1Toripalimab plus chemotherapy as second-line treatment in previously EGFR-TKI treated patients with EGFR-mutant-advanced NSCLC:a multicenter phase-II trial显示文摘This multicenter phase-II trial aimed to investigate the efficacy,safety,and predictive biomarkers of toripalimab plus chemotherapy as second-line treatment in patients with EGFR-mutant-advanced NSCLC.Patients who failed from first-line EGFR-TKIs and did not harbor T790M mutation were enrolled.Toripalimab plus carboplatin and pemetrexed were administrated every three weeks for up to six cycles,followed by the maintenance of toripalimab and pemetrexed.The primary endpoint was objective-response rate(ORR).Integrated biomarker analysis of PD-L1 expression,tumor mutational burden(TMB),CD8+tumor-infiltrating lymphocyte(TIL)density,whole-exome,and transcriptome sequencing on tumor biopsies were also conducted.Forty patients were enrolled with an overall ORR of 50.0%and disease-control rate(DCR)of 87.5%.The median progression free survival(PFS)and overall survival were 7.0 and 23.5 months,respectively.The most common treatment-related adverse effects were leukopenia,neutropenia,anemia,ALT/AST elevation,and nausea.Biomarker analysis showed that none of PD-L1 expression,TMB level,and CD8+TIL density could serve as a predictive biomarker.Integrated analysis of whole-exome and transcriptome sequencing data revealed that patients with DSPP mutation had a decreased M2 macrophage infiltration and associated with longer PFS than those of wild type.Toripalimab plus chemotherapy showed a promising anti-tumor activity with acceptable safety profiles as the second-line setting in patients with EGFR-mutant NSCLC.DSPP mutation might serve as a potential biomarker for this combination.A phase-III trial to compare toripalimab versus placebo in combination with chemotherapy in this setting is ongoing(NCT03924050).Tao Jiang Pingyang Wang Jie Zhang Yanqiu Zhao Jianying Zhou Yun Fan Yongqian Shu Xiaoqing Liu Helong Zhang Jianxing He Guanghui Gao Xiaoqian Mu Zhang Bao Yanjun Xu Renhua Guo Hong Wang Lin Deng Ningqiang Ma Yalei Zhang Hui Feng Sheng Yao Jiarui Wu Luonan Chen Caicun Zhou Shengxiang Ren 2021Signal Transduction and Targeted Therapy2021,6,11:8
2Cloning and expression analysis of a dirigent protein gene from the resurrection plant Boea hygrometrica显示文摘Resurrection plants are tolerant to extreme dehydration and are useful model systems to study genes that play a role in drought tolerance. A gene fragment encoding a dirigent protein that is predicted to function in lignin biosynthesis was identified from leaves of the resurrection plant Boea hygrometrica via cDNA microarray screening. A cDNA, designated BhDIR1, containing the complete predicted open reading frame, was obtained by 50-RACE. BhDIR1 transcripts were found to be accumulated in response to changes in plant dehydration status, exogenously applied phytohormones and signaling molecules, and temperature stresses. BhDIR1 encodes a protein of 199 amino acids, which shows 20-40% similarity to dirigent proteins reported from other plants. BhDIR1 is predicted to contain a cleavable signal peptide at the N-terminal, and its plasma membrane/cell wall localization was confirmed using a GFP fusion protein assay. Consistent with this discovery, the acid-soluble lignin content decreased in dehydrated B. hygrometrica leaves. 3High VHL Expression Reverses Warburg Phenotype and Enhances Immunogenicity in Kidney Tumor Cells显示文摘Clear cell renal cell carcinoma(ccRCC)is a frequently occurring renal cancer.The Von Hippel-Lindau disease tumor suppressor VHL,a known tumor suppressor gene,is frequently mutated in about 50%of patients with ccRCC.However,it is unclear whether VHL influences the progression of ccRCC tumors expressing wild-type VHL.In the present study,we found that higher expression of VHL was correlated with the better disease-free survival(DFS)in ccRCC patients using The Cancer Genome Atlas(TCGA)datasets.We revealed that VHL overexpression in ccRCC cells inhibited epithelial-mesenchymal transition(EMT),sterol regulatory elementbinding protein 1(SREBP1)-regulated triglyceride synthesis,and cell proliferation.Proteomic analysis provided us a global view that VHL regulated four biological processes,including metabolism,immune regulation,apoptosis,and cell movement.Importantly,we found that VHL overexpression led to up-regulated expression of proteins associated with antigen processing and interferonresponsive proteins,thus rendering ccRCC cells more sensitive to interferon treatment.We defined an interferon-responsive signature(IRS)composed of ten interferon-responsive proteins,whose m RNA expression levels were positively correlated with DFS in ccRCC patients.Taken together,our results propose that the subset of ccRCC patients with high VHL expression benefit from immunotherapy.Songbiao Zhu Wenxi Ding Yuling Chen Weixuan Wang Renhua Xu Chongdong Liu Xiaohui Liu Haiteng Deng 2022Genomics, Proteomics & Bioinformatics2022,20,4:1

6Polymersome formation by solvent annealing-induced structural reengineering under 3D soft confinement显示文摘A solvent annealing-induced structural reengineering approach is exploited to fabricate polymersomes from block copolymers that are hard to form vesicles through the traditional solution self-assembly route.More specifically,polystyrene-b-poly(4-vinyl pyridine)(PS-b-P4VP)particles with sphere-within-sphere structure(SS particles)are prepared by three-dimensional(3D)soft-confined assembly through emulsion-solvent evaporation,followed by 3D soft-confined solvent annealing upon the SS particles in aqueous dispersions for structural engineering.A water-miscible solvent(e.g.,THF)is employed for annealing,which results in dramatic transitions of the assemblies,e.g.,from SS particles to polymersomes.This approach works for PS-b-P4VP in a wide range of block ratios.Moreover,this method enables effective encapsulation/loading of cargoes such as fluorescent dyes and metal nanoparticles,which offers a new route to prepare polymersomes that could be applied for cargo release,diagnostic imaging,and nanoreactor,etc.Xi Mao Hao Li Jinwoo Kim Shuai Deng Renhua Deng Bumjoon J.Kim Jintao Zhu 2021Nano Research2021,14,12:0
7Kinetically Controlled Supramolecular Block Copolymer Self-Assembly:Multicolor Photonic Crystal Patterns from a Single Formulation显示文摘Photonic crystal(PC)patterns with tunable and changeable nonvolatile structural colors printed from a single ink are of great interest for optical products but have rarely been reported because most inks can only output one respective structural color.Herein,we propose a facile yet effective kinetically controlled self-assembly strategy to address this challenge.An ink formulation containing supramolecular block copolymers(SBCPs)is developed.SBCP patterns were printed by direct-ink-writing followed by solvent annealing to generate different structural colors by simply controlling the annealing time.The self-assembly kinetic regime suggests that different colors result from various kinetically trapped metastable states.In turn,the variation in structural color enables“visualization”of the self-assembly dynamics.Furthermore,we demonstrate that these kinetically trapped structures exhibit different responsive color-change behaviors.In addition,this kinetic control strategy can be synergistic with thermodynamic control to extend the color range.This study provides a facile yet effective solution for well-designed PC patterns with tunable,responsive,and unfading colors printed from the simplest single-nozzle printer with a single colorless ink,presenting great potential in broad applications,including information storage,encryption,and anti-fake.Shuai Deng Renhua Deng Xi Mao Bijin Xiong Mian Wang Senbin Chen Wolfgang H.Binder Jintao Zhu Zhenzhong Yang 2023CCS Chemistry2023,5,10:0
