| 2 | Echinacoside Protects Against MPP+-Induced Neuronal Apoptosis via ROS/ATF3/CHOP Pathway Regulation显示文摘Echinacoside(ECH) is protective in a mouse model of Parkinson's disease(PD) induced by 1-methyl-4-phenylpyridinium ion(MPP^+). To investigate the mechanisms involved, SH-SY5Y neuroblastoma cells were treated with MPP^+or a combination of MPP^+and ECH,and the expression of ATF3(activating transcription factor3), CHOP(C/EBP-homologous protein), SCNA(synuclein alpha), and GDNF(glial cell line-derived neurotrophic factor) was assessed. The results showed that ECH significantly improved cell survival by inhibiting the generation of MPP^+-induced reactive oxygen species(ROS). In addition, ECH suppressed the ROS and MPP^+-induced expression of apoptotic genes(ATF3, CHOP, and SCNA). ECH markedly decreased the MPP^+-induced caspase-3 activity in a dose-dependent manner. ATF3-knockdown also decreased the CHOP and cleaved caspase-3 levels and inhibited the apoptosis induced by MPP^+.Interestingly, ECH partially restored the GDNF expression that was down-regulated by MPP^+. ECH also improved dopaminergic neuron survival during MPP^+treatment and protected these neurons against the apoptosis induced by MPTP. | 3 | A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. | 4 | Preparation and evaluation of poly(L-histidine) based pH-sensitive micelles for intracellular delivery of doxorubicin against MCF-7/ADR cells显示文摘In this study, a p H-sensitive micelle self-assembled from poly(L-histidine) based triblock copolymers of poly(ethylene glycol)–poly(D,L-lactide)–poly(L-histidine)(mPEG-PLA-PHis) was prepared and used as the intracellular doxorubicin(Dox) delivery for cancer chemotherapy. Dox was loaded into the micelles by thin-film hydration method and a Box–Behnken design for three factors at three levels was used to optimize the preparations. The optimized mPEG-PLA-Phis/Dox micelles exhibited good encapsulation efficiency of 91.12%,a mean diameter of 45 nm and narrow size distribution with polydispersity index of 0.256.In vitro drug release studies demonstrated that Dox was released from the micelles in a p Hdependent manner. Furthermore, the cellular evaluation of Dox loaded micelles displayed that the micelles possessed high antitumor activity in vitro with an IC50 of 35.30 μg/ml against MCF-7/ADR cells. The confocal microscopy and flow cytometry experiments indicated that m PEG-PLA-Phis micelles mediated efficient cytoplasmic delivery of Dox with the aid of poly(Lhistidine) mediated endosomal escape. In addition, blank m PEG-PLA-Phis micelles were shown to be nontoxic to MCF-7/ADR cells even at a high concentration of 200 μg/ml. | 5 | Arsenic methylation by an arsenite S-adenosylmethionine methyltransferase from Spirulina platensis显示文摘Arsenic-contaminated water is a serious hazard for human health. Plankton plays a critical role in the fate and toxicity of arsenic in water by accumulation and biotransformation.Spirulina platensis(S. platensis), a typical plankton, is often used as a supplement or feed for pharmacy and aquiculture, and may introduce arsenic into the food chain, resulting in a risk to human health. However, there are few studies about how S. platensis biotransforms arsenic. In this study, we investigated arsenic biotransformation by S. platensis. When exposed to arsenite(As(Ⅲ)), S. platensis accumulated arsenic up to 4.1 mg/kg dry weight.After exposure to As(Ⅲ), arsenate(As(Ⅴ)) was the predominant species making up 64% to86% of the total arsenic. Monomethylarsenate(MMA(Ⅴ)) and dimethylarsenate(DMA(Ⅴ))were also detected. An arsenite S-adenosylmethionine methyltransferase from S. platensis(Sp Ars M) was identified and characterized. Sp Ars M showed low identity with other reported Ars M enzymes. The Escherichia coli AW3110 bearing Spars M gene resulted in As(Ⅲ) methylation and conferring resistance to As(Ⅲ). The in vitro assay showed that Sp Ars M exhibited As(Ⅲ) methylation activity. DMA(Ⅴ) and a small amount of MMA(Ⅴ) were detected in the reaction system within 0.5 hr. A truncated Sp Ars M derivative lacking the last 34 residues still had the ability to methylate As(Ⅲ). The three single mutants of Sp Ars M(C59S, C186 S, and C238S) abolished the capability of As(Ⅲ) methylation, suggesting the three cysteine residues are involved in catalysis. | 6 | Photoinduced large polaron transport and dynamics in organic-inorganic hybrid lead halide perovskite with terahertz probes显示文摘Organic-inorganic hybrid metal halide perovskites(MHPs)have attracted tremendous attention for optoelectronic applications.The long photocarrier lifetime and moderate carrier mobility have been proposed as results of the large polaron formation in MHPs.However,it is challenging to measure the effective mass and carrier scattering parameters of the photogenerated large polarons in the ultrafast carrier recombination dynamics.Here,we show,in a one-step spectroscopic method,that the optical-pump and terahertz-electromagnetic probe(OPTP)technique allows us to access the nature of interplay of photoexcited unbound charge carriers and optical phonons in polycrystalline CH_(3)NH_(3)PbI_(3)(MAPbI_(3))of about 10μm grain size.Firstly,we demonstrate a direct spectral evidence of the large polarons in polycrystalline MAPbI_(3).Using the Drude-Smith-Lorentz model along with the Frӧhlich-type electron-phonon(e-ph)coupling,we determine the effective mass and scattering parameters of photogenerated polaronic carriers.We discover that the resulting moderate polaronic carrier mobility is mainly influenced by the enhanced carrier scattering,rather than the polaron mass enhancement.While,the formation of large polarons in MAPbI_(3)polycrystalline grains results in a long charge carrier lifetime at room temperature.Our results provide crucial information about the photo-physics of MAPbI3 and are indispensable for optoelectronic device development with better performance. | Zuanming Jin Yan Peng Yuqing Fang Zhijiang Ye Zhiyuan Fan Zhilin Liu Xichang Bao Heng Gao Wei Ren Jing Wu Guohong Ma Qianli Chen Chao Zhang Alexey VBalakin Alexander PShkurinov Yiming Zhu Songlin Zhuang | 2022 | Light(Science & Applications)2022,11,8: | 4 |

| 9 | 调强适形放疗在乳腺癌保乳术后应用的系统评价(英文)显示文摘Objective:The aim of this study was to assess the long-term effect of breast conservation with breast intensitymodulated radiotherapy (IMRT) for early-stage breast cancers.Methods:Pub Med,EMBASE,Cochrane Library,Chinese Biomedical Literature Database were searched to identify relevant original published trails,and the references of eligible studies were manually screened.Randomized controlled trails reported in any language,comparing breast IMRT with conventional radiotherapy (RT) in patients with early-stage breast cancer were eligible for inclusion.Two investigators independently assessed the quality of included trials and extracted data.The RevMan 5.0 software was used for statistical analysis.Results:Four trials involving 1440 patients were included.The results showed that in the IMRT group,the occurrence of acute moist dermatitis,the edema of the mammary gland and the occurrence of hyperpigmentation were lower than those of RT group,the pooled risk ratio value and 95% confidence interval were 0.28 (0.17-0.48),0.03 (0.00-0.21) and 0.06 (0.02-0.15) respectively.The use of IMRT did not correlate with pain and quality of life,but the presence of moist desquamation did significantly correlate with pain and reduced quality of life.Conclusion:The currently available evidence showed that IMRT significantly reduces the occurrence of moist acute dermatitis anywhere in the breast and alleviates the suffering of patients and improves the quality of life of patients.Future large-scale,high-quality,double-blind trials are needed. | Xiupeng Ye Ling Su Xiaohu Wang Kehu Yang inhui Tian Qiuning Zhang Qiang Wang Xiaojun Li Yuqing Kang Liyun Guo Ruifeng Liu Shifang Feng | 2010 | The Chinese-German Journal of Clinical Oncology2010,9,12: | 2 |
