1Biological characteristics of an improved triploid crucian carp显示文摘An improved triploid crucian carp(ITCC) was produced by crossing improved tetraploids(G1×AT,) with improved red crucian carp(IRCC,),which were obtained by distant crossing and gynogenesis.The biological characteristics of ITCC,including the number and karyotype of chromosomes,gonadad and pituitary structures,phenotype,and growth rate are reported.ITCC possessed 150 chromosomes with the karyotype 33m+51sm+33st+33t.In the breeding season,both ovary-like and testis-like gonads of ITCC were unable to produce normal mature gametes.The ultrastructure of the pituitary of ITCC showed that most of the endocrine granules in gonadotrophic hormone(GTH) cells had not been released,providing endocrinological evidence for the sterility of ITCC.Compared with triploid crucian carp(TCC) produced by mating Japanese crucian carp with allotetraploid hybrids,ITCC not only retained the excellent traits of fast growth rate and sterility,but also acquired improved morphological characteristics,including higher body,shorter tail and smaller head.CHEN Song,WANG Jing,LIU ShaoJun,QIN QinBo,XIAO Jun,DUAN Wei,LUO KaiKun,LIU JinHui & LIU Yun Key Laboratory of Protein Chemistry and Developmental Biology of State Education Ministry of China,College of Life Sciences,Hunan Normal University,Changsha 410081,China 2009Science China(Life Sciences)2009,52,8:14

3Transfer matrix method for multibody systems(Rui method) and its applications显示文摘The transfer matrix method for multibody systems, namely the 'Rui method', is a new method for studying multibody system dynamics, which avoids the global dynamics equations of the system, keeps high computational speed, and allows highly formalized programming. It has been widely applied to scientific research and key engineering of lots of complex mechanical systems in 52 research directions. 4Formation and biological characterization of three new types of improved crucian carp显示文摘The improved tetraploids (G1×AT) were obtained by distant crossing and gynogenesis and the high-body individuals accounted for 2% among G1×AT. After mating with each other, the high-body in- dividuals produced three kinds of bisexual fertile diploid fishes: high-body red crucian carp, high-body fork-like-tails goldfish and gray common carp. The high-body red crucian carp mating with each other formed three types of improved crucian carp (ICC) including improved red crucian carp (IRCC), im- proved color crucian carp (ICCC) and improved gray crucian carp (IGCC). The phenotypes, chromo- some numbers, gonadal structure and fertility of the three kinds of ICC and their offspring were observed. All the three kinds of ICC possessed some improved phenotypes such as higher body, smaller head and shorter tail. The ratios of the body height to body length of IRCC, ICCC and IGCC were 0.54, 0.51 and 0.54, respectively. All of them were obviously higher than that of red crucian carp 0.41 (P<0.01). Three kinds of ICC had the same chromosome number as red crucian carp with 100 chromosomes. All the ICC possessed normal gonads producing mature eggs or sperm, which was important for the production of an improved diploid population. Compared with red crucian carp, all the ICC had stronger fertility such as higher gametes production, higher fertilization rate and higher hatchery rate. Three types of improved diploid fish population were generated from the three kinds of ICC by self-crossing, respectively. The ICC can serve as ornamental fish and edible fish. They are also ideal parents to produce triploids by mating with tetraploids. 5Polyploid organisms显示文摘Polyploids are organisms with three or more complete chromosome sets. Polyploidization is widespread in plants and animals, and is an important mechanism of speciation. Genome sequencing and related molecular systematics and bioinformatics studies on plants and animals in recent years support the view that species have been shaped by whole genome duplication during evolution. The stability of polyploids depends on rapid genome recombination and changes in gene expression after formation. The formation of polyploids and subsequent diploidization are important aspects in long-term evolution. Polyploids can be formed in various ways. Among them, hybrid organisms formed by distant hybridization could produce unreduced gametes and thus generate offspring with doubled chromosomes, which is a fast, efficient method of polyploidization. The formation of fertile polyploids not only promoted the interflow of genetic materials among species and enriched the species diversity, but also laid the foundation for polyploidy breeding. 6Establishment and application of distant hybridization technology in fish显示文摘Hybridization is widely used. However, for a long time, systematic theories and technologies related to hybridization in fish have been lacking. In this study, through long-term systematic research, we investigated and obtained the main rules regarding inheritance and reproduction related to fish distant hybridization. Furthermore, we established one-step and multistep breeding technologies that were suitable for interspecific hybridization and intraspecific hybridization. Simultaneously, we used these two breeding technologies to produce a batch of diploid fish lineages and tetraploid fish lineages and improved fishes. In addition, we widely discuss the methods, technologies and results of hybridization breeding, referring to the domestic and foreign literature on fish hybridization. 7The chimeric genes in the hybrid lineage of Carassius auratus cuvieri(♀)×Carassius auratus red var.(♂)显示文摘Hybridization can combine the genomes of different strains or species, which leads to changes of genotype and phenotype in the hybrids. In this study, we aimed to investigate the genetic variations of hybrids(WR-F1 and WR-F2) derived from the intraspecific hybridization of white crucian carp(Carassius auratus cuvieri, WCC, ♀) and red crucian carp(Carassius auratus red var., RCC, ♂). Here, we compared the orthologous genes in the liver transcriptomes of hybrids with those of WCC and RCC, and classified the orthologous genes into eight gene patterns within three categories(chimera, mutant, and biparental origin genes).The results revealed 19.04%, 4.17% chimeric genes and 6.90%, 5.05% mutations of orthologous genes in WR-F1 and WR-F2 respectively. Seventeen of twenty-three characterized genes(77%) were confirmed to be the chimeras at the genomic DNA level.The GO classification discovered that some chimeric and mutant genes were related to metabolic process, immune system and developmental process in WR-F1. Our results provide the new evidence that hybridization can combine the parental genomes,leading to changes in the genotype of the resultant hybrids. 8Characteristics and Preliminary Causes of Tropical Cyclone Extreme Rainfall Events over Hainan Island显示文摘The characteristics of tropical cyclone(TC) extreme rainfall events over Hainan Island from 1969 to 2014 are analyzed from the viewpoint of the TC maximum daily rainfall(TMDR) using daily station precipitation data from the Meteorological Information Center of the China Meteorological Administration, TC best-track data from the Shanghai Typhoon Institute,and NCEP/NCAR reanalysis data. The frequencies of the TMDR reaching 50, 100 and 250 mm show a decreasing trend[-0.7(10 yr)^(-1)], a weak decreasing trend [-0.2(10 yr)^(-1)] and a weak increasing trend [0.1(10 yr)^(-1)], respectively. For seasonal variations, the TMDR of all intensity grades mainly occurs from July to October, with the frequencies of TMDR 50 mm and 100 mm peaking in September and the frequency of TMDR 250 mm [TC extreme rainstorm(TCER) events]peaking in August and September. The western region(Changjiang) of the Island is always the rainfall center, independent of the intensity or frequencies of different intensity grades. The causes of TCERs are also explored and the results show that topography plays a key role in the characteristics of the rainfall events. TCERs are easily induced on the windward slopes of Wuzhi Mountain, with the coordination of TC tracks and TC wind structure. A slower speed of movement, a stronger TC intensity and a farther westward track are all conducive to extreme rainfall events. A weaker northwestern Pacific subtropical high is likely to make the 500-h Pa steering flow weaker and results in slower TC movement, whereas a stronger South China Sea summer monsoon can carry a higher moisture flux. 10Targeted disruption of tyrosinase causes melanin reduction in Carassius auratus cuvieri and its hybrid progeny显示文摘The white crucian carp(Carassius auratus cuvieri,WCC) not only is one of the most economically important fish in Asia,characterized by strong reproductive ability and rapid growth rates,but also represents a good germplasm to produce hybrid progenies with heterosis.Gene knockout technique provides a safe and acceptant way for fish breeding.Achieving gene knockout in WCC and its hybrid progeny will be of great importance for both genetic studies and hybridization breeding.Tyrosinase(TYR) is a key enzyme in melanin synthesis.Depletion of tyr in zebrafish and mice results in mosaic pigmentation or total albinism.Here,we successfully used CRISPR-Cas9 to target tyr in WCC and its hybrid progeny(WR) derived from the cross of WCC(♀) and red crucian carp(Carassius auratus red var.,RCC,♂).The level of TYR protein was significantly reduced in mutant WCC.Both the mutant WCC and the mutant WR showed different degrees of melanin reduction compared with the wild-type sibling control fish,resulting from different mutation efficiency ranging from 60% to 90%.In addition,the transcriptional expression profiles of a series of pivotal pigment synthesis genes,i.e.tyrp1,mitfa,mitfb,dct and sox10,were down-regulated in tyr-CRISPR WCC,which ultimately caused a reduction in melanin synthesis.These results demonstrated that tyr plays a key role in melanin synthesis in WCC and WR,and CRISPR-Cas9 is an effective tool for modifying the genome of economical fish.Furthermore,the tyr-CRISPR models could be valuable in understanding fundamental mechanisms of pigment formation in non-model fish.Qingfeng Liu Yanhua Qi Qiuli Liang Jia Song Junmei Liu Wuhui Li Yuqin Shu Min Tao Chun Zhang Qinbo Qin Jing Wang Shaojun Liu 2019Science China(Life Sciences)2019,62,9:4
11The formation of improved tetraploid population of red crucian carp×common carp hybrids by androgenesis显示文摘Bisexual fertile diploid androgenetic individuals (A0) (2n=100) were formed by androgenesis. In this way, the diploid spermatozoa from male allotetraploid hybrids (AT) (4n=200) of red crucian carp (Carassius auratus red var.) (♀) × common carp (Cyprinus carpio L.) (♂) were used to fertilize the UV-treated hap- loid eggs of goldfish (Carassius auratus), and living androgenetic diploid fish were developed. The A0 became sexually mature at the age of 2 years, and they fertilized with each other to form their offspring (A1). In this study, we observed the chromosomal number, gonadal structure and appearance of A1 fish. The results are as follows: (1) In A1, there were 85% tetraploids (A1-4n), 10% triploids (A1-3n) and 5% diploids (A1-2n), suggesting that diploid A0 could produce diploid gametes. It was concluded that the formation of diploid gametes generated from diploid A0 was probably related to the mechanism of pre-meiotic endoreduplication. (2) Among A1, only A1-4n possessed normal ovaries and testes. The mature males of A1-4n produced white semen. Under the electron microscope, the head of diploid sperm generated by A1-4n was bigger than that of haploid sperm generated by red crucian carp. In the testes of the A1-4n, there were many mature normal spermatozoa with a head bearing plasma mem- brane and a tail having the typical structure of '9+2' microtubules. Between the head and the tail, there were some mitochondria. The ovaries of A1-4n developed well and mainly contained II, III and IV-stage oocytes. The IV-stage oocytes were surrounded by inner and outer follicular cells. The micropyle was observed on the oolemma of follicular cells. There were abundant yolks and plenty of endoplasmic reticulum in the cytoplasm of IV-stage oocytes. Because A1-2n and A1-3n were distant crossing diploid hybrids and triploid hybrids respectively, they possessed abnormal gonads, and no mature semen and eggs were observed. (3) Compared with allotetraploids, the A1-4n fish not only had advantages such as fast growth rate and strong resistibility but also showed some new good performances such as high ratio of body width to body length, smaller heads and shorter tails. 12Transfer matrix method for determination of the natural vibration characteristics of elastically coupled launch vehicle boosters显示文摘The analysis of natural vibration characteristics has become one of important steps of the manufacture and dynamicdesignintheaerospaceindustry.Thispaperpresents a new scenario called virtual cutting in the context of the transfer matrix method of linear multibody systems closedlooptopologyforcomputingthefreevibrationcharacteristics of elastically coupled fl xible launch vehicle boosters. In this approach, the coupled system is idealized as a triple-beam system-like structure coupled by linear translational springs,where a non-uniform free-free Euler–Bernoulli beam is used.A large thrust-to-weight ratio leads to large axial accelerations that result in an axial inertia load distribution from nose to tail. Consequently, it causes the development of significan compressive forces along the length of the launch vehicle.Therefore, it is important to take into account this effect in the transverse vibration model. This scenario does not need the global dynamics equations of a system, and it has high computational efficien y and low memory requirements. 13A comparative study of distant hybridization in plants and animals显示文摘Distant hybridization refers to crosses between two different species, genera, or higher-ranking taxa, which can break species limits, increase genetic variation, and combine the biological characteristics of existing species. It is an important way of creating genetic variation, fertile strains, and excellent characteristics in new strains and populations. Combining analyses and summaries from many inter-related documents in plants and animals, both domestic and international, including examples and long-standing research on distant hybridization in fish from our laboratory, we summarize and compare the similarities and differences in plant and animal distant hybridization. In addition, we analyze and review the biological characteristics of their different ploidy progenies and the possible causes of disparity in survival rates. Mechanisms of sterility in animal and plant distant hybrids are also discussed, and research methods for the study of biological characteristics of hybrids, including morphology, cytology,and molecular cytogenetics are presented. This paper aims to provide comprehensive research materials and to systematically compare the general and specific characteristics of plant and animal hybrids with regards to reproduction, genetics, growth traits,and other biological characteristics. 14Comparison of diploid and triploid Carassius auratus provides insights into adaptation to environmental change显示文摘Focusing on adaptation of aquatic organisms, especially fish, can help elucidate complex dynamics in freshwater ecology. The differences in genetic and epigenetic regulation between diploid and triploid Carassius auratus affect survival under eutrophication. To identify the underlying mechanisms that lead to better adaption of triploids than diploids, we compared mRNA and microRNA(miRNA) expressions in liver tissue of diploid and triploid individuals obtained from the Dongting lake water system in central China. Differential expression analysis revealed that 566 transcripts were significantly up-regulated, whereas758 were down-regulated in triploids; of these differentially expressed transcripts, 33 transcripts including cacna1 d, nfkb2, hspa1 and fgfr4 were involved in the MAPK signaling pathway, and eight transcripts were determined to be regulated by seven miRNAs. Additionally, four of 25 differential expressed(DE) transcripts(mhc1, irf7, nfkb2 and pik3 c) involving the viral carcinogenesis pathway were regulated by four miRNAs. Furthermore, genetic polymorphisms analysis showed that more heterozygous mutations were detected in triploids than diploids. The d N/d S results revealed that 21 genes were under positive selection(d N/d S>1) in C. auratus complex. 15The R1947X mutation of NF1 causing autosomal dominant neurofibromatosis type 1 in a Chinese family显示文摘Neurofibromatosis type 1 is a common autosomal dominant disorder with a high rate of penetrance. It is caused by the mutation of the tumor suppressor gene NF1, which encodes neurofibromin. The main function of neurofibromin is down-regulating the biological activity of the proto-oncoprotein Ras by acting as a Ras-specific GTPase activating protein. In this study, we identified a Chinese family affected with neurofibromatosis type 1. The known gene NF1 associated with NF1 was studied by linkage analysis and by direct sequencing of the entire coding region and exon-intron boundaries of the NF1 gene. The R1947X mutation of NF1 was identified, which was co-segregated with affected individuals in the Chinese family, but not present in unaffected family members. 16Autotriploid origin of Carassius auratus as revealed by chromosomal locus analysis显示文摘In the Dongting water system, the Carassius auratus(Crucian carp) complex is characterized by the coexistence of diploid forms(2n=100, 2n CC) and polyploid forms. Chromosomal and karyotypic analyses have suggested that the polyploid C. auratus has a triploid(3n=150, 3n CC) and a tetraploid origin(4n=200), respectively. However, there is a lack of direct genetic evidence to support this conclusion. In this paper, analysis of the 5S r DNA chromosomal locus revealed that the 3n CC is of triploid origin. Analysis of the species-specific chromosomal centromere locus revealed that 3n CC individuals possess three sets of C. auratus-derived chromosomes. Our results provide direct cytogenetic evidence suggesting that individuals with 150 chromosomes are of autotriploid origin within the C. auratus complex. 17A composite of Ni-Fe-Zn layered double hydroxides/biochar for atrazine removal from aqueous solution显示文摘This study assembled corn stalk-derived biochar(BC)with layered double hydroxide(LDH)through a rapid coprecipitation of biochar and metal(Ni/Fe/Zn)hydroxide precipitates.A BC and LDH composite(BC-LDH)and modified BC-LDH material after heating(BC-LDH-P)were prepared successfully for atrazine adsorptive removal.The physicochemical properties of the synthesized samples were analyzed by X-ray diffraction(XRD),scanning electron microscopy(SEM),Fourier transform infrared(FT-IR),and X-ray photoelectron spectroscopy(XPS).Batch experiments were conducted to study the sorption of atrazine onto BC,BC-LDH,and BC-LDH-P from the aqueous solution.The adsorption of atrazine onto BC-LDH and BC-LDH-P were performed following with the pseudo-second-order kinetic model,and the sorption isotherms agreed well with the Freundlich fitting model.The adsorption amounts of the three materials are arranged in descending order is as follows:BC-LDH-P>BC-LDH>BC(143.15>123.10>95.93 mg g−1,respectively).Due to the high crystallinity of the LDH material and the high binding degree with biochar,Ni/Fe/Zn-LDH biochar composites offer a potential alternative to a carbon-based adsorbent for atrazine removal from aqueous solution.Yifan Wang Jiaming Kang Simeng Jiang Hui Li Zheyi Ren Qinbo Xu Qun Jiang Wenzhu Liu Ruizhen Li Ying Zhang 2020Biochar2020,2,4:2
18Analysis of an Ensemble of High-Resolution WRF Simulations for the Rapid Intensification of Super Typhoon Rammasun(2014)显示文摘Diagnostics are presented from an ensemble of high-resolution forecasts that differed markedly in their predictions of the rapid intensification(RI)of Typhoon Rammasun.We show that the basic difference stems from subtle differences in initializations of(a)500-850-h Pa environmental winds,and(b)midlevel moisture and ventilation.We then describe how these differences impact on the evolving convective organization,storm structure,and the timing of RI.As expected,ascent,diabatic heating and the secondary circulation near the inner-core are much stronger in the member that best forecasts the RI.The evolution of vortex cloudiness from this member is similar to the actual imagery,with the development of an inner cloud band wrapping inwards to form the eyewall.We present evidence that this structure,and hence the enhanced diabatic heating,is related to the tilt and associated dynamics of the developing inner-core in shear.For the most accurate ensemble member:(a)inhibition of ascent and a reduction in convection over the up-shear sector allow moistening of the boundary-layer air,which is transported to the down-shear sector to feed a developing convective asymmetry;(b)with minimal ventilation,undiluted clouds and moisture from the down-shear left quadrant are then wrapped inwards to the up-shear left quadrant to form the eyewall cloud;and(c)this process seems related to a critical down-shear tilt of the vortex from midlevels,and the vertical phase-locking of the circulation over up-shear quadrants.For the member that forecasts a much-delayed RI,these processes are inhibited by stronger vertical wind shear,initially resulting in poor vertical coherence of the circulation,lesser moisture and larger ventilation.Our analysis suggests that ensemble prediction is needed to account for the sensitivity of forecasts to a relatively narrow range of environmental wind shear,moisture and vortex inner-structure.Xun LI Noel EDAVIDSON Yihong DUAN Kevin JTORY Zhian SUN Qinbo CAI 2020Advances in Atmospheric Sciences2020,37,2:2
