1Enclosed stigma contributes to higher spikelet fertility for rice(Oryza sativa L.) subjected to heat stress显示文摘With global warming, rice plants may be subjected to heat stress more regularly during the heatsensitive flowering stage, causing spikelet sterility and grain yield loss.Stigma exsertion is considered to increase pollen reception and promote female reproductive success.The aim of this study was to investigate the role of stigma exsertion on spikelet fertility at high temperatures.Five rice cultivars(Liangyoupeijiu, Shanyou 63, Huanghuazhan, Nagina 22, and IR64) with differing degrees of stigma exsertion were cultivated and exposed to high temperature at anthesis.Heat-tolerant cultivars did not always show a high percentage of spikelets with exserted stigmas, and vice versa.Irrespective of the presence of more pollen grains on exserted stigmas, spikelets with exserted stigmas did not show greater spikelet fertility than spikelets with fewer exserted stigmas or hidden stigmas under heat stress.GA3 application augmented the percentage of spikelets with exserted stigmas;however, it did not increase spikelet fertility under heat stress.Spikelet fertility of whole panicles was negatively correlated with the percentage of spikelets with exserted stigmas, but positively with that with hidden stigmas.Viability of the hidden stigmas was less reduced than that of exserted stigmas under heat stress, suggesting that hidden stigmas have an advantage in maintaining viability.Heat stress delayed anther dehiscence and reduced the viabilities of both exserted stigmas and pollens, thereby causing low spikelet fertility.Together, these results suggest that high spikelet fertility does not depend on stigma exsertion and that enclosed stigma generally contributes to higher spikelet fertility and heat tolerance under high-temperature conditions during flowering in rice.Chao Wu Kehui Cui Qiuqian Hu Wencheng Wang Lixiao Nie Jianliang Huang Shaobing Peng 2019The Crop Journal2019,7,3:4
2SHRIMP zircon U-Pb dating for two episodes of migmatization in the Dabie orogen显示文摘Zircon CL imaging and SHRIMP U-Pb dating were carried out for migmatite in the Dabie orogen. Zircons from the Manshuihe migmatite show clear core-rim structures. The cores display sector or weak zoning and low Th/U ratios of 0.01 to 0.17, indicating their precipitation from metamorphic fluid. They yield a weighted mean age of 137±5 Ma. By contrast, the rims exhibit planar or nebulous zoning with relatively high Th/U ratios of 0.35 to 0.69, suggesting their growth from metamorphic melt. They give a weighted mean age of 124±2 Ma. Zircons from the Fenghuangguan migmatite also display core-rim structures. The cores are weakly oscillatory zoned or unzoned with high Th/U ratios of 0.21 to 3.03, representing inherited zircons of magmatic origin that experienced different degrees of solid-state recrystallization. SHRIMP U-Pb analyses obtain that its protolith was emplaced at 768±12 Ma, consistent with middle Neoproterozoic ages for protoliths of most UHP metaigneous rocks in the Dabie-Sulu orogenic belt. By contrast, the rims do not show significant zoning and have very low Th/U ratios of 0.01 to 0.09, typical of zircon crystallized from metamorphic fluid. They yield a weighted 206Pb/238U age of 137±4 Ma. Taking the two case dates together, it appears that there are two episodes of zircon growth and thus migmati-tization at 137±2 Ma and 124±2 Ma, respectively, due to metamorphic dehydration and partial melting. The appearance of metamorphic dehydration corresponds to the beginning of tectonic extension thus to the tectonic switch from crustal compression to extension in the Dabie orogen. On the other hand, the partial melting is responsible for the extensional climax, resulting in formation of coeval migmatite, granitoid and granulite. 4Tracing of dye molecules in living plants through NaGdF_4:Yb^(3+),Er^(3+)fluorescent nanoprobes显示文摘In this paper, we demonstrated trace of dye molecules in living plants. The NaGdF4:Yb^(3+),Er^(3+) nanoparticles probe was used to detect the rhodamine B(RhB) in bean sprout. It is found that the fluorescencedye can be efficiently imbibed during the growing process and the absorbance presented a position dependence effect, which was supported by the upconversion spectra and the fluorescent image characterization. In addition, the concentration of the residual RhB in bean sprout can be efficiently traced by the synthesized probe based on the fluorescent resonant energy transfer. Finally, the relation between the excitation power, concentration and the ratio of yellow to green emission are discussed in detail. 13Response of spikelet water status to high temperature and its relationship with heat tolerance in rice显示文摘In rice, high-temperature stress(HT) during flowering results in decreased grain yield via a reduction in spikelet fertility;however, the effect of plant water status on spikelet fertility under HT remains unknown. To investigate the relationship between spikelet water status and spikelet fertility under HT, two experiments were performed under temperature-controlled conditions using four genotypes with varying tolerance to HT. Rice plants were exposed to HT for seven consecutive days during the flowering stage under three soil water treatments(soil water potential 0, -20, and -40 kPa), as well as under hydroponic conditions in a separate experiment. HT significantly decreased spikelet fertility, pollen fertility, and anther dehiscence under each of the three water treatments. HT significantly increased the spikelet transpiration rate, and this change was accompanied by a significant decrease in the internal temperature of the spikelets. HT decreased pollen grain diameter in heat-sensitive genotypes. HT had varying effects on the water potential of panicles and anthers but increased anther soluble-sugar concentration. Different aquaporin genes showed different expression profiles under HT, and the expression levels of PIPs for plasma membrane intrinsic proteins and TIPs for tonoplast intrinsic proteins increased in anthers but decreased in glumes.Correlation analyses showed that anther dehiscence and pollen(spikelet) fertility were tightly associated with anther water status, and the expression levels of almost all anther aquaporin genes were significantly correlated with anther dehiscence under HT. In summary, an increased spikelet transpiration rate and decreased internal spikelet temperature were associated with alleviation of the effects of HT in rice genotypes with varying degrees of heat tolerance, and the response of spikelet water status to HT, involving increased total expression of aquaporins and soluble sugar content, thereby improved pollen fertility,anther dehiscence, and spikelet fertility, especially in heat-resistant genotypes. 14MicroRNA319-mediated gene regulatory network impacts leaf development and morphogenesis in poplar显示文摘MicroRNA319(miR319)has been implicated in leaf development in a number of plant species.Here we study the roles of miR319a and its regulated network in leaf development in poplars.Over-expression of miR319a in Populus alba×Populus glandulosa caused dwarf statures,narrow leaf blades and serrated leaf margins.The vascular bundles and bundle sheaths in transgenic leaves had more layers of cells than those in the leaves of control plants,indicating enhanced lignification in these cells.Among the 93 putative targets of miR319a predicted with the psRNATarget tool,only three genes,TCP(TEOSINTE BRANCHED1,CYCLOIDEA,and PROLIFERATING CELL NUCLEAR ANTIGEN BINDING FACTOR),were differentially expressed in the leaves of MIR319a-over-expression transgenic lines.With the RNA-seq data sets from multiple MIR319a over-expression transgenic lines,we built a three-layered gene regulatory network mediated by miR319a using Top-down graphic Gaussian model(GGM)algorithm that is capable of capturing causal relationships from transcriptomic data.The results support that miR319a primarily regulates the lignin biosynthesis,leaf development and differentiation as well as photosynthesis via miR319-MEE35/TCP4,miR319-TCP2 and miR319-TCP2-1 regulatory modules.Yanxia Cheng Lihu Wang Manzar Abbas Xiong Huang Qiao Wang Aimin Wu Hairong Wei Shaobing Peng Xinren Dai Quanzi Li 2021Forestry Research2021,1,1:0
15Control of rice ratooning ability by a nucleoredoxin that inhibits histidine kinase dimerization to attenuate cytokinin signaling in axillary buds显示文摘Rice ratooning,the fast outgrowth of dormant buds on stubble,is an important cropping practice in rice production.However,the low ratooning ability(RA)of most rice varieties restricts the application of this cost-efficient system,and the genetic basis of RA remains unknown.In this study,we dissected the genetic architecture of RA by a genome-wide association study in a natural rice population.Rice ratooning ability 3(RRA3),encoding a hitherto not characterized nucleoredoxin involved in reduction of disulfide bonds,was identified as the causal gene of a major locus controlling RA.Overexpression of RRA3 in rice significantly accelerated leaf senescence and reduced RA,whereas knockout of RRA3 significantly delayed leaf senescence and increased RA and ratoon yield.We demonstrated that RRA3 interacts with Oryza sativa histidine kinase 4(OHK4),a cytokinin receptor,and inhibits the dimerization of OHK4 through disulfide bond reduction.This inhibition ultimately led to decreased cytokinin signaling and reduced RA.In addition,variations in the RRA3 promoter were identified to be associated with RA.Introgression of a superior haplotype with weak expression of RRA3 into the elite rice variety Guichao 2 significantly increased RA and ratoon yield by 23.8%.Collectively,this study not only uncovers an undocumented regulatory mechanism of cytokinin signaling through de-dimerization of a histidine kinase receptor-but also provides an eximious gene with promising value for ratoon rice breeding.Yilong Yao Denghao Xiang Nai Wu Yao Wang Yu Chen Yang Yuan Ying Ye Dan Hu Chang Zheng Yu Yan Qingya Lv Xiaokai Li Guoxing Chen Honghong Hu Haiyan Xiong Shaobing Peng Lizhong Xiong 2023Molecular Plant2023,16,12:0
