Enhanced secondary pollution offset reduction of primary emissions during COVID-19 lockdown in China显示文摘To control the spread of the 2019 novel coronavirus(COVID-19),China imposed nationwide restrictions on the movement of its population(lockdown)after the Chinese New Year of 2020,leading to large reductions in economic activities and associated emissions.Despite such large decreases in primary pollution, there were nonetheless several periods of heavy haze pollution in eastern China, raising questions about the well-established relationship between human activities and air quality. Here, using comprehensive measurements and modeling,we show that the haze during the COVID lockdown was driven by enhancements of secondary pollution. In particular, large decreases in NOx emissions from transportation increased ozone and nighttime NO_(3)radical formation, and these increases in atmospheric oxidizing capacity in turn facilitated the formation of secondary particulate matter. Our results, afforded by the tragic natural experiment of the COVID-19 pandemic, indicate that haze mitigation depends upon a coordinated and balanced strategy for controlling multiple pollutants.Xin Huang Aijun Ding Jian Gao Bo Zheng Derong Zhou Ximeng Qi Rong Tang Jiaping Wang Chuanhua Ren Wei Nie Xuguang Chi Zheng Xu Liangduo Chen Yuanyuan Li Fei Che Nini Pang Haikun Wang Dan Tong Wei Qin Wei Cheng Weijing Liu Qingyan Fu Baoxian Liu Fahe Chai Steven JDavis Qiang Zhang Kebin He 2021National Science Review2021,8,2:47
Modes of genetic adaptations underlying functional innovations in the rumen显示文摘The rumen is the hallmark organ of ruminants and hosts a diverse ecosystem of microorganisms that facilitates efficient digestion of plant fibers. We analyzed 897 transcriptomes from three Cetartiodactyla lineages:ruminants, camels and cetaceans,as well as data from ruminant comparative genomics and functional assays to explore the genetic basis of rumen functional innovations. We identified genes with relatively high expression in the rumen,of which many appeared to be recruited from other tissues. These genes show functional enrichment in ketone body metabolism, regulation of microbial community, and epithelium absorption, which are the most prominent biological processes involved in rumen innovations.Several modes of genetic change underlying rumen functional innovations were uncovered, including coding mutations, genes newly evolved, and changes of regulatory elements. We validated that the key ketogenesis rate-limiting gene(HMGCS2)with five ruminant-specific mutations was under positive selection and exhibits higher synthesis activity than those of other mammals. Two newly evolved genes(LYZ1 and DEFB1)are resistant to Gram-positive bacteria and thereby may regulate microbial community equilibrium.Furthermore,we confirmed that the changes of regulatory elements accounted for the majority of rumen gene recruitment. These results greatly improve our understanding of rumen evolution and organ evo-devo in general.Xiangyu Pan Yudong Cai Zongjun Li Xianqing Chen Rasmus Heller Nini Wang Yu Wang Chen Zhao Yong Wang Han Xu Songhai Li Ming Li Cunyuan Li Shengwei Hu Hui Li Kun Wang Lei Chen Bin Wei Zhuqing Zheng Weiwei Fu Yue Yang Tingting Zhang Zhuoting Hou Yueyang Yan Xiaoyang Lv Wei Sun Xinyu Li Shisheng Huang Lixiang Liu Shengyong Mao Wenqing Liu Jinlian Hua Zhipeng Li Guojie Zhang Yulin Chen Xihong Wang Qiang Qiu Brian PDalrymple Wen Wang Yu Jiang 2021Science China(Life Sciences)2021,64,1:3

The F-box protein SHORT PRIMARY ROOT modulates primary root meristem activity by targeting SEUSS-LIKE protein for degradation in rice显示文摘Root meristem activity is essential for root morphogenesis and adaptation, but the molecular mechanism regulating root meristem activity is not fully understood. Here,we identify an F-box family E3 ubiquitin ligase named SHORT PRIMARY ROOT(SHPR) that regulates primary root(PR)meristem activity and cell proliferation in rice.SHPR loss-of-function mutations impair PR elongation in rice.SHPR is involved in the formation of an SCF complex with the Oryza sativa SKP1-like protein OSK1/20.We show that SHPR interacts with Oryza sativa SEUSS-LIKE(OsSLK) in the nucleus and is required for OsSLK polyubiquitination and degradation by the ubiquitin 26S-proteasome system(UPS).Transgenic plants overexpressing OsSLK display a shorter PR phenotype, which is similar to the SHPR loss-of-function mutants.Genetic analysis suggests that SHPR promotes PR elongation in an OsSLK-dependent manner.Collectively, our study establishes SHPR as an E3 ubiquitin ligase that targets OsSLK for degradation, and uncovers a protein ubiquitination pathway as a mechanism for modulating root meristem activity in rice.Nini Ma Nian Li Zhongmao Yu Chunli Chen Dao-Xiu Zhou Yu Zhao 2023Journal of Integrative Plant Biology2023,65,8:0
Novel flavin-containing monooxygenase protein FMO1 interacts with CAT2 to negatively regulate drought tolerance through ROS homeostasis and ABA signaling pathway in tomato显示文摘Drought stress is themajor abiotic factor that can seriously affect plant growth and crop production. The functions of flavin-containing monooxygenases(FMOs)are known in animals. They addmolecular oxygen to lipophilic compounds or produce reactive oxygen species(ROS).However, little information on FMOs in plants is available. Here,we characterized a tomato drought-responsive gene that showed homology to FMO,and it was designated as FMO1.FMO1 was downregulated promptly by drought and ABA treatments.Transgenic functional analysis indicated that RNAi suppression of the expression of FMO1(FMO1-Ri)improved drought tolerance relative to wild-type(WT)plants,whereas overexpression of FMO1(FMO1-OE)reduced drought tolerance. The FMO1-Ri plants exhibited lower ABA accumulation, higher levels of antioxidant enzyme activities, and less ROS generation comparedwith theWTand FMO1-OE plants under drought stress.RNA-seq transcriptional analysis revealed the differential expression levels of many drought-responsive genes thatwere co-expressed with FMO1,including PP2Cs,PYLs,WRKY,and LEA.Using Y2H screening,we found that FMO1 physically interacted with catalase 2(CAT2),which is an antioxidant enzyme and confers drought resistance. Our findings suggest that tomato FMO1 negatively regulates tomato drought tolerance in the ABA-dependent pathway and modulates ROS homeostasis by directly binding to SlCAT2.Lulu Wang Yinlian Zhou Yin Ding Chunrui Chen Xueting Chen Nini Su Xingguo Zhang Yu Pan Jinhua Li 2023Horticulture Research2023,10,4:0
CNEReg Interprets Ruminant-specific Conserved Non-coding Elements by Developmental Gene Regulatory Network显示文摘The genetic information coded in DNA leads to trait innovation via a gene regulatory network(GRN)in development. Here,we developed a conserved non-coding element interpretation method to integrate multi-omics data into gene regulatory network(CNEReg)to investigate the ruminant multi-chambered stomach innovation. We generated paired expression and chromatin accessibility data during rumen and esophagus development in sheep, and revealed 1601 active ruminantspecific conserved non-coding elements(active-RSCNEs).To interpret the function of these activeRSCNEs,we defined toolkit transcription factors(TTFs)and modeled their regulation on rumenspecific genes via batteries of active-RSCNEs during development. Our developmental GRN revealed 18 TTFs and 313 active-RSCNEs regulating 7 rumen functional modules.Notably,6 TTFs(OTX1,SOX21,HOXC8,SOX2,TP63,and PPARG),as well as 16 active-RSCNEs,functionally distinguished the rumen from the esophagus. Our study provides a systematic approach to understanding how gene regulation evolves and shapes complex traits by putting evo-devo concepts into practice with developmental multi-omics data.Xiangyu Pan Zhaoxia Ma Xinqi Sun Hui Li Tingting Zhang Chen Zhao Nini Wang Rasmus Heller Wing Hung Wong Wen Wang Yu Jiang Yong Wang 2023Genomics, Proteomics & Bioinformatics2023,21,3:0
A novel lytic phage potentially effective for phage therapy against Burkholderia pseudomallei in the tropics显示文摘Background:Burkholderia pseudomallei is a tropical pathogen that causes melioidosis. Its intrinsic drug-resistance is a leading cause of treatment failure, and the few available antibiotics require prolonged use to be effective. This study aimed to assess the clinical potential of B.pseudomallei phages isolated from Hainan,China. Methods:Burkholderia pseudomallei strain(HNBPoo1)was used as the isolation host, and phages were recovered from domestic environmental sources, which were submitted to the host range determination, lytic property assays, and stability tests. The best candidate was examined via the transmission electron microscope for classification. With its genome sequenced and analyzed, its protective efficacy against B.pseudomallei infection in A549 cells and Caenorhabditis elegans was evaluated,in which cell viability and survival rates were compared using the one-way ANOVA method and the log-rank test. Results:A phage able to lyse 24/25 clinical isolates was recovered. It was classified in the Podoviridae family and was found to be amenable to propagation.Under the optimal multiplicity of infection(MOl)of o.1,an eclipse period of around 20 min and a high titer(io12 PFU/ml)produced within 1 h were demonstrated. This phage was found stabile at a wide range of temperatures(24,37,40,50,and 60 C)and pH values(3-12).After being designated as vB_BpP_HN01,it was fully sequenced, and the 71,398 bp linear genome, containing 93 open reading frames and a tRNA-Asn,displayed a low sequence similarity with known viruses.Additionally, protective effects of applications of vB_BpP_HNo1(MOl=0.1 and MOl=1)alone or in combination with antibiotics were found to improve viability of infected cells(70.6±6.8%,85.8±5.7%,91.9±1.8%,and 96.8±1.8%,respectively).A significantly reduced mortality(10%)and a decreased pathogen load were demonstrated in infected C.elegans following the addition of this phage. Conclusions:As the frst B.pseudomallei phage was isolated in Hainan,China, phage vB_BpP_HNO1 was characterized by promising lytic property, stability, and effciency of bacterial elimination during the in vitro/vivo experiments. Therefore,we can conclude that it is a potential alternative agent for combating melioidosis.Yanshuang Wang Xuemiao Li David A.B.Dance Han Xia Chen Chen Nini Luo Anyang Li Yanmei Li Qiao Zhu Qinghui Sun Xingyong Wu Yingfei Zeng Lin Chen Shen Tian Qianfeng Xia 2022Infectious Diseases of Poverty2022,11,4:0
Analysis of Clinical Features and Influencing Factors of Depression and Cognitive Dysfunctionin Parkinson's Patients显示文摘Objective:To explore the clinical characteristics and analyze the influencing factors of depression and cognitive impairment in Parkinson's patients.Methods:127 patients with Parkinson's disease who were treated in two hospitals from March 2021 to March 2023 were selected for this study. The assessment of depression was conducted using the Geriatric Depression Self-Rating Scale(GDS).In addition, cognitive function was evaluated through the Montreal Cognitive Assessment(MOCA),and the clinical attributes of patients with cognitive impairment were examined. The study also aimed to analyze the factors that influence cognitive impairment in this patient population. Results:The progression of the disease, presence of cognitive impairment, and the Hoehn-Yahr stage were identified as significant influencing factors for depression among the 127 patients with Parkinson's disease(P<0.05).The factors that influence cognitive impairment included the level of education,Hoehn-Yahr stage, and depression(P<0.05).Conclusion:Depression and cognitive dysfunction is relatively common in patients with Parkinson's disease. The occurrence of depression and cognitive dysfunction varies significantly based on factors such as the duration of the disease, varying drug dosages, and the severity of the condition. Therefore,it is necessary to treat patients with Parkinson's disease.Nini Li Dongdong Zhang 2023Journal of Clinical and Nursing Research2023,7,5:0
