1Life History Recorded in the Vagino-cervical Microbiome Along with Multi-omes显示文摘The vagina contains at least a billion microbial cells,dominated by lactobacilli.Here we perform metagenomic shotgun sequencing on cervical and fecal samples from a cohort of 516 Chinese women of reproductive age,as well as cervical,fecal,and salivary samples from a second cohort of 632 women.Factors such as pregnancy history,delivery history,cesarean section,and breastfeeding were all more important than menstrual cycle in shaping the microbiome,and such information would be necessary before trying to interpret differences between vagino-cervical microbiome data.Greater proportion of Bifidobacterium breve was seen with older age at sexual debut.The relative abundance of lactobacilli especially Lactobacillus crispatus was negatively associated with pregnancy history.Potential markers for lack of menstrual regularity,heavy flow,dysmenorrhea,and contraceptives were also identified.Lactobacilli were rare during breastfeeding or post-menopause.Other features such as mood fluctuations and facial speckles could potentially be predicted from the vagino-cervical microbiome.Gut and salivary microbiomes,plasma vitamins,metals,amino acids,and hormones showed associations with the vagino-cervical microbiome.Our results offer an unprecedented glimpse into the microbiota of the female reproductive tract and call for international collaborations to better understand its long-term health impact other than in the settings of infection or pre-term birth.Zhuye Jie Chen Chen Lilan Hao Fei Li Liju Song Xiaowei Zhang Jie Zhu Liu Tian Xin Tong Kaiye Cai Zhe Zhang Yanmei Ju Xinlei Yu Ying Li Hongcheng Zhou Haorong Lu Xuemei Qiu Qiang Li Yunli Liao Dongsheng Zhou Heng Lian Yong Zuo Xiaomin Chen Weiqiao Rao Yan Ren Yuan Wang Jin Zi Rong Wang Na Liu Jinghua Wu Wei Zhang Xiao Liu Yang Zong Weibin Liu Liang Xiao Yong Hou Xun Xu Huanming Yang Jian Wang Karsten Kristiansen Huijue Jia 2022Genomics, Proteomics & Bioinformatics2022,20,2:2

8A collision detection approach in virtual environment of micromanipulation robot显示文摘Operators suffer much difficulty in manipulating micro-size objects without the assistance of friendly interfaces due to the scaling effects in micro world. The paper presented a general framework for micromanipulation robot based on virtual reality technology. With the framework we brought forward a FDH (Fixed Direction Hulls) based bounding box method to handle the collision detection of the peg-in-hole microassembly. The collision response model for the collision between micro needle and hole was presented. The virtual force and corresponding displacement were calculated with the model of bending deformation and pressing deformation. 10Magnetic Actuated Shape-memory Helical Microswimmers with Programmable Recovery Behaviors显示文摘Inspired by bacterial flagella in nature,magnetic helical microswimmer is an ideal model to perform complex task in a low Reynolds number environments.Shape Memory Polymers(SMPs)with desirable properties are considered as one of the most preferred options for the development of small-scale robots.However,fabricating and programming strategies are still challenging.Here,we report an approach to fabricate helical microswimmers based on thermoplastic SMP(polylactic acid).Melt-spun polylactic acid fibers containing magnetic particles were enwound to form helical microstructures.Their shape recovery behaviors were programmed by annealing and pre-deformation.Three forms of helical microswimmers(constant-helix-angle conical helix,constant-pitch conical helix,and straight helix)with controlled morphological parameters were tailored.The obtained microswimmers showed 3D locomotion capability under rotating magnetic fields.The maximum swimming velocity of microswimmers was nearly six body lengths per second,and the near-wall swimming of conical helixes along their sharp end exhibited a smaller drift.Moreover,we demonstrated programmed shape-switching processes(spring-like contraction and elongation,coiling and uncoiling)and self-repairing of the microswimmers.As demonstrations of potential applications,tasks of mobile microstent,cargo delivery,and minimally invasive injection were carried out.The multifunctional shape-memory microswimmers have immense potential in a variety of applications.Fu Zhao Weibin Rong Lefeng Wang Lining Sun 2021Journal of Bionic Engineering2021,18,4:1

12Discovery of a highly specific ^(18)F-labeled PET ligand for phosphodiesterase 10A enabled by novel spirocyclic iodonium ylide radiofluorination显示文摘As a member of cyclic nucleotide phosphodiesterase(PDE)enzyme family,PDE10A is in charge of the degradation of cyclic adenosine(cAMP)and guanosine monophosphates(cGMP).While PDE10A is primarily expressed in the medium spiny neurons of the striatum,it has been implicated in a variety of neurological disorders.Indeed,inhibition of PDE10A has proven to be of potential use for the treatment of central nervous system(CNS)pathologies caused by dysfunction of the basal ganglia–of which the striatum constitutes the largest component.A PDE10A-targeted positron emission tomography(PET)radioligand would enable a better assessment of the pathophysiologic role of PDE10A,as well as confirm the relationship between target occupancy and administrated dose of a given drug candidate,thus accelerating the development of effective PDE10A inhibitors.In this study,we designed and synthesized a novel ^(18)F-aryl PDE10A PET radioligand,codenamed[^(18)F]P10A-1910([^(18)F]9),in high radiochemical yield and molar activity via spirocyclic iodonium ylide-mediated radiofluorination.[^(18)F]9 possessed good in vitro binding affinity(IC_(50)=2.1 nmol/L)and selectivity towards PDE10A.Further,[^(18)F]9 exhibited reasonable lipophilicity(logD=3.50)and brain permeability(P_(app)>10×10^(−6) cm/s in MDCK-MDR1 cells).PET imaging studies of[^(18)F]9 revealed high striatal uptake and excellent in vivo specificity with reversible tracer kinetics.Preclinical studies in rodents revealed an improved plasma and brain stability of[^(18)F]9 when compared to the current reference standard for PDE10A-targeted PET,[^(18)F]MNI659.Further,dose–response experiments with a series of escalating doses of PDE10A inhibitor 1 in rhesus monkey brains confirmed the utility of[^(18)F]9 for evaluating target occupancy in vivo in higher species.In conclusion,our results indicated that[^(18)F]9 is a promising PDE10A PET radioligand for clinical translation.Zhiwei Xiao Huiyi Wei Yi Xu Ahmed Haider Junjie Wei Shiyu Yuan Jian Rong Chunyu Zhao Guocong Li Weibin Zhang Huangcan Chen Yuefeng Li Lingling Zhang Jiyun Sun Shaojuan Zhang Hai-Bin Luo Sen Yan Qijun Cai Lu Hou Chao Che Steven H.Liang Lu Wang 2022Acta Pharmaceutica Sinica B2022,12,4:0
13基于磁驱动正交悬臂探针的三维原子力显微镜技术开发显示文摘本文介绍了一种基于磁驱动正交悬臂探针(magnetically driven-orthogonal cantilever probes,MDOCP)的三维原子力显微镜(three-dimensional atomic force microscopy,3D-AFM)表征方法,该方法采用两个独立的三自由度纳米扫描器,能够实现探针沿可控矢量角度跟踪扫描样品表面。该3D-AFM系统还配备了高精度旋转台,可实现360°全向成像。定制的MD-OCP包含水平悬臂、垂直悬臂和磁球三部分,其中磁球可在磁场中机械驱动OCP实现激振。垂直悬臂具有一个突出的尖端,可检测深槽和具有悬垂/凹边特征的结构。首先,对MD-OCP的设计、模拟、制造和性能分析进行了描述;其次,详细介绍了探针振幅补偿和360°旋转原点定位的方法。通过使用标准AFM阶梯光栅进行对比实验,验证了所提出方法对于陡峭侧壁和拐角处细节的表征能力,其中采用了三维地形重建方法将图像整合。通过对具有微梳结构的微机电系统(MEMS)器件进行3D表征,进一步证实了所提出基于MD-OCP的3D-AFM技术的有效性。最后,该技术被用于确定微阵列芯片的关键尺寸(critical dimensions,CD)。实验结果表明,所提出的方法可以高精度地获取三维结构的CD信息,相比于难以获得侧壁信息的二维技术,在三维微纳制造检测领域具有更好的潜力。Hao Zhang Junyuan Geng Haibo Gao Weibin Rong Hui Xie 2023Engineering2023,,5:0
14Multi-sensor control for precise assembly of optical components显示文摘In order to perform an optical assembly accurately, a multi-sensor control strategy is developed which includes an attitude measurement system, a vision system, a loss measurement system and a force sensor. A 3-DOF attitude measuring method using linear variable differential transformers(LVDT) is designed to adjust the relation of position and attitude between the spherical mirror and the resonator. A micro vision feedback system is set up to extract the light beam and the diaphragm, which can achieve the coarse positioning of the spherical mirror in the optical assembly process. A rapid self-correlation method is presented to analyze the spectrum signal for the fine positioning. In order to prevent the damage of the optical components and realize sealing of the resonator, a hybrid force-position control is constructed to control the contact force of the optical components. The experimental results show that the proposed multi-sensor control strategy succeeds in accomplishing the precise assembly of the optical components, which consists of parallel adjustment, macro coarse adjustment, macro approach, micro fine adjustment, micro approach and optical contact.
