1Immune secondary response and clonal selection inspired optimizers显示文摘The immune system's ability to adapt its B cells to new types of antigen is powered by processes known as clonal selection and affinity maturation. When the body is exposed to the same antigen,immune system usually calls for a more rapid and larger response to the antigen,where B cells have the function of negative adjustment. Based on the clonal selection theory and the dynamic process of immune response,two novel artificial immune system algorithms,secondary response clonal programming algorithm (SRCPA) and secondary response clonal multi-objective algorithm (SRCMOA),are presented for solving single and multi-objective optimization problems,respectively. Clonal selection operator (CSO) and secondary response operator (SRO) are the main operators of SRCPA and SRCMOA. Inspired by the clonal selection theory,CSO reproduces individuals and selects their improved maturated progenies after the affinity mat-uration process. SRO copies certain antibodies to a secondary pool,whose members do not participate in CSO,but these antibodies could be activated by some external stimulations. The update of the secondary pool pays more attention to maintain the population diversity. On the one hand,decimal-string representation makes SRCPA more suitable for solving high-dimensional function optimiza-tion problems. Special mutation and recombination methods are adopted in SRCPA to simulate the somatic mutation and receptor edit-ing process. Compared with some existing evolutionary algorithms,such as OGA/Q,IEA,IMCPA,BGA and AEA,SRCPA is shown to be able to solve complex optimization problems,such as high-dimensional function optimizations,with better performance. On the other hand,SRCMOA combines the Pareto-strength based fitness assignment strategy,CSO and SRO to solve multi-objective optimization problems. 2Blockade of Tim-3 Pathway Ameliorates Interferon-γ Production from Hepatic CD8^+ T Cells in a Mouse Model of Hepatitis B Virus Infection显示文摘T cell immunoglobulin-and mucin-domain-containing molecule-3(Tim-3) has been reported to participate in the pathogenesis of inflammatory diseases. However,whether Tim-3 is involved in hepatitis B virus(HBV) infection remains unknown. Here,we studied the expression and function of Tim-3 in a hydrodynamics-based mouse model of HBV infection. A significant increase of Tim-3 expression on hepatic T lymphocytes,especially on CD8+ T cells,was demonstrated in HBV model mice from day 7 to day 18. After Tim-3 knockdown by specific shRNAs,significantly increased IFN-γ production from hepatic CD8+ T cells in HBV model mice was observed. Very interestingly,we found Tim-3 expression on CD8+ T cells was higher in HBV model mice with higher serum anti-HBs production. Moreover,Tim-3 knockdown influenced anti-HBs production in vivo. 4Ginsenoside Rb1 attenuates activated microglia-induced neuronal damage显示文摘The microglia-mediated inflammatory reaction promotes neuronal damage under cerebral ischemia/hypoxia conditions. We therefore speculated that inhibition of hypoxia-induced microglial activation may alleviate neuronal damage. To test this hypothesis, we co-cultured ginsenoside Rb1, an active component of ginseng, and cortical neurons. Ginsenoside Rb1 protected neuronal morphology and structure in a single hypoxic culture system and in a hypoxic co-culture system with microglia, and reduced neuronal apoptosis and caspase-3 production. The protective effect was observable prior to placing in co-culture. Additionally, ginsenoside Rb1 inhibited levels of tumor necrosis factor-α in a co-culture system containing activated N9 microglial cells. Ginsenoside Rb1 also significantly decreased nitric oxide and superoxide production induced by N9 microglia. 5Evidences of rapid erosion driven by climate in the Yarlung Zangbo (Tsangpo) Great Canyon,the eastern Himalayan syntaxis显示文摘Climate and tectonism are both particularly intense in the Yarlung Zangbo (Tsangpo) Great Canyon in the eastern Himalayan syntaxis,which is characterized by the most rapid landscape evolution of anywhere in the world.Thus,the eastern Himalayan syntaxis is one of the best locations to study the interactions between climate and tectonics.This paper investigates the cooling ages of the Doxong La-Baibung profile using apatite fission track (AFT) dating on 11 bedrock samples at elevations ranging from 4210 to 710 m.There are topographic,climatic,metamorphic,and thermochronological gradients in the profile,providing good conditions to study interactions between climate and tectonics.AFT ages ranged from 4.6±0.6 Ma to 1.7±0.3 Ma,and the mean fission track lengths ranged from 11.0 to 12.4 μm.It was found that the cooling rates revealed by AFT ages increased with decreasing elevation.However,the tendency of the cooling rates revealed by the 40 Ar-39 Ar ages was different from that indicated by the AFT ages.Moreover,for most districts of the eastern Himalayan syntaxis,the compiled AFT age distribution correlates well with the annual average precipitation,indicating the coupling of the cooling and erosion rates of the near-surface rock and precipitation.The geothermal history modeling results indicate an obvious increase in the cooling and erosion rate between 1.0 and 0.5 Ma.This age is consistent with other research findings for this time,when the vapor channel of the Yarlung Zangbo Great Canyon began to take effect.These evidences suggest that climate,especially precipitation,has acting as a key factor influencing the rapid cooling and erosion in the Yarlung Zangbo Great Canyon since 1-0.5 Ma.YU XiangJiang JI JianQing GONG JunFeng SUN DongXia QING JianChun WANG LiNing ZHONG DaLai ZHANG ZhiCheng 2011Chinese Science Bulletin2011,56,11:11

9OSBPL2-disrupted pigs recapitulate dual features of human hearing loss and hypercholesterolaemia显示文摘Oxysterol binding protein like 2(OSBPL2), an important regulator in cellular lipid metabolism and transport, was identified as a novel deafness-causal gene in our previous work. To resemble the phenotypic features of OSBPL2 mutation in animal models and elucidate the potential genotypephenotype associations, the OSBPL2-disrupted Bama miniature(BM) pig model was constructed using CRISPR/Cas9-mediated gene editing, somatic cell nuclear transfer(SCNT) and embryo transplantation approaches, and then subjected to phenotypic characterization of auditory function and serum lipid profiles. The OSBPL2-disrupted pigs displayed progressive hearing loss(HL) with degeneration/apoptosis of cochlea hair cells(HCs) and morphological abnormalities in HC stereocilia, as well as hypercholesterolaemia. High-fat diet(HFD) feeding aggravated the development of HL and led to more severe hypercholesterolaemia. 10Synergistic therapy of magnetism-responsive hydrogel for soft tissue injuries显示文摘Soft tissue injury is very common and associated with pain,tissue swelling and even malformation if not treated on time.Treating methods include cryotherapy,electrical therapy,ultrasound therapy and anti-inflammatory drug,but none of them is completely satisfying.In this work,for a better therapeutic effect,drug therapy and pulsed electromagnetic field(PEMF)therapy were combined.We constructed a drug delivery system using the tetra-PEG/agar hydrogel(PA).By incorporating Fe3O4 NPs into the hydrogel network,a magnetism-responsive property was achieved in the system.The cytotoxicity and in vivo study showed a good biocompatibility of the PA/Fe3O4 hydrogel.A magnetism-controlled release was attained by the incorporation of Fe3O4.Finally,in vivo study showed a better performance of the DS-loaded PA/Fe3O4 compared with the commercially available DS ointment regarding the recovery of the injured soft tissue.Therefore,this magnetism-responsive hydrogel may represent a promising alternative to treat soft tissue injury.Lining Zhang Xiuqin Zuo Shengjie Li Mi Sun Huimin Xie Kai Zhang Jikun Zhou Liyun Che Junxuan Ma Zishan Jia Fei Yang 2019Bioactive Materials2019,4,1:7
11An epidermal sEMG tattoo-like patch as a new human–machine interface for patients with loss of voice显示文摘Throat cancer treatment involves surgical removal of the tumor,leaving patients with facial disfigurement as well as temporary or permanent loss of voice.Surface electromyography(sEMG)generated from the jaw contains lots of voice information.However,it is difficult to record because of not only the weakness of the signals but also the steep skin curvature.This paper demonstrates the design of an imperceptible,flexible epidermal sEMG tattoo-like patch with the thickness of less than 10μm and peeling strength of larger than 1N cm−1 that exhibits large adhesiveness to complex biological surfaces and is thus capable of sEMG recording for silent speech recognition.When a tester speaks silently,the patch shows excellent performance in recording the sEMG signals from three muscle channels and recognizing those frequently used instructions with high accuracy by using the wavelet decomposition and pattern recognization.The average accuracy of action instructions can reach up to 89.04%,and the average accuracy of emotion instructions is as high as 92.33%.To demonstrate the functionality of tattoo-like patches as a new human–machine interface(HMI)for patients with loss of voice,the intelligent silent speech recognition,voice synthesis,and virtual interaction have been implemented,which are of great importance in helping these patients communicate with people and make life more enjoyable.Huicong Liu Wei Dong Yunfei Li Fanqi Li Jiangjun Geng Minglu Zhu Tao Chen Hongmiao Zhang Lining Sun Chengkuo Lee 2020Microsystems & Nanoengineering2020,6,1:6

13Fine particulate matter(PM_(2.5)/PM_(1.0)) in Beijing,China: Variations and chemical compositions as well as sources显示文摘Particulate matter(i.e.,PM_(1.0) and PM_(2.5)),considered as the key atmospheric pollutants,exerts negative effects on visibility,global climate,and human health by associated chemical compositions.However,our understanding of PM and its chemical compositions in Beijing under the current atmospheric environment is still not complete after witnessing marked alleviation during 2013–2017.Continuous measurements can be crucial for further air quality improvement by better characterizing PM pollution and chemical compositions in Beijing.Here,we conducted simultaneous measurements on PM in Beijing during 2018–2019.Results indicate that annualmean PM_(1.0) and PM_(2.5) concentrations were 35.49±18.61μg/m^(3) and 66.58±60.17μg/m^(3),showing a positive response to emission controls.The contribution of sulfate,nitrate,and ammonium(SNA)played an enhanced role with elevated PM loading and acted as the main contributors to pollution episodes.Discrepancies observed among chemical species between PM_(1.0) and PM_(2.5) in spring suggest that sand particles trend to accumulate in the range of 1–2.5μm.Pollution episodes occurred accompanied with southerly clusters and high formation of SNA by heterogeneous reactions in summer and winter,respectively.Results from positive matrix factorization(PMF)combined with potential source contribution function(PSCF)models showed that potential areas were seasonal dependent,secondary and vehicular sources became much more important compared with previous studies in Beijing.Our study presented a continuous investigation on PM and sources origins in Beijing,which provides a better understanding for further emission control as well as a reference for other cities in developing countries.Lining Luo Xiaoxuan Bai Shuhan Liu Bobo Wu Wei Liu Yunqian Lv Zhihui Guo Shumin Lin Shuang Zhao Yan Hao Jiming Hao Kai Zhang Aihua Zheng Hezhong Tian 2022Journal of Environmental Sciences2022,34,11:3
14Antigenicity of tissues and organs from GGTA1/CMAH/β4GalNT2 triple gene knockout pigs显示文摘Clinical xenotransplantations have been hampered by human preformed antibody-mediated damage of the xenografts.To overcome biological incompatibility between pigs and humans,one strategy is to remove the major antigens[Gal,Neu5 Gc,and Sd(a)]present on pig cells and tissues.Triple gene(GGTAI,CMAH,and β4 GalNT2)knockout(TKO)pigs were produced in our laboratory by CRISPR-Cas9 targeting.To investigate the antigenicity reduction in the TKO pigs,the expression levels of these three xenoantigens in the cornea,heart,liver,spleen,lung,kidney,and pancreas tissues were examined.The level of human IgG/IgM binding to those tissues was also investigated,with wildtype pig tissues as control.The results showed that aGal,Neu5 Gc,and Sd(a)were markedly positive in all the examined tissues in wildtype pigs but barely detected in TKO pigs.Compared to wildtype pigs,the liver,spleen,and pancreas of TKO pigs showed comparable levels of human IgG and IgM binding,whereas corneas,heart,lung,and kidney of TKO pigs exhibited significantly reduced human IgG and IgM binding.These results indicate that the antigenicity of TKO pig is significantly reduced and the remaining xenoantigens on porcine tissues can be eliminated via a gene targeting approach.Ronggen Wang Miaomiao Ruan Runjie Zhang Lei Chen Xiaoxue Li Bin Fang Chu Li Xueyang Ren Jiying Liu Qiang Xiong Lining Zhang Yong Jin Lin Li Rongfeng Li Ying Wang Haiyuan Yang Yifan Dai 2019The Journal of Biomedical Research2019,33,4:3

17Overexpressing dominant-negative FGFR2-IIIb impedes lung branching morphogenesis in pigs显示文摘Genetic studies with mouse models have shown that fibroblast growth factor receptor 2-IIIb(FGFR2-IIIb)plays crucial roles in lung development and differentiation. To evaluate the effect of FGFR2-IIIb in pig lung development, we employed somatic cell nuclear transfer(SCNT) technology to generate transgenic pig fetuses overexpressing the transmembrane(dn FGFR2-IIIb-Tm) and soluble(dn FGFR2-IIIb-HFc) forms of the dominant-negative human FGFR2-IIIb driven by the human surfactant protein C(SP-C) promoter,which was specifically expressed in lung epithelia. Eight dn FGFR2-IIIb-Tm transgenic and twelve dn FGFR2-IIIb-HFc transgenic pig fetuses were collected from three and two recipient sows, respectively.Repression of FGFR2-IIIb in lung epithelia resulted in smaller lobes and retardation of alveolarization in both forms of dn FGFR2-IIIb transgenic fetuses. Moreover, the dn FGFR2-IIIb-HFc transgenic ones showed more deterioration in lung development. Our results demonstrate that disruption of FGFR2-IIIb signaling in the epithelium impedes normal branching and alveolarization in pig lungs, which is less severe than the results observed in transgenic mice. 19Structural geology and favorable exploration prospect belts in northwestern Sichuan Basin, SW China显示文摘The northwestern Sichuan region has experienced multi-stage tectonic evolution including marine cratonic basin from the Sinian to the Middle Triassic and intra-continental basin from the Late Triassic to the Cenozoic. Several regional tectonic activities caused complicated stratigraphic distribution and structural deformations in the deep-buried layers. During the key tectonic periods, some characteristic sedimentary and deformation structures were formed, including the step-shaped marginal carbonate platform of Dengying Formation, the western paleo-high at the end of Silurian, and the passive continental margin of the Late Paleozoic–Middle Triassic. The Meso-Cenozoic intra-continental compressional tectonic processes since the Late Triassic controlled the formation of complex thrusting structures surrounding and inside the northwestern basin. The northern Longmenshan fold-thrust belt has a footwall in-situ thrust structure,which is controlled by two sets of detachments in the Lower Triassic and Lower Cambrian and presents as a multi-level deformation structure with the shallow folds, the middle thin-skin thrusts and the deeper basement-involved folds. The thrust belt in front of the Micangshan Mountain shows a double-layer deformation controlled by the Lower Triassic salt detachment, which is composed by the upper monocline and deep-buried imbricate thrust structures. The interior of the basin is characterized by several rows of large-scale basement-involved folds with NEE strike direction. From the perspective of structural geology, the favorable exploration reservoirs and belts in northwestern Sichuan have obvious zoning characteristics. The favorable exploration layers of Dengying Formation of Upper Sinian are mainly distributed in the eastern and northern areas of the northwestern Sichuan Basin, in which the Jiulongshan structural belt, Zitong syncline and Yanting slope are the most favorable. The Lower Paleozoic was transformed by Caledonian paleo-uplift and late Cenozoic folding, and the midwest area such as the Zitong syncline is a potential area for hydrocarbon exploration.
