2Universal FRFT-based algorithm for parameter estimation of chirp signals显示文摘The realization of the parameter estimation of chirp signals using the fractional Fourier transform (FRFT) is based on the assumption that the sampling duration of practical observed signals would be equal to the time duration of chirp signals contained in the former. However, in many actual circumstances, this assumption seems unreasonable. On the basis of analyzing the practical signal form, this paper derives the estimation error of the existing parameter estimation method and then proposes a novel and universal parameter estimation algorithm. Furthermore, the proposed algorithm is developed which allows the estimation of the practical observed Gaussian windowed chirp signal. 3A novel cancer vaccine with the ability to simultaneously produce anti-PD-1 antibody and GM-CSF in cancer cells and enhance Th1-biased antitumor immunity显示文摘Tumor escape from immune-mediated destruction has been associated with immunosuppressive mechanisms that inhibit T-cell activation.A promising strategy for cancer immunotherapy is to disrupt key pathways regulating immune tolerance,such as program death-1(PD-1/PD-L1)pathway in the tumor environment.However,the determinants of response to anti-PD-1 monoclonal antibodies(mAbs)treatment remain incompletely understood.In murine models,PD-1 blockade alone fails to induce effective immune responses to poorly immunogenic tumors,but is successful when combined with additional interventions,such as cancer vaccines.Novel cancer vaccines combined with antibody may offer promising control of cancer development and progression.In this investigation,we generated a novel tumor cell vaccine simultaneously expressing anti-PD-1 mAbs and granulocyte-macrophage colony stimulating factor(GM-CSF)in CT26 colon cancer and B16-F10 melanoma.The antitumor effect of the vaccine was verified by therapeutic and adoptive animal experiments in vivo.The antitumor mechanism was analyzed using Flow cytometry,Elispot and in vivo intervention approaches.The results showed that tumor cell vaccine secreting PD-1 neutralizing antibodies and GM-CSF induced remarkable antitumor immune effects and prolonged the survival of tumor-bearing animals compared with animals treated with either PD-1 mAbs or GM-CSF alone.Antitumor effects and prolonged survival correlated with strong antigen-specific T-cell responses by analyzing CD11c^(+)CD86^(+)DC,CD11b^(+)F4/80^(+)MΦcells,increased ratio of Teff/Treg in the tumor microenvironment,and higher secretion levels of Th1 proinflammatory cytokines in serum.Furthermore,the results of ELISPOT and in vivo blocking strategies further confirmed that the antitumor immune response is acquired by CD4 and CD8 T immune responses,primarily dependent on CD4 Th1 immune response,not NK innate immune response.The combination of PD-1 blockade with GM-CSF secretion potency creates a novel tumor cell vaccine immunotherapy,affording significantly improved antitumor responses by releasing the state of immunosuppressive microenvironment and augmenting the tumor-reactive T-cell responses.Hongwei Tian Gang Shi Qin Wang Yiming Li Qianmei Yang Chunlei Li Guoyou Yang Min Wu Qian Xie Shuang Zhang Yang Yang Rong Xiang Dechao Yu Yuquan Wei Hongxin Deng 2016Signal Transduction and Targeted Therapy2016,1,1:3
4LHCGR and ALMS1 defects likely cooperate in the development of polycystic ovary syndrome indicated by double-mutant mice显示文摘Polycystic ovary syndrome(PCOS)is a heterogeneous disorder with evidence of polygenetic components,and obesity may be a risk factor for hyperandrogen ism.Previous studies have shown that LHCGR is en riched in the ovary and LHCGR deficiency causes infertility without typical PCOS phenotypes.ALMS1 is implicated in obesity and hyperandrogenism,the common phenotypes among PCOS patients.Through whole-exome sequencing of 22 PCOS families and targeted candidate gene sequencing of additional 65 sporadic PCOS patients,we identified potential causative mutations in LHCGR and ALMS1 in a sibling-pair PCOS family and three sporadic PCOS patients.The expression of LHCGRL638 P in granulosa-like tumor cell line(KGN)cells promoted cyclic adenosine monophosphate production and granulosa cell proliferation,indicating that LHCGRL638 P is an activating mutation.Lhcgr~(L642 P/L642 P)mice showed an irregular estrous cycle,reduced follicles with dynamic folliculogenesis,and increased testosterone(T),estradiol(E2),and dehydroepiandrosterone.Lhcgr~(+/L642 P)AIms1~(+/PB)mice displayed increased T and E2 but decreased late secondary and preovulatory follicles.We showed that activating mutation of LHCGR likely plays important roles in the pathophysiology of PCOS involving abnormal reproductive physiology,whereas ALMS1 deficiency may promote anovulatory infertility via elevated androgens,suggesting that the disturbed LHCGR and ALMS1 cooperatively induce PCOS phenotypes,characterized as anovulation and hyperandrogenemia frequently observed in PCOS patients with obesity.Li Yu Lina Wang Wufan Tao Wenxiang Zhang Shuanghao Yang Jian Wang Jia Fei Rui Peng Yiming Wu Xiumei Zhen Hong Shao Weiyue Gu Rong Li Bai-Lin Wu Hongyan Wang 2021Journal of Genetics and Genomics2021,48,5:3

18Derivation of totipotent-like stem cells with blastocyst-like structure forming potential显示文摘It is challenging to derive totipotent stem cells in vitro that functionally and molecularly resemble cells from totipotent embryos.Here,we report that a chemical cocktail enables the derivation of totipotent-like stem cells,designated as totipotent potential stem(TPS)cells,from 2-cell mouse embryos and extended pluripotent stem cells,and that these TPS cells can be stably maintained long term in vitro.TPS cells shared features with 2-cell mouse embryos in terms of totipotency markers,transcriptome,chromatin accessibility and DNA methylation patterns.In vivo chimera formation assays show that these cells have embryonic and extraembryonic developmental potentials at the single-cell level.Moreover,TPS cells can be induced into blastocyst-like structures resembling preimplantation mouse blastocysts.Mechanistically,inhibition of HDAC1/2 and DOT1L activity and activation of RARγsignaling are important for inducing and maintaining totipotent features of TPS cells.Our study opens up a new path toward fully capturing totipotent stem cells in vitro.Yaxing Xu Jingru Zhao Yixuan Ren Xuyang Wang Yulin Lyu Bingqing Xie Yiming Sun Xiandun Yuan Haiyin Liu Weifeng Yang Yenan Fu Yu Yu Yinan Liu Rong Mu Cheng Li Jun Xu Hongkui Deng 2022Cell Research2022,32,6:1
