1The transport of water in subduction zones显示文摘The transport of water from subducting crust into the mantle is mainly dictated by the stability of hydrous minerals in subduction zones. The thermal structure of subduction zones is a key to dehydration of the subducting crust at different depths. Oceanic subduction zones show a large variation in the geotherm, but seismicity and arc volcanism are only prominent in cold subduction zones where geothermal gradients are low. In contrast, continental subduction zones have low geothermal gradients, resulting in metamorphism in cold subduction zones and the absence of arc volcanism during subduction. In very cold subduction zone where the geothermal gradient is very low(?5?C/km), lawsonite may carry water into great depths of ?300 km. In the hot subduction zone where the geothermal gradient is high(>25?C/km), the subducting crust dehydrates significantly at shallow depths and may partially melt at depths of <80 km to form felsic melts, into which water is highly dissolved. In this case, only a minor amount of water can be transported into great depths. A number of intermediate modes are present between these two end-member dehydration modes, making subduction-zone dehydration various. Low-T/low-P hydrous minerals are not stable in warm subduction zones with increasing subduction depths and thus break down at forearc depths of ?60–80 km to release large amounts of water. In contrast, the low-T/low-P hydrous minerals are replaced by low-T/high-P hydrous minerals in cold subduction zones with increasing subduction depths, allowing the water to be transported to subarc depths of 80–160 km. In either case, dehydration reactions not only trigger seismicity in the subducting crust but also cause hydration of the mantle wedge. Nevertheless, there are still minor amounts of water to be transported by ultrahigh-pressure hydrous minerals and nominally anhydrous minerals into the deeper mantle. The mantle wedge overlying the subducting slab does not partially melt upon water influx for volcanic arc magmatism, but it is hydrated at first with the lowest temperature at the slab-mantle interface, several hundreds of degree lower than the wet solidus of hydrated peridotites. The hydrated peridotites may undergo partial melting upon heating at a later time. 2Tectonic evolution of convergent plate margins and its geological effects显示文摘Oceanic lithosphere is generated at divergent plate boundaries and disappears at convergent plate boundaries.Seafloor spreading and plate subduction together constitute the physical coupling and mass conservation relationships to the movement of lithospheres on Earth.Subduction zones are a key site for the transfer of both matter and energy at converging plate boundaries,and their study has been the hot spot and frontier of Earth system science since the development of plate tectonics theory.As far as the dynamic regime and geothermal gradient of convergent plate margins are concerned,they have different properties in different stages of the subduction zone evolution.In general,the early low-angle subduction leads to compressional tectonism dominated by low geothermal gradients at the plate interface,and the late high-angle subduction results in extensional tectonism dominated by high geothermal gradients at the plate interface and its hanging wall.Active rifts are produced along suture zones through not only slab rollback or slab breakoff in the terminal stage of oceanic subduction but also foundering and thinning of the lithosphere in the post-subduction stage.Due to the differences and changes in the geometric and thermobaric structures of convergent plate margins,a series of changes in the type of metamorphism and magmatism can occur in active and fossil subduction zones.Dehydration and melting of the subducting oceanic crust are prominent at subarc depths,giving rise to fluids that dissolve different concentrations of fluid-mobile incompatible elements.The subduction zone fluids at subarc depths would chemically react with the overlying mantle wedge peridotite,generating metasomatites as the mantle sources of mafic magmas in oceanic and continental arcs.However,these metasomatites did not partially melt immediately upon the fluid metasomatism to trigger arc magmatism,and they did not melt until they were heated by asthenospheric convection due to rollback of the subducting slab.Therefore,recognition of the changes in the dynamic regime and geothermal gradient of subduction zones in different stages of plate convergence not only provides insights into geodynamic mechanisms of the tectonic evolution from subduction zones to orogenic belts,but also places constraints on the formation and evolution of different types of metamorphic and magmatic rocks within the advanced framework of plate tectonics.Yongfei ZHENG Yixiang CHEN Renxu CHEN Liqun DAI 2022Science China Earth Sciences2022,65,7:6
3Water contents and hydrogen isotopes in nominally anhydrous minerals from UHP metamorphic rocks in the Dabie-Sulu orogenic belt显示文摘A continuous flow method,by a combination of thermal conversion elemental analyzer(TC/EA)with isotope ratio mass spectrometry(MS),was developed to determine both H isotope composition and H2O concentration of ultrahigh-pressure(UHP)metamorphic rocks in the Dabie-Sulu orogenic belt.By using the developed step-heating technique,we have studied H2O concentration and H isotope composition of the different forms of water(structural OH and molecular H2O)in garnet.The quantitative measurements of H2O concentration and H isotope composition of minerals in UHP metamorphic rocks from several typical outcrops indicate that the gneisses can release more amounts of water than the eclogites during exhumation of the deeply subducted continental crust.Therefore,by decompression dehydration at the contact between eclogite and gneiss,the released water could flow from the gneiss to the eclogite and result in significant hydration of the eclogite adjacent to the gneiss.The measured maximum water contents of minerals in eclogites indicate that garnet and omphacite have the maximum water solubilities of 2500and 3500 ppm,respectively,under the peak UHP metamorphic conditions.GONG Bing CHEN RenXu ZHENG YongFei 2013Chinese Science Bulletin2013,58,35:5
4Maxingxiongting mixture attenuates hypoxia pulmonary arterial hypertension to improve right ventricular hypertrophy by inhibiting the rho-kinase signaling pathway显示文摘OBJECTIVE:To explore the mechanism of Maxingxiongting mixture(MXXTM)on pulmonary hypertension in a rat model established by intraperitoneal injection of monocrotaline solution,smoking and forced swimming.METHODS:A total of 30 male Sprague-Dawley rats were randomly divided into five groups:control group,model group,high-dose of MXXTM group(HM),low-dose of MXXTM group(LM),and fasudil group.The mean pulmonary artery pressure(m PAP)was measured by using a miniature catheter.Lung tissue and right ventricular tissue sections were stained with hematoxylin-eosin.The right ventricle(RV)and left ventricle+septum(LV+S)were weighted.RV/(LV+S)was calculated to reflect the degree of right ventricular hypertrophy.Rho/Rho-kinase signaling pathway key proteins(Rho A,ROCKⅠand ROCKⅡ)in rat right ventricular tissue were measured by Western blot analysis.The levels of serum hypoxia-inducible factor-1α(HIF-1α),vascular endothelial growth factor(VEGF)and the levels of plasma renin activity(PRA),angiotensinⅡ(ANG-Ⅱ),aldosterone(ALD)in rat anticoagulated plasma were all measured by enzyme-linked immunosorbent assay.RESULTS:Compared with the control group,the m PAP and RV/(LV+S)in the model group were significantly increased.Administration of fasudil resulted in a significant decrease of m PAP and RV/(LV+S).In the HM group and LM group,m PAP and RV/(LV+S)were significantly lower than the model group.Compared with the control group,the contents of HIF-1α,VEGF,PRA,ANG-Ⅱand ALD in the model group were significantly increased.The administration of fasudil and high-dose MXXTM significantly reduced the contents of HIF-1α,VEGF,PRA,ANG-II and ALD.Compared with the control group,the expression of Rho A,ROCKⅠand ROCKⅡin the right ventricle of the model group were significantly increased.The administration of fasudil and high-dose MXXTM significantly reduced the expression of Rho A and RockⅡproteins.Our results indicated that high-dose of MXXTM had similar effects on reducing pulmonary artery pressure and improving right ventricular remodeling to fasudil.However,MXXTM was unable to restore parameters above to control levels.CONCLUSIONS:MXXTM attenuates hypoxia pulmonary arterial hypertension to improve right ventricular hypertrophy by inhibiting the Rho-kinase signaling pathway.An Xing Li Songtao Weng Xiangwen Wang Xian Wu Hao Zhang Xinyue Gao Jian Yang Renxu Peng Bo 2020Journal of Traditional Chinese Medicine2020,40,6:4
5Crystal structure of plant PLDα1 reveals catalytic and regulatory mechanisms of eukaryotic phospholipase D显示文摘Phospholipase D(PLD)hydrolyzes the phosphodiester bond of glycerophospholipids and produces phosphatidic acid(PA),which acts as a second messenger in many living organisms.A large number of PLDs have been identified in eukaryotes,and are viewed as promising targets for drug design because these enzymes are known to be tightly regulated and to function in the pathophysiology of many human diseases.However,the underlying molecular mechanisms of catalysis and regulation of eukaryotic PLD remain elusive.Here,we determined the crystal structure of full-length plant PLDα1 in the apo state and in complex with PA.The structure shows that the N-terminal C2 domain hydrophobically interacts with the C-terminal catalytic domain that features two HKD motifs.Our analysis reveals the catalytic site,substrate-binding mechanism,and a new Ca2+-binding site that is required for the activation of PLD.In addition,we tested several efficient small-molecule inhibitors against PLDα1,and suggested a possible competitive inhibition mechanism according to structure-based docking analysis.This study explains many long-standing questions about PLDs and provides structural insights into PLD-targeted inhibitor/drug design.Jianxu Li Fang Yu Hui Guo Renxue Xiong Wenjing Zhang Fangyuan He Minhua Zhang Peng Zhang 2020Cell Research2020,30,1:3
6Fabrication of a monolithic 4H-SiC junction barrier schottky diode with the capability of high current显示文摘There is a great interest in monolithic 4H-Si C Junction Barrier Schottky(JBS) diodes with the capability of a high forward current for industrial power applications.In this paper,we report large-area monolithic 4H-Si C JBS diodes fabricated on a 10 μm 4H-Si C epitaxial layer doped to 6×1015 cm3.JBS diodes with an active area of 30 mm2 had a forward current of up to 330 A at a forward voltage of 5 V,which corresponds to a current density of 1100 A/cm2.A near ideal breakdown voltage of 1.6 k V was also achieved for a reverse current of up to 100 A through the use of an optimum multiple floating guard rings(MFGR) termination,which is about 87.2% of the theoretical value.The differential specific-on resistance(RSP-ON) was measured to be 3.3 m cm2,leading to a FOM(VB2/RSP-ON) value of 0.78 GW/cm2,which is very close to the theoretical limit of the tradeoff between the specific-on resistance and breakdown voltage for 4H-Si C unipolar devices.SONG QingWen YUAN Hao HAN Chao ZHANG YuMing TANG XiaoYan ZHANG YiMeng GUO Hui ZHANG YiMen JIA RenXu WANG YueHu 2015Science China(Technological Sciences)2015,58,8:2

10Metamorphic evolution of the East Tethys tectonic domain and its tectonic implications显示文摘The China Central Orogenic System(CCOS),extending in an east-west direction in the middle part of China,is composed of the Early Paleozoic Altyn-North Qilian-North Qaidam-East Kunlun-North Qinling-North Tongbai orogens in the west and the Late Paleozoic to Early Mesozoic South Tongbai-Hong'an-Dabie-Sulu orogens in the east.They were produced by oceanic subduction and continental subduction/collision during the closure of the Proto-Tethys and the Paleo-Tethys oceans,respectively.Different types of metamorphic rocks with various ages are extensively exposed in these orogens,and they were produced at different geothermal gradients in different stages during the tectonic evolution of convergent continental margins,making them ideal targets to reconstruct the spatiotemporal evolution of the Eastern Tethys tectonic domain.In this article,an integrated study of metamorphic temperature(T)-pressure(P)-time(t)records is presented for metamorphic rocks along the CCOS,aiming to ascertain the change of metamorphic T/P ratios in both time and space,and then shed light on the tectonic evolution of the East Tethys tectonic domain in association with the thermal state change of convergent continental margins.The results indicate that despite the difference in metamorphic ages,metamorphic rocks in different orogens show a common trend with clockwise P-T-t paths.With respect to plate convergence for subduction and collision,regional metamorphism is categorized into three stages:(1)an early convergent stage,corresponding to low T/P Alpine-type blueschist-to eclogite-facies high-P to ultrahigh-P metamorphism;(2)a later convergent stage,corresponding to the medium T/P Barrovian-type medium-P amphibolite to high-P granulite-facies metamorphism;and(3)a post-convergent stage,corresponding to the high T/P Buchan-type lowP amphibolite to MP granulite-facies metamorphism.Nonetheless,a few metamorphic rocks only record a two-sage metamorphic evolution,with an early Barrovian-type high-P granulite-facies metamorphism and a late Buchan-type low-P granulitefacies metamorphic overprinting.In modern convergent plate margins,Alpine-type metamorphism mainly occurs in the stages of oceanic subduction and continental collision,Barrovian-type metamorphism takes pace in both stages of crustal thickening during continental hard collision and slab exhumation when continental subduction zones have evolved from compressional to extensional regimes,and Buchan-type metamorphism occurs in intracontinental rifting stage after the plate convergence.Therefore,the tectonic evolution of convergent continental margins can be reconstructed by combining metamorphic T/P ratios with their corresponding metamorphic facies series and metamorphic timing of metamorphic rocks.Based on the reported metamorphic rocks of different types and ages along the CCOS,it appears that the continental subduction/collision occurred at 500–490 Ma in the Altyn-North Qinling-North Tongbai orogens but 450–430 Ma in the North Qaidam-East Kunlun orogens,and the intracontinental rifting occurred at 460–450 Ma in the Altyn-North Qinling-North Tongbai orogens but 410–400 Ma in the North Qaidam-East Kunlun orogens,respectively,in the western Proto-Tethys domain.For the eastern Paleo-Tethys domain,in contrast,the continental subduction/collision occurred at 250–220 Ma and post-collisional intracontinental rifting occurred at 140–120 Ma.Furthermore,metamorphic evolution from low T/P ratios in the subduction/collision stage to high T/P ratios in the intracontinental rifting stage needs 40–60 Myr in the Proto-Tethys domain but about 110 Myr in the Paleo-Tethys domain.For the two different orogenic domains,therefore,the convergent continental margins underwent a common tectonic evolution from warm collision/cold subduction to hot rifting,which starts from continental subduction/collision characterized by the formation of medium-P amphibolite to high-P granulite facies series or high-P to ultrahigh-P eclogite facies series in compressional regimes,through exhumation of the deeply subducted crustal rocks,and terminates with intracontinental rifting featured by highT to ultrahigh-T granulite facies series in extensional regimes.Qiangqiang ZHANG Xiaoying GAO Renxu CHEN Yongfei ZHENG 2023Science China Earth Sciences2023,66,12:1

14EXPRESSION AND SUBCELLULAR LOCALIZATION OF P9-ZFD PROTEIN IN PATIENTS WITH MYASTHENIA GRAVIS显示文摘To express and purify the protein coded by the TRAF-type zinc finger domain of myasthenia gravis (MG)-re-lated gene P9 ( P9-ZFD ) and to prepare P9-ZFD antiserum for detecting expression and subcellular distribution of P9-ZFD protein in the skeletal muscles of patient with MG. Methods The cDNA encoding P9-ZFD was amplified by RT-PCR. The cloned P9-ZFD cDNA was ligated into pET-24a, and the P9-ZFD recombinant protein was induced via E.coli. BL21 (DE3) and purified by histidine affinity chromato-graphy. P9-ZFD antiserum was prepared and its titer and specificity were determined by ELISA and Western blot. Expres-sion and subcellular distribution of P9-ZFD protein in the skeletal muscles of MG and control were studied. Results The molecular weight of purified P9-ZFD protein was about 30 kD. Its purity was more than 95%. Antiserum specific for P9-ZFD was excellent. P9-ZFD protein is fully confined to the cytoplasm membrane of skeletal muscle cell of MG, obvious immunostaining was absent in the A, I, and Z bands of cytoplasm and no immunoreactivity was observed in the skeletal muscle cell of control. 18Transcription factor EB(TFEB)-mediated autophagy protects bovine mammary epithelial cells against H_(2)O_(2)-induced oxidative damage in vitro显示文摘Background:Bovine mammary epithelial cells after calving undergo serious metabolic challenges and oxidative stress both of which could compromise autophagy.Transcription factor EB(TFEB)-mediated autophagy is an important cytoprotective mechanism against oxidative stress.However,effects of TFEB-mediated autophagy on the oxidative stress of bovine mammary epithelial cells remain unknown.Therefore,the main aim of the study was to investigate the role of TFEB-mediated autophagy in bovine mammary epithelial cells experiencing oxidative stress.Results:H_(2)O_(2) challenge of the bovine mammary epithelial cell MAC-T increased protein abundance of LC3-II,increased number of autophagosomes and autolysosomes while decreased protein abundance of p62.Inhibition of autophagy via bafilomycin A1 aggravated H_(2)O_(2)-induced reactive oxygen species(ROS)accumulation and apoptosis in MAC-T cells.Furthermore,H_(2)O_(2) treatment triggered the translocation of TFEB into the nucleus.Knockdown of TFEB by siRNA reversed the effect of H_(2)O_(2) on protein abundance of LC3-II and p62 as well as the number of autophagosomes and autolysosomes.Overexpression of TFEB activated autophagy and attenuated H_(2)O_(2)-induced ROS accumulation.Furthermore,TFEB overexpression attenuated H_(2)O_(2)-induced apoptosis by downregulating the caspase apoptotic pathway.Conclusions:Our results indicate that activation of TFEB mediated autophagy alleviates H_(2)O_(2)-induced oxidative damage by reducing ROS accumulation and inhibiting caspase-dependent apoptosis.Xudong Sun Renxu Chang Yan Tang Shengbin Luo Chunhui Jiang Hongdou Jia Qiushi Xu Zhihao Dong Yusheng Liang Juan J.Loor Chuang Xu 2021Journal of Animal Science and Biotechnology2021,12,3:1
