1CD147 antibody specifically and effectively inhibits infection and cytokine storm of SARS-CoV-2 and its variants delta,alpha,beta,and gamma显示文摘SARS-CoV-2 mutations contribute to increased viral transmissibility and immune escape,compromising the effectiveness of existing vaccines and neutralizing antibodies.An in-depth investigation on COVID-19 pathogenesis is urgently needed to develop a strategy against SARS-CoV-2 variants.Here,we identified CD147 as a universal receptor for SARS-CoV-2 and its variants.Meanwhile,Meplazeumab,a humanized anti-CD147 antibody,could block cellular entry of SARS-CoV-2 and its variants-alpha,beta,gamma,and delta,with inhibition rates of 68.7,75.7,52.1,52.1,and 62.3%at 60μg/ml,respectively.Furthermore,humanized CD147 transgenic mice were susceptible to SARS-CoV-2 and its two variants,alpha and beta.When infected,these mice developed exudative alveolar pneumonia,featured by immune responses involving alveoli-infiltrated macrophages,neutrophils,and lymphocytes and activation of IL-17 signaling pathway.Mechanistically,we proposed that severe COVID-19-related cytokine storm is induced by a'spike protein-CD147-CyPA signaling axis':Infection of SARS-CoV-2 through CD147 initiated the JAK-STAT pathway,which further induced expression of cyclophilin A(CyPA);CyPA reciprocally bound to CD147 and triggered MAPK pathway.Consequently,the MAPK pathway regulated the expression of cytokines and chemokines,which promoted the development of cytokine storm.Importantly,Meplazumab could effectively inhibit viral entry and inflammation caused by SARS-CoV-2 and its variants.Therefore,our findings provided a new perspective for severe COVID-19-related pathogenesis.Furthermore,the validated universal receptor for SARS-CoV-2 and its variants can be targeted for COVID-19 treatment.Jiejie Geng Liang Chen Yufeng Yuan Ke Wang Youchun Wang Chuan Qin Guizhen Wu Ruo Chen Zheng Zhang Ding Wei Peng Du Jun Zhang Peng Lin Kui Zhang Yongqiang Deng Ke Xu Jiangning Liu Xiuxuan Sun Ting Guo Xu Yang Jiao Wu Jianli Jiang Ling Li Kun Zhang Zhe Wang Jing Zhang Qingguo Yan Hua Zhu Zhaohui Zheng Jinlin Miao Xianghui Fu Fengfan Yang Xiaochun Chen Hao Tang Yang Zhang Ying Shi Yumeng Zhu Zhuo Pei Fei Huo Xue Liang Yatao Wang Qingyi Wang Wen Xie Yirong Li Mingyan Shi Huijie Bian Ping Zhu Zhi-Nan Chen 2021Signal Transduction and Targeted Therapy2021,6,10:6
2Tribological properties of CrN coating deposited on 20CrMo against tin bronze显示文摘A CrN coating was deposited on the piston pin material 20CrMo. The tribological properties of 20CrMo and CrN coating against tin bronze QSn7-0.2 were compared by pin-on-disk tests under dry friction and oil lubrication, respectively. The cross-sectional morphologies and structures of the coating were characterized through scanning electron microscopy and X-ray diffraction. The hardness, elastic modulus, and adhesion strength of the coating were tested through nano-hardness and scratch tests. The wear tracks were tested with an optical profilometer, and wear volume was calculated. Under dry friction conditions, the CrN coating exhibited a significantly stable friction coefficient and a relatively lower wear rate against QSn7-0.2 at various temperatures compared with 20CrMo. The wear rate decreased by 98.5% at 700°C. Under poly-alpha-olefine lubrication, the CrN coating reduced the load effect on the friction coefficient. Under engine oil 5 W40 lubrication, the CrN coating improved the tribological properties under high speed and heavy load conditions. 3Antifouling mechanism of natural product-based coatings investigated by digital holographic microscopy显示文摘Using natural product-based antifouling coatings has proven to be an effective strategy to combat biofouling.However,their antifouling mechanisms are still unclear.In this study,the antifouling mechanism of natural product-based coatings consisting of bio-sourced poly(lactic acid)-based polyurethane and ecofriendly antifoulant(butenolide)derived from marine bacteria was revealed by observing 3D bacterial motions utilizing a 3D tracking technique-digital holographic microscopy(DHM).As butenolide content increases,the density of planktonic marine bacteria(Pseudomonas sp.)near the surface decreases and thus leads to a reduced adhesion,indicating that butenolide elicits the adaptive response of Pseudomonas sp.to escape from the surface.Meanwhile,among these remained cells,an increased percentage is found to undergo subdiffusive motions compared with the case of smaller dose of butenolide.Further experiments show that butenolide can accelerate their swimming velocity and reduce flick frequency.Antibacterial assay confirms that butenolide-based coating shows high efficacy of antifouling performance against Pseudomonas sp.but without killing them like 4,5-dichloro-2-n-octyl-4-isothiazolin-3-one(DCOIT).Jiansen Pan Qingmei Peng Guoliang Zhang Qingyi Xie Xiangjun Gong Pei-Yuan Qian Chunfeng Ma Guangzhao Zhang 2021Journal of Materials Science & Technology2021,,25:2

8Metal-phenolic networks modified polyurethane as periosteum for bone regeneration显示文摘Treatment of bone defects still poses a great challenge in orthopedic clinics, and the vital role of periosteum in such processes has attracted widespread attention. However, studies focusing on the oxidative stress micro-environment with an artificial periosteum at the site of defect have been scarce. The intrinsic anti-oxidative properties and therapeutic potential for bone defects of metal-phenolic networks(MPNs)have provided a potential solution to this.
