1Disentangling the effects of topography and space on the distributions of dominant species in a subtropical forest显示文摘Topography and space are two important factors determining plant species assemblages in forest communities.Quantification of the contribution of these two factors in determining species distribution helps us to evaluate their relative importance in determining species assemblages.This study aims to disentangle the effect of topography and space on the distributions of 14 dominant species in a subtropical mixed forest.Spearman correlation analysis and the torustranslation test were used to test the species–habitat associations.Variation partitioning was used to quantify the relative contributions of topography and space at three sampling scales and three life stages.Correlation analyses and torus-translation tests showed species abundance was mostly correlated with topographic wetness index,vertical distance from the channel network and convexity.Variation partitioning showed that pure topography,pure space and spatially structured topography explained about 2.1 %,41.2 % and 13.8 %of the variation in species distributions,respectively.For nine species,total topography fractions peaked in 20 m quadrats.For ten species,the pure space fractions peaked in 50 m quadrats.For many species,the total topography fraction andthe pure space fraction were larger for the most abundant life stages,which reflected the importance of sampling effect.However,some cases did not follow this trend suggesting that the effects of ecological processes such as habitat filtering,density dependence or dispersal limitation may exceed the sampling effects.In conclusion,we found that spatially structured topography and pure space primarily shaped the distribution of dominant tree species.Furthermore,their effects were both scale- and life stage-dependent.Qinggang Wang Yaozhan Xu Zhijun Lu Dachuan Bao Yili Guo Junmeng Lu Kuihan Zhang Haibo Liu Hongjie Meng Xiujuan Qiao Handong Huang Mingxi Jiang 2014Chinese Science Bulletin2014,59,35:16
2Density dependence and habitat preference shape seedling survival in a subtropical forest in central China显示文摘Aims seedlings are vulnerable to many kinds of fatal abiotic and biotic agents,and examining the causes of seedling dynamics can help understand mechanisms of species coexistence.To disentangle the relative importance of neighborhood densities,habitat factors and phylogenetic relatedness on focal seedling survival,we monitored the survival of 5306 seedlings of 104 species>15 months.We address the following questions:(i)How do neighborhood densities,habitat variables and phylogenetic relatedness affect seedling survival?What is the relative importance of conspecific densities,habitat variables and phylogenetic relatedness to seedling survival?(ii)Does the importance of the neighborhood densities,habitat variables and phylogenetic relatedness vary among growth forms,leaf habits or dispersal modes?specially,does the conspecific negative density dependence inhibit tree and decidu-ous seedlings more compared with shrub and evergreen species?Does density dependence affect the wind and animal-dispersed species equally?Methods We established 135 census stations to monitor seedling dynamics in a 25-ha subtropical forest plot in central China.Conspecific and heterospecific seedling density in the 1-m2 seedling plot and adult basal area within a 20-m radius provided neighborhood density var-iables.mean elevation,convexity and aspect of every 5-×5-m grid with seedling plots were used to quantify habitat characteristics.We calculated the relative average phylodiversity between focal seed-ling and heterospecific neighbors to quantify the species related-ness in the neighborhood.Eight candidate generalized linear mixed models with binominal error distribution were used to compare the relative importance of these variables to seedling survival.akaike’s information criteria were used to identify the most parsimonious models.Important Findingsat the community level,both the neighborhood densities and phylogenetic relatedness were important to seedling survival.We found negative effects of increasing conspecific seedlings,which suggested the existence of species-specific density-dependent mortality.Phylodiversity of heterospecific neighbors was negatively related to survival of focal seedlings,indicating similar habitat preference shared among phylogenetically closely related species may drive seedling survival.The relative importance of neighborhood densities,habitat variables and phylogenetic relatedness varied among ecological guilds.Conspecific densities had significant negative effect for deciduous and wind-dispersed species,and marginally significant for tree seedlings>10 cm tall and animal-dispersed species.Habitat variables had limited effects on seedling survival,and only elevation was related to the sur-vival of evergreen species in the best-fit model.We conclude that both negative density-dependent mortality and habitat preference reflected by the phylogenetic relatedness shape the species coex-istence at seedling stage in this forest.Junmeng Lu Daniel J.Johnson Xiujuan Qiao Zhijun Lu Qinggang Wang Mingxi Jiang 2015Journal of Plant Ecology2015,8,6:12
3Preparation and evaluation of poly(L-histidine) based pH-sensitive micelles for intracellular delivery of doxorubicin against MCF-7/ADR cells显示文摘In this study, a p H-sensitive micelle self-assembled from poly(L-histidine) based triblock copolymers of poly(ethylene glycol)–poly(D,L-lactide)–poly(L-histidine)(mPEG-PLA-PHis) was prepared and used as the intracellular doxorubicin(Dox) delivery for cancer chemotherapy. Dox was loaded into the micelles by thin-film hydration method and a Box–Behnken design for three factors at three levels was used to optimize the preparations. The optimized mPEG-PLA-Phis/Dox micelles exhibited good encapsulation efficiency of 91.12%,a mean diameter of 45 nm and narrow size distribution with polydispersity index of 0.256.In vitro drug release studies demonstrated that Dox was released from the micelles in a p Hdependent manner. Furthermore, the cellular evaluation of Dox loaded micelles displayed that the micelles possessed high antitumor activity in vitro with an IC50 of 35.30 μg/ml against MCF-7/ADR cells. The confocal microscopy and flow cytometry experiments indicated that m PEG-PLA-Phis micelles mediated efficient cytoplasmic delivery of Dox with the aid of poly(Lhistidine) mediated endosomal escape. In addition, blank m PEG-PLA-Phis micelles were shown to be nontoxic to MCF-7/ADR cells even at a high concentration of 200 μg/ml. 4Co-delivery of resveratrol and docetaxel via polymeric micelles to improve the treatment of drug-resistant tumors显示文摘Co-delivery of anti-cancer drugs is promising to improve the efficacy of cancer treatment.This study was aiming to investigate the potential of concurrent delivery of resveratrol(RES)and docetaxel(DTX)via polymeric nanocarriers to treat breast cancer.To this end,methoxyl poly(ethylene glycol)-poly(D,L-lactide)copolymer(mPEG-PDLA)was prepared and characterized using FTIR and 1H NMR,and their molecular weights were determined by GPC.Isobologram analysis and combination index calculation were performed to find the optimal ratio between RES and DTX to against human breast adenocarcinoma cell line(MCF-7 cells).Subsequently,RES and DTX were loaded in the mPEG-PDLA micelles simultaneously,and the morphology,particle size distribution,in vitro release,pharmacokinetic profiles,as well as cytotoxicity to the MCF-7 cells were characterized.IC50 of RES and DTX in MCF-7 cells were determined to be 23.0μg/ml and 10.4μg/ml,respectively,while a lower IC50 of 4.8μg/ml of the combination of RES and DTX was obtained.The combination of RES and DTX at a ratio of 1:1(w/w)generated stronger synergistic effect than other ratios in the MCF-7 cells.RES and DTX loaded mPEG-PDLA micelles exhibited prolonged release profiles,and enhanced cytotoxicity in vitro against MCF-7 cells.The AUC(0→t)of DTX and RES in mPEG-PDLA micelles after i.v.administration to rats were 3.0-fold and 1.6-fold higher than that of i.v.injections of the individual drugs.These findings indicated that the co-delivery of RES and DTX using mPEG-PDLA micelles could have better treatment of tumors.Xiong Guo Zhiyue Zhao Dawei Chen Mingxi Qiao Feng Wan Dongmei Cun Yi Sun Mingshi Yang 2019Asian Journal of Pharmaceutical Sciences2019,14,1:4
5Combining immune checkpoint blockade with ATP-based immunogenic cell death amplifier for cancer chemo-immunotherapy显示文摘Amplifying“eat me signal”during tumor immunogenic cell death(ICD)cascade is crucial for tumor immunotherapy.Inspired by the indispensable role of adenosine triphosphate(ATP,a necessary“eat me signal”for ICD),a versatile ICD amplifier was developed for chemotherapy-sensitized immunotherapy.Doxorubicin(DOX),ATP and ferrous ions(Fe^(2+))were co-assembled into nanosized amplifier(ADO-Fe)throughπ‒πstacking and coordination effect.Meanwhile,phenylboric acid-polyethylene glycol-phenylboric acid(PBA-PEG-PBA)was modified on the surface of ADO-Fe(denoted as PADO-Fe)by the virtue of d-ribose unit of ATP.PADO-Fe could display active targetability against tumor cells via sialic acid/PBA interaction.In acidic microenvironment,PBA-PEG-PBA would dissociate from amplifier.Moreover,high H_(2)O_(2)concentration would induce hydroxyl radical(·OH)and oxygen(O_(2))generation through Fenton reaction by Fe^(2+).DOX and ATP would be released from the amplifier,which could induce ICD effect and“ICD adjuvant”to amplify this process.Together with programmed death ligands 1(PD-L1)checkpoint blockade immunotherapy,PADO-Fe could not only activate immune response against primary tumor,but also strong abscopal effect against distant tumor.Our simple and multifunctional ICD amplifier opens a new window for enhancing ICD effect and immune checkpoint blockade therapy.Jiulong Zhang Xiaoyan Sun Xiufeng Zhao Chunrong Yang Menghao Shi Benzhuo Zhang Haiyang Hu Mingxi Qiao Dawei Chen Xiuli Zhao 2022Acta Pharmaceutica Sinica B2022,12,9:2

7Effect of surface ligand density on cytotoxicity and pharmacokinetic profile of docetaxel loaded liposomes显示文摘Various biotin-modified liposomes incorporated with docetaxel(DTX) were prepared to study the effect of surface biotin density on the pharmacokinetic profile of the liposome. Four types of liposomes such as PEG modified liposome(PDL), 0.5%(mol) biotin modified liposome(0.5 BDL), 1%(mol) biotin modified liposome(1 BDL) and 2%(mol) biotin modified liposome(2 BDL) were prepared using thin film dispersion method. The prepared liposomes were characterized by measuring encapsulation efficiency(EE), particle size, Zeta-potential, physical stability and drug release profiles in vitro. MTT assay was performed to elevate the cytotoxicity of liposomes on MCF-7 cells. In vivo evaluation was further performed to investigate the effect of biotin surface density on the pharmacokinetic profiles. All the prepared liposomes exhibited high encapsulation efficiency, small particle size, narrow particle distribution and sustained release profiles in vitro. In MTT assay, 0.5 BDL showed largest tumor cell toxicity, compared with DTX solution. All liposomes containing DTX showed prolonged blood circulation in vivo, and 0.5 BDL showed the longest circulation time among the biotin modified liposome. Surface modification of liposome had a negative impact on the circulation of liposomes in the blood, which needs to be considered when designing the ligand mediated targeting delivery systems. 10Effects of topography on structuring species assemblages in a subtropical forest显示文摘Aims Topography has long been recognized as an important factor in shaping species distributions.Many studies revealed that species may show species-habitat associations.However,few studies inves-tigate how species assemblages are associated with local habitats,and it still remains unclear how the community-habitat associa-tions vary with species abundance class and life stage.In this study,we analyzed the community-habitat associations in a subtropical montane forest.Methods The fully mapped 25-ha(500×500 m)forest plot is located in Badagongshan Nature Reserve in Hunan Province,Central China.It was divided into 625(20×20 m)quadrats.Habitat types were classified by multivariate regression tree analyses that cluster areas with similar species composition according to the topographic characteristics.Indicator species analysis was used to identify the most important species for structuring species assemblages.We also compared the community-habitat associations for two levels of species abundances(i.e.abundant and rare)and three different life stages(i.e.saplings,juveniles and adults),while accounting for sample size effects.Important Findings The Badagongshan plot was divided into five distinct habitat types,which explained 34.7%of the variance in tree species composi-tion.Even with sample size taken into account,community-habi-tat associations for rare species were much weaker than those for abundant species.Also when accounting for sample size,very small differences were found in the variance explained by topography for the three life stages.Indicator species of habitat types were mainly abundant species,and nearly all adult stage indicator species were also indicators in juvenile and sapling stages.Our study manifested that topographical habitat filtering was important in shaping over-all local species compositions.However,habitat filtering was not important in shaping rare species’distributions in this forest.The community-habitat association patterns in this forest were mainly shaped by abundant species.In addition,during the transitions from saplings to juveniles,and from juveniles to adults,the relative importance of habitat filtering was very weak.Qinggang Wang Ruwan Punchi-Manage Zhijun Lu Scott B.Franklin Zhiheng Wang Yaoqi Li Xiulian Chi Dachuan Bao Yili Guo Junmeng Lu Yaozhan Xu Xiujuan Qiao Mingxi Jiang 2017Journal of Plant Ecology2017,10,3:1

13Reversing ferroptosis resistance by MOFs through regulation intracellular redox homeostasis显示文摘As a non-apoptotic cell death form,ferroptosis offers an alternative approach to overcome cancer chemotherapy resistance.However,accumulating evidence indicates cancer cells can develop ferroptosis resistance by evolving antioxidative defense mechanisms.To address this issue,we prepared a Buthionine-(S,R)-sulfoximine(BSO)loaded metal organic framework(MOF)of BSO-MOF-HA(BMH)with the combination effect of boosting oxidative damage and inhibiting antioxidative defense.MOF nanoparticle was constructed by the photosensitizer of[4,4,4,4-(porphine-5,10,15,20-tetrayl)tetrakis(benzoic acid)](TCPP)and the metal ion of Zr6,which was further decorated with hyaluronic acid(HA)in order to impart active targeting to CD44 receptors overexpressed cancer cells.BMH exhibited a negative charge and spherical shape with average particle size about 162.5nm.BMH was found to restore the susceptibility of 4T1 cells to ferroptosis under irradiation.This was attributed to the combination of photodynamic therapy(PDT)andγ-glutamylcysteine synthetase inhibitor of BSO,shifting the redox balance to oxidative stress.Enhanced ferroptosis also induced the release of damage associated molecular patterns(DAMPs)to maturate dendritic cells and activated T lymphocytes,leading to superior anti-tumor performance in vivo.Taken together,our findings demonstrated that boosting oxidative damage with photosensitizer serves as an effective strategy to reverse ferroptosis resistance.Chengcheng Wang JiaoWang Xue Pan Shuang Yu Meiqi Chen Yan Gao Zilin Song Haiyang Hu Xiuli Zhao Dawei Chen Fei Han Mingxi Qiao 2023Asian Journal of Pharmaceutical Sciences2023,18,1:0
14Effects of chemotherapeutic agent bendamustine for nonhodgkin lymphoma on spermatogenesis in mice显示文摘Non-Hodgkin lymphoma(NHL) is one of the most common cancers affecting men of reproductive age. The high response rate of bendamustine as first-line treatment for NHL, coupled with young age of patients, makes elucidation of the impact of treatment on male reproduction important. Our aim was to determine the effects of bendamustine on male reproduction by animal model. Male mice were treated with bendamustine(40 mg/kg) through tail vein injection while cisplatin was given as a standard(3 mg/kg) through intraperitoneal injection. After 3 weeks, bendamustine induced weight loss and sperm morphology abnormalities were compared to the control. Additionally, sperm with folded tails were the most frequent abnormality in bendamustine-treated mice. But the mechanism of sperm abnormality induced by bendamustine remains to be elucidated.
