| 4 | KRAS-G12D mutation drives immune suppression and the primary resistance of anti-PD-1/PD-L1 immunotherapy in non-small cell lung cancer显示文摘Background:Although immune checkpoint inhibitors(ICIs)against programmed cell death protein 1(PD-1)and its ligand PD-L1 have demonstrated potency towards treating patients with non-small cell lung carcinoma(NSCLC),the potential association between Kirsten rat sarcoma viral oncogene homolog(KRAS)oncogene substitutions and the efficacy of ICIs remains unclear.In this study,we aimed to find point mutations in the KRAS gene resistant to ICIs and elucidate resistance mechanism.Methods:The association between KRAS variant status and the efficacy of ICIs was explored with a clinical cohort(n=74),and confirmed with a mouse model.In addition,the tumor immune microenvironment(TIME)of KRASmutant NSCLC,such as CD8+tumor-infiltrating lymphocytes(TILs)and PD-L1 level,was investigated.Cell lines expressing classic KRAS substitutions were used to explore signaling pathway activation involved in the formation of TIME.Furthermore,interventions that improved TIME were developed to increase responsiveness to ICIs.Results:We observed the inferior efficacy of ICIs in KRAS-G12D-mutant NSCLC.Based upon transcriptome data and immunostaining results from KRAS-mutant NSCLC,KRAS-G12D point mutation negatively correlated with PD-L1 level and secretion of chemokines CXCL10/CXCL11 that led to a decrease in CD8+TILs,which in turn yielded an immunosuppressive TIME.The analysis of cell lines overexpressing classic KRAS substitutions further revealed that KRAS-G12D mutation suppressed PD-L1 level via the P70S6K/PI3K/AKT axis and reduced CXCL10/CXCL11 levels by down-regulating high mobility group protein A2(HMGA2)level.Notably,paclitaxel,a chemotherapeutic agent,upregulated HMGA2 level,and in turn,stimulated the secretion of CXCL10/CXCL11.Moreover,PD-L1 blockade combined with paclitaxel significantly suppressed tumor growth compared with PD-L1 inhibitor monotherapy in a mouse model with KRAS-G12D-mutant lung adenocarcinoma.Further analyses revealed that the combined treatment significantly enhanced the recruitment of CD8+TILs via the up-regulation of CXCL10/CXCL11 levels.Results of clinical study also revealed the superior efficacy of chemo-immunotherapy in patients with KRAS-G12D-mutant NSCLC compared with ICI monotherapy.Conclusions:Our study elucidated the molecular mechanism by which KRASG12D mutation drives immunosuppression and enhances resistance of ICIs in NSCLC.Importantly,our findings demonstrate that ICIs in combination with chemotherapy may be more effective in patients with KRAS-G12D-mutant NSCLC. | Chengming Liu Sufei Zheng Zhanyu Wang Sihui Wang Xinfeng Wang Lu Yang Haiyan Xu Zheng Cao Xiaoli Feng Qi Xue Yan Wang Nan Sun Jie He | 2022 | Cancer Communications2022,42,9: | 4 |

| 6 | Tectonic Evolution of the North Depression of the South Yellow Sea Basin Since Late Cretaceous显示文摘On the basis of subsidence history analysis and balanced cross-section analysis, the vertical uplift/subsidence history and horizontal extension/compression history of the north depression of the south Yellow Sea basin are quantitatively studied. The results show that the tectonic evolution of the north depression of the south Yellow Sea basin since late Cretaceous can be divided into a rifting phase(late Cretaceous to Paleogene) and a post-rifting phase(Neogene to Quaternary). The rifting phase can be further subdivided into an initial rifting stage(late Cretaceous), an intensive rifting stage(Paleocene), a rifting termination stage(Eocene), and an inversion-uplifting stage(Oligocene). Together, this division shows the characteristics of an episodic-evolved intracontinental rift-depression basin. The deformation of the north depression of the south Yellow Sea basin since late Cretaceous was mainly fault-related. The horizontal extension and tectonic subsidence were controlled by the activity of faults. The differential evolution of faults also caused variations in local uplift/subsidence movements and the regional heterogeneity in extension. The late Cretaceous initial rifting of the north depression of the south Yellow Sea basin is related to the Pacific-Eurasia convergence. | 7 | Possibility of women treated with fertility-sparing surgery fornon-epithelial ovarian tumors to safely and successfully becomepregnant--a Chinese retrospective cohort study among 148cases显示文摘This study was performed to evaluate the oncological and reproductive outcomes of childbearing-agewomen treated with fertility-sparing surgery (FSS) for non-epithelial ovarian tumors in China. One hundred andforty eight non-epithelial ovarian tumor women treated with FSS between January I, 2000 and August 31, 2015from two medical centers in China were identified. Progression-free survival (PFS) was 88.5%, whereas overallsurvival (OS) was 93.9%. Univariate analysis suggested that delivery after treatment is related to PFS (P = 0.023),whereas histology significantly influenced OS. Cox regression analysis suggested that only histology was associatedwith PFS and OS (P 〈 0.05). Among the 129 women who completed adjuvant chemotherapy (ACT), nonedeveloped amenorrhea. Among the 44 women who desired pregnancy, 35 (79.5%) successfully had 51 gestationsincluding 35 live births without birth defects. Non-epithelial ovarian tumors can achieve fulfilling prognosis afterFSS and chemotherapy. Histology might be the only independent prognostic factor for PFS and OS. FSS followedby ACT appeared to have little or no effect on fertility. Meanwhile, postoperative pregnancy did not increase thePFS or OS. | 8 | Retrospective Studies on Clinical Features of Melanosis of the Colon and Its Correlation with the Polypus and Carcinoma of Large Intestine显示文摘OBJECTIVE To investigate the clinical characteristics ofmelanosis of the colon (MC) and its correlation with polypus andcarcinoma of the large intestine.METHODS Clinical feature and colonoscopic findings of 156MC patients were retrospectively analyzed. Final diagnosis of MCcase was made after colonoscopic and pathological examination,and data of the case history was recorded.RESULTS The overall detection rate of MC was 1.12%, with0.46% in males, 0.66% in females. An 82.69% of the patientssuffered various constipations and had a history of takingcathartics. The clinical manifestation of total-colon melanosisoccurred in 83.97% of the patients. There was no significantdifference between the rate of concomitant polypus (27.56%) inthe MC patients and the detection rate of polypus in the patientswho underwent a synchronous colonoscopy (x^2 = 1.205, P = 0.298).Also there was no significant difference between the detectionrate of carcinoma of the large intestine (8.97%) in MC patientsand the detection rate of the same disease in the patients whounderwent synchronous colonoscopy (x^2 = 0.268, P = 0.604). Alogistic regression analysis has shown that the older the age of MCpatients is, the more opportunities of MC and the concomitantpolypus would occur.CONCLUSION MC may be related to constipations and along-term administration of cathartics, among which total-colonmelanosis ranks first in the cases. There is an earlier age of onsetin the females, with a higher detection rate. | 10 | Spatial diff erence in net growth rate of Yesso scallop Patinopecten yessoensis revealed by an aquaculture ecosystem model显示文摘Identifying the main factors on spatial diff erences in net growth rate of Yesso scallop(Patinopecten yessoensis)in culture system is the key to eff ective aquaculture management and development.Coupling a 3D ecosystem model(ROMS-CoSiNE)with a dynamic energy budget model for scallops,a Yesso scallop culture ecosystem(YeSCE)model was established with which scallop growth was simulated with real seeding density and juvenile size from local aquaculture experiments from December 1,2012 to November 30,2013.Results show that the YeSCE model has reasonably simulated the environmental variation and scallop net growth rate in the Changhai sea area.The growth of scallops was slow in winter and midsummer and was limited mainly by temperature.Food availability was a key factor that contributed to the fast growth of the scallops during spring to early summer and in autumn.Generally,the scallops cultured in the north part of the Changhai sea area grew faster than those in the south;and the net growth rate for scallops cultured near the island was signifi cantly higher compare to the others,which is probably correlated to the spatial distribution of food availability.Based on the correlation analysis,the spatial diff erences of the net growth rate were largely aff ected by the length of the match timing of temperatures and food availability.The results of this study provide a scientifi c support for optimizing bottom culture planning and adjusting bottom culture methods. | Xueliang NAN Hao WEI Haiyan ZHANG Hongtao NIE | 2022 | Journal of Oceanology and Limnology2022,40,1: | 1 |

| 17 | A two-year(2020–2021)observation of marine phycotoxins in phytoplankton in typical mariculture areas of East China Sea显示文摘To learn the marine phycotoxins(MPTs)contamination status in mariculture areas in the East China Sea(ECS),from May 2020 to October 2021,80 net-concentrated phytoplankton samples were collected from 12 stations in three typical areas with high incidence of red tides,namely,Gouqi Island,Sandu’ao-Lianjiang,and Zhangzhou-Dongshan Island in ECS,and MPTs of the samples were detected.Six types of toxins were detected in 24 samples from 9 stations.Lipophilic marine toxins(LMTs)were more common and diverse in these areas.Pectenotoxin 2(PTX2)was the main lipophilic marine toxin(LMT)in the concentrated phytoplankton samples and the occurrence showed seasonal differences from north to south.According to the potential risks of pectenotoxin(PTXs)to seafood safety,it is suggested to execute regular monitoring on PTXs in ECS and a mandatory standard should be formulated based on the comprehensive analysis of in-situ monitoring and lab research.Meanwhile,contamination risks of cyclic imine toxins in the north and domoic acid(DA)in the south of ECS should also be paid with attention to.Only 2 paralytic shellfish toxins(PSTs),N-sulfocarbamoyl toxin C2(C2),and decarbamoyl gonyatoxin 3(dcGTX3),were found from spring samples in the north of ECS.As the biggest mussel culture county of China,Gouqi Island showed higher toxin diversity and the toxin detection rate was higher than the other two areas.In Gouqi Island area,PSTs were a serious potential threat in spring,and LMTs instead of PSTs became the main risk in summer-autumn seasons.To ensure the safety of seafood and marine environmental health,it is recommended to conduct long-term targeted tracking and monitoring of MPTs in this and similar mariculture areas. | Xiaoqing TIAN Chengqi FAN Yunyu TANG Haiyan ZHANG Wei KANG Sha CHEN Chongbin LI Ya’nan LU | 2022 | Journal of Oceanology and Limnology2022,40,6: | 1 |
| 18 | Evaluating Tislelizumab, Lenvatinib, and FOLFOX4-HAIC as a Conversion Therapy for Unresectable Hepatocellular Carcinoma显示文摘Background and aims:Conversion therapy downstages tumors and renders patients with unresectable hepatocel-lular carcinoma(HCC)eligible for radical resection.This study aimed to evaluate the efficacy and safety of tislelizumab plus lenvatinib and hepatic artery infusion chemotherapy with oxaliplatin,fluorouracil,and leuco-vorin(FOLFOX4-HAIC)as a first-line conversion therapy.Methods:Clinical data from HCC patients who were treated with the triple therapy between April 2021 and April 2022 were retrospectively analyzed.The primary outcome included objective response rate(ORR),disease control rate(DCR),conversion resection rate(CRR),and treatment-related adverse events(TRAEs).Results:A total of 18 patients completed conversion therapy assessment,which ended on March 27,2023.The patients had a median age of 55.5(37–72)years,and 94.4%were male.According to mRECIST,tumor shrinkage was observed in all patients,with an ORR of 94.4%(17/18),a DCR of 94.4%(17/18),and a median time to response of 1.4(0.7–3.0)months.Successful conversion was observed in 61.1%(11/18)of patients(mRECIST).The CRR and pathological complete response were 38.9%(7/18)and 57.1%(4/7),respectively.The median progression-free survival(PFS)was 17.8 months,while median overall survival was not reached.The 6-and 9-month PFS rates were 83.3%and 66.7%,respectively.The most common TRAE(16/18 patients,88.9%)was an increase in aspartate aminotransferase levels.Conclusion:Tislelizumab combined with lenvatinib and FOLFOX4-HAIC achieved a high conversion rate and acceptable toxicity in patients with unresectable HCC,suggesting that this combination may represent a new conversion strategy for this population. | Kai Tan Xiaojun He Haoran Yuan Shoujie Zhao Chenyu Gong Yaoyao Zhao Haiyan Nan Li Zang Zhonghua Luo Xilin Du | 2023 | iLIVER2023,2,3: | 0 |
