| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 2 | Gastric cancer: Epidemiology, risk factors and prevention strategies显示文摘Gastric cancer(GC) is a global health problem, with more than 1 million people newly diagnosed with GC worldwide each year. GC is more prevalent in less developed countries than in more developed countries. About half of all GC cases worldwide occur in East Asia, notably China. Globally, overall incidence rates of GC are declining, which is potentially attributed to a decrease in Helicobacter pylori(H. pylori) infection and the use of refrigeration to preserve foods rather than salt. GC is a multifactorial disease, and its occurrence and development were impacted by environmental and genetic factors. H. pylori infection is the primary risk factor for GC, especially for non-cardia. The prognosis of GC is poor due to stages at the first diagnosis. The 5-year survival rate is less than 10% when patients are diagnosed at an advanced stage, but the rate is as high as 85% if patients are detected at an earlier stage. Endoscopic screening can potentially prevent GC by early diagnosis and early treatment and has been widely adopted in screening programs in East Asian countries, such as Japan and Korea. | 3 | Effects of culm carbohydrate partitioning on basal stem strength in a high-yielding rice population显示文摘Rice culm carbohydrate transport can simultaneously affect grain filling and stem lodging resistance by regulating non-structural carbohydrate(NSC) and structural carbohydrate(SC) contents. However, the relationship between carbohydrate transposition and culm strength is not well documented. Accordingly, a high-yielding hybrid rice cultivar(Y Liangyou 2) was tested under different N fertilization regimes at two locations, Taoyuan(a special high-yield eco-site), Yunnan province and Danyang(a representative eco-site of the middle and lower Yangtze), Jiangsu province, China. Significantly higher grain yield and basal stem strength were found at Taoyuan than Danyang under all N rates throughout the two-year experiment. At heading stage, soluble sugars, starch, cellulose and lignin contents of the basal culm at Taoyuan were significantly 132.0%, 73.7%, 1.2%, and 62.7% higher than those at Danyang, respectively. At 20 days after heading, soluble sugars and starch content at Taoyuan decreased significantly compared to Danyang, but lignin content remained higher. Culm carbohydrate transport to kernels at Taoyuan was significantly greater than that at Danyang, and the proportion of soluble sugars and starch was correspondingly 62.9%lower. However, the proportion of lignin and cellulose was 22.7% higher at Taoyuan than that at Danyang. Soluble sugars and starch partitioning were significantly reduced under an increased nitrogen application rate, but SC partitioning was little affected. Two sequences of S1 on the eastern and west-ern sides of the Liupan Mountain were both composed of five paleosol layers and four loess layers,indicating that there were five strong summer monsoon events and four strong winter monsoon events in MIS5. This corresponds with other records of the East Asian Monsoon,along with NGRIP and the North Atlantic records,implying that the climate of the Northern Hemisphere was very instable during the last interglacial. | 10 | Occurrence, spatial distribution and ecological risks of antibiotics in soil in urban agglomeration显示文摘Antibiotics in soil environment are regarded as emerging pollutants and have introduced increasing risks to soil ecosystem and human health in rapid urbanization areas. Identifying the occurrence and spatial variability of antibiotics in soils is an urgent issue in sustaining soil security. In this study, antibiotics in soils were investigated and analyzed in BeijingTianjin-Hebei urban agglomeration. The occurrence, spatial distribution, and related affecting factors of antibiotics in soils were identified and ecological risks of antibiotics in soil environment were assessed. Results showed that(1) The mean concentration of soil antibiotics in Beijing-Tianjin-Hebei urban agglomeration was 21.79 μg/kg. Land use substantially affected the occurrence and concentration of antibiotics in soils. Concentrations of antibiotics in cropland and orchard soils were 2-3 times higher than the other land use types.(2)The concentrations of antibiotics in soils in Beijing-Tianjin-Hebei urban agglomeration presented a spatial pattern of high values in southeast, and low values in northwest. Spatial variability of antibiotics in soils was closely related to the application of organic fertilizer and wastewater irrigation as well as topographical features. Furthermore, soil properties and land management policy had substantial influences on soil antibiotics, and soil heavy metals may aggravate the accumulation of antibiotics in soils.(3) Ecological risks assessment of antibiotics in soils demonstrated that erythromycin(ERY), sulfamethoxazole(SMX), and doxycycline(DOX) may introduce high risks to soil ecosystem health, and more attention should be paid to the areas with intensive human activities that had potential high risk to soil ecosystem health. | 13 | Selenium deficiency induces spleen pathological changes in pigs by decreasing selenoprotein expression,evoking oxidative stress,and activating inflammation and apoptosis显示文摘Background:The immune system is one aspect of health that is affected by dietary selenium(Se)levels and selenoprotein expression.Spleen is an important immune organ of the body,which is directly involved in cellular immunity.However,there are limited reports on Se levels and spleen health.Therefore,this study established a Se-deficient pig model to investigate the mechanism of Se deficiency-induced splenic pathogenesis.Methods:Twenty-four pure line castrated male Yorkshire pigs(45 days old,12.50±1.32 kg,12 full-sibling pairs)were divided into two equal groups and fed Se-deficient diet(0.007 mg Se/kg)or Se-adequate diet(0.3 mg Se/kg)for 16 weeks.At the end of the trial,blood and spleen were collected to assay for erythroid parameters,the osmotic fragility of erythrocytes,the spleen index,histology,terminal deoxynucleotidyl transferase nick-end labeling(TUNEL)staining,Se concentrations,the selenogenome,redox status,and signaling related inflammation and apoptosis.Results:Dietary Se deficiency decreased the erythroid parameters and increased the number of osmotically fragile erythrocytes(P<0.05).The spleen index did not change,but hematoxylin and eosin and TUNEL staining indicated that the white pulp decreased,the red pulp increased,and splenocyte apoptosis occurred in the Se deficient group.Se deficiency decreased the Se concentration and selenoprotein expression in the spleen(P<0.05),blocked the glutathione and thioredoxin antioxidant systems,and led to redox imbalance.Se deficiency activated the NF-κB and HIF-1αtranscription factors,thus increasing pro-inflammatory cytokines(IL-1β,IL-6,IL-8,IL-17,and TNF-α),decreasing anti-inflammatory cytokines(IL-10,IL-13,and TGF-β)and increasing expression of the downstream genes COX-2 and iNOS(P<0.05),which in turn induced inflammation.In addition,Se-deficiency induced apoptosis through the mitochondrial pathway,upregulated apoptotic genes(Caspase3,Caspase8,and Bak),and downregulated antiapoptotic genes(Bcl-2)(P<0.05)at the mRNA level,thus verifying the results of TUNEL staining.Conclusions:These results indicated that Se deficiency induces spleen injury through the regulation of selenoproteins,oxidative stress,inflammation and apoptosis. | Shuang Li Wenjuan Sun Kai Zhang Jiawei Zhu Xueting Jia Xiaoqing Guo Qingyu Zhao Chaohua Tang Jingdong Yin Junmin Zhang | 2021 | Journal of Animal Science and Biotechnology2021,12,4: | 4 |

| 15 | Targeting PFKL with penfluridol inhibits glycolysis and suppresses esophageal cancer tumorigenesis in an AMPK/FOXO3a/BIM-dependent manner显示文摘As one of the hallmarks of cancer,metabolic reprogramming leads to cancer progression,and targeting glycolytic enzymes could be useful strategies for cancer therapy.By screening a small molecule library consisting of 1320 FDA-approved drugs,we found that penfluridol,an antipsychotic drug used to treat schizophrenia,could inhibit glycolysis and induce apoptosis in esophageal squamous cell carcinoma(ESCC).Gene profiling and Ingenuity Pathway Analysis suggested the important role of AMPK in action mechanism of penfluridol.By using drug affinity responsive target stability(DARTS)technology and proteomics,we identified phosphofructokinase,liver type(PFKL),a key enzyme in glycolysis,as a direct target of penfluridol.Penfluridol could not exhibit its anticancer property in PFKL-deficient cancer cells,illustrating that PFKL is essential for the bioactivity of penfluridol.High PFKL expression is correlated with advanced stages and poor survival of ESCC patients,and silencing of PFKL significantly suppressed tumor growth.Mechanistically,direct binding of penfluridol and PFKL inhibits glucose consumption,lactate and ATP production,leads to nuclear translocation of FOXO3a and subsequent transcriptional activation of BIM in an AMPK-dependent manner.Taken together,PFKL is a potential prognostic biomarker and therapeutic target in ESCC,and penfluridol may be a new therapeutic option for management of this lethal disease. | Cancan Zheng Xiaomei Yu Yiyao Liang Yidong Zhu Yan He Long Liao Dingkang Wang Yanming Yang Xingfeng Yin Ang Li Qingyu He Bin Li | 2022 | Acta Pharmaceutica Sinica B2022,12,3: | 4 |

| 17 | Serum uric acid is a risk factor for large-artery atherosclerosis cerebral infarction显示文摘Using the Trial of Org 10172 in Acute Stroke Treatment (TOAST) classification for acute ischemic stroke, 371 patients with either acute large-artery atherosclerosis or small-artery oc-clusion cerebral infarction were recruited to investigate the potential impact of elevated serum uric acid on cerebrovascular disorders. The results showed that patients who have suffered from large-artery atherosclerosis, relative to small-artery occlusion patients, were characterized by elevated serum uric acid but reduced high-density lipoprotein cholesterol and triglyceride levels. Logistic regression showed that elevated uric acid and lower triglyceride levels were the main risk factors for patients with large-artery atherosclerosis. The main difficulty of this problem is to accurately and comprehensively reflect the time-series characteristics and spatial-temporal correlation of wind power generation. In this paper, the marginal distribution model and the dependence structure are combined to describe these complex characteristics. On this basis, a scenario generation method for multiple wind farms is proposed. For the marginal distribution model, the autoregressive integrated moving average-generalized autoregressive conditional heteroskedasticity-t (ARIMA-GARCH-t) model is proposed to capture the time-series characteristics of wind power generation. For the dependence structure, a time-varying regular vine mixed Copula (TRVMC) model is established to capture the spatial-temporal correlation of multiple wind farms. Based on the data from 8 wind farms in Northwest China, sufficient scenarios are generated. The effectiveness of the scenarios is evaluated in 3 aspects. | 19 | Low Sulfur Low Olefin Gasoline Production by RIDOS Technology显示文摘By analysis of the component and structure of a real high olefinic FCC naphtha, a technology of deep desulfurization and olefin saturation with minimal octane loss is developed, which is named RIDOS. The multi functions of RIDOS technology are achieved by using a series of new catalysts, which show the good feed adaptability and operation flexibility. The results of the first commercial RIDOS unit showed that an ultra low sulfur (<10 μg/g), low olefin(<20%) gasoline was produced, while the road octane number lost only 1.3 unit. | 20 | Depositional System of the Carboniferous Huanglong Formation,Eastern Sichuan Basin:Constraints from Sedimentology and Geochemistry显示文摘Using analyses of the lithology,sequences,paleoenvironment,and tectonic setting,the depositional system of the Carboniferous Huanglong Formation in the eastern Sichuan Basin was identified.The lithological characteristics of the Lower Member,Middle Member,and Upper Member were analyzed and classified.Before the use of carbon,oxygen,and strontium isotopes in the analysis,all of the geochemical data were tested for validity.On the basis of the Z values obtained from carbon and oxygen isotopes,the paleoenvironments of the three members were elucidated.Lower Member was dominantly an enclosed marine environment with intense evaporation and little freshwater input into the sea.Middle Member developed in a semi-enclosed to normal marine environment with many rivers.Upper Member was formed in a normal marine environment.The east Sichuan Basin was enclosed by paleouplifts before the deposition of the Huanglong Formation,forming a relatively enclosed depositional setting.Paleogullies developed in the Silurian strata that underlie the Carboniferous rocks;these paleogullies can be identified.On the basis of a comprehensive analysis,we propose that the Huanglong Formation developed in a platform system.Four microfacies were identified:supratidal flat,dolostone flat,grain shoal,and shelf microfacies.The high-permeability and high-porosity characteristics of the grain shoal microfacies are favorable for hydrocarbon accumulation,while the supratidal flat and shelf microfacies developed very few high-quality reservoirs.The paleogullies,in which increased amounts of grain shoal microfacies developed,controlled the distribution of high-quality reservoirs. | WANG Kun HU Suyun LI Wei LIU Wei HUANG Qingyu MA Kui SHI Shuyuan | 2016 | Acta Geologica Sinica(English Edition)2016,90,5: | 3 |
