| 1 | Ribosome profiling analysis identified a KRAS-interacting microprotein that represses oncogenic signaling in hepatocellular carcinoma cells显示文摘The roles of concealed microproteins encoded by long noncoding RNAs(lncRNAs)are gradually being exposed,but their functions in tumorigenesis are still largely unclear.Here,we identify and characterize a conserved 99-amino acid microprotein named KRASIM that is encoded by the putative lncRNA NCBP2-AS2.KRASIM is differentially expressed in normal hepatocytes and hepatocellular carcinoma(HCC)cells and can suppress HCC cell growth and proliferation.Mechanistically,KRASIM interacts and colocalizes with the KRAS protein in the cytoplasm of human HuH-7 hepatoma cells.More importantly,the overexpression of KRASIM decreases the KRAS protein level,leading to the inhibition of ERK signaling activity in HCC cells.These results demonstrate a novel microprotein repressor of the KRAS pathway for the first time and provide new insights into the regulatory mechanisms of oncogenic signaling and HCC therapy. | Wenli Xu Bing Deng Penghui Lin Chang Liu Bin Li Qiaojuan Huang Hui Zhou Jianhua Yang Lianghu Qu | 2020 | Science China(Life Sciences)2020,63,4: | 11 |
| 2 | The Late Carboniferous–Early Permian Ocean-Continent Transition in the West Junggar,Central Asian Orogenic Belt: Constraints from Columnar Jointed Rhyolite显示文摘The West Junggar of the western Central Asian Orogenic Belt is one of the typical regions in the term of ocean subduction, contraction and continental growth in the Late Paleozoic. However, it is still controversial on the exact time of ocean-continent transition so far. This study investigates rhyolites with columnar joint in the West Junggar for the first time.Based on zircon U-Pb dating, we determined that the ages of the newly-discovered rhyolites are between 303.6 and 294.5 Ma, belonging to Late Carboniferous–Early Permian, which is the oldest rhyolite with columnar joint preserved in the world at present. Geochemical results show that the characteristics of the major element compositions include a high content of SiO_2(75.78–79.20 wt%) and a moderate content of Al_2O_3(12.21–13.19 wt%). The total alkali content(K_2O +Na_2O) is 6.14–8.05 wt%, among which K_2O is 2.09–4.72 wt% and the rate of K_2O/Na_2O is 0.38–3.05. Over-based minerals such as Ne, Lc, and Ac do not appear. The contents of TiO_2(0.09–0.24 wt%), CaO(0.15–0.99 wt%) and MgO(0.06–0.18 wt%) are low. A/CNK=0.91–1.68, A/NK=1.06–1.76, and as such, these are associated with the quasi-aluminum-weak peraluminous high potassium calc-alkaline and some calc-alkaline magma series. These rhyolites show a significant negative Eu anomaly with relative enrichment of LREE and LILE(Rb, Ba, Th, U, K) and depletion of Sr, HREE and HFSE(Nb, Ta, Ti, P). These rhyolites also have the characteristics of an A2-type granite, similar to the Miaoergou batholith,which indicates they both were affected by post-orogenic extension. | 3 | Diagenesis and Diagenetic Evolution of Deltaic and Neritic Gas-Bearing Sandstones in the Lower Mingyuefeng Formation of Paleogene, Lishui Sag, East China Sea Shelf Basin: Implications for Depositional Environments and Sequence Stratigraphy Controls显示文摘Gas-bearing deposits in the Lower Mingyuefeng Formation of Paleogene, Lishui Sag, East China Sea Shelf Basin consist of shoreface sandstones of the highstand systems tract(HST) and transgressive systems tract(TST), and deltaic sandstones of the lowstand systems tract(LST) and falling stage systems tract(FSST).Detailed petrographic observations suggest that the diagenetic features and related evolution of these deposits cannot be simply characterized and demonstrated in the depth domain.However, the occurrence of diagenetic minerals systematically depends on the studied interval within the HST, TST, LST, and FSST; therefore, diagenesis in this region can be better constrained when studied in the context of the depositional environments and sequence stratigraphic framework.The eogenetic processes in such settings include:(1) microcrystalline siderite precipitated as concretions in almost all environments and systems tracts, which inhibited further mechanical compaction;(2) grain dissolution and kaolinitization occurred in shoreface HST sandstones and deltaic LST and FSST sandstones;(3) glaucony was locally observed, which did not clearly reflect the controls of facies or sequence stratigraphy; and(4) cementation by pyrite aggregates occurred in the shoreface HST sandstones and deltaic LST sandstones.The mesogenetic diagenesis includes:(1) partial conversion of kaolinite into dickite in deltaic LST sandstones, and minor chlorite cementation in deltaic FSST sandstones;(2) transformation of kaolinite into illite and quartz cementation in deltaic LST and FSST sandstones;(3) frequent precipitation of ankerite and ferroan calcite in shoreface TST sandstones and early HST sandstones, forming baffles and barriers for fluid flow, with common calcite in shoreface HST sandstones as a late diagenetic cement; and(4) formation of dawsonite in the deltaic LST and FSST sandstones, which is interpreted to be a product of the invasion of a CO2-rich fluid, and acts as a good indicator of CO2-bearing reservoirs.This study has thus constructed a reliable conceptual model to describe the spatial and temporal distribution of diagenetic alterations.The results may provide an entirely new conceptual framework and methodology for successful gas exploration in the continental margins of offshore China, thus allowing us to predict and unravel the distribution and quality evolution of clastic reservoirs at a more detailed and reliable scale. | XU Fa ZHANG Penghui ZHANG Jinliang LIU Jinshui HOU Guowei ZHANG Ming LI Jingzhe LIU Shasha GUO Jiaqi MENG Ningning | 2015 | Acta Geologica Sinica(English Edition)2015,89,5: | 5 |
| 4 | Effects of growth temperature on μc-Si:H films prepared by plasma assistant magnetron sputtering显示文摘Hydrogenated microcrystalline silicon thin films (μc-Si:H) were deposited by plasma assistant magnetron sputtering in Ar-H 2 gas mixture. The effects of growth temperature from 150 to 450 C on properties of deposited Si films were investigated at two different hydrogen diluted gases with [H 2 ]/([Ar]+[H 2 ]) of 10% and 50% at 3 Pa. The crystallinity of Si films examined by Raman scattering exhibited higher degrada- tion by lowering growth temperature from 250 to 150 C in low hydrogen diluted gas of 10% than that in high hydrogen diluted gas of 50%. The IR absorption band around 845 and 890 cm 1 as well as calculated concentration of bonded hydrogen showed more obvious decrease of samples deposited in low hydrogen diluted gas of 10% than that in high hydrogen diluted gas of 50%. | 5 | CsTCPs regulate shoot tip development and catechin biosynthesis in tea plant (Camellia sinensis)显示文摘The growth of leaves and biosynthesis of characteristic secondary metabolites are critically important for tea production and quality control.However,little is known about the coordinated regulation of leaf development and catechin biosynthesis in tea plants.Here,we reported that TCP TFs are involved in both catechin biosynthesis and leaf development.An integrated analysis of catechin profiling and CsTCP expression in different tissues of plants under various environmental conditions at different developmental stages indicated significant correlations between the transcript levels of CIN-type TCPs and catechin production.CIN-type CsTCP3 and CsTCP4 and PCF-type CsTCP14 interacted with the MYB-bHLH-WD40 repeat(MBW)complex by forming a CsTCP3-CsTT8 heterodimer and modulating the transactivation activity of the promoters of anthocyanin synthase(CsANSl)and anthocyanidin reductase(CsANRl).Four types of microRNA/target modules,miR319b/CsTCP3-4,miR164b/CsCUC,miR396/CsGRF-GIF,and miR165b/HD-ZIPIII ones,were also identified and characterized for their functions in the regulation of the development of teaplant shoot tips and leaf shape.The results of these modules were reflected by their different expression patterns in developing buds and leaves that had distinctly different morphologies in three different teaplant varieties.Their roles in the regulation of catechin biosynthesis were also further verified by manipulation of microRNA319b(miR319b),which targets the transcripts of CsTCP3 and CsTCP4.Thus,CsTCPs represent at least one of these important groups of TFs that can integrate tea plant leaf development together with secondary metabolite biosynthesis.Our study provides new insight into shoot tip development and catechin production in tea plants and lays a foundation for further mechanistic understanding of the regulation of tea plant leaf development and secondary metabolism. | Shuwei Yu Penghui Li Xuecheng Zhao Mangmang Tan Muhammad Zulfiqar Ahmad Yujie Xu Million Tadege Jian Zhao | 2021 | Horticulture Research2021,8,1: | 2 |

| 8 | Cytosolic TGM2 promotes malignant progression in gastric cancer by suppressing the TRIM21-mediated ubiquitination/degradation of STAT1 in a GTP binding-dependent modality显示文摘Background:Previous studies have revealed the critical role of transglutaminase 2(TGM2)as a potential therapeutic target in cancers,but the oncogenic roles and underlying mechanisms of TGM2 in gastric cancer(GC)are not fully understood.In this study,we examined the role and potential mechanism of TGM2 in GC.Methods:Western blotting,immunohistochemistry,CCK8,colony formation and transwell assays were used to measure TGM2 expression in the GC cells and tissues and to examine the in vitro role of TGM2 in GC.Xenograft and in vivo metastasis experiments were performed to examine the in vivo role of TGM2 in GC.Gene set enrichment analysis,quantitative PCR and western blotting were conducted to screen for potential TGM2 targets involved in GC.Gain/loss-offunction and rescue experiments were conducted to detect the biological roles of STAT1 in GC cells in the context of TGM2.Co-immunoprecipitation,mass spectrometry,quantitative PCR and western blotting were conducted to identify STAT1-interacting proteins and elucidate their regulatory mechanisms.Mutations in TGM2 and two molecules(ZM39923 and A23187)were used to identify the enzymatic activity of TGM2 involved in the malignant progression of GC and elucidate the underlying mechanism.Results:In this study,we demonstrated elevated TGM2 expression in the GC tissues,which closely related to pathological grade,and predicted poor survival in patients with GC.TGM2 overexpression or knockdown promoted(and inhibited)cell proliferation,migration,and invasion,which were reversed by STAT1 knockdown or overexpression.Further studies showed that TGM2 promoted GC progression by inhibiting STAT1 ubiquitination/degradation.Then,tripartite motif-containing protein 21(TRIM21)was identified as a ubiquitin E3 ligase of STAT1 in GC.TGM2 maintained STAT1 stability by facilitating the dissociation of TRIM21 and STAT1 with GTP-binding enzymatic activity.A23187 abolished the role of TGM2 in STAT1 and reversed the pro-tumor role of TGM2 in vitro and in vivo.Conclusions:This study revealed a critical role and regulatory mechanism of TGM2 on STAT1 in GC and highlighted the potential of TGM2 as a therapeutic target,which elucidates the development of medicine or strategies by regulating the GTP-binding activity of TGM2 in GC. | Lu Zhang Qingya Li Jing Yang Penghui Xu Zhe Xuan Jianghao Xu Zekuan Xu | 2023 | Cancer Communications2023,43,1: | 1 |

| 10 | Power spectral density analysis for nonlinear systems based on Volterra series显示文摘A consequence of nonlinearities is a multi-harmonic response via a monoharmonic excitation.A similar phenomenon also exists in random vibration.The power spectral density(PSD)analysis of random vibration for nonlinear systems is studied in this paper.The analytical formulation of output PSD subject to the zero-mean Gaussian random load is deduced by using the Volterra series expansion and the conception of generalized frequency response function(GFRF).For a class of nonlinear systems,the growing exponential method is used to determine the first 3 rd-order GFRFs.The proposed approach is used to achieve the nonlinear system’s output PSD under a narrow-band stationary random input.The relationship between the peak of PSD and the parameters of the nonlinear system is discussed.By using the proposed method,the nonlinear characteristics of multi-band output via single-band input can be well predicted.The results reveal that changing nonlinear system parameters gives a one-of-a-kind change of the system’s output PSD.This paper provides a method for the research of random vibration prediction and control in real-world nonlinear systems. | Penghui WU Yan ZHAO Xianghong XU | 2021 | Applied Mathematics and Mechanics(English Edition)2021,42,12: | 1 |
| 11 | Spectral characteristics of dissolved organic matter in sediment pore water from Pearl River Estuary显示文摘As key parts of land-sea transition zones,estuary ecosystems play a very important role in the ocean carbon cycle processes.The sources,degradation,and preservation of dissolved organic matter(DOM)in estuaries have long been the subject of intense study.To examine the aforementioned issues,this study examined three-dimensional fluorescence spectroscopy and ultraviolet-visible absorption spectroscopy to determine the spatial distribution and sources of DOM in the pore water of three sedimentary cores from the Pearl River Estuary(S1,S2 and S3,with increasing salinity).Using the parallel factor analysis(PARAFAC)method to analyze the three-dimensional fluorescence spectrum data,five fluorescent components were obtained—three humic-like components(C1,C3,and C4),and two protein-like components(C2 and C5).C2 exhibited a significant positive correlation with the sediment microbial deoxyribose nucleic acid(DNA)concentration(R~2=0.69,P<0.01),indicating that the protein-like component C2 might be derived from the catabolism of in situ microbes.C5 displayed a relatively weak correlation with DNA concentration(R~2=0.40,P<0.05),presumably due to the incorporation of phenolic compounds,which have a fluorescence peak very similar to that of protein-like components.The source of humic-like fluorescent components is extremely complex.The content at station S1 was relatively high(1.45–8.83 R.U.),which implies that terrestrial inputs had a significant influence.The three humic-like components showed similar distributions at S2 and S3,and the fluorescence intensity was rather low;this result indicates that the DOM at these two stations was more likely affected by the metabolism of algae and microorganisms.The humification index(HIX)and the fluorescence intensity of protein-like components increased and decreased,respectively,with depth.There was a significant positive correlation between the relative content of protein-like components and the spectral slope ratio(SR),which indicates that DOM transitioned from low-molecular-weight protein-like components in the surface sediment to high-molecular-weight humic-like components in the subsurface.This study provides valuable information for understanding the pore water size/reactivity(PWSR)model of DOM and its biochemical processes occurring in estuary sediments. | Yang XU Penghui LI Chuanlun ZHANG Peng WANG | 2021 | Science China Earth Sciences2021,64,1: | 1 |

| 16 | N6-methyladenosine modification of CENPF mRNA facilitates gastric cancer metastasis via regulating FAK nuclear export显示文摘Background:N6-methyladenosine(m^(6)A)modification is the most common modification that occurs in eukaryotes.Although substantial effort has been made in the prevention and treatment of gastric cancer(GC)in recent years,the prognosis of GC patients remains unsatisfactory.The regulatory mechanism between m^(6)A modification and GC development needs to be elucidated.In this study,we examined m^(6)A modification and the downstream mechanism in GC.Methods:Dot blotting assays,The Cancer Genome Atlas analysis,and quantitative real‑time PCR(qRT-PCR)were used to measure the m^(6)A levels in GC tissues.Methylated RNA-immunoprecipitation sequencing and RNA sequencingwere performed to identify the targets ofm^(6)Amodification.Western blotting,Transwell,wound healing,and angiogenesis assays were conducted to examine the role of centromere protein F(CENPF)in GC in vitro.Xenograft,immunohistochemistry,and in vivo metastasis experiments were conducted to examine the role of CENPF in GC in vivo.Methylated RNA-immunoprecipitation-qPCR,RNA immunoprecipitation-qPCR and RNA pulldown assays were used to verify the m^(6)A modification sites of CENPF.Gain/loss-of-function and rescue experiments were conducted to determine the relationship between CENPF and the mitogen-activated protein kinase(MAPK)signaling pathway in GC cells.Coimmunoprecipitation,mass spectrometry,qRT-PCR,and immunofluorescence assays were performed to explore the proteins that interact with CENPF and elucidate the regulatory mechanisms between them.Results:CENPF was upregulated in GC and facilitated the metastasis of GC both in vitro and in vivo.Mechanistically,increasedm^(6)A modification of CENPF was mediated by methyltransferase 3,and this modified molecule could be recognized by heterogeneous nuclear ribonucleoprotein A2/B1(HNRNPA2B1),thereby promoting its mRNA stability.In addition,the metastatic phenotype of CENPF was dependent on the MAPK signaling pathway.Furthermore,CENPF could bind to FAK and promote its localization in the cytoplasm.Moreover,we discovered that high expression of CENPF was related to lymphatic invasion and overall survival in GC patients.Conclusions:Our findings revealed that increased m^(6)A modification of CENPF facilitates the metastasis and angiogenesis of GC through the CENPF/FAK/MAPK and epithelial-mesenchymal transition axis.CENPF expression was correlated with the clinical features of GC patients;therefore,CENPF may serve as a prognostic marker of GC. | Penghui Xu Jing Yang Zetian Chen Xing Zhang Yiwen Xia Sen Wang Weizhi Wang Zekuan Xu | 2023 | Cancer Communications2023,43,6: | 0 |

| 19 | Enhancing tylosin production by combinatorial overexpression of efflux,SAM biosynthesis,and regulatory genes in hyperproducing Streptomyces xinghaiensis strain显示文摘Tylosin is a 16-membered macrolide antibiotic widely used in veterinary medicine to control infections caused by Gram-positive pathogens and mycoplasmas.To improve the fermentation titer of tylosin in the hyperproducing Streptomyces xinghaiensis strain TL01,we sequenced its whole genome and identified the biosynthetic gene cluster therein.Overexpression of the tylosin efflux gene tlrC,the cluster-situated S-adenosyl methionine(SAM)synthetase gene metK_(cs),the SAM biosynthetic genes adoK_(cs)-metFcs,or the pathway-specific activator gene tylR enhanced tylosin production by 18%,12%,11%,and 11%in the respective engineered strains TLPH08-2,TLPH09,TLPH10,and TLPH12.Co-overexpression of metK_(cs)and adoK_(cs)-metFcs as two transcripts increased tylosin production by 22%in the resultant strain TLPH11 compared to that in TL01.Furthermore,combinational overexpression of tlrC,metK_(cs),adoK_(cs)-metFcs,and tylR as four transcripts increased tylosin production by 23%(10.93g/L)in the resultant strain TLPH17 compared to that in TL01.However,a negligible additive effect was displayed upon combinational overexpression in TLPH17 as suggested by the limited increment of fermentation titer compared to that in TLPH08-2.Transcription analyses indicated that the expression of tlrC and three SAM biosynthetic genes in TLPH17 was considerably lower than that of TLPH08-2 and TLPH11.Based on this observation,the five genes were rearranged into one or two operons to coordinate their overexpression,yielding two engineered strains TLPH23 and TLPH24,and leading to further enhancement of tylosin production over TLPH17.In particular,the production of TLPH23 reached 11.35 g/L.These findings indicated that the combinatorial strategy is a promising approach for enhancing tylosin production in high-yielding industrial strains. | Penghui Dai Yuyao Qin Luyuan Li Haidi Li Lihuo Lv Danying Xu Yuqing Song Tingting Huang Shuangjun Lin Zixin Deng Meifeng Tao | 2023 | Synthetic and Systems Biotechnology2023,8,3: | 0 |
