| 8 | Blockade of Tim-3 Pathway Ameliorates Interferon-γ Production from Hepatic CD8^+ T Cells in a Mouse Model of Hepatitis B Virus Infection显示文摘T cell immunoglobulin-and mucin-domain-containing molecule-3(Tim-3) has been reported to participate in the pathogenesis of inflammatory diseases. However,whether Tim-3 is involved in hepatitis B virus(HBV) infection remains unknown. Here,we studied the expression and function of Tim-3 in a hydrodynamics-based mouse model of HBV infection. A significant increase of Tim-3 expression on hepatic T lymphocytes,especially on CD8+ T cells,was demonstrated in HBV model mice from day 7 to day 18. After Tim-3 knockdown by specific shRNAs,significantly increased IFN-γ production from hepatic CD8+ T cells in HBV model mice was observed. Very interestingly,we found Tim-3 expression on CD8+ T cells was higher in HBV model mice with higher serum anti-HBs production. Moreover,Tim-3 knockdown influenced anti-HBs production in vivo. | 9 | D2 dissection in laparoscopic and open gastrectomy for gastric cancer显示文摘AIM: To evaluate the radicalness and safety of laparoscopic D2 dissection for gastric cancer. METHODS: Clinicopathological data from 209 patients with gastric cancer, who underwent radical gastrectomy with D2 dissection between January 2007 and February 2011, were analyzed retrospectively. Among these patients, 131 patients underwent laparoscopyassisted gastrectomy (LAG) and 78 underwent open gastrectomy (OG). The parameters analyzed included operative time, blood loss, blood transfusion, morbidity, mortality, the number of harvested lymph nodes (HLNs), and pathological stage.RESULTS: There were no significant differences in sex, age, types of radical resection [radical proximal gastrectomy (PG + D2), radical distal gastrectomy (DG + D2) and radical total gastrectomy (TG + D2)], and stages between the LAG and OG groups (P > 0.05). Among the two groups, 127 cases (96.9%) and 76 cases (97.4%) had 15 or more HLNs, respectively. The average number of HLNs was 26.1 ± 11.4 in the LAG group and 24.2 ± 9.3 in the OG group (P = 0.233). In the same type of radical resection, there were no signifi cant differences in the number of HLNs between the two groups (PG + D2: 21.7 ± 7.5 vs 22.4 ± 9.3; DG + D2: 25.7 ± 11.0 vs 22.3 ± 7.9; TG + D2: 30.9 ± 13.4 vs 29.3 ± 10.4; P > 0.05 for all comparisons). Tumor free margins were obtained in all cases. Compared with OG group, the LAG group had signifi cantly less blood loss, but a longer operation time (P < 0.001). The morbidity of the LAG group was 9.9%, which was not signifi cantly different from the OG group (7.7%) (P = 0.587). The mortality was zero in both groups. CONCLUSION: Laparoscopic D2 dissection is equivalent to OG in the number of HLNs, regardless of tumor location. | 15 | Therapeutic potential of intravenous Xuebijing on transforming growth factor β_1 and procollagen type III peptide in patients with acute paraquat poisoning显示文摘OBJECTIVE:Paraquat(PQ) poisoning-induced pulmonary fibrosis causes asphyxiation and death.The therapeutic potential of intravenous Xuebijing therapy in PQ poisoning patients and its underlying immunomodulatory effects on transforming growth factor(TGF)-β1 and procollagen type III peptide(PIIIP) were investigated.METHODS:Thirty-six acute PQ poisoning patients were randomly assigned to conventional therapy(Group A) and intravenous Xuebijing administration plus conventional therapy(Group B).Twenty volunteers served as controls(Group C).Blood samples were collected upon admission(day 0) and at post-treatment days 5,10,and 14.TGF-β1 and PIIIP concentrations were determined by ELISA and analyzed for intra-and inter-group differences over time.One-month follow-up was conducted for determining the mortality rate.RESULTS:TGF-β1 and PIIIP levels were significantly higher in PQ poisoning patients and increased over time(Groups A and B vs C,P<0.01).However,the TGF-β1 and PIIIP levels were consistently significantly lower in Group B compared with those of Group A(P<0.01).The 1-month mortality rate was also lower in Group B compared with that of Group A(P<0.05).PQ poisoning patients showed remarkably high levels of TGF-β1 and PIIIP,which increased as PQ-induced pulmonary fibrosis progressed.CONCLUSION:Treatment with intravenous Xuebijing plus conventional therapy significantly lowered TGF-β1 and PIIIP levels,which indicates therapeutic efficacy in the treatment of PQ poisoning patients. | Xuebing Zheng Xiaoli Sun Piyong Ma Zhongming Liu Nan Jiang | 2012 | Journal of Traditional Chinese Medicine2012,32,4: | 11 |

| 17 | Graft-accelerated virus-induced gene silencing facilitates functional genomics in rose flowers显示文摘Rose has emerged as a model ornamental plant for studies of flower development, senescence, and morphology, as well as the metabolism of floral fragrances and colors.Virus-induced gene silencing(VIGS) has long been used in functional genomics studies of rose by vacuum infiltration of cuttings or seedlings with an Agrobacterium suspension carrying TRV-derived vectors. However, VIGS in rose flowers remains a challenge because of its low efficiency and long time to establish silencing. Here we present a novel and rapid VIGS method that can be used to analyze gene function in rose,called ‘graft-accelerated VIGS', where axil ary sprouts are cut from the rose plant and vacuum infiltrated with Agrobacterium. The inoculated scions are then grafted back onto the plants to flower and silencing phenotypes can be observed within 5 weeks, post-infiltration. Using this new method, we successfully silenced expression of the RhDFR_1, RhA G, and RhNUDX_1 in rose flowers, and affected their color, petal number, as well as fragrance, respectively. This grafting method will facilitate high-throughput functional analysis of genes in rose flowers.
