| 1 | Bph30confers resistance to brown planthopperby fortifying sclerenchyma in rice leaf sheaths显示文摘Phloem-feeding insects cause massive losses in agriculture and horticulture.Host plant resistance to phloem-feeding insects is often mediated by changes in phloem composition,which deter insect settling and feeding and decrease viability.Here,we report that rice plant resistance to the phloem-feeding brown planthopper(BPH)is associated with fortification of the sclerenchyma tissue,which is located just beneath the epidermis and a cell layer or two away from the vascular bundle in the rice leaf sheath.We found that BPHs prefer to feed on the smooth and soft region on the surface of rice leaf sheaths called the long-cell block.We identified Bph30 as a rice BPH resistance gene that prevents BPH stylets from reaching the phloem due to the fortified sclerenchyma.Bph30 is strongly expressed in sclerenchyma cells and enhances cellulose and hemicellulose synthesis,making the cell walls stiffer and sclerenchyma thicker.The structurally fortified sclerenchyma is a formidable barrier preventing BPH stylets from penetrating the leaf sheath tissues and arriving at the phloem to feed.Bph30 belongs to a novel gene family,encoding a protein with two leucine-rich domains.Another member of the family,Bph40,also conferred resistance to BPH.Collectively,the fortified sclerenchyma-mediated resistance mechanism revealed in this study expands our understanding of plant-insect interactions and opens a new path for controlling planthoppers in rice. | Shaojie Shi Huiying Wang Lingyun Nie Di Tan Cong Zhou Qian Zhang YiLi Bo Du Jianping Guo Jin Huang DiWu Xiaohong Zheng Wei Guan Junhan Shan Lili Zhu Rongzhi Chen Longjian Xue Linda L.Walling Guangcun He | 2021 | Molecular Plant2021,14,10: | 7 |
| 2 | Real world effectiveness of PCSK-9 inhibitors combined with statins versus statins-based therapy among patients with very high risk of atherosclerotic cardiovascular disease in China(RWE-PCSK study)显示文摘BACKGROUND The efficacy and safety of proprotein convertase subtilisin/kexin type 9(PCSK-9) inhibitors were confirmed by several clinical trials, but its effectiveness in routine clinical practice in China has not been evaluated. This study aims to describe the real world effectiveness of PCSK-9 inhibitors combined with statins compared with statins-based therapy among patients with very high risk of atherosclerotic cardiovascular disease(ASCVD).METHODS This is a multi-center observational study, enrolled patients from 32 hospitals who underwent percutaneous coronary intervention(PCI) from January to June in 2019. There are 453 patients treated with PCSK-9 inhibitors combined with statins in PCSK-9 inhibitor group and 2,610 patients treated with statins-based lipid lowering therapies in statins-based group. The lipid control rate and incidence of major adverse cardiovascular events(MACE) over six months were compared between two groups.A propensity score-matched(PSM) analysis was used to balance two groups on confounding factors. Survival analysis using Kaplan-Meier methods was applied for MACE.RESULTS In a total of 3,063 patients, 89.91% of patients had received moderate or high-intensity statins-based therapy before PCI, but only 9.47% of patients had low-density lipoprotein cholesterol(LDL-C) levels below 1.4 mmol/L at baseline. In the PSM selected patients, LDL-C level was reduced by 42.57% in PCSK-9 inhibitor group and 30.81%(P < 0.001) in statins-based group after six months. The proportion of LDL-C ≤ 1.0 mmol/L increased from 5.29% to 29.26% in PCSK-9 inhibitor group and 0.23% to 6.11% in statins-based group, and the proportion of LDL-C ≤ 1.4 mmol/L increased from 10.36% to 47.69% in PCSK-9 inhibitor group and 2.99% to 18.43% in statins-based group(P < 0.001 for both). There was no significant difference between PCSK-9 inhibitor and statins-based treatment in reducing the risk of MACE(hazard ratio = 2.52, 95% CI: 0.49-12.97, P = 0.250).CONCLUSIONS In the real world, PCSK-9 inhibitors combined with statins could significantly reduce LDL-C levels among patients with very high risk of ASCVD in China. | 4 | Characterization of particle size distribution of mainstream cigarette smoke generated by smoking machine with an electrical low pressure impactor显示文摘Cigarette smoking is a particle-related exposure. Studying the characteristics of the particle size distribution of cigarette smoke can aid in providing knowledge of smoke aerosol attributes. We used an electrical low pressure impactor(ELPI) to measure the particle size distribution of mainstream cigarette smoke generated by a smoking machine and provided a continuum of particle sizes of cigarette smoke from a whole cigarette. The results showed that the aerodynamic diameters(D, geometric mean of a channel) of particles ranged from 0.021 to 1.956 μm, and the number concentrations were on the order of 105–109cm-3for different sizes of particles. The particle number of the size category below 0.1 μm approximated that of the category 0.1–2.0 μm, and the particles in the size category of 0.1–2.0 μm contributed extremely heavily to total particulate mass. | 6 | Tracing of dye molecules in living plants through NaGdF_4:Yb^(3+),Er^(3+)fluorescent nanoprobes显示文摘In this paper, we demonstrated trace of dye molecules in living plants. The NaGdF4:Yb^(3+),Er^(3+) nanoparticles probe was used to detect the rhodamine B(RhB) in bean sprout. It is found that the fluorescencedye can be efficiently imbibed during the growing process and the absorbance presented a position dependence effect, which was supported by the upconversion spectra and the fluorescent image characterization. In addition, the concentration of the residual RhB in bean sprout can be efficiently traced by the synthesized probe based on the fluorescent resonant energy transfer. Finally, the relation between the excitation power, concentration and the ratio of yellow to green emission are discussed in detail. | 7 | Pterygial body epithelium domination of pterygial proliferation with TCF4 as a potential key factor显示文摘AIM: To characterize the proliferative capacity of pterygial epithelium in different regions(head, neck and body) of pterygium and explore the function of transcription factor 4(TCF4) in pterygium proliferation.METHODS: Thirty pterygium tissues and 10 normal conjunctival tissues were obtained from Zhongshan Ophthalmic Center(ZOC) and Guangdong Eye Bank, respectively. Proliferative capacity of head, neck and body in pterygial epithelium was measured using clonal analysis, fold growth analysis and expression profile of proliferative markers revealed by immunofluorescent staining and real-time PCR. The expression of TCF4 was highlighted by double immunofluorescent staining with other proliferation related markers such as proliferating cell nuclear antigen(PCNA) and ATP-binding cassette subfamily G member 2(ABCG2).RESULTS: The proliferative potential of pterygial epithelium was higher than that of normal conjunctival epithelium. High expression levels of proliferative markers(P63α, PCNA and ABCG2) in pterygial body epithelium were observed in immunofluorescent staining and real-time PCR(P<0.05). Also, epithelial cells isolated from pterygial body demonstrated higher proliferative capacity in clonal analysis and fold growth analysis, than those isolated from the head and neck regions. The TCF4 expression in pterygial epithelium was similar to other proliferative markers(P63α, PCNA and ABCG2), as higher in pterygial body than head and neck. | 15 | Synthesis of Quaternary Phosphonium Salts with Ester Functional Group and Its Application to Improve the Thermal Stability of Modified Montmorillonite显示文摘Both of quaternary ammonium and quaternary phosphonium salts of bis-hydroxyethyl terephthalate (BHET) were successfully synthesized and characterized by fourier transform infrared spectroscopy (FT-IR). These two kinds of salts were used to intercalate Na-MMT to yield two kinds of respective organo-modified MMTs. Basal spacing and thermal stability were investigated by using X-ray diffraction (XRD) and thermogravimetric analysis (TGA), respectively. The experimental results show that, as compared with Na-MMT, basal spacings of both of MMTs modified by the quaternary phosphonium salt of BHET (BHETPP) and the quaternary ammonium salt of BHET (BHEA), increase from 12.4 ? to 19.7 ? and 31.3 ?, respec-tively. Thermal stability of BHETPP-modified MMT is much better than that of BHEA-modified MMT, i e, Tonset of BHETPP-modified MMT is around 400 ℃ while Tonset of BHEA-modified MMT is near 250 ℃. | 20 | SDCBP-AS1 destabilizesβ-catenin by regulating ubiquitination and SUMOylation of hnRNP K to suppress gastric tumorigenicity and metastasis显示文摘Background:Gastric cancer(GC)is among the most malignant tumors,yet the pathogenesis is not fully understood,especially the lack of detailed information about the mechanisms underlying long non-coding RNA(lncRNA)-mediated post-translational modifications.Here,the molecular mechanisms and clinical significance of the novel lncRNA syndecan-binding protein 2-antisense RNA 1(SDCBP2-AS1)in the tumorigenesis and progression of GC were investigated.Methods:The expression levels of SDCBP2-AS1 in 132 pairs of GC and adjacent normal tissues were compared,and the biological functions were assessed in vitro and in vivo.RNA pull-down and immunoprecipitation assays were conducted to clarify the interactions of SDCBP2-AS1 and heterogeneous nuclear ribonucleoprotein(hnRNP)K.RNA-sequencing,immunoprecipitation,immunofluorescence,and luciferase analyses were performed to investigate the functions of SDCBP2-AS1.Results:SDCBP2-AS1 was significantly downregulated in GC tissues and pre-dictive of poor patient prognosis.Silencing of SDCBP2-AS1 promoted the proliferation and migration of GC cells both in vitro and in vivo.Mechanically,SDCBP2-AS1 physically bound to hnRNP K to repress SUMOylation of hnRNP K and facilitated ubiquitination of hnRNP K andβ-catenin,thereby promoting the degradation ofβ-catenin in the cytoplasm.Silencing of SDCBP2-AS1 caused SUMOylation of hnRNP K and stabilizedβ-catenin activity,which altered tran-scription of downstream genes,resulting in tumorigenesis and metastasis of GC.Moreover,the knockdown of hnRNP K partially abrogated the effects of SDCBP2-AS1.Conclusions:SDCBP2-AS1 interacts with hnRNP K to suppress tumorigenesis and metastasis of GC and regulates post-transcriptional modifications of hnRNP K to destabilizeβ-catenin.These findings suggest SDCBP2-AS1 as a potential target for the treatment of GC. | Jing Han Menglin Nie Cong Chen Xiaojing Cheng Ting Guo Longtao Huangfu Xiaomei Li Hong Du Xiaofang Xing Jiafu Ji | 2022 | Cancer Communications2022,42,11: | 1 |
