1Newborn screening with targeted sequencing:a multicenter investigation and a pilot clinical study in China显示文摘Different newborn screening(NBS) programs have been practiced in many countries since the 1960 s. It is of considerable interest whether next-generation sequencing is applicable in NBS. We have developed a panel of 465 causative genes for 596 early-onset, relatively high incidence, and potentially actionable severe inherited diseases in our Newborn Screening with Targeted Sequencing(NESTS) program to screen 11,484 babies in 8 Women and Children’s hospitals nationwide in China retrospectively. The positive rate from preliminary screening of NESTS was 7.85%(902/11,484). With 45.89%(414/902) follow-up of preliminary positive cases, the overall clinically confirmative diagnosis rate of monogenic disorders was 12.07%(50/414), estimating an average of 0.95%(7.85% × 12.07%) clinical diagnosis rate, suggesting that monogenic disorders account for a considerable proportion of birth defects. The disease/gene spectrum varied in different regions of China. NESTS was implemented in a hospital by screening 3923 newborns to evaluate its clinical application. The turn-around time of a primary report, including the sequencing period of < 7 days, was within 11 days by our automatic interpretation pipeline. 2Comparing the regeneration potential between PLLA/Aragonite and PLLA/ Vaterite pearl composite scaffolds in rabbit radius segmental bone defects显示文摘Mussel-derived nacre and pearl,which are natural composites composed CaCO3 platelets and interplatelet organic matrix,have recently gained interest due to their osteogenic potential.The crystal form of CaCO3 could be either aragonite or vaterite depending on the characteristics of mineralization template within pearls.So far,little attention has been paid on the different osteogenic capacities between aragonite and vaterite pearl.In the current work,aragonite or vaterite pearl powders were incorporated into poly-L-lactic acid(PLLA)scaffold as bio-functional fillers for enhanced osteogenesis.In intro results revealed that PLLA/aragonite scaffold possessed stronger stimulatory effect on SaOS-2 cell proliferation and differentiation,evidenced by the enhanced cell viability,alkaline phosphatase activity,collagen synthesis and gene expressions of osteogenic markers including osteocalcin,osteopotin and bone sialoprotein.The bone regeneration potential of various scaffolds was evaluated in vivo employing a rabbit critical-sized radial bone defect model.The X-ray and micro-CT results showed that significant bone regeneration and bridging were achieved in defects implanted with composite scaffolds,while less bone formation and non-bridging were found for pure PLLA group.Histological evaluation using Masson's trichrome and hematoxylin/eosin(H&E)staining indicated a typical endochondral bone formation process conducted at defect sites treated with composite scaffolds.Through three-point bending test,the limbs implanted with PLLA/aragonite scaffold were found to bear significantly higher bending load compared to other two groups.Together,it is suggested that aragonite pearl has superior osteogenic capacity over vaterite pearl and PLLA/aragonite scaffold can be employed as a potential bone graft for bone regeneration.Qianli Huang Yuansheng Liu Zhengxiao Ouyang Qingling Feng 2020Bioactive Materials2020,5,4:7
3Surgical Therapeutic Strategy for Non-small Cell Lung Cancer with Mediastinal Lymph Node Metastasis (N2)显示文摘Background and objective Approximately 30% of patients who are diagnosed with non-small cell lung cancer (NSCLC) are classified as N2 on the basis of metastasis to the mediastinal lymph nodes. The effectiveness of surgery for these patients remains controversial. Although surgeries in recent years are proved to be effective to some extent,yet due to many reasons,5-year survival rate after surgery varies greatly from patient to patient. Thus it is necessary to select patients who have a high probability of being be cured through an operation,who are suitable to receive surgery and the best surgical methods so as to figure out the conditions under which surgical treatment can be chosen and the factors that may influence prognosis. Methods 165 out of 173 patients with N2 NSCLC were treated with surgery in our department from January 1999 to May 2003,among whom 130 were male,43 female and the sex ratio was 3:1,average age 53,ranging from 29 to 79. The database covers the patients’ complete medical history including the information of their age,sex,location and size of tumor,date of operation,surgical methods,histologic diagnosis,clinical stage,post-operative TNM stage,neoadjuvant treatment and chemoradiotherapy. The methods of clinical stage verification include chest X-ray,chest CT,PET,mediastinoscopy,bronchoscope (+?),brain CT or MRI,abdominal B ultrasound (or CT),and bone ECT. The pathological classification was based on the international standard for lung cancer (UICC 1997). Survival time was analyzed from the operation date to May 2008 with the aid of SPSS (Statistical Package for the Social Sciences) program. Kaplan-Meier survival analysis,Log-rank test and Cox multiplicity were adopted respectively to obtain patients’ survival curve,survival rate and the impact possible factors may have on their survival rate. Results The median survival time was 22 months,with 3-year survival rate reaching 28.1% and 5-year survival rate reaching 19.0%. Age,sex,different histological classification and postoperative chemoradiotherapy seem to have no correlation with 5-year survival rate. In all N2 subtypes,5-year survival rate is remarkably higher for unexpected N2 discovered at thoractomy and proven N2 stage before preoperative work-up and receive a mediastinal down-staging after induction therapy (P<0.01),reaching 30.4% and 27.3% respectively. 5-year survival rate for single station lymph node metastasis were 27.8%,much higher compared with 9.3% for multiple stations (P<0.001). Induction therapy which downstages proven N2 in 73.3% patients gains them the opportunity of surgery. The 5-year survival rate were 23.6% and 13.0% for patients who had complete resection and those who had incomplete resection (P<0.001). Patients who underwent lobectomy (23.2%) have higher survival rate,less incidence rate of complication and mortality rate,compared with pneumonectomy (14.8%) (P<0.01). T4 patients has a 5-year survival rate as low as 11.1%,much less than T1 (31.5%) and T2 (24.3%) patients (P=0.01). It is noted through Cox analysis that completeness of resection,number of positive lymph node stations and primary T status have significant correlativity with 5-year survival rate. Conclusion It is suggested that surgery (lobectomy preferentially) is the best solution for T1 and T2 with primary tumor have not invaded pleura or the distance to carina of trachea no less than 2 cm,unexpected N2 discovered at thoractomy when a complete resection can be applied,and proven N2 discovered during preoperative work-up and is down-staged after induction therapy. Surgical treatment is the best option,lobectomy should be prioritized in operational methods since ite rate of complication and morality are lower than that of pneumonectomy. Patients’ survival time will not benefit from surgery if they are with lymph nodes metastasis of multiple stations (Bulky N2 included) and T4 which can be partially removed. Neoadjuvant chemotherapy increases long-term survival rate of those with N2 proven prior to surgery. 4Morphology,composition,and sources of individual aerosol particles at a regional background site of the YRD,China显示文摘Aerosol samples were collected at Lin'an, a background site of Yangtze River Delta(YRD).Morphology, size, composition, and mixing state of individual aerosol particles were characterized by transmission electron microscopy(TEM) coupled with energy dispersive X-ray spectroscopy(EDS), and the soluble ions of PM_(1.0) were studied by aerosol mass spectrometer(AMS). The daily average AMS mass concentrations of sulfate, nitrate, and ammonium were about 5.8, 8.6, and 5.6 μg/m^3, respectively. Individual aerosol particles were classified into seven types: S-rich, K-rich, organic matter(OM), soot, fly ash, metal, and mineral. S-rich particles were dominant in all size bins, and 51%(by number) of S-rich particles were internally mixed with other particles. The fraction of organic coating particles was 13.7% in morning, 25.2% in afternoon, and 11% in evening, suggesting that the strong photochemical process during afternoon produced more secondary organic aerosols(SOA) on the surface of inorganic particles. Fly ash and metal particles were abundant during the day, suggesting the influence of emissions from coal-fired power plants and steel plants. The results indicate that the intense industrial emissions in the YRD significantly transported to the background areas. 6Incorporation of silica nanoparticles to PLGA electrospun fibers for osteogenic differentiation of human osteoblast-like cells显示文摘The development of bone tissue engineering scaffolds still remains a challenging field,although various biomaterials have been developed for this purpose.Electrospinning is a promising approach to fabricate nanofibers with an interconnected porous structure,which can support cell adhesion,guide cell proliferation and regulate cell differentiation.The aim of this study is to fabricate composite fibers composed of poly(lactic-co-glycolic acid)(PLGA)and silica nanoparticles(NPs)via electrospinning and investigate the effect of PLGA/SiO_(2)composite fibers on the cellular response of osteoblast-like cells(SaOS-2 cells).SEM and EDX analysis showed that silica NPs were homogenously dispersed in the composite fibers.The mechanical behavior of the fibers showed that silica NPs acted as reinforcements at concentrations of 2.5 and 5 mg/ml.The incorporation of silica NPs led to enhancement of cell attachment and spreading on PLGA/SiO_(2)composite fibers.SaOS-2 cells cultured on PLGA/SiO_(2)composite fibers exhibited increased alkaline phosphatase activity,collagen secretion and bone nodules formation.The bone nodules formation of SaOS-2 cells increased along with the amount of incorporated silica NPs.The present findings indicate that PLGA/SiO_(2)composite fibers can stimulate osteogenic differentiation of SaOS-2 cells and may be a promising candidate scaffold for bone tissue engineering.Xing Yang Yuanyuan Li Xujie Liu Qianli Huang Ranran Zhang Qingling Feng 2018Regenerative Biomaterials2018,5,4:4

8Photoinduced large polaron transport and dynamics in organic-inorganic hybrid lead halide perovskite with terahertz probes显示文摘Organic-inorganic hybrid metal halide perovskites(MHPs)have attracted tremendous attention for optoelectronic applications.The long photocarrier lifetime and moderate carrier mobility have been proposed as results of the large polaron formation in MHPs.However,it is challenging to measure the effective mass and carrier scattering parameters of the photogenerated large polarons in the ultrafast carrier recombination dynamics.Here,we show,in a one-step spectroscopic method,that the optical-pump and terahertz-electromagnetic probe(OPTP)technique allows us to access the nature of interplay of photoexcited unbound charge carriers and optical phonons in polycrystalline CH_(3)NH_(3)PbI_(3)(MAPbI_(3))of about 10μm grain size.Firstly,we demonstrate a direct spectral evidence of the large polarons in polycrystalline MAPbI_(3).Using the Drude-Smith-Lorentz model along with the Frӧhlich-type electron-phonon(e-ph)coupling,we determine the effective mass and scattering parameters of photogenerated polaronic carriers.We discover that the resulting moderate polaronic carrier mobility is mainly influenced by the enhanced carrier scattering,rather than the polaron mass enhancement.While,the formation of large polarons in MAPbI_(3)polycrystalline grains results in a long charge carrier lifetime at room temperature.Our results provide crucial information about the photo-physics of MAPbI3 and are indispensable for optoelectronic device development with better performance.Zuanming Jin Yan Peng Yuqing Fang Zhijiang Ye Zhiyuan Fan Zhilin Liu Xichang Bao Heng Gao Wei Ren Jing Wu Guohong Ma Qianli Chen Chao Zhang Alexey VBalakin Alexander PShkurinov Yiming Zhu Songlin Zhuang 2022Light(Science & Applications)2022,11,8:4
9Genomic characterizations of H4 subtype avian influenza viruses from live poultry markets in Sichuan province of China, 2014–2015显示文摘Dear Editor,Avian influenza viruses (AIVs) have posed a serious threat to poultry production and public health. To date, more than fourteen AIV subtypes that are able to infect human beings have been documented. Also, it is suggested that new subtypes may be reported in the future, owing to the migration of wild birds and live poultry transportation (Gao, 2018).Poultry may act as a potential incubator for novel subtypes of avian influenza virus (Bi et al., 2016a; Bi et al., 2016b; Liu et al., 2014a; Su et al., 2017). 10A Survey on Low Complexity Detectors for OTFS Systems显示文摘The newly emerging orthogonal time frequency space(OTFS)modulation can ob⁃tain delay-Doppler diversity gain to significantly improve the system performance in high mobility wireless communication scenarios such as vehicle-to-everything(V2X),high-speed railway and unmanned aerial vehicles(UAV),by employing inverse symplectic finite Fouri⁃er transform(ISFFT)and symplectic finite Fourier transform(SFFT).However,OTFS modu⁃lation will dramatically increase system complexity,especially at the receiver side.Thus,de⁃signing low complexity OTFS receiver is a key issue for OTFS modulation to be adopted by new-generation wireless communication systems.In this paper,we review low complexity OTFS detectors and provide some insights on future researches.We firstly present the OTFS system model and basic principles,followed by an overview of OTFS detector structures,classifications and comparative discussion.We also survey the principles of OTFS detection algorithms.Furthermore,we discuss the design of hybrid OTFS and orthogonal frequency di⁃vision multiplexing(OFDM)detectors in single user and multi-user multi-waveform commu⁃nication systems.Finally,we address the main challenges in designing low complexity OT⁃FS detectors and identify some future research directions.ZHANG Zhengquan LIU Heng WANG Qianli FAN Pingzhi 2021ZTE Communications2021,19,4:3

12Investigation of frost heave prevention using permeable subgrade structure显示文摘This paper set up a series of comprehensive targets based on the concept of 'anti-freeze filler', which include reasonable water retention rate, frost heave characteristics, and compaction characteristics of filling material. Then, a type of permeable graded gravel is proposed, suitable for high-speed railway subgrade. A series of in-door water retention, permeability, and frost heave tests were performed under different graded conditions. Water retention, permeability, and frost heave characteristic of different graded filling materials can be determined, in order to define the gradation range of permeable graded gravel. Relying on the frost-heave monitoring record of high speed railway in Northeast China, a series of experimental studies were performed, which included on-site filler production, compaction test, and the anti-frost effect test, in order to improve the production and compaction techniques of permeable graded gravel. 13Calcium carbonate nanoparticles promote osteogenesis compared to adipogenesis in human bone-marrow mesenchymal stem cells显示文摘Rhombohedron-like and fusiform calcium carbonate nanoparticles were fabricated using a new method. Their geometry was controlled by varying the mixing speed and ratio of ethanol versus water in reaction system. The calcium carbonate nanoparticles(CCNPs) have slight effect on viability of human bone-marrow mesenchymal stem cells(hBMSCs) with dose-dependent and shape-dependent, but they can significantly promote osteogenic differentiation of hBMSCs in vitro by 10–37% increase of alkaline phosphatase(ALP) activity, 9–36% growth of collagen secretion and 1.13–1.83 folds upregulation of osteogenesis-related genes, even at lower dose ranges(5–20 μg/ml). The efficacity of promoting osteogenesis depends on the shape and dose of CCNPs. Furthermore,adipogenesis was inhibited by less accumulation of lipid droplets, lower triglyceride(TG) secretion and downregulation of adipogenesis-related genes. 14High-precision uranium-series dating of lacustrine carbonates from Daihai Lake显示文摘Ages of small aragonite samples retrieved from sediments from the Daihai Lake are determined by using the Thermal Ionization Mass Spectrometry (TIMS) technique. The ages of the samples range from (8740±601) to (10446±489) a, with a precision of 2%-3% for non-corrected dating ages and 4.7%-9.8% for corrected ages respectively, and are consistent within the deviation range. At about 10.5 ka BP, the grain-size of the sediments coarsens sharply, the frequency curves of grain-size distribution show coarse grain dominated single-double kurtosis, indicating cooling, decreased hydrodynamic conditions and increased wind influence procedure. This is in good agreement with previous results obtained from the Daihai Lake, which suggests that the climate was cold and the lake surface was shrinking. 15Quambalaria species associated with eucalypt diseases in southern China显示文摘The genus Quambalaria includes several important pathogens of species of Eucalyptus and Corymbia, mainly causing leaf and shoot blight. Recently,extensive shoot and leaf dieback and stem cankers suspected to be Quambalaria diseases have been found on young Eucalyptus urophylla ? E. grandis trees in Guangdong and Hainan Provinces. The occurrence of Quambalaria species and their association with eucalypt hosts within China needs to be investigated for tree diseases management. The isolates from the diseased samples were identified based on their morphological structures and phylogenetic analyses with DNA sequence data for the internal transcribed spacer region and large ribosome subunit RNA of the nuclear r DNA. This work revealed that three species of Quambalaria were present: Quambalaria pitereka from Corymbia citriodora,Q. eucalypti from E. urophylla ? E. grandis, both isolated from young eucalypt leaves and shoots in Guangdong Province, and Quambalaria simpsonii, which was isolated from stem cankers of E. urophylla ? E. grandis at four different sites across Guangdong and Hainan Provinces.These results confirmed that Quambalaria agents were associated with the diseases occurring on eucalypt hosts in South China. 18Nanoparticles and their effects on differentiation of mesenchymal stem cells显示文摘Over the past decades,advancements in nanoscience and nanotechnology have resulted in numerous nanomedicine platforms.Various nanoparticles,which exhibit many unique properties,play increasingly important roles in the field of biomedicine to realize the potential of nanomedicine.Due to the capacity of self-renewal and multilineage mesenchymal differentiation,mesenchymal stem cells(MSCs)have been widely used in the area of regenerative medicine and in clinical applications due to their potential to differentiate into various lineages.There are several factors that impact the differentiation of MSCs into different lineages.Many types of biomaterials such as polymers,ceramics,and metals are commonly applied in tissue engineering and regenerative therapies,and they are continuously refined over time.In recent years,along with the rapid development of nanotechnology and nanomedicine,nanoparticles have been playing more and more important roles in the fields of biomedicine and bioengineering.The combined use of nanoparticles and MSCs in biomedicine requires greater knowledge of the effects of nanoparticles on MSCs.This review focuses on the effects of four inorganic or metallic nanoparticles(hydroxyapatite,silica,silver,and calcium carbonate),which are widely used as biomaterials,on the osteogenic and adipogenic differentiation of MSCs.In this review,the cytotoxicity of these four nanoparticles,their effects on osteogenic/adipogenic differentiation of MSCs and the signalling pathways or transcription factors involved are summarized.In addition,the chemical composition,size,shape,surface area,surface charge and surface chemistry of nanoparticles,have been reported to impact cellular behaviours.In this review,we particularly emphasize the influence of their size on cellular responses.We envision our review will provide a theoretical basis for the combined application of MSCs and nanoparticles in biomedicine.Xing Yang Yuanyuan Li Xujie Liu Wei He Qianli Huang Qingling Feng 2020Biomaterials Translational2020,1,1:1
