was achieved under conditions of 180℃ and 10 min,170℃ and 85 min,respectively. Interestingly,the maximum yield of hemicellulose via ethanol precipitation was achieved in a much shorter time,compared with the maximum yield of oligomer from the hydrolysate by autohydrolysis. A 6. 66 g / L quantity of hemicellulose was obtained by intensifying the conditions of ethanol precipitation. 20Effect of fructooligosaccharides on the colonization of Lactobacillus rhamnosus AS 1.2466^(T) in the gut of mice显示文摘Lactobacillus rhamnosus and fructooligosaccharides(FOS)have been widely studied so far.However,the effects of L.rhamnosus on the intestinal microecological environment at the species level and the effect of different proportions of FOS on L.rhamnosus colonization in different parts of mice intestine are still unclear.The study results indicated that the specific bands of enterobacterial repetitive intergenic consensus polymerase chain reaction(ERIC-PCR)in the L.rhamnosus(LR)group significantly increased at 7 days.Although the number of bands was similar to the natural recovery(NR)group,the brightness of few bands significantly enhanced in the later stage of recovery.Besides,Southern-blot maps showed strong signals,indicating that the ERIC-PCR fingerprint could accurately reflect the changes in the mouse gut microbiota diversity.Further,the high-throughput results confirmed that the Lactobacillus and Akkermansia had different changes at different periods,but all of them showed an upward trend,while the Klebsiella were inhibited,thereby maintaining the intestinal microecology balance.Moreover,FOS exerted a positive effect on L.rhamnosus colonization in the gut.Zhihua Niu Meijuan Zou Tingting Bei Na Zhang Dongyao Li Miaoshu Wang Chen Li Hongtao Tian 2023Food Science and Human Wellness2023,12,2:3
