| 1 | Does Helicobacter pylori infection play a role in iron deficiency anemia? | 2 | ShRNA-mediated gene silencing of β-catenin inhibits growth of human colon cancer cells显示文摘AIM: To observe the gene silencing mediated by the specific shRNA targeted against β-catenin and its effect on cell proliferation and cycle distribution in the human colon cancer cell line Colo205. METHODS: Two shRNA plasmid vectors against β-catenin were constructed and transfected into Colo205 cells with LipofectamineTM2000. The down-regulations of β-catenin, c-myc and cyclinD1 expressions were detected by RT-PCR and western blot analysis. The cell proliferation inhibitions were determined by MTT assay and soft agar colony formation assay. The effect of these two β-catenin shRNAs on cell cycle distribution and apoptosis was examined by flow cytometry. RESULTS: These two shRNA vectors targeted against β-catenin efficiently suppressed the expression of β-catenin and its down stream genes, c-myc and cyclinD1. The expression inhibition rates were around 40%-50% either at the mRNA or at the protein level. The shRNA-mediated gene silencing of β-catenin resulted in significant inhibition of cell growth both on the culture plates and in the soft agar. Moreover, the cancer cells showed significant G0/G1 arrest and increased apoptosis at 72 h post transfection due to gene silencing. CONCLUSION: These specific shRNAs targeted against β-catenin could have a gene silencing effect and block the WNT signaling pathway. They could inhibit cell growth, increase apoptosis, and induce cell cycle arrest in Colo205 cells. | 4 | Influence of temperature and solution composition on the formation of calcium sulfates显示文摘Calcium sulfates (anhydrite and hydrates) were synthesized by mixing CaCl 2 and Na 2 SO 4 solutions at room temperature followed by aging the resulting slurries at elevated temperatures. The variation of the morphology and structure of the calcium sulfates with aging temperature was investigated. Experimental results indicated that CaSO 4 ·2H 2 O plates, CaSO 4 ·0.5H 2 O whiskers and CaSO 4 spindles were formed at ≤100 °C, 130–160 °C and ≥170 °C, respectively. The formation and conversion of the calcium sulfates were discussed on the basis of characterization of the products and chemical analysis of the solutions. | 11 | Risk factors for sporadic colorectal cancer in southern Chinese显示文摘AIM:To investigate the role of smoking, alcohol drinking, family history of cancer, and body mass index(BMI) in sporadic colorectal cancer in southern Chinese.METHODS:A hospital-based case-control study was conducted from July 2002 to December 2008.There were 706 cases and 723 controls with their sex and age(within 5 years) matched.An unconditional logistic regression model was used to analyze the association between smoking, alcohol drinking, family history of cancer, BMI and sporadic colorectal cancer.RESULTS:No positive association was observed between smoking status and sporadic colorectal cancer risk.Compared with the non alcohol drinkers, the current and former alcohol drinkers had an increased risk of developing sporadic colorectal cancer(CRC)(adjusted OR = 8.61 and 95% CI = 6.15-12.05;adjusted OR = 2.30, 95% CI = 1.27-4.17).Moreover, the increased risk of developing sporadic CRC was significant in those with a positive family history of cancer(adjusted OR = 1.62, 95% CI = 1.12-3.34) and in those with their BMI ≥ 24.0 kg/m2(adjusted OR = 1.39, 95% CI = 1.10-1.75).Stratification analysis showed that the risk of developing both colon and rectal cancers was increased in current alcohol drinkers(adjusted OR = 7.60 and 95% CI = 5.13-11.25;adjusted OR = 7.52 and 95% CI = 5.13-11.01) and in those with their BMI ≥ 24.0 kg/m2(adjusted OR = 1.38 and 95% CI = 1.04-1.83;adjusted OR = 1.35 and 95% CI = 1.02-1.79).The risk of developing colon cancer, but not rectal cancer, was found in former alcohol drinkers and in those with a positive family history of cancer(adjusted OR = 2.51 and 95% CI = 1.24-5.07;adjusted OR = 1.82 and 95% CI = 1.17-2.82).CONCLUSION:Alcohol drinking, high BMI(≥ 24.0 kg/m2) and positive family history of cancer are the independent risk factors for colorectal cancer in southern Chinese. | Yi-Sheng Wei Jia-Chun Lu Lei Wang Ping Lan Hong-Jun Zhao Zhi-Zhong Pan Jun Huang Jian-Ping Wang | 2009 | World Journal of Gastroenterology2009,15,20: | 11 |
| 12 | Towards Kinetic Modeling of Global Metabolic Networks: Methylobacterium extorquens AM1 Growth as Validation显示文摘Here we report a systematic method for constructing a large scale kinetic metabolic model and its initial application to the modeling of central metabolism of Methylobacterium extorquens AM1, a methylotrophic and environmental important bacterium. Its central metabolic network includes formaldehyde metabolism, serine cycle, citric acid cycle, pentose phosphate pathway, gluconeogensis, PHB synthesis and acetyl-CoA conversion pathway, respiration and energy metabolism. Through a systematic and consistent procedure of finding a set of parameters in the physiological range we overcome an outstanding difficulty in large scale kinetic modeling: the requirement for a massive number of enzymatic reaction parameters. We are able to construct the kinetic model based on general biological considerations and incomplete experimental kinetic parameters. Our method consists of the following major steps: 1) using a generic enzymatic rate equation to reduce the number of enzymatic parameters to a minimum set while still preserving their characteristics; 2) using a set of steady state fluxes and metabolite concentrations in the physiological range as the expected output steady state fluxes and metabolite concentrations for the kinetic model to restrict the parametric space of enzymatic reactions; 3) choosing enzyme constants K's and K'eqs optimized for reactions under physiological concentrations, if their experimental values are unknown; 4) for models which do not cover the entire metabolic network of the organisms, designing a dynamical exchange for the coupling between the metabolism represented in the model and the rest not included. | Ping Ao Lik Wee Lee Mary E. | 13 | Influence of high-fluorine environmental background on crops and human health in hot spring-type fluorosis-diseased areas显示文摘Drinking water-type fluorosis is the most harmful endemic disease in China with the largest number of sufferers. Although the implementation of the policy to alter water sources to lower fluoride level has effectively controlled the spread of this kind of endemic disease,its prevalence could not thoroughly be stopped because the high-fluoride environmental background in these endemically diseased areas could still do harm to human health through food chain. Therefore,it is necessary to conduct a more deep-going study on the drinking water-type fluoro-sis. To investigate the effect of high fluorine environmental background on crops and human health in the hot spring-type fluorosis-diseased areas,local water,paddy soil,rice,whole vegetables and soils around their roots were sampled for analysis. The results were compared with those of the control groups in fluorosis-free areas which are similar to the fluorosis-diseased areas both in natural background and in social background. It is indicated that rice and vegetables can accumulate water-soluble fluorine either in soils or in irrigating water,and different crops have different abilities of fixing fluorine. The contents of fluorine in different parts of vegetables in the fluorosis-diseased and fluorosis-free areas were statistically categorized. The results showed that the fluorine contents of roots,tubers,leaves and flowers of vegetables in the fluorosis-diseased areas are 3.56,1.17,3.07 and 3.23 mg/kg,respectively. However,comparisons showed that in the fluorosis-free areas,the fluorine contents are 2.17,0.70,1.91 and 2.52 mg/kg,respectively. Moreover,different parts of a crop also show significantly different fluorine fixation abilities. It is demonstrated that the fluorine contents of the strongly metabolic parts are relatively high. For example,the fluo-rine contents of roots,leaves and flowers of vegetables are much higher than those of stems. The fluorine fixation ability of seeds is very weak. In order to reduce the risk of human body’s exposure to fluoride,the impact of hot spring water on the capability of crops to fix fluorine should be reduced as much as possible. | 15 | Autophagy Accompanied with Bisdemethoxycurcumin-induced Apoptosis in Non-small Cell Lung Cancer Cells显示文摘Objective To investigate the effects of bisdemethoxycurcumin(BDMC) on non-small cell lung cancer(NSCLC) cell line, A549, and the highly metastatic lung cancer 95 D cells. Methods CCK-8 assay was used to assess the effect of BDMC on cytotoxicity. Flow cytometry was used to evaluate apoptosis. Western blot analysis, electron microscopy, and quantification of GFP-LC3 punctuates were used to test the effect of BDMC on autophagy and apoptosis of lung cancer cells. Results BDMC inhibited the viability of NSCLC cells, but had no cytotoxic effects on lung small airway epithelial cells(SAECs). The apoptotic cell death induced by BDMC was accompanied with the induction of autophagy in NSCLC cells. Blockage of autophagy by the autophagy inhibitor 3-methyladenine(3-MA) repressed the growth inhibitory effects and induction of apoptosis by BDMC. In addition, BDMC treatment significantly decreased smoothened(SMO) and the transcription factor glioma-associated oncogene 1(Gli1) expression. Furthermore, depletion of Gli1 by si RNA and cyclopamine(a specific SMO inhibitor) induced autophagy. Conclusion Aberrant activation of Hedgehog(Hh) signaling has been implicated in several human cancers, including lung cancers. | 18 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. China has seen an increasing prevalence of childhood asthma in recent decades[1]. Earlier studies have shown that air particulate matter (PM), particularly fine particulate matter (PM2.5)[2], is an important factor triggering childhood asthma. Since national PM2.5 data were not available until 2013, the effects of PM2.5 on childhood asthma are less known, and most of the related research has been conducted primarily in some metropolises in China. Moreover, the majority of these studies focus on the impact of a unit increase (1 or 10 μg/m3) or an interquartile range increase in PM2.5 on childhood asthma based on the same pollutant background concentrations. Assessment of the impact of air quality grades (AQGs) based on PM2.5 concentration related to health effects is rarely considered[3].
